Evaluation of Analgesic and Anti-Inflammatory Activities on Ethanolic Extract of Terminalia chebulaFruits in Experimental Animal Models

The present study was undertaken to evaluate the analgesic and anti-inflammatory activities of ethanolic extract of Terminalia chebula (commonly known as Haritaki) fruits in experimental animal models. The study was carried out using Swiss Albino mice (20-25 g) and Long Evans rats (100-150 g) of either sex. The analgesic activity of Terminalia chebula was assessed by using hot plate method. For the determination of analgesic effect, doses of ethanolic extract of Terminalia chebula used in the present study were 250 mg/kg and 500 mg/kg body weight (BW). Anti-inflammatory effect was analyzed by carrageenan induced paw edema method with the administration dose of 300 mg/kg BW of animals. The analysis of experimental data was performed by statistical process of ANOVA to determine the variability of sample, while Dunnet’s test was performed for evaluation of comparative analgesic and anti-inflammatory activity of Terminalia chebula with control and standard. The animals were divided into four treatment groups of six animals each and the “Mean ± SEM” is the statistical identifiable value of the data and P values <0.05 was considered statistically significant. Hot plate test showed a significant increase in the mean reaction time to heat stimuli in hot plate method at both 250 mg/kg and 500 mg/kg BW doses throughout the observation period in 30 minutes and 60 minutes after treatment, which was comparable to the standard ketorolac and control group. In carrageenan induced paw edema method, considerable results were found after determining the percentage change in paw volume in extract. In both cases of analgesic and anti-inflammatory study, % inhibition of pain and inflammation were evaluated. Comparing with control, largest inhibition was found in inhibiting inflammation 5 hours after treatment, while the largest inhibition of pain was obtained in 30 minutes and 60 minutes after treatment of doses. Simvastatin Potentiates the Antihyperglycemic, Antidyslipidimic and Antioxidative Effect of Glibenclamide on Alloxan-Induced Diabetic Rats

The aim of the current study is to investigate the effect of combination of glibenclamide;an antidiabetic drug and simvastatin;a HMG-CoA reductase inhibitor on long-term (four weeks) alloxan-induced diabetes rats (ADRs). Methods: Alloxan (120 mg/kg body weight, BW) was injected intra-peritonially (i.p.) in rats. At first alloxan (120 mg/kg BW) induced diabetic rats were treated with single dose of glibenclamide (1.2 mg/70kg BW) and simvastatin (10 mg/70kg BW) for two weeks. Then fixed dose combinations of glibenclamide (0.6 mg/70kg BW) and simvastatin (5 mg/70kg BW) were injected along with those of two drugs for four weeks. Results: At first it was found that glibenclamide reduced significant amount of glucose in blood, but it had no significant effect on lipid profile on short term (two weeks) ADRs. In contrast, simvastatin had no effect on blood glucose level, whereas it significantly reduced total cholesterol (TC), triglycerides (TG) and low density lipoprotein cholesterol (LDL-C) and increased significant amount of high density lipoprotein cholesterol (HDL-C). However, pathological changes of pancreas’s Islets of Langerhans were observed only after long-term (four weeks) induction of alloxan in rats. The inhibitory effect of combination therapy on blood glucose, TC, TG and LDL-C level was higher than those of monotherapy alone on long term ADRs. In addition, treatment with combination therapy on long term ADRs showed higher amount of HDL-C level and super oxide dismutase and catalase enzyme activity than those with monotherapy. They also decreased serum glutamic pyruvic transaminase (SGPT) and Serum glutamic oxaloacetic transaminase (SGOT) level. Administration of simvastatin recovered Langerhans cells from shrinkage whereas glibenclamide displayed slight recovery. But the combination therapy showed complete recovery of Langerhans cells as compared with diabetic rats. Social media is an essential component of our personal and professional lives. We use it extensively to share various things, including our opinions on daily topics and feelings about different subjects. This sharing of posts provides insights into someone’s current emotions. In artificial intelligence (AI) and deep learning (DL), researchers emphasize opinion mining and analysis of sentiment, particularly on social media platforms such as Twitter (currently known as X), which has a global user base. This research work revolves explicitly around a comparison between two popular approaches: Lexicon-based and Deep learning-based Approaches. To conduct this study, this study has used a Twitter dataset called sentiment140, which contains over 1.5 million data points. The primary focus was the Long Short-Term Memory (LSTM) deep learning sequence model. In the beginning, we used particular techniques to preprocess the data. The dataset is divided into training and test data. We evaluated the performance of our model using the test data. Simultaneously, we have applied the lexicon-based approach to the same test data and recorded the outputs. Finally, we compared the two approaches by creating confusion matrices based on their respective outputs. This allows us to assess their precision, recall, and F1-Score, enabling us to determine which approach yields better accuracy. This research achieved 98% model accuracy for deep learning algorithms and 95% model accuracy for the lexicon-based approach.

Hydrobiological variables as a regulatory factor on the abundance of heterotrophic flagellates in an urban pond

The seasonal abundance of flagellates has been monitored over a period of 1 year from December 2013 to November 2014(divided into 4 conjugative seasons namely winter,spring,summer,and autumn)in an experimental pond located in Rajshahi City Corporation area,*** our knowledge,this study is the fi rst to shed light on the occurrence and possible interrelationships among heterotrophic flagellates(HF),bacteria and zooplankton in Bangladesh and the result obtained by this study will be beneficial for similar water ecosystem all over the *** methods were used to determine the prescribed hydrological parameters and zooplankton cell *** HF abundance(14 346.00 cells/mL)was found in the spring and the minimum(5 215.00 cells/mL)occurred in the *** to HF,significantly(P<0.05)higher zooplankton abundance was found during the winter(782.00±47.62 cells/mL)and the lowest value was found in the autumn(448.00±39.15 cells/mL).Whereas similar to the HF,total bacterial abundance was significantly higher during the spring((2.25±1.05)×10^5 cells/mL)and lower in the summer((0.79±0.06)×10^5 cells/mL).Multivariate analyses(ANOSIM and MDS)have shown significant seasonal differences for cell numbers where MDS ordination plot and cluster analysis based on similarity in the genera abundance of HF revealed overlapping condition between winter and *** correspondence analysis(CCA)also showed a distinct separation among the genera based on the prevailing hydrological situation and indicated that temperature,pH,BOD5,and NO3^-were the most important environmental variables in determining the observed variation in HF community *** the biological factors,zooplankton showed negative but total bacteria were positively correlated with HF abundance.

Investigation of Antidiabetic Effect of Ethanolic Extract of Phyllanthus emblicaLinn. Fruits in Experimental Animal Models

Hypoglycemic effects were investigated in the ethanolic extract of fruits of Phyllanthus emblica Linn. with long evans rats (100-120 g). Residual gut sucrose content, gut perfusion and disaccharidase activity studies were carried out to assess these activities. The residual gut sucrose content study was carried out to reveal the amount of sucrose remaining in the gastro intestinal tract (GIT) at six different segments. The ethanolic fruit extract of Phyllanthus emblica Linn. significantly increases the residual sucrose content throughout the gut after sucrose ingestion. Study on intestinal enzyme activity showed that the extract caused a significant (p 0.05), dose dependent inhibition of intestinal disaccharidase enzyme activity. Evaluation of the effect of the plant extract on intestinal enzyme activity showed significant inhibition of intestinal disaccharidase activity, suggesting that reduction of sucrose absorption may be partly related to the inhibition of disaccharidase activity in the gut. The gut perfusion analysis showed that the extract significantly reduced intestinal glucose absorption. The results of the present study suggest that ethanolic extract of Phyllanthus emblica Linn. has significant antidiabetic effects in a dose dependent manner and this may be effective in the treatment of diabetes.

In current decade, pharmaceutical industries of Bangladesh are giving much emphasize on the formulation of time release preparation to treat various chronic diseases in order to decrease the frequency of administration and to improve patient compliance. Objectives: The objective of this investigation is to design and evaluate sustained release matrix tablet of Gliclazide by direct compression method employing polymers of hydroxypropylmethyl cellulose (HPMC) derivatives (K15M CR and K4M CR) and to select the optimized formulations and compression process by performing a comparative release kinetic study with a reference product, Diamicron MR (one of the worldwide brand of Gliclazide sustain released tablet manufactured by Servier one of the French pharmaceutical company) tablet. Methods: Release kinetics of Gliclazide matrix tablets were determined using USP paddle method at Phosphate buffer (pH 7.4). The release mechanism was explored and explained with zero order, first order, Higuchi and Korsmeyer model. Result: It is found that formulation with lower polymeric concentration follows Higuchi release kinetics and that the formulation with higher concentration best fits with zero order release kinetics. Among the formulations, F1 and F6 show almost similar dissolution profile with Diamicron MR Tablet, which can be suitable candidates for further in-vivo bioequivalence study. Conclusion: Findings of this investigation suggest that F1 and F6 formulations are potential candidates for further bioequivalence study among other formulations.
