Microscopic Characteristics, Chromatographic Profiles and Inhibition of Peroxidase Activity of the Leaves of <i>Manihot esculenta</i>and <i>Manihot glaziovii</i>, Consumed as Traditional Vegetables

Methanolic extracts from the leaves of Manihot esculenta (Two cultivars) and Manihot glaziovii, consumed as traditional vegetables in DR. Congo was chemically characterized by Thin layer Chromatography and High Performance Liquid Chromatography. In vitro biochemical activities of extracts against Radical Oxidative Species (ROS) production were assessed in cellular models, on enzymes, Myeloperoxidase (MPO) and Horseradish Peroxidase (HRP) involved in inflammation. The microscopic analysis of the powder of leaves showed that each species displays specific and discriminating botanical microscopic features. Varieties of M. esculenta had a chemical fingerprint different from M. glaziovii. The majority of compounds were polyphenols, represented mainly by rutin, kaempferol-3-O-rutinoside, amentoflavone, phenolic acids such as gallic acid. All extracts exhibited high cellular antioxidant activity in the range of 0.1 to 10 μg·mL-1 using lucigenin with neutrophils, but a moderate cellular antioxidant activity ranging between 10 and 100 μg·mL-1 with DCFDA on HL60 monocytes. Extracts from Manihot leaves showed a pronounced inhibitory effect on the production of extracellular ROS, on HRP and myeloperoxidase activity. Cellular antioxidant activities, the inhibitory effect on HRP of extracts from M. glaziovii, M. esculenta cultivar Mwambu were significantly higher, but their inhibitory effect on the activity of MPO was lower than those of M. esculenta cultivar TEM 419. The biological activities of Manihot esculenta and Manihot glaziovii were well correlated to their phytochemicals that could justify their traditional use as vegetables, potential functional foods or nutraceutical resources and medicines.

Intraperitoneally Administered Lidocaine Attenuates Thermal Allodynia in a Murine “Two-Hit” Chronic Constriction Injury Model

Background: Mechanical ventilation (hit one) during surgery (hit two) is often needed and both induce an inflammatory response. Dysregulation of the inflammatory response can cause chronic postoperative pain. Methods: Healthy C57BL6 mice (n = 56) were mechanically ventilated (MV) and allocated to receive sham (MV-sham) or mechanically ventilation with chronic constriction injury (MV-CCI) surgery in the left hind paw. Plasma interleukin (IL)-1β, IL-6, IL-10, keratinocyte derived chemokine (KC) and tumor necrosis factor (TNF)-α were determined on day 0 and 16. Sensory testing was performed on day 0, 3, 7 and 16 by cold plate test (number of lifts (NOL) and cumulative reaction time (CRT)) and von Frey test. The effect of lidocaine on cytokines and sensory testing was analyzed. Results: MV-Sham showed an increase in IL-1β and TNF-α, and MV- CCI-lido increased levels of KC compared with MV on day 0. No difference in cytokine levels was observed on day 16. NOL of the left paw versus the right was increased in MV-CCI on day 7, and in MV-CCI-lido on day 7 and 16. The NOL of the left paw was decreased in MV-sham and MV-CCI-lido compared with MV-CCI on day 16. The CRT of the left paw was increased for MV-CCI on day 3 and 7, and for MV-CCI-lido on day 7. On day 16, MV-sham and MV-CCI-lido showed a decreased CRT of the left paw compared with MV-CCI. Conclusion: Nerve injury and not systemic inflammatory response seems mandatory for development of neuropathic pain in this “two-hit” model. Lidocaine attenuates cold allodynia in mice.

Twin Anemia Polycythemia Sequence: Knowledge and Insights After 15 Years of Research

Twin anemia polycythemia sequence (TAPS) is a chronic form of unbalanced feto-fetal transfusion through minuscule placental anastomoses in monochorionic twin pregnancies, leading to anemia in the donor twin and polycythemia in the recipient twin. TAPS can occur spontaneously in up to 5% of monochorionic twins or can arise in 2%-16% of cases after incomplete laser surgery for twin-twin transfusion syndrome. TAPS can develop across the entire second and third trimester. Antenatal diagnosis for TAPS is reached via Doppler measurement of the fetal middle cerebral artery peak systolic velocity, showing an increased velocity in the donor, combined with a decreased velocity in the recipient. Treatment options for TAPS include expectant management, preterm delivery, intrauterine blood transfusion with or without a partial exchange transfusion, fetoscopic laser surgery and selective feticide. The best treatment option is unclear and is currently being investigated in an international multicenter randomized trial (the TAPS trial). Spontaneous fetal demise occurs in 5%-11% of TAPS twins, more often in donors (8%-18%) than in recipients (2%-5%). Severe long-term neurodevelopmental impairment is seen in 9% of TAPS twins, with donors having an increased risk for cognitive impairment and hearing problems (15%).

Background: IgG4-related disease is a rare autoimmune condition that presents with lymphoplasmacytic infiltrate and fibrosis in the organ affected. Isolated testicle involvement is uncommon and there are only a few cases reported in the literature. Case Presentation: We report a case of isolated chronic orchitis due to IgG4 hypersecretion in a 61-year-old patient that evolved with asymptomatic tumor-like mass growth and was treated with left orchiectomy. Histopathological study revealed orchitis related to IgG4 hypersecretion disease. Conclusion: IgG4-related disease can be manifested as a multi or single-organ disorder. Most diagnoses are made after surgery with histopathological analysis. Most of the cases in literature stand out the difficulty in diagnosis and necessity of high suspicion due to this condition’s similarity with neoplasm presentation.

The aim of this review is to investigate the application and latest developments of the Diffusive Gradients in-thin films (DGT) with a focus on the mobility and bioavailability of heavy metals in soil. Soil chemical extractions are extensively used to predict nutrients elements in the soil. However, these measurements have their weaknesses and shortcomings. Comparing DGT with conventional extraction methods, DGT is a sampling technique with significant advantages;including speciation capabilities, sensitivity, time-in- tegrated signal, low risk of contamination and time averaged concentrations. These findings have strengthened the usefulness of the DGT technique as a potential monitoring tool for soil with heavy metal contamination. Studies which have used the DGT technique to evaluate processes important to bioavailability have been booming in the last 13 years, especially its application in soils science. Some recent studies have shown a good relationship between the measurement of metals concentrations in soil and plant by DGT, and cohesive results have been obtained from these measurements when they are based on the DGT technique. DGT is a newly established procedure to assess the bioavailability of trace elements in sediments and soils, and its applications are still in the early stage of testing. Therefore, future application of DGT is likely to include the studies of HMs contamination in soil for risk assessment and transfer rates to the food chain, as some studies have indicated the potential of DGT in these areas.

Cosmeceuticals are the fastest growing products in the cosmetics field and represent a hybrid between cosmetics and drugs. The main benefits reported for cosmeceutical herbal extracts, used in skin care, include antioxidant and antimicrobial activities and the tyrosinase inhibiting effect. The aim of this study was to evaluate the antioxidant and antibacterial activities of extracts from Chromolaena odorata, Mitracarpus villosus, Senna alata, Tetradenia riparia, Cannabis sativa and Pterocarpus soyauxii used traditionally in the management of skin and skin annexes pathologies;additionally to determine their microscopical characteristics and their chromatographic fingerprints. Microscopic analyses showed the specific botanical microscopic characters for each plant that constitute the database useful for the identification and authentication of these botanicals. To the best of our knowledge, our work is the first to describe the microscopic histological characters of studied plants except for Cannabis sativa. The results of the TLC chromatographic analysis revealed the richness in phenolic compounds, terpenes in all extracts and Pterocarpus soyauxii contained coumarins. All extracts displayed high ABTS and DPPH radical-scavenging activities connected with their IC50 values at the concentration range of 1 - 125 μg/mL with Cannabis sativa and Pterocarpus soyauxii as the most active. All plant extracts exhibited antibacterial activity on Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus to varying degrees. Cannabis sativa exhibited strong effect on Staphylococcus aureus and Escherichia coli. Chromolaena odorata and Tetradenia riparia showed a marked effect on Escherichia coli. Standardized quantitative measurements of wrist cartilage in healthy humans using 3T magnetic resonance imaging

AIM: To quantify the wrist cartilage cross-sectional area in humans from a 3D magnetic resonance imaging(MRI) dataset and to assess the corresponding reproducibility. METHODS: The study was conducted in 14 healthy volunteers(6 females and 8 males) between 30 and 58 years old and devoid of articular pain. Subjectswere asked to lie down in the supine position with the right hand positioned above the pelvic region on top of a home-built rigid platform attached to the scanner bed. The wrist was wrapped with a flexible surface coil. MRI investigations were performed at 3T(Verio-Siemens) using volume interpolated breath hold examination(VIBE) and dual echo steady state(DESS) MRI sequences. Cartilage cross sectional area(CSA) was measured on a slice of interest selected from a 3D dataset of the entire carpus and metacarpalphalangeal areas on the basis of anatomical criteria using conventional image processing radiology software. Cartilage cross-sectional areas between opposite bones in the carpal region were manually selected and quantified using a thresholding ***: Cartilage CSA measurements performed on a selected predefined slice were 292.4 ± 39 mm2 using the VIBE sequence and slightly lower, 270.4 ± 50.6 mm2, with the DESS sequence. The inter(14.1%) and intra(2.4%) subject variability was similar for both MRI methods. The coefficients of variation computed for the repeated measurements were also comparable for the VIBE(2.4%) and the DESS(4.8%) sequences. The carpus length averaged over the group was 37.5 ± 2.8 mm with a 7.45% between-subjects coefficient of variation. Of note, wrist cartilage CSA measured with either the VIBE or the DESS sequences was linearly related to the carpal bone length.
