The effect of radio-frequency （RF） or low-frequency （LF） bias voltage on the for- mation of amorphous hydrogenated carbon （a-C：H） films was studied on silicon substrates with a low methane （CH4） concentration （2-10 vol.%） in CH4＋Ar mixtures. The bias substrate was applied either by RF （13.56 MHz） or by LF （150 kHz） power supply. The highest hardness values （~18-22 GPa） with lower hydrogen content in the fihns （~20 at.%） deposited at 10 vol.% CH4, was achieved by using the RF bias, However, the films deposited using the LF bias, under similar RF plasma generation power and CH4 concentration （50 W and 10 vol.%, respectively）, displayed lower hardness （~6-12 GPa） with high hydrogen content （~40 at.%）. The structures analyzed by Fourier Transform Infrared （FTIR） and Raman scattering measurements provide an indication of trans-polyacetylene structure formation. However, its excessive formation in the films deposited by the LF bias method is consistent with its higher bonded hydrogen concentration and low level of hardness, as compared to the film prepared by the RF bias method. It was found that the effect of RF bias on the film structure and properties is stronger than the effect of the low-frequency （LF） bias under identical radio-frequency （RF） powered electrode and identical PECVD （plasma enhanced chemical vapor deposition） system configuration.

Uridine-diphosphate glucuronosyltransferase 1 A1(UGT1 A1) is an important conjugative enzyme in mammals that is responsible for the conjugation and detoxification of both endogenous and xenobiotic compounds. Strong inhibition of UGT1 A1 may trigger adverse drug/herb–drug interactions, or result in metabolic disorders of endobiotic metabolism. Therefore, both the US Food and Drug Administration(FDA)and the European Medicines Agency(EMA) have recommended assaying the inhibitory potential of drugs under development on the human UGT1 A1 prior to approval. This review focuses on the significance,progress and challenges in discovery and characterization of UGT1 A1 inhibitors. Recent advances in the development of UGT1 A1 probes and their application for screening UGT1 A1 inhibitors are summarized and discussed in this review for the first time. Furthermore, a long list of UGT1 A1 inhibitors, including information on their inhibition potency, inhibition mode, and affinity, has been prepared and analyzed. Challenges and future directions in this field are highlighted in the final section. The information and knowledge that are presented in this review provide guidance for rational use of drugs/herbs in order to avoid the occurrence of adverse effects via UGT1 A1 inhibition, as well as presenting methods for rapid screening and characterization of UGT1 A1 inhibitors and for facilitating investigations on UGT1 A1–ligand interactions.

Therapeutic targeting of metastatic breast cancer still remains a challenge as the tumor cells are highly heterogenous and exploit multiple pathways for their growth and metastatic spread that cannot always be targeted by a single-agent monotherapy ***,a rational approach through simultaneous targeting of several pathways may provide a better anti-cancer therapeutic effect We tested this hypothesis using a combination of two nutraceutical agents S-adenosylmethionine(SAM)and Vitamin D(Vit.D)prohormone[25-hydroxyvitamin D;/25(OH)D,]that are individually known to exert distinct changes in the expression of genes involved in tumor growth and *** results show that both SAM and 25(OH)D monotherapy significantly reduced proliferation and clonogenic survival of a panel of breast cancer cell lines in vitro and inhibited tumor growth,lung metastasis,and breast tumor cell colonization to the skeleton in ***,these effects were significantly more pronounced in the combination ***-Sequencing revealed that the transcriptomic footprint on key cancer-related signaling pathways is broader in the combination setting than any of the ***,comparison of the differentially expressed genes from our transcriptome analyses with publicly available cancer-related dataset demonstrated that the combination treatment upregulates genes from immune-related pathways that are otherwise downregulated in bone metastasis in *** SAM and Vit.D are both approved nutraceuticals with known safety profiles,this combination treatment may serve as a novel strategy to reduce breast cancer-associated morbidity and mortality。

BACKGROUND Clinically significant post-endoscopic retrograde cholangiopancreatography(ERCP) bacteremia(PEB) occurs in up to 5% of cases, while antibiotic prophylaxis is recommended only when an ERCP is unlikely to achieve complete biliary drainage. However, the current recommendations may not cover all potential risk factors for *** To identify novel risk factors for PEB and evaluate appropriateness of antibiotic *** A retrospective study of 1082 ERCP procedures performed between January 2012-December 2013 in a single tertiary medical center. Data collection included: Demographic and clinical characteristics such as pre and post procedure antibiotic treatment and bacterial blood cultures. Exclusion criteria were:(1) Age < 18 years;(2) Positive bacterial blood culture before ERCP;(3) Scheduled antibiotic treatment prior to ERCP;(4) Hospitalization longer than 14 d before ERCP;and(5) missing critical data. Stepwise Logistic Regression analysis and Decision Tree algorithms were used for prediction modeling of *** A total of 626 ERCPs performed in 434 patients were included. Mean age 66.49 ± 15.4 years and 46.5% were males. PEB prevalence was 3.7%. Antibiotic prophylaxis was administrated in 139/626(22.2%) cases but was indicated according to the guidelines only in 44/626(7%) cases. In all the PEB cases, prophylaxis was deemed not indicated. A stepwise logistic regression [receiver operating characteristic(ROC), 0.766], identified 3 variables as independent risk factors for PEB: Age at ERCP ≥ 75 years(OR, 3.780, 95%CI: 1.519-9.408, P = 0.004);Tandem EUS/ERCP with fine needle aspiration(FNA)(OR, 14.528, 95%CI: 3.571-59.095, P < 0.001);ERCP duration longer than 60 min(OR, 5.396, 95%CI: 1.86-15.656, P = 0.002). Aims Acorn weevils(Curculio spp.),endoparasitic granivorous insects,impede recruitment of fagaceous trees,and in turn affect community structure and ecosystem *** studies have made considerable progress in elucidating local factors that contribute to seed predation by acorn weevils,but it is still not clear how habitat configurations interplay with local factors in influencing Curculio *** this study,we assessed the roles of crop size,landscape configurations(area and isolation)and their interactions on the predation rate of seeds by acorn *** We studied acorn weevils feeding on the seeds of Castanopsis sclerophylla(Fagaceae)on two peninsulas and nine islands with varying areas and isolation levels in the Thousand-island Lake in Chun’an County,Zhejiang Province of ***,crop size was estimated for 130 trees and predation status was assessed for 26207 seeds from these trees during two *** linear mixed models were performed to clarify how island area and isolation interplayed with crop size to determine predation rate on a single *** Findings A negative relationship was detected between crop size and seed predation rate,indicating predator satiation at the tree *** suggests that acorn weevils tend to stay sedentary once they have arrived at a suitable *** fragmentation had significant effects on seed predation such that predation rate was higher on larger,less isolated ***,the relationship between crop size and predation rate was significantly changed by both island area and *** finding highlighted that the effects of habitat isolation might be overlooked relative to those of habitat loss.

Background It has long been an interesting question of whether withdrawal seizures in epileptic patients differ from habitual seizures in terms of semiology and *** presentation Here,we addressed this issue in a 40 year-old woman with drug-resistant focal epilepsy monitored by presurgical intracranial *** a part of this routine pre-operative investigation,anti-seizure medications(ASMs)were halted;as a result,multiple withdrawal seizures were recorded before ASM *** 4 days of invasive monitoring,we noticed three different phases in seizure organization:Acute withdrawal seizure(AWS):The first recorded seizure 10h after the implantation;the stabilized withdrawal seizures(SWS):seven habitual seizures recorded from 24h post implantation to readministration of ASMs;and the Non-withdrawal seizures(NWS):ten seizures recorded 24h after readministration of *** and SWS had the same semiology and same epileptic network,but the propagation time from the temporal pole to the para-hippocampal gyrus(PHG)and hippocampus ranged from no latency in AWS to up to 50 s in *** were electrographic seizures,without any apparent clinical *** onset in this type of seizure,as in the first two types,was in the temporal ***,NWS could last up to 3 min without involving the PHG or *** We concluded that in acute withdrawal seizures the propagation time of epileptic activity is significantly reduced without affecting ictal organization network or ***,ASM in this case had a remarkable influence on propagation rather than initiation of epileptic activity.

Drug-metabolizing enzymes(DMEs),a diverse group of enzymes responsible for the metabolic elimination of drugs and other xenobiotics,have been recognized as the critical determinants to drug safety and *** and understanding the key roles of individual DMEs in drug metabolism and toxicity,as well as characterizing the interactions of central DMEs with xenobiotics require reliable,practical and highly specific tools for sensing the activities of these enzymes in biological *** the last few decades,the scientists have developed a variety of optical substrates for sensing human DMEs,parts of them have been successfully used for studying target enzyme(s)in tissue preparations and living ***,molecular design principals and recent advances in the development and applications of optical substrates for human DMEs have been reviewed ***,the challenges and future perspectives in this field are also *** presented information offers a group of practical approaches and imaging tools for sensing DMEs activities in complex biological systems,which strongly facilitates high-throughput screening the modulators of target DMEs and studies on drug/herb-drug interactions,as well as promotes the fundamental researches for exploring the relevance of DMEs to human diseases and drug treatment outcomes.
