Binding kinetics enhancement of a microfluidic biosensor into a micro-channel through the application of a supplementary mechanism has received tremendous attention because of the obtained significant enhancement factor. However, biosensor’s performance enhancement using only simple channel engineering is still rarely realized. Herein, we present a novel design of a complex reactive protein (CRP) biosensor into a U-shaped channel with a sensitive membrane located in the middle of the bent zone. Various critical factors affecting the equilibrium binding time are numerically investigated. The turn geometry is then optimized when the arc length along the inner and outer radii is almost the same, which leads to locally minimizing the channel height overhead the reaction surface and improves the analyte transport towards the sensing area. The numerical studies reveal that applying a local narrowing above the reaction surface can notably enhance the trapping and the surface formation of complex antibody-antigen, thus upgrading the biosensor performance. This work puts a significant advance towards microfluidic channel engineering and the exploration of micro-flow injection experimental studies.

Ions Sensitive Field Effect Transistors (ISFETs) are becoming the platform sensors for important chemical and biomedical applications. However, the accuracy of ISFET output measurement is greatly affected by the presences of low-frequency noise, drift and slow response of the device. This requires more safety in measured results and the tools of analysis. In this paper, we present fundamental limits on the sensitivity of ISFETs micro-sensors, arising from intrinsic and extrinsic noise sources. We developed an algorithm in MATLAB in order to model the frequency analysis of the 1/f noise in ISFET sensor using Hooge theory. We have shown that the 1/f noise of the ISFETs sensors is due to both the electrochemical system (pH solution) and the MOS component (canal size, insulator thickness). The temperature effect on the ISFET noise and the signal conditioning are also performed.

We question whether,in men with an abnormal rate of sperm DNA fragmentation,the magnetic-activated cell sorting(MACS)could select spermatozoa with lower rates of DNA fragmentation as well as spermatozoa with unbalanced chromosome content.Cryopreserved spermatozoa from six males were separated into nonapoptotic and apoptotic populations.We determined the percentages of spermatozoa with(i)externalization of phosphatidylserine(EPS)by annexin V-Fluorescein isothiocyanate(FITC)labeling,(ii)DNA fragmentation by TdT-mediated-dUTP nick-end labeling(TUNEL),and(iii)numerical abnormalities for chromosomes X,Y,13,18,and 21 by fluorescence in situ hybridization(FISH),on the whole ejaculate and selected spermatozoa in the same patient.Compared to the nonapoptotic fraction,the apoptotic fraction statistically showed a higher number of spermatozoa with EPS,with DNA fragmentation,and with numerical chromosomal abnormalities.Compared to the whole ejaculate,we found a significant decrease in the percentage of spermatozoa with EPS and decrease tendencies of the DNA fragmentation rate and the sum of disomy levels in the nonapoptotic fraction.Conversely,we observed statistically significant higher rates of these three parameters in the apoptotic fraction.MACS may help to select spermatozoa with lower rates of DNA fragmentation and unbalanced chromosome content in men with abnormal rates of sperm DNA fragmentation.

Growing evidence suggests that oxidative stress is a major cause of male infertility.Spermatozoa are highly sensitive cells due to the vulnerability of their membrane fatty acids and the diminished antioxidant defense.Reactive oxygen species(ROS)impact multiple signaling pathways involved in mitochondrial dysfunction,hormonal unbalance and semen quality decline.The origin of ROS is diverse,including generated normal cellular metabolism,environmental exposure,advanced paternal age and inflammation.Research has indicated that antioxidant supplementation can improve oxidative stress and reduce the risk of chronic diseases.Moreover,it is widely known that antioxidant supplementation can enhance sperm quality and increase the pregnancy rate in couples undergoing fertility treatments.The current study highlights the deleterious effect of ROS and its impact on semen function.In addition,it contributes to the development of a clinical approach for the use of antioxidants in the management of male infertility.

Application of four machine-learning methods to predict short-horizon wind energy

Renewable energy has garnered attention due to the need for sustainable energy sources.Wind power has emerged as an alternative that has contributed to the transition towards cleaner energy.As the importance of wind energy grows,it can be crucial to provide forecasts that optimize its performance potential.Artificial intelligence(AI)methods have risen in prominence due to how well they can handle complicated systems while enhancing the accuracy of prediction.This study explored the area of AI to predict wind-energy production at a wind farm in Yalova,Turkey,using four different AI approaches:support vector machines(SVMs),decision trees,adaptive neuro-fuzzy inference systems(ANFIS)and artificial neural networks(ANNs).Wind speed and direction were considered as essential input parameters,with wind energy as the target parameter,and models are thoroughly evaluated using metrics such as the mean absolute percentage error(MAPE),coefficient of determination(R~2),and mean absolute error(MAE).The findings accentuate the superior performance of the SVM,which delivered the lowest MAPE(2.42%),the highest R~2(0.95),and the lowest MAE(71.21%)compared with actual values,while ANFIS was less effective in this context.The main aim of this comparative analysis was to rank the models to move to the next step in improving the least efficient methods by combining them with optimization algorithms,such as metaheuristic algorithms.

Dimethoate Induced Oxidative Damage and Histopathological Changes in lung of Adult rats:Modulatory Effects of Selenium and/or Vitamin E

Objective To determine the efficiency of selenium and/or vitamin E to alleviate lung oxidative damage induced by dimethoate, an organophosphorus compound. Methods Adult Wistar rats were exposed during 30 days either to dimethoate （0.2 g/L of drinking water）, dimethoate＋selenium （0.5 mg/kg of diet）, dimethoate＋vitamin E （100 mg/kg of diet）, or dimethoate＋selenium＋vitamin E. Results Exposure to dimethoate caused oxidative stress in lung evidenced by an increase of malondialdehyde, protein carbonyl groups and advanced oxidation protein products. An increase in glutathione peroxidase, superoxide dismutase, catalase and a decrease in acetylcholinesterase and butyrylcholinesterase activities, glutathione, non-protein thiols and vitamins C levels were observed. Histopathological changes in lung tissue were noted as emphysema, hemorrhages and hemosiderin deposits. Co-administration of selenium or vitamin E to the diet of dimethoate treated rats ameliorated the biochemical parameters as well as histological impairments. The joint effect of these elements was more powerful in antagonizing dimethoate-induced lung oxidative damage. Conclusion We concluded that selenium and vitamin E ameliorated the toxic effects of this pesticide in lung tissue suggesting their role as potential antioxidants.

Objective The current study aimed to elucidate the effect of vanillin on behavioral changes, oxidative stress, and histopathological changes induced by potassium bromate （KBrO3）, an environmental pollutant, in the cerebellum of adult mice.Methods The animals were divided into four groups： group 1 served as a control, group 2 received KBrO3, group 3 received KBrO3 and vanillin, and group 4 received only vanillin. We then measured behavioral changes, oxidative stress, and molecular and histological changes in the cerebellum.Results We observed significant behavioral changes in KBrO3-exposed mice. When investigating redox homeostasis in the cerebellum, we found that mice treated with KBrO3 had increased lipid peroxidation and protein oxidation in the cerebellum. These effects were accompanied by decreased Na＋-K＋ and Mg2＋ ATPase activity and antioxidant enzyme gene expression when compared to the control group. Additionally, there was a significant increase in cytokine gene expression in KBrO3-treated mice. Microscopy revealed that KBrO3 intoxication resulted in numerous degenerative changes in the cerebellum that were substantially ameliorated by vanillin supplementation. Co-administration of vanillin blocked the biochemical and molecular anomalies induced by KBrO3.Conclusion Our results demonstrate that vanillin is a potential therapeutic agent for oxidative stress associated with neurodegenerative diseases.

Study of gonadic growth factors: Seminal transforming growth factor-β1, epidermal growth factor and insulin-like growth factor-I and their relationship with male infertility

Background: The aim of this study was to establish normal reference values for several growth factors in semen and provide a basis for understanding their physiological significance in fertile and infertile subjects. Methods: 110 infertile men semen was evaluated according to WHO standards and by enzyme immunoassay (ELISA kit protocol) to determine the rate of growth factors (TGFβ1, IGFI, EGF). We have also listed neutrophils by staining with peroxidase to examine the association of inflammation with the levels of these factors. The determination of the epididymal (alpha-1,4-glucosidase) marker was directed according to the technical recommended by the WHO in 2010. Each patient received a hormonal balance including the determination of plasma FSH, LH and Testosterone. Results: Concentrations of TGFβ1 were significantly higher in (severe and extreme azoospermic, oligospermic) pathological groups compared to the control group (P 0.05) between control and pathological groups. The IGFI is partly testicular and/or epididymal as it is lowered in some patients with obstructive azoospermia. Positive correlations were observed between the seminal IGFI, mobility (r = 0.487) and sperm morphology (r = +0.187). Immunological sperm abnormalities and/or idiopathic do not alter the rate of EGF seminal. There is no significant correlation between seminal EGF or with sperm parameters or with plasma levels of FSH and LH. Conclusion: The growth factors are primarily peptides, handling and identification in a complex environment as the seminal plasma, but it requires a lot of precautions if we do not want to expose to the risk of bad farms/interpretations of data obtained.

Biobased smart packaging is been the most interesting research field.In this study,pH-sensitive films based on anthocyanin molecules from red cabbage and sepiolite mineral nano-clay as functional additives were studied.The films were fabricated by casting method of blend polymer Chitosan/Poly(Vinyl Alcohol)at two fillers loading(5wt.%and 10wt.%).The structural,morphological,thermal,optical,hygroscopic,and mechanical properties as well as the pH sensitivity of films were investigated.The thermal properties confirm that the amount of the anthocyanin in sepiolite was around 6.4%.The tensile strength and the young’s modulus of produced composites at 10wt.%sepiolite–anthocyanin content was increased to 8.90 MPa and 163 MPa,respectively.The maximum water absorption that the films may absorb was 12.3%.pH sensitivity analysis showed that the Smart films change the color by changing the pH level,from pink at pH lower than 6 to yellow at pH higher than 6.Thus,the developed pH-sensitive films show band gaps in visible light that make them useful for monitoring milk spoilage.

Haemophagocytic lymphohistiocytosis secondary to Plasmodium falciparum malaria: Case report and review of the literature

Rationale:Haemophagocytic lymphohistiocytosis is a rare complication of malaria,which is often misdiagnosed.Patient concerns:A 30-year-old male was admitted to our department for persistent fever,which began after returning from a stay in Guinea-Conakry.The laboratory investigations revealed a pancytopenia and an elevated C-reactive protein.Peripheral smear examination showed Plasmodium falciparum,therefore confirming the diagnosis of malaria.The laboratory tests showed a worsening pancytopenia.Bone marrow aspiration and biopsy revealed images of hemophagocytosis.Diagnosis:The diagnosis of haemophagocytic lymphohistiocytosis complicating malaria infection was established.Interventions:The patient was treated with artemether-lumefantrine.No immunosuppressant treatment was delivered to the patient.He received antipyretic and antimalarial treatment only.Outcomes and lessons:We report a case of haemophagocytic lymphohistiocytosis trigged by malaria infection and we review all reported cases secondary to Plasmodium falciparum malaria by searching PubMed publications till October 2019.Haemophagocytic lymphohistiocytos secondary to malaria should be suspected even in non-severe cases of malaria.
