Localization of AKAP4 and tubulin proteins in sperm with reduced motility

Aim： To perform screening, related to A-kinase anchoring proteins 4 （AKAP4） and tubulin proteins, in spermatozoa with absent or severely reduced motility in order to detect the status of the fibrous sheath and the axonemal structure. Methods： An immunocytochemical study of tubulin, used as a positive control, and AKAP4 was carded out to detect the presence and the distribution of these proteins in different sperm samples. The morphological characteristics of sperm were studied by transmission electron microscope （TEM） and the results were elaborated using a formula reported in previous studies. PCR was carried out on DNA extracted from peripheral blood lymphocytes to analyse partial sequences of the Akap4 and Akap3 genes. Results： Immunolabelling of tubulin and AKAP4 showed different patterns, which led us to divide the patients into groups. In group I, the absence of AKAP4 and tubulin was revealed, although these patients did not show alterations in the Akap4/Akap3 binding site. TEM evaluation highlighted that a high presence of necrosis was associated with total sperm immotility. In group Ⅱ, a regular AKAP4 and tubulin signal was present, although motility was reduced and TEM analysis revealed the presence of immaturity. In group Ⅲ, in which a weak AKAP4 label associated with normal tubulin staining and reduced motility was observed, a severe disorganization of the fibrous sheath was highlighted by TEM. Conclusion： While the role of AKAP4 in sperm motility is unclear, absent or weak AKAP4-1abelling seems to be associated with absent or weak sperm motility.

Building Information Modelling(BIM)is becoming increasingly present in every stage of assets’*** and more,the BIM approach is related to the sustainability assessment protocols which play an important role in reducing the impact on the *** the design and construction phase,the products’sustainability certification is a key issue to be managed and controlled,to achieve higher efficiency in *** this research,a BIM-based methodology to automate the sustainability certification process in construction phase has been *** to the proposed method,the contractor proposes a building component to the work supervisor,by uploading the related technical datasheet in a Common Data Environment(CDE).If the component meets the performance requirements defined for the construction stage and agreed with the involved parties(work supervisor,client and sustainability accredited professional),it is validated and uploaded in the BIM model by the BIM *** methodology has been tested in a case study,confirming the effectiveness of the proposed ***,it should be further ***,it can be improved and a higher level of automation can be achieved,in order to cope with product dictionaries and templates under development in CEN technical committee 442.

18,X,Y aneuploidies and transmission electron microscopy studies in spermatozoa from five carriers of different reciprocal translocations

We analysed ejaculated spermatozoa from five infertile men with different balanced reciprocal translocations to contribute to the study of meiotic segregation of chromosomes 18, X and Y and also to evaluate sperm morphology by transmission electron microscopy （TEM） analysis. Conventional lymphocyte karyotype analyses highlighted dif- ferent reciprocal balanced translocations： t（12; 13）, t（4;9）, t（X;8）, t（8; 10） and t（3; 16）. Semen analysis was performed by light and TEM. Fluorescence in situ hybridization was performed directly on sperm nuclei using centromeric probes for chromosomes 18, X and Y. The carriers of the balanced reciprocal translocations considered in the present study showed a very similar pattern of sperm pathologies： diffused presence of apoptosis and immaturity. All patients showed meiotic segregation derangements, highlighted by the presence of sperm diploidies and sex chromosome disomies particularly related to the failure of the first meiotic division. However, an increased incidence of chromosome 18 aneuploidy was detected in spermatozoa from t（X;8） and t（8;10） carriers. We have also reported values from sex chromosomes such as t（X;8）, although the X chromosome was involved in translocation. Since patients with reciprocal translocations and spermatogenetic impairment are candidates for intracytoplasmic sperm injection cycles, the study of sperm parameters, and particularly of the level of aneuploidy rates, would provide better information for couples at risk and would contribute to the data in the literature for a better understanding of the effects of chromosomal rearrangement on the whole meiotic process and, in particular, on chromosomes not involved in translocation.

Monitoring Landslide-Induced Displacements with TerraSAR-X Persistent Scatterer Interferometry (PSI): Gimigliano Case Study in Calabria Region (Italy)

This work provides a detailed detection of landslide-induced displacements at local scale on Gimigliano site (Italy), by means of PSI (Persistent Scatterers Interferometry) analysis, exploiting TerraSAR-X data acquired in November 2010-October 2011. In February-March 2010, several landslides affected Gimigliano, following high-intensity precipitation, and causing damages to structures and roads. In order to assess any spatial and temporal patterns of deformation, the present X-band PS data were compared with historical motion rates derived from ERS1/2 and ENVISAT satellites, and with geological and geomorphological evidences resulting from auxiliary data such as landslide databases and orthophotos referred to different dates, finally validated with recent field checks. The PSI analysis of the historical ground motion rates highlighted that the modern built-up area located downhill was already affected by surface ground deformation since 1993. A significant enlargement of the instability phenomena is detected across time. The recent PS data analysis and the in situ observations permitted us to accurately update the ground movements in the investigated site: their evidence allowed assessing a reactivation of the large deep-seated landslide zone on which the new urbanized area had built-up, with an average deformation rate of about 8 - 9 mm/yr. The higher instability phenomenon is affecting the eastern part of Gimigliano old village, with a mean displacement velocity reaching up values of -30 mm/yr, in the radar temporal acquisition of only 10 months. These outcomes can be taken into account for further hazard-reduction analysis and to support risk mitigation design within the investigated area.

Airborne Particles during a Firework Festival in Belvedere ***, South-Western Italian Coast

The effects of a Summer Firework Festival on the air quality were studied for the first time in a rural area of Southern Italy. The case study analyzes the physical-chemical properties of airborne particles collected during the Note di Fuoco (NDF) Festival, which took place in Belvedere *** in 2016. The sampling period was peculiar since in one week three different kinds of events have succeeded: three days during the NDF Festival with the concurrence of the festival and the firework displays, one day with the typical street Market involving food stalls and specialty shops, and three days with no specific events, considered as background conditions. Particulate Matter in different two size fractions (PM2.5 and PM10) was simultaneously collected on a daily basis and then chemically analyzed for major and rare metal content as well as for organic and elemental carbon determination. Levels of particles were even below the European air quality limit values. However, the day with the country market and during the three days of the NDF festival, the finer and respirable particulate fraction, PM2.5, showed an increase of 46% and 84%, respectively, over the mean concentration values observed during the background days. Both elemental and organic carbon, even in the finer fraction showed an increment up to 30%. All major, trace and rare elements were found in higher concentrations during the festival with respect to those recorded in days with no events. In our case study, K was recognized as the best fireworks tracer because its level doubled during the festival. Typical firework tracers like Fe, Ti, Mn, Pb and Sr resulted in greater concentrations, up to 50%.
