Non-ribosomal peptides are a group of structurally diverse natural products with various important therapeutic and agrochemical *** pyrrolizidine alkaloids(PAs),containing a scaffold of two fused five-membered ring system with a nitrogen atom at the bridgehead,have been found to originate from a multidomain non-ribosomal peptide synthetase to generate indolizidine intermediates,followed by multistep oxidation,catalysed by single Bayer-Villiger(BV)enzymes,to yield PA *** bacterial PAs are rare in natural product inventory,bioinformatics analysis suggested that the biosynthetic gene clusters(BGCs)that are likely to be responsible for the production of PA-like metabolites are widely distributed in bacterial ***,most of the strains containing PA-like BGCs are not deposited in the public domain,therefore preventing further assessment of the chemical spaces of this group of bioactive ***,we report a genomic scanning strategy to assess the potential of PA metabolites production in our culture collection without prior knowledge of genome *** the strains tested,we found fifteen contain the key BV enzymes that are likely to be involved in the last step of PA ring *** one-strain-many-compound(OSMAC)method,supported by a combination of HR-MS,NMR,SMART 2.0 technology,and GNPS analysis,allowed identification and characterization of a new[5+7]heterobicyclic carbamate,legoncarbamate,together with five known PAs,bohemamine derivatives,from Streptomyces ***37,a Ghanaian soil *** absolute stereochemistry of legoncarbamate was determined by comparison of measured and calculated ECD *** displays antibacterial activity against *** ATCC 25922 with an MIC value of 3.1μg/***,a biosynthetic model of legoncarbamate and other bohemamines was proposed based on the knowledge we have gained so far.

This study tries to understand in which ways stereotypes negatively affect women's development and thus wonder if it is possible to develop an identity that is resilient to these gendered stereotypes. The study first looks at theoretical approaches to stereotypes and identity politics for women. It is established that stereotypes can restrain women's freedom by limiting the way they perceive their future path and ambitions, through self-criticism, demotivation and disengagement. The study also considers stereotypes within structures of power-relations which define standards of acceptance and exclusion, and shows that stereotyping discourses determine women's identity. Then, the study wonders how this theoretical framework applies to female entrepreneurs — who have been considered has women challenging stereotyping structures in certain social contexts. It is found that stereotypes pressurise female entrepreneurs to adapt to one mould, which causes women who do not fit this model to be overlooked as entrepreneurs by society — denying them the freedom to develop their identity as such. The study's solution to counteract the negative effect of stereotypes is the development of role models. It is argued that because women entrepreneurs essentially represent resilience and resistance in the face of sexist stereotypes given the implications of entrepreneurial life, they inevitably become role models, regardless of their different paths and individual identities. Apart from showing women they can confidently follow their ambitions, role models are also vital in today's society because they create new dynamisms within power-relations. Female entrepreneur role models promote notions of empowerment and collective power distinct from that of men's power-play and control. Therefore, through the promotion of this new definition of power, role models influence the transformation of the social structure in ways which do not foster stereotyped discourses and enable women to develop

Texture is a complex trait and a major component of fruit quality in *** the major effect of MdPG1,a gene controlling firmness,has already been exploited in elite cultivars,the genetic basis of crispness remains poorly *** further improve fruit texture,harnessing loci with minor effects via genomic selection is therefore *** this study,we measured acoustic and mechanical features in 537 genotypes to dissect the firmness and crispness components of fruit *** of across-year phenotypic values for these components were calculated using a model calibrated with 8,294 SNP *** best prediction accuracies following cross-validations within the training set of 259 genotypes were obtained for the acoustic linear distance(0.64).Predictions for biparental families using the entire training set varied from low to high accuracy,depending on the family *** adding siblings or half-siblings into the training set did not clearly improve predictions,we performed an optimization of the training set size and composition for each validation *** allowed us to increase prediction accuracies by 0.17 on average,with a maximal accuracy of 0.81 when predicting firmness in the‘Gala’×‘Pink Lady’*** results therefore identified key genetic parameters to consider when deploying genomic selection for texture in *** particular,we advise to rely on a large training population,with high phenotypic variability from which a‘tailored training population’can be extracted using a priori information on genetic relatedness,in order to predict a specific target population.

Rubisco and fructose-1,6-bisphosphate aldolases （FBAs） are involved in CO2 fixation in chloroplasts. Both enzymes are trimethylated at a specific lysine residue by the chloroplastic protein methyltransferase LSMT. Genes coding LSMT are present in all plant genomes but the methylation status of the substrates varies in a species-specific manner. For example, chloroplastic FBAs are naturally trimethylated in both Pisum sati- vum and Arabidopsis thaliana, whereas the Rubisco large subunit is trimethylated only in the former spe- cies. The in vivo methylation status of aldolases and Rubisco matches the catalytic properties of AtLSMT and PsLSMT, which are able to trimethylate FBAs or FBAs and Rubisco, respectively. Here, we created chimera and site-directed mutants of monofunctional AtLSMT and bifunctional PsLSMT to identify the molecular determinants responsible for substrate specificity. Our results indicate that the His-Ala/Pro- Trp triad located in the central part of LSMT enzymes is the key motif to confer the capacity to trimethylate Rubisco. Two of the critical residues are located on a surface loop outside the methyltransferase catalytic site. We observed a strict correlation between the presence of the triad motif and the in vivo methylation status of Rubisco. The distribution of the motif into a phylogenetic tree further suggests that the ancestral function of LSMT was FBA trimethylation. In a recent event during higher plant evolution, this function evolved in ancestors of Fabaceae, Cucurbitaceae, and Rosaceae to include Rubisco as an additional substrate to the archetypal enzyme. Our study provides insight into mechanisms by which SET-domain protein methyltransferases evolve new substrate specificity.

This article describes the FAIRification process(which involves making data Findable, Accessible, Interoperable and Reusable—or FAIR—for both machines and humans) for data related to the impact of COVID-19 on migrants, refugees and asylum seekers in Tunisia, Libya and Niger, according to the scheme adopted by GO FAIR. This process was divided into three phases: pre-FAIRification, FAIRification and postFAIRification. Each phase consisted of seven steps. In the first phase, 118 in-depth interviews and 565 press articles and research reports were collected by students and researchers at the University of Sousse in Tunisia and researchers in Niger. These interviews, articles and reports constitute the dataset for this research. In the second phase, the data were sorted and converted into a machine actionable format and published on a FAIR Data Point hosted at the University of Sousse. In the third phase, an assessment of the implementation of the FAIR Guidelines was undertaken. Certain barriers and challenges were faced in this process and solutions were found. For FAIR data curation, certain changes need to be made to the technical process. People need to be convinced to make these changes and that the implementation of FAIR will generate a long-term return on investment. Although the implementation of FAIR Guidelines is not straightforward, making our resources FAIR is essential to achieving better science together.

Shaping the eye from embryonic stem cells: Biological and medical implications

Organogenesis is regulated by a complex network of intrinsic cues, diffusible signals and cell/cell or cell/matrix interactions that drive the cells of a prospective organ to differentiate and collectively organize in three dimensions. Generating organs in vitro from embryonic stem (ES) cells may provide a simplified system to decipher how these processes are orchestrated in time and space within particular and between neighboring tissues. Recently, this field of stem cell research has also gained considerable interest for its potential applications in regenerative medicine. Among human pathologies for which stem cell-based therapy is foreseen as a promising therapeutic strategy are many retinal degenerative diseases, like retinitis pigmentosa and age-related macular degeneration. Over the last decade, progress has been made in producing ES-derived retinal cells in vitro, but engineering entire synthetic retinas was considered beyond reach. Recently however, major breakthroughs have been achieved with pioneer works describing the extraordinary self-organization of murine and human ES cells into a three dimensional structure highly resembling a retina. ES-derived retinal cells indeed assemble to form a cohesive neuroepithelial sheet that is endowed with the intrinsic capacity to recapitulate, outside an embryonic environment, the main steps of retinal morphogenesis as observed in vivo. This represents a tremendous advance that should help resolving fundamental questions related to retinogenesis. Here, we will discuss these studies, and the potential applications of such stem cell-based systems for regenerative medicine.

Dear Editor,Infertility affects about 15%of couples of childbearing age.i About half of these cases can be attributed predominantly to a male factor,such as a quantitative or qualitative impairment in *** etiology of male infertility is related to anatomic,genetic,and environmental causes,being sometimes multifactorial and of unknown or idiopathic *** present,genetic screening for nonobstructive azoospermia(NOA)is generally limited to karyotyping for the identification of chromosome abnormalities such as 47,XXY,46,X,der(X)t(X;Y)(p22.3;p11.2),chromosome rearrangements,and Y chromosome ***-genome analyses have shown that over 100 gene variants are associated with a male infertility phenotype,and the list continues to grow;hence,a large number of genes are involved in *** continuing identification of genes responsible for male infertility in general and azoospermia,in particular,is a major challenge in both research and the diagnosis ofmale infertility.
