Gene cloning in repeat-rich polyploid genomes remains ***,we describe a strategy for overcoming major bottlenecks in cloning of the powdery mildew resistance gene(R-gene)Pm69 derived from tetraploid wild emmer wheat.A conventional positional cloning approach was not effective owing to suppressed *** sorting was compromised by insufficient purity.A Pm69 physical map,constructed by assembling Oxford Nanopore Technology(ONT)long-read genome sequences,revealed a rapidly evolving nucleotide-binding leucine-rich repeat(NLR)R-gene cluster with structural variations.A single candidate NLR was identified by anchoring RNA sequencing reads from susceptible mutants to ONT contigs and was validated by virus-induced gene ***69 is likely a newly evolved NLR and was discovered in only one location across the wild emmer wheat distribution range in ***69 was successfully introgressed into cultivated wheat,and a diagnostic molecular marker was used to accelerate its deployment and pyramiding with other R-genes.

目的探讨数字化胸部体层摄影术（dTS）检测偶发肺部实性肺结节及其特征。材料与方法本前瞻性研究,随机选择1 111例行胸部CT检查的病人（年龄50～64岁）。其中125例受试者偶发肺部实性结节（≥5 mm）。在2012年3月—2014年10月期间对该125名受试者进行dTS和CT随访。按照结节大小,将肺部结节分为5 mm组（125名受试者）、随访后结节增大至6 mm组（55名受试者）,由4名胸部影像诊断医生在不知道结节数量的情况下评估dTS影像所显示的肺结节。医生阅片时在影像上直接标记肺结节,并对结节存在的可信度和随访建议进行分级。采用Jackknife受试者操作特征（ROC）检验法、ROC曲线以及Cohenκ系数进行统计学分析。结果 CT所证实的≥5 mm和≥6 mm的肺结节dTS整体检出率分别为49%58%和48%62%。Jackknife ROC特征值分别为0.47和0.51,曲线下面积分别为0.62和0.65。结论常规使用dTS会降低肺结节检出率,导致肺小结节随访中漏诊。

deactivation of Streptococcus mutans Biofilms on a Tooth Surface Using He dielectric Barrier discharge at Atmospheric Pressure

This paper presents a study of the effect of the low temperature atmospheric helium dielectric barrier discharge（dBd） on the Streptococcus mutans biofilms formed on tooth surface. Pig jaws were also treated by plasma to detect if there is any harmful effect on the *** plasma was characterized by using optical emission *** data indicated that the discharge is very effective in deactivating Streptococcus mutans *** can destroy them with an average decimal reduction time（d-time） of 19 s and about 98%of thein were killed after a treatment time of 30 *** to the survival curve kinetic an overall 32 s treatment time would be necessary to perform a complete *** experimental results presented in this study indicated that the helium dielectric barrier discharge,in plan-parallel electrode configuration,could be a very effective tool for deactivation of oral bacteria and might be a promising technique in various dental clinical applications.

A case of interstitial pneumonitis in a patient with ulcerative colitis treated with azathioprine

The early hypersensitivity reaction and late bone marrow depression are well-known side-effects of azathioprine, whereas interstitial pneumonia is a rare complication. A 40-year old male patient had been treated with azathioprine in consequence of extensive ulcerative colitis for 10 years. He then complained of 7 d of fever, cough and catarrhal signs, without symptoms of active colitis. Opportunistic infections were ruled out. The chest X-ray, CT and lung biopsy demonstrated the presence of interstitial inflammation. Azathioprine therapy was discontinued as a potential source of the pulmonary infiltrate. In response to steroid therapy, and intensive care, the pulmonary infiltrate gradually decreased within 4 wk. Three months later, his ulcerative colitis relapsed, and ileo-anal pouch surgery was performed. In cases of atypical pneumonia, without a proven infection, azathioprine-associated interstitial pneumonitis may be present, which heals after withdrawal of the drug.

Bioremediation perspective of historically contaminated sediment with polycyclic aromatic hydrocarbons

The purpose of the current study is to compare the influence of different aerobic conditions(biostimulation(BS), bioaugmentation(BA), and a combination of biostimulation and bioaugmentation(BB))on polycyclic aromatic hydrocarbons(PAH) degradation and compare the degraded amount with single step XAd-4 extraction as a new tool for bioavailability assessment for chronically contaminated sediment samples obtained from territory of Autonomous Province Vojvodina of Serbia(S1, S2, and S3). A great number of papers dealing with biodegradation of PAHs in spiked sediment or soil have been published,but to the authors' knowledge, a limited number of papers studied aged, historically polluted sediment and a sum of chosen U.S. Environmental Protection Agency(USEPA) PAHs. A significant reduction(up to67%) in PAH concentration was observed, while the percentage of reduction varied depending on the sediment sample and treatment used. BS treatment successfully stimulated growth of indigenous bacteria. Further, PAH-degrading strain Sphingomonas paucimobilis F8 inoculated in BA and BB treatment survived for up to 7 weeks after it was suppressed by unfavorable conditions or native *** amounts generally showed good correlation with results obtained from XAd-4 *** obtained in the current study represent a good start for standardizing a XAd-4 extraction technique as a simplified, easier, and lower cost method for bioavailability assessment.

AIM: To determine common NOd2/CARd15 mutations and TLR4 d299G polymorphism in Hungarian patients with Cd. METHOdS: A total of 527 unrelated patients with Cd (male/female: 265/262, age: 37.1 (Sd 7.6) years) and 200 healthy subjects were included. dNA was screened for possible NOd2/CARd15 mutations by denaturing high-performance liquid chromatography (confirmed by direct sequencing). TLR4 d299G was tested by PCR-RFLP. RESULTS: NOd2/CARd15 mutations were found in 185 patients (35.1%) and in 33 controls (16.5%,Pd exon4 R703C (2.1% vs 0%, P= 0.02) mutations were more frequent in Cd, while the frequency of SNP12/G908R was not increased. The frequency of TLR4 d299G was not different (Cd: 9.9% vs controls: 12.0%). Variant NOd2/CARd15 allele was associated with an increased risk for Cd (ORhet=1.71, 95%CI=1.12-2.6, P= 0.0001, ORtwo-risk alleles = 25.2, 95%CI =4.37- ,Pdisease onset (carrier: 26.4 years vs non-carrier: 29.8 years, P=0.0006), ileal disease (81.9% (?) 69.5%, OR = 1.99, 95%CI = 1.29-3.08, P= 0.02, presence of NOd2/CARd15 and TLR4: 86.7% vs 64.8%), stricturing behavior (OR = 1.69,95%CI = 1.13-2.55, P= 0.026) and increased need for resection (OR=1.71, 95%CI: 1.13-2.62, P= 0.01), but not with duration, extra-intestinal manifestations, familial disease or smoking. TLR4 exhibited a modifier effect: age of onset in wt/TLR4 d299G carriers: 27.4 years vs NOd2mut/TLR d299G: 23 years (P = 0.06), in NOd2mut/wt: 26.7 years. CONCLUSION: These results confirm that variant NOd2/ CARd15 (R702W, R703C and 3020insC) alleles are associated with earlier disease onset, ileal disease, stricturing disease behavior in Hungarian Cd patients. In contrast, although the frequency of TLR4 d299G polymorphism was not different from controls, NOd2/TLR4 mutation carriers tended to present at earlier age.

利用普通小麦(TriticumaestivumL.)“小偃6号”与黑麦(SecalecerealeL．)品种“德国白粒”杂交，选育出“小偃6号”类型带有黑麦性状的种质材料。应用总基因组原位杂交(GISH)进行检测，在8份材料中探测到黑麦染色质的存在，其中附加系3个，代换系1个，易位系4个；进一步用荧光绿标记探针pSc119．2及荧光红标记探针pAs1的双色荧光原位杂交(FISH)技术，对其中部分品系的染色体组成进行分析鉴定，结果表明：易位系BC116-1是1RS／1BL小麦／黑麦易位系，BC152-1是涉及一条1B染色体的1Rs／1BL易位系，代换系BC97-2是2R(2d)二体代换系；附加系BC122-3附加了一条6R黑麦染色体，一条6B染色体的长臂缺失。同时，对连续的总基因组原位杂交和双色荧光原位杂交技术在小麦育种中的应用进行了讨论。

Tuberculosis terminal ileitis:A forgotten entity mimicking Crohn's disease

Intestinal tuberculosis(TB) is an uncommon lesion for which differential diagnosis can be difficult. We present a case of a 53-year-old male and a systematic review of the literature, from clinical symptoms to differential diagnosis, unusual complications and therapy. The patient was admitted to the hospital with signs of acute abdomen as a result of a perforated terminal ileitis. Based on the skip lesions of the terminal ileum and cecum, Crohn's disease(Cd) was clinically suspected. An emergency laparotomy and right colectomy with terminal ileum resection was performed and systematic antibiotherapy was prescribed. The patient's status deteriorated and he died 4 d after the surgical intervention. At the autopsy, TB ileotyphlitis was discovered. The clinical criteria of the differential diagnosis between intestinal TB and Cd are not very well established. despite the large amount of published articles on this subject, only 50 papers present new data regarding intestinal TB. Based on these studies and our experience, we present an update focused on the differential diagnosis and therapy of intestinal TB. We highlight the importance of considering intestinal TB as a differential diagnosis for inflammatory bowel disease. despite the modern techniques of diagnosis and therapy, the fulminant evolution of TB can still lead to a patient's death.
