AIM: To find the point mutations meaningful for inactivationof liver-related putative tumor suppressor gene (LPTS) gene,a human novel liver-related putative tumor suppressor geneand telomerase inhibitor in hepatocellular ***: The entire coding sequence of LPTS genewas examined for mutations by single strand conformationpolymorphism (SSCP) assay and PCR products directsequencing in 56 liver cancer cell lines, 7 ovarian cancerand 7 head & neck tumor cell lines and 70 pairs of HCCtissues samples. The cDNA fragment coding for the mostfrequent mutant protein was subcloned into GST fusionexpression vector. The product was expressed in E. coliand purified by glutathione-agarose column. Telomericrepeat amplification protocol (TRAP) assays wereperformed to study the effect of point mutation totelomerase inhibitory ***: SSCP gels showed the abnormal shifting bandsand DNA sequencing found that there were 5 differentmutations and/or polymorphisms in 12 tumor cell lineslocated at exon2, exon5 and exon7. The main alterationswere A(778)A/G and A(880)T in exon7. The change in siteof 778 could not be found in HCC tissue samples, while themutation in position 880 was seen in 7 (10 %) cases. Themutation in the site of 880 had no effect on telomeraseinhibitory ***: Alterations identified in this study arepolymorphisms of LPTS gene. LPTS mutations occur in HCCbut are infrequent and of little effect on the telomeraseinhibitory function of the protein. Epigenetics, such asmethylation, acetylation, may play the key role in inactivationof LPTS.

Transcription analysis of peloric mutants of Phalaenopsis orchids derived from tissue culture

Tissue culture has been widely used for mass propagation of Phalaenopsis. However, somaclonal variation occurred during micropropagation process posed a severe problem by affecting product quality. In this study, wild type and peloric flower buds of Phalaenopsis hybrids derived from flower stalk nodal culture were used for cDNA-RAPD and cDNA suppression subtractive hybridization analyses in order to study their genetic difference in terms of expressed sequence tags. A total of 209 ESTs from normal flower buds and 230 from mutants were sequenced. These ESTs sequences can be grouped into several functional categories involved in different cellular processes including metabolism, signal transduction, transcription, cell growth and division, protein synthesis, and protein localization, and into a subcat- egory of proteins with unknown function. Cymbidium mosaic virus transcript was surprisingly found expressed fre- quently in the peloric mutant of P. Little Mary. Real-time RT-PCR analysis on selected ESTs showed that in mutant flower buds, a bZIP transcription factor (TGA1a-like protein) was down-regulated, while up-regulated genes include auxin-regulated protein kinase, cyclophilin, and TCP-like genes. A retroelement clone was also preferentially expressed in the peloric mutant flowers. On the other hand, ESTs involved in DNA methylation, chromatin remodeling and post- transcriptional regulation, such as DNA methyltransferase, histone acetyltransferase, ERECTA, and DEAD/DEAH RNA helicase, were enriched in normal flower buds than the mutants. The enriched transcripts in the wild type indicate the down regulation of these transcripts in the mutants, and vice versa. The potential roles of the analyzed transcripts in the development of Phalaenopsis flowers are discussed.

Objective:To investigate the efficacy of frankincense and myrrha in the treatment of acute interstitial cystitis/painful bladder syndrome(IC/PBS).Methods:The effects of frankincense and myrrha on the proliferation and migration of primary human urothelial cells(HUCs)were assessed in *** the animal study,48 virgin female rats were randomized into 4 groups(12 in each group):(1)control group(saline-injected control);(2)cyclophosphamide(CYP)group(intraperitoneal injected 150 mg/kg CYP);(3)CYP+pentosan polysulfate sodium group(orally received 50 mg/kg pentosan polysulfate sodium);and(4)CYP+frankincense and myrrha group[orally received frankincense(200 mg/kg)and myrrha(200 mg/kg)].Rats orally received pentosan polysulfate sodium or frankincense and myrrha on day 1,2,and *** experiments were performed on day *** and cystometry assessment behavior test were *** interval values were assessed in rats under ***,immunohistochemistry and Western blot were used to confirm the location and level,respectively,of cell junction-associated protein zonula occludens-2(ZO-2)***:Low dose frankincense and myrrha increased cell proliferation and migration in HUCs compared with control(P<0.05).Rats with acute IC/PBS rats exhibited lower voiding interval values,pain tolerance,and ZO-2 expression(P<0.05).Voiding interval values and pain tolerance were higher in the frankincense and myrrha group than CYP group(P<0.05).ZO-2 expression in the bladder was increased in the CYP+pentosan polysulfate and frankincense+myrrha groups compared with the CYP-induced acute IC/PBS group(P<0.05).Conclusion:frankincense and myrrha modulate urothelial wound healing,which ameliorates typical features of acute IC/PBS in rats.

Estimating sediment deposition volume in a reservoir using the physiographic soil erosion-deposition model

A traditional way to estimate the amount of sediment deposition in a reservoir is simply based on the variation in the elevation measurements made at different *** the present study,a physiographic soil erosion-deposition(PSED) model is applied ***-Wen reservoir watershed to compute the discharge and sediment concentration into the reservoir,and the deposition *** PSED model is verified using three storm events and then compared to the historic data recorded at the hydrological stations in the reservoir watershed,which shows close agreement between the simulated and recorded values of the flow discharge into the reservoir and the corresponding sediment yield. The numerical simulation was also conducted for all rainfall events from 1996 to 2001 to estimate the annual sediment deposition volume in the ***,the simulated result was compared to that obtained from the cross-section elevation *** is found that some measured data points had large uncertainty or were *** the contrary,the model results and the measured data that renders the model ***,regardless of the estimation for the flow discharge into the reservoir induced by a single rainfall event,the sediment yield,or the sediment deposition,the PSED model has successfully demonstrated its ability.

Improvement of extraction efficiency for GaN-based light emitting diodes

A simple, low cost method for mass production to enhance the light extraction efficiency of GaN-related LEDs was proposed. With appropriate process parameters, the nature lithography of nanosphere can be used to fabricate two-dimensional nanostructures, including the nanomesh ZnO layer, photonic crystal (PhC) patterned p-GaN, and patterned sapphire substrates. Based on preliminary results, the extraction efficiencies of LEDs with these nanostructures can thus be improved and the nature lithography is demonstrated to be a promising method to be widely exploited in the manufacture of all kinds of LED devices.

Diffusion characteristics of HTO and 99TcO_4^- in compacted Gaomiaozi(GMZ) bentonite

The characteristics of diffusion are essential to the transport of radionuclides through buffer/backfill materials, such as bentonite, which are commonly found in waste repositories. This study used through-diffusion techniques to investigate the diffusion behavior of HTO and ^(99)TcO_4^- on GMZ bentonite of various densities. Diffusion rates were calculated by measuring the diffusion coefficients(De, Da), plotting breakthrough curves and interpreting experiment data. The apparent and effective diffusion coefficients of HTO ranged from(1.68 ± 0.40) 9 × 10^(-11) to(2.80 ± 0.62) 9 × 10^(-11) m^2/s and from(4.61 ±1.28) 9 × 10^(-12) to (16.2 ± 2.50) 9 × 10^(-12) m^2/s, *** apparent and effective diffusion coefficients of^(99)TcO_4^-ranged from(5.26 ± 0.16) 9 × 10^-12to(7.78 ± 0.43) 9× 10^-12m^2/s and from(1.49 ± 0.002) 9 × 10^(-12) to(4.16 ±0.07) 9 × 10^(-12) m^2/s, respectively. The distribution coefficients of HTO and^(99)TcO_4^-ranged from(0.70 ± 0.12) 9× 10^(-2) to(1.36 ± 0.53) 9 × 10^(-2) mL/g and from(1.12 ±0.06) 9 × 10^(-2) to(5.79 ± 2.22) 9 × 10^(-2) mL/g, *** Deand Kdvalues were shown to decrease with an increase in the bulk dry density of compacted bentonite. Our results show that HTO and ^(99)Tc could be considered nonsorbent radionuclides. The data obtained in this studyprovide a valuable reference for the safety assessment of waste repositories.

Fabrication and Characterization of Poly(Styrene-Butadiene-Styrene)/Thermoplastic Polyurethane Blends

This investigation presents thermoplastic elastomers (TPEs) based on poly (styrene-butadiene-styrene) (SBS) and thermoplastic polyurethane (TPU) materials were prepared with varying compositions. A series of works were conducted on the relationships between rheological, optical properties, morphology, mechanical properties, abrasion resistance and thermostability given. The results showed that the shear viscosity of SBS not obvious effect with TPU content. The optical properties of the SBS/TPU blend that its uniform transparency. The morphology characteristics indicating the phase diversion and the variation in the size of the SBS domains from large to small as the TPU contents increased, with heterogeneous domain dispersions. Additionally, the mechanical properties, abrasion resistance and thermal resistance are improved as the amount of added TPU is increased, suggesting that the blending of SBS with TPU is consistent with the compound rule.

An analytical solution is undertaken to describe the wave-induced flow field and the surge motion of a permeable platform structure with fuzzy controllers in an oceanic *** the design procedure of the controller,a parallel distributed compensation（PDC） scheme is utilized to construct a global fuzzy logic controller by blending all local state feedback controllers.A stability analysis is carried out for a real structure system by using Lyapunov *** corresponding boundary value problems are then incorporated into scattering and radiation *** are analytically solved,based on separation of variables,to obtain series solutions in terms of the harmonic incident wave motion and surge *** dependence of the wave-induced flow field and its resonant frequency on wave characteristics and structure properties including platform width,thickness and mass has been thus drawn with a parametric *** which mathematical models are applied for the wave-induced displacement of the surge motion.A nonlinearly inverted pendulum system is employed to demonstrate that the controller tuned by swarm intelligence method can not only stabilize the nonlinear system,but has the robustness against external disturbance.
