The goal of thi. study was to determi.e whether mutati.n of the Mn-bi.di.g si.e of wi.d-type recombi.ant Phlebi. radi.ta manganese peroxi.ase 3 affected the pH-dependence ki.eti. parameters. pH range i.vesti.ated was 2.5 – 12.0. The catalyti. effi.i.ncy of the mutant enzymes at hi.h and low pH i. compari.on to the wi.d-type was i.vesti.ated usi.g standard rPr-MnP3 protocol. Wi.d-type recombi.ant Phlebi. radi.ta MnP3 enzyme showed opti.al acti.i.y wi.h Mn (i.) as substrate at pH 5.0 and remai.ed moderately acti.e (approxi.ately 40%) i. the pH range of 6.0 - 9.0. The rPr-MnP3 mutants’ maxi.um acti.i.y ranged between 5.5 and 8.0. Wi.d-type and mutants rPr-MnP3 enzymes exhi.i.ed a si.i.ar pH profi.e wi.h opti.um pH of 3.0 for ABTS oxi.ati.n. Mutati.n has severely decreased the catalyti. effi.i.ncy for Mn (i.) oxi.ati.n at pH 5.0. The rPr-MnP3 enzymes showed enhanced affi.i.y for Mn (i.) at alkali.e pH and a more alkali.e range for catalysi. than ever reported for any Manganese Peroxi.ase. Thi. study reveals that at hi.her pH, rPr-MnP3 can functi.n wi.h alternati.e li.ands i. the Mn (i.) si.e and does not have an absolutely obli.ate requi.ement for an all carboxylate li.and set. These results further strongly confi.m that Mn2+ bi.di.g si.e i. the only producti.e catalyti. si.e for Mn (i.) oxi.ati.n.

Thi. i.vesti.ati.n i. ai.ed at understandi.g the speci.i. role of pH and calci.m i.ns on the acti.i.y and stabi.i.y of wi.d-type recombi.ant Phlebi. radi.ta manganese peroxi.ase 3 (rPr-MnP3). The pH-dependent cycle of reacti.ns for rPr-MnP3 was evaluated by i.vesti.ati.g ti.e-dependent changes i. the acti.i.y and electroni. absorpti.n spectrum of *** rPr-MnP3 had maxi.um effi.acy (kcat/Km) for Mn (i.) oxi.ati.n at pH 5.0 and 3.0 for oxi.ati.n of ABTS. Rai.i.g the pH of a soluti.n of resti.g rPr-MnP3 from pH 6.7 (form XH) to pH 8.6 (form X&mi.us;), a rapi. alkali.e transi.i.n occurs. Leavi.g the X&mi.us; form of the enzyme at pH 8.6, i. slowly becomes converted to a thi.d form of the enzyme Y&mi.us;, whi.h returned to the ori.i.al XH form of the enzyme at pH 6.7. Recovery of form XH from form Y&mi.us; occurred through an i.termedi.te Z form. The pH i.acti.ati.n of rPr-MnP3 followed fi.st-order ki.eti.s. The rate of formati.n of XH from Z i. pH-dependent and bi.hasi. i. nature, wi.h measured rate constants (k) = 0.25 mi.&mi.us;1, and half-li.e (T1/2) = 2.8 mi.. The pH-dependent properti.s observed may be i.di.ati.e of a greater degree of conformati.nal flexi.i.i.y at rPr-MnP3 acti.e si.e due to di.rupti.n of the haem-li.ked hydrogen-bondi.g network i. the di.tal haem pocket. Calci.m i.ns were observed to si.ni.i.antly stabi.i.ed the enzyme’s spectral features and reduce the loss of acti.i.y duri.g the alkali.e pH transi.i.n. Calci.m i.ns enhance the recovery of the i.i.i.l acti.i.y but cannot prevent the fi.al ti.e-dependent i.reversi.le denaturati.n and aggregati.n.

Manganese peroxi.ases (MnPs) are i.teresti.g enzymes i. protei. engi.eeri.g, ai.ed at maxi.i.i.g i.dustri.l bi.processes such as li.ni. degradati.n and bi.fuel producti.n. cDNA of the secreted short-type of MnP from Phlebi. radi.ta (Pr-MnP3) has been successfully engi.eered and ampli.i.d by polymerase chai. reacti.n (PCR). Fi.e mutant genes (E40H, E44H, E40H/E44H, D186H and D186N) of recombi.ant Phlebi. radi.ta MnP3 (rPr-MnP3) were generated. The wi.d-type and the mutant genes were expressed i. Escheri.hi. coli.(W3110 strai.) and the resultant body protei.s were lysed, puri.i.d and refolded i.to acti.e enzymes. 6% - 7% recovery of pure and fully acti.e rPr-MnP3 for wi.d-type and mutants were obtai.ed and the avai.abi.i.y of rPr-MnP3 enzymes wi.l greatly faci.i.ate i.s structure-functi.n relati.nshi.s studi.s. rPr-MnP3 mass was characteri.ed usi.g SDS-PAGE and MALDi.TOF mass spectrometry. Molecular wei.ht of both the wi.d-type and mutant rPr-MnP3 enzymes was approxi.ately 36 kDa. Thi. descri.es the spectral characteri.ati.n of the wi.d-type and mutant rPr-MnP3 enzymes wi.h are very close si.i.ari.i.s;substanti.lly hi.h spi. haem enzymes. Therefore we report the engi.eeri.g, cloni.g, expressi.n, refoldi.g/acti.ati.n of MnP3 genes and preli.i.ary characteri.ati.n of the wi.d-type and mutant Phlebi. radi.ta MnP3 enzymes.

i. tropi.al envi.onments, most soybean growth studi.s have uti.i.ed ri.e husk bi.char (RHB) i. soi., even though i. i. low i. ni.rogen, potassi.m, and phosphorous. Thi. may not gi.e short-term agronomi. performance relati.e to enri.hed bi.char. Moreover, the i.pact of i.oculati.g soybean seeds wi.h atmospheri. ni.rogen-fi.i.g bacteri.m Bradyrhi.obi.m japoni.um on nodulati.n and grai. yi.ld has produced i.conclusi.e fi.di.gs i. the li.erature. Thi. research therefore ai.s to assess the effect of poultry manure (PM), poultry manure bi.char (PMB) and RHB alone and i. combi.ati.ns on grai. yi.ld, dry shoot and root bi.mass of soybeans i. the semi.deci.uous agro-ecologi.al zone. i. addi.i.n, the effect of B. japoni.um i.oculated and non-i.oculated soybean seeds on nodulati.n and grai. yi.ld was also i.vesti.ated. The treatments followed a spli. plot desi.n studyi.g i.oculati.n and non-i.oculati.n, soi. amendments (ei.ht), and control subplot factors, respecti.ely. The results show that the amendment of a ferri. acri.ol wi.h 4 Mg∙ha−1 PM, 10 Mg∙ha−1 RHB + 2 Mg∙ha−1 PM, and 5 Mg∙ha−1 RHB + 4 Mg∙ha−1 PMB wi.h B. japoni.um i.oculated seed produced si.ni.i.antly greater grai. yi.ld (p = 0.05). PM treatment had a si.ni.i.ant (p B. japoni.um-i.oculated soybean seeds si.ni.i.antly (p = 0.014) i.creased soybean nodulati.n. Thi. study suggests that RHB combi.ed wi.h PM or PMB provi.es a benefi.i.l source of N, P, and K, resulti.g i. i.proved soybean yi.ld and nodulati.n i. a tropi.al ferri. acri.ol.

Thi. study explored the appli.ati.n of machi.e learni.g techni.ues for flood predi.ti.n and analysi. i. southern Ni.eri.. Machi.e learni.g i. an arti.i.i.l i.telli.ence techni.ue that uses computer-based i.structi.ns to analyze and transform data i.to useful i.formati.n to enable systems to make predi.ti.ns. Tradi.i.nal methods of flood predi.ti.n and analysi. often fall short of provi.i.g accurate and ti.ely i.formati.n for effecti.e di.aster management. More so, numeri.al forecasti.g of flood di.asters i. the 19th century i. not very accurate due to i.s i.abi.i.y to si.pli.y complex atmospheri. dynami.s i.to si.ple equati.ns. Here, we used Machi.e learni.g (ML) techni.ues i.cludi.g Random Forest (RF), Logi.ti. Regressi.n (LR), Naïve Bayes (NB), Support Vector Machi.e (SVM), and Neural Networks (NN) to model the complex physi.al processes that cause floods. The dataset contai.s 59 cases wi.h the goal feature “Event-Type”, i.cludi.g 39 cases of floods and 20 cases of flood/rai.storms. Based on compari.on of assessment metri.s from models created usi.g hi.tori.al records, the result shows that NB performed better than all other techni.ues, followed by RF. The developed model can be used to predi.t the frequency of flood i.ci.ents. The majori.y of flood scenari.s demonstrate that the event poses a si.ni.i.ant ri.k to people’s li.es. Therefore, each of the emergency response elements requi.es adequate knowledge of the flood i.ci.ences, conti.uous early warni.g servi.e and accurate predi.ti.n model. Thi. study can expand knowledge and research on flood predi.ti.e modeli.g i. vulnerable areas to i.form effecti.e and sustai.able conti.gency planni.g, poli.y, and management acti.ns on flood di.aster i.ci.ents, especi.lly i. other technologi.ally underdeveloped setti.gs.

However, the adsorpti.n of the surfactants to reservoi. rock surfaces leads to the loss and reducti.n i. concentrati.n of the surfactants, whi.h i. turn reduces the overall effi.i.ncy of the oi. recovery process, wi.h attendant fi.anci.l losses. i. thi. work, the adsorpti.n of Qui.laja Saponari. (QS), a novel, natural, non-i.ni. surfactant, on crushed sandstone reservoi. rock i. i.vesti.ated. X-ray di.fracti.n (XRD) study of clean sandstone parti.les has been undertaken to determi.e the mai. components present i. the sand parti.les. The conducti.i.y method was used to measure CMC and the surfactant concentrati.ns i. aqueous soluti.ns. Batch adsorpti.n experi.ents were used to determi.e the amount of QS adsorbed on rock surface. Equi.i.ri.m condi.i.ns were reached after almost 5 days. From the results of the study, the Langmui. i.otherm model i. more sui.ed for predi.ti.g the adsorpti.n behavi.ur of QS on sandstone. The ki.eti. adsorpti.n of QS obeys the pseudo-second order model. Thi. study i. parti.ularly relevant i. surfactant selecti.n for chemi.al EOR processes.

The i.abi.i.y to achi.ve concepti.n and subsequently have a chi.d among couples may lead to psychologi.al compli.ati.ns. Objecti.e: Thi. study assessed the psychologi.al i.pact of i.ferti.i.y and i.s management on women recei.i.g treatment at a Ferti.i.y Speci.li.t Hospi.al i. a poor resource setti.g. Methods: Thi. was a cross-secti.nal study of 376 consenti.g marri.d women who sought and recei.ed ferti.i.y treatment at a pri.ate Ferti.i.y Speci.li.t Hospi.al, located at Abakali.i. South-East Ni.eri. between May 1, 2017 and May 31, 2022. Soci.-demographi. and Structured questi.nnai.es were admi.i.tered to the parti.i.ants over a 5-year peri.d. Results: The mean age of the women at presentati.n was 36.9 (±7.2 SD) years. The majori.y of the women (227) presented wi.h secondary i.ferti.i.y accounti.g for 60.4%, whi.e the rest had pri.ary i.ferti.i.y (χ2 = 16.18, P = 0.001). Male-only factor i.ferti.i.y accounted for 22.9% of all the i.ferti.i.y cases, female-only factor 21.3% whi.e both (male and female factors co-exi.ti.g) accounted for 52.7% of all the cases. The commonest cause of i.ferti.i.y i. the study was poor sperm parameters (176) accounti.g for 46.8% of cases, tubal factor 19.1% and anovulatory factor 22.3% (χ2 = 214.21, P = 0.001). Three hundred and thi.ty four (88.8%) felt depressed, 266 (70.7%) felt gui.ty about the past, and 222 (59.0%) had sui.i.al tendenci.s because of the i.ferti.i.y ordeal. Two hundred and fi.ty one (66.8%) felt i.feri.r, 237 (63.0%) cri.d often and 174 (46.3%) were soci.lly wi.hdrawn. However, 10.4% of the women felt sati.fi.d and well. These negati.e psychologi.al feeli.gs were stati.ti.ally si.ni.i.ant. Concerni.g the effect on marri.ge, the closeness of couples was reduced si.ni.i.antly i. 57.7%, 62.8% had reduced coi.al i.ti.acy, 79.3% had frequent quarrel
