AIM To establish a permanent piwi like RNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CRISPR-Cas9 system and analyze phenotypic modifications as well as gene expression *** CRISPR-Cas9 system used was purchased from Dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, Santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. RESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. Moreover, functional analysis based on grouping of all differentially expressed m RNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(DOCK2, ZNF503, PDE4 D, ABL1, ABL2, LPAR1, SMAD2, WASF3 and DACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). CONCLUSION Taken together, these data reinforce the idea that PIWIL1 plays a crucial role in the signaling pathway of gastric cancer, regulating several genes involved in migration and invasion processes; therefore, its use as a therapeutic target may generate promising results in the treatment of gastric cancer.

T-lymphoblastic lymphoma(T-LBL)is a rare and aggressive form of non-Hodgkin’s lymphoma and little is known about their molecular ***,complex karyotypes were already related to this group of malignancy and associated with poor ***,we describe a 17-year-old female being diagnosed with T-LBL and a normal karyotype after standard G-banding with trypsin-Giemsa(GTG)-***,further analyses including high-resolution molecular approaches,array-comparative genomic hybridization(aCGH),multiplex ligation-dependent probe amplification,fluorescence in situ hybridization and multicolor chromosome banding revealed a cryptic complex karyotype,NUP214-ABL1 gene fusion,episomes and intra-tumor genetic *** addition,homozygous loss of CDKN2A,as well as amplification of oncogene TLX1(HOX11)were ***,NUP214-ABL1 fusion gene replicated autonomously in this case as ***,highly amplification of NUP214-ABL1 fusion gene defines possibly a new subgroup of T-LBL patients which accordingly could benefit from treatment with tyrosine kinase *** episomes are missed in standard karyotyping aCGH should be performed routinely in T-LBL to possibly detect more of such cases.

为了提高人体肌电信号对于下肢动作识别的准确率,提出一种基于遗传算法(GA)优化的径向基(RBF)神经网络分类模型。通过采集人体日常8种下肢动作的表面肌电信号并选择“sym6”系小波函数对肌电信号进行滤波预处理,使用主成分分析法(PCA)对时频域特征降维,把特征向量输入GA算法优化的RBF神经网络进行训练和识别。实验结果表明,该方法对同一受试者8种下肢动作的平均识别率为94.00%±0.45%;对15位不同受试下肢动作识别率达到89.30%,比传统BP神经网络的识别准确率提高11.8%,预测时间缩短6 s。所提出的方法为肌电信号应用于下肢智能康复机器人的意图识别研究提供参考,有助于病人的康复。

Based on Maxwell’s constraint counting theory, rigidity percolation in GexSe1-x glasses occurs when the mean coordination number reaches the value of 2.4. This corresponds to Ge0.20Se0.80 glass. At this composition, the number of constraints experienced by an atom equals the number of degrees of freedom in three dimensions. Hence, at this composition, the network changes from a floppy phase to a rigid phase, and rigidity starts to percolate. In this work, we use reverse Monte Carlo (RMC) modeling to model the structure of Ge0.20Se0.80 glass by simulating its experimental total atomic pair distribution function (PDF) obtained via high energy synchrotron radiation. A three-dimensional configuration of 2836 atoms was obtained, from which we extracted the partial atomic pair distribution functions associated with Ge-Ge, Ge-Se and Se-Se real space correlations that are hard to extract experimentally from total scattering methods. Bond angle distributions, coordination numbers, mean coordination numbers and the number of floppy modes were also extracted and discussed. More structural insights about network topology at this composition were illustrated. The results indicate that in Ge0.20Se0.80 glass, Ge atoms break up and cross-link the Se chain structure, and form structural units that are four-fold coordinated (the GeSe4 tetrahedra). These tetrahedra form the basic building block and are connected via shared Se atoms or short Se chains. The extent of the intermediate ranged oscillations in real space (as extracted from the width of the first sharp diffraction peak) was found to be around 19.6 ?. The bonding schemes in this glass are consistent with the so-called “8-N” rule and can be interpreted in terms of a chemically ordered network model.

Anomalous structural characteristics of the so-called first sharp diffraction peak (FSDP) that arises in the total static structure functions of network-forming glasses and liquids at around 1-2 A-1 have been reviewed and discussed in details. Unlike other peaks in the static structure functions, the FSDP has anomalous dependencies on temperature, pressure and composition. Despite the fact that the FSDP is considered as a signature of intermediate range order (IRO) in network-forming glasses and liquids, its structural origin remains unclear and till now, it forms a subject of debate. A brief account for some anomalous characteristics of the FSDP followed by the different controversial interpretations about its structural origin has been reviewed and discussed. Some of the interpretations that seem to be inconsistent with recent experimental results have been ruled out. The most likely structural origins for the occurrence of the FSDP have been highlighted and discussed in details.

Malignant glioma remains one of the most intractable of human cancers principally due to the highly infiltrative nature of these neoplasms. The use of neural precursor cells (NPC) has received considerable attention based on their ability to selectively migrate towards disseminated areas of tumor in vivo and their described ability to deliver tumor-directed therapies specifically to infiltrating tumor cells. Fundamental to optimizing the use of these cells for potential clinical translation is the development of an understanding regarding the biologic cues that govern their ability to migrate towards infiltrative glioma foci. To this end, in this paper we detail that NPC selected for double-expression of the glial-precursor marker A2B5 and the cell-surface chemokine receptor, CXCR4, demonstrate enhanced in vitro gliomadirected tropism. These findings demonstrate the relevance of these markers for the phenotypic segregation of an optimally tumor-tropic NPC sub-population as a means of enhancing NPC-based therapeutic strategies for the treatment of glioma.
