Effect of an amalgamated antibiotic and its connection to cytogenotoxicity and histo-architectural malformations in stinging catfish

Intensive fish farming has an excessive prevalence of infection and is typically controlled by the administration of *** amalgamated antibiotics are a relatively novel therapeutic idea and more effective than traditional antibiotic monotherapy,they can also have a toxic effect on the fish body when it is administered *** study investigated the cyto-genotoxic effects on erythrocytes and histo-architectural malformations in the liver and kidneys of stinging catfish(Heteropneustes fossilis)in relation to Enrocip plus use as an amalgamated antibiotic *** experimental fish with an initial average weight of 17.38±1.94 g were divided into four treatment groups with antibiotic doses:according to the recommendation of the manufacturer 0.167 mg/ml Enrocip plus was used as a standard dose(x)and it was treated as T3,while 0%of the standard dose(0×mg/ml),1/2×(0.083 mg/ml),and 2×(0.333 mg/ml)were treated as T1,T2 and T4,respectively for a period of 30 *** observed erythrocyte cellular deformities(ECD)were twin,tear-drop,serrated,tail budded and de-membranated cells,whereas the erythrocyte nuclear deformities(END)were a nuclear bridge,bi-nucleus,nuclear termination,karyopyknosis,and micronucleus at different concentrations of Enrocip *** ECD and END percentages experienced the interaction of antibiotic dose and exposure time,and were significantly different(P<0.01).Significant changes in hepatocytes,mild to severe necrosis,vacuole formation,and hepatopancreas damage were also observed in the liver of the treated fish whereas highly degraded renal tubules and hematopoietic tissue,glomerular occlusion,and vacuolation were evident in the *** current investigation fully emphasizes the adverse effects of amalgamated antibiotics on the cytogenotoxicity and the histomorphology of the kidneys and liver of ***,the use of an amalgamated antibiotic in aquaculture must be carefully evaluated.

Striped snakehead(Channa striata)has enormous fisheries and aquaculture importance due to its high market demand and food ***,Vietnam strain of the species has been introduced to Bangladesh for ***,it is essential to develop a reliable molecular technique that can distinguish the Bangladesh strain from the Vietnam ones as biological invasion by feral exotic fish to the wild may cause enduring damage to ecosystem and species *** clips from snakehead samples were collected from six locations and total DNA was *** cytochrome oxidase c 1(COI)gene fragment was amplified by PCR using the Fish F1/R1 primer set and sequenced for analysis of intra-and inter-strain genetic diversity and detection of single nucleotide polymorphism to distinguish the Vietnam strain from Bangladesh ones.A total of 15 haplotypes were identified with an overall haplotype diversity of *** median-joining network created based on COI gene sequences clearly differentiated the Bangladesh strain from the strain of Vietnamese origin by a total of 18 single nucleotide *** In silico restriction analysis revealed AluI was the most suitable restriction enzyme that produced diagnostic restriction profiles of the two strains of ***-RFLP analysis by the restriction enzyme AluI created distinguishing restriction profile similar to that in the In silico *** work illustrates a simple,reliable and inexpensive molecular tool for the identification of the Bangladesh and Vietnam strains of *** by one-step PCR and restriction of the COI gene fragment.

Role of Polyethylene Glycol and Silica for Dissolution Enhancement of Cefuroxime Axetil: In-Vitro Performance Evaluation and Characterization

Cefuroxime Axetil (CA) a widely used cephalosporin antibiotic displays low aqueous solubility and high membrane penetrability. This results in its solubility driven variable and/or low oral bioavailability and therapeutic efficacy as a major drawback. Thus, most of the goal of our study was to increase the solubility as well as dissolution rate of CA using the simple and cost-effective solid dispersion (SD) method. At first, the SD formulations of CA were prepared at various weight ratios of Carplex-67 and PEG-4000 by solvent evaporation technique. These new formulations were then subjected to an in-vitro drug release performance study and tested for physicochemical characterization to distinguish the thermal behavior, crystallinity, interactions phenomena, and surface morphology. Among the formulated Cefuroxime Axetil Solid Dispersion (CSD), CSD-8 which contained CA, Carplex-67, and PEG-4000 at the weight ratio 1:3:2, respectively showed the most significant (p in-vitro dissolution in water, Gastric Simulated Fluid (GSF), and Intestinal Simulated Fluid (ISF). This study also showed a significant (p < 0.001) increase in drug release compared to the marketed product. Therefore, it is supposed to be a promising alternative to conventional antimicrobial therapy.

Development and Evaluation of Stable Paracetamol Loaded Solid Dispersion with Enhanced Analgesic and Hepatoprotective Property

Paracetamol (PCM) is enlisted in the WHO model list as an essential medicine for pain and palliative care, but at overdose, it causes hepatic damage. This study was designed to assess the analgesic efficacy and hepatoprotective property of a solid dispersion (SD) loaded with PCM. A number of PCM loaded formulations (PSDs) were fabricated using silica alone or in combination with polyethylene glycol and/or Na-citrate followed by in-vitro dissolution profiling. Selected PSDs with improved dissolution profile were subjected to solid-state characterization (DSC, PXRD, FTIR, and SEM), stability study along with investigation of in-vivo analgesic efficacy and effect on hepatocytes. Among these, PSD10 showed a rapid and significantly higher in-vitro drug release than pure PCM. This improvement was distinct to other PSDs also. Solid-state characterization of PSD10 authenticated the conversion of crystalline PCM to amorphous form upon formulation. Subsequent oral administration of PSD10 in Swiss albino mice showed 1.44-fold greater analgesic efficacy than pure PCM at dose 30 mg/kg. Besides, at acute toxic dose, liver histology of PSD10 mice was comparable with NC mice indicating hepatic protection upon formulation, whereas the PCM mice showed extensive hepatic necrosis which was also endorsed by significantly higher values of SGPT, SGOT, and ALP than PSD10 mice. Finally, an accelerated stability study of PSD10 performed according to the guideline of ICH noticed no remarkable deviation in its dissolution performance as well as crystalline nature. Thus, this newly developed PSD10 may be a safe and promising alternative for pain management and palliative care.

Impacts of Different Sources of Biochar on Plant Growth Characteristics

A net-house experiment was conducted to assess the impacts of different sources of biochar on growth and development of plant. Three different types of biomasses viz., cow dung, poultry manure and sewage sludge produced were used for the study. An upland leafy vegetable commonly known as kalmi (Ipomoea aquatic) was used for pot culture experiment. The biomass or biochar was applied to the soil at a rate of 5 t/ha and the pots were arranged following completely randomized design. A number of physical, chemical and physico-chemical properties of the soil, biochar, biomass and plant samples were analyzed in the laboratory. Biochar treated soils showed higher CEC than the corresponding biomass treated ones. Organic carbon in biochar treated soils was also higher. Total nitrogen contents of biomass treatments were higher than biochar treatments. Phosphorus, potassium and sulphur concentrations of the treated soils increased after harvesting. The paper discusses plant growth and agricultural crop production vis-à-vis biochar application, indicating that the materials might not exert similar positive effects as their biomass sources and thereby likely to jeopardize crop production and soil health.

Simulating Transport Capacity, Delivery Speed, and Routing Efficiency to Predict Economic Growth

This study uses a simulation-based approach to investigate the impact of delivery delays due to constraints on transport capacity, transit speed, and routing efficiencies on an economy with various levels of interdependency among firms. The simulation uses object-oriented programming to create specialized production, consumption, and transportation classes. A set of objects from each class is distributed randomly on a 2D plane. A road network is then established between fixed objects using Prim’s mst (Minimum Spanning Tree) algorithm, followed by construction of an all-pair shortest path matrix using the Floyd Warshall algorithm. A genetic algorithm-based vehicle routing problem solver employs the all-pair shortest path matrix to best plan multiple pickup and delivery orders. Production units utilize economic order quantities (EOQ) and reorder points (ROP) to manage inventory levels. Hicksian and Marshallian demand functions are utilized by consumption units to maximize personal utility. The transport capacity, transit speed, routing efficiency, and level of interdependence serve as 4 factors in the simulation, each assigned 3 distinct levels. Federov’s exchange algorithm is used to generate an orthogonal array to reduce the number of combination replays from 34 to just 9. The simulation results of a 9-run orthogonal array on an economy with 6 mining facilities, 12 industries, 8 market centers, and 8 transport hubs show that the level of firm interdependence, followed by transit speed, has the most significant impact on economic productivity. The principal component analysis (PCA) indicates that interdependence and transit speed can explain 90.27% of the variance in the data. According to the findings of this research, a dependable and efficient regional transportation network among various types of industries is critical for regional economic development.

Location Optimization of a Coal Power Plant to Balance Costs against Plant’s Emission Exposure

The goal of the research is to develop a methodology to minimize the public’s exposure to harmful emissions from coal power plants while maintaining minimal operational costs related to electric distribution losses and coal logistics. The objective is achieved by combining EPA Screen3, ISC3 and Japanese METI-LIS model equations with minimum spanning tree (mst) algorithm. Prim’s mst algorithm is used to simulate an electric distribution system and coal transportation pathways. The model can detect emission interaction with another source and estimate the ground level concentrations of emissions up to distances of 25 kilometers. During a grid search, the algorithm helps determine a candidate location, for a new coal power plant, that would minimize the operational cost while ensuring emission exposure is below the EPA/NIOSH thresholds. The proposed methodology has been coded in form of a location analysis simulation. An exhaustive search strategy delivers a final candidate location for a new coal power plant to ensure minimum operational costs as compared to the random or greedy search strategy. The simulation provides a tool to industrial zone planners, environmental engineers, and stakeholders in coal-based power generation. Using operational and emissions perspectives, the tool helps ascertain a list of compromise locations for a new coal power plant facility.
