Renal function assessment in atrial fibrillation:Usefulness of chronic kidney disease epidemiology collaboration vs reexpressed 4 variable modification of diet in renal disease

AIM: To compare the performance of the re-expressed Modification of Diet in Renal Disease equation vs the new Chronic Kidney Disease Epidemiology Collaboration equation in patients with non-valvular atrial ***: We studied 911 consecutive patients with non-valvular atrial fibrillation on vitamin-K antagonist. The performance of the re-expressed Modification of Diet in Renal Disease equation vs the new Chronic Kidney Disease Epidemiology Collaboration equation in patients with non-valvular atrial fibrillation with respect to either a composite endpoint of major bleeding, thromboembolic events and all-cause mortality or each individual component of the composite endpoint was assessed using continuous and categorical ≥ 60, 59-30, and stimated glomerular filtration ***: During 10 ± 3 mo, the composite endpoint occurred in 98(10.8%) patients: 30 patients developed major bleeding, 18 had thromboembolic events, and 60 died. The new equation provided lower prevalence of renal dysfunction ssed equation(34.1%). Estimated glomerular filtration rate from both equations was independent predictor of composite endpoint(HR = 0.98 and 0.97 for the re-expressed and the new equation, respectively; P se mortality(HR = 0.98 for both equations, P strong association with thromboembolic events was observed only when estimated glomerular filtration rate was s 5.1 for the re-expressed equation, and HR = 5.0 for the new equation. No significant association with major bleeding was observed for both ***: The new equation reduced the prevalence of renal dysfunction. Both equations performed similarly in predicting major adverse outcomes.

In this study,a hydrophobic synthetic zeolite,namely ZsM-5 is chosen as an adsorbent/catalyst for toluene *** results showed that toluene adsorption onto ZsM-5 was favourable,following a Langmuir adsorption isotherm ***-5 zeolite was regenerated using gaseous ozone at low *** toluene was oxidised,releasing mainly CO_2 and H_*** of oxidation by-products such as acetic acid and acetaldehyde were formed and remained adsorbed after the oxidativate regeneration with *** four successive cycles of adsorption/ozonation,the adsorption efficiency was not affected（92%-99%）.These results showed that volatile organic compound（VOC）removal by adsorption onto ZsM-5 zeolite followed by ozone regeneration could be used as a promising hybrid process for the control of VOC emissions in terms of efficiency.

One of the biggest challenges in the biocompatibility of implantable metals is the prevention of the stress shielding effect,which is related to the coupling of the bone-metal mechanical *** stress shielding phenomenon provokes bone resorption and the consequent adverse effects on prosthesis ***,it can be inhibited by adapting the stiffness of the implant *** the use of titanium(Ti)porous structures is a great alternative not only to inhibit this effect but also to improve the osteointegration of orthopedic and dental implants,a brief description of the techniques used for their manufacturing and a review of the current commercialized implants produced from porous Ti assemblies are compiled in this *** powder metallurgy(PM)with space holder(sH)is a powerful technology used to produce porous Ti structures,it is here discussed its potential for the fabrication of medical devices from the perspectives of both design and *** most important parameters of the technique such as the size and shape of the initial metallic particles,the sH and binder type of materials,the compaction pressure of the green form,and in the sintering stage,the temperature,atmosphere,and time are reviewed according to the bibliography ***,the importance of the porosity and its types together with the influence of the mentioned parameters in the final porosity and,consequently,in the ultimate mechanical properties of the structure are ***,a few examples of the PM-sH application for the manufacturing of orthopedic implants are presented.

Background:Internal tandem duplications(ITD)within the juxtamembrane domain of FMs-like tyrosine kinase 3(FLT3)represent a poor prognostic indicator in acute myeloid leukemia(AML).Therapeutic benefits of tyrosine kinase inhibitors,such as sorafenib,are limited due to the emergence of drug *** investigations have been con-ducted to improve the understanding of the molecular mechanisms underlying the resistance to this FLT3 inhibitor,a profile of cell functioning at the metabolite level and crosstalk between metabolic pathways has yet to be *** study aimed to elucidate the alteration of metabolomic profile of leukemia cells resistant to the FLT3 ***:We established two sorafenib-resistant cell lines carrying FLT3/ITD mutations,namely the murine BaF3/ITD-R and the human MV4-11-R cell *** performed a global untargeted metabolomics and stable isotope-labe-ling mass spectrometry analysis to identify the metabolic alterations relevant to the therapeutic ***:The resistant cells displayed fundamentally rewired metabolic profiles,characterized by a higher demand for glucose,accompanied by a reduction in glucose flux into the pentose phosphate pathway(PPP);and by an increase in oxidative stress,accompanied by an enhanced glutathione *** demonstrated that the highest scoring network of altered metabolites in resistant cells was related to nucleotide degradation.A stable isotope tracing experiment was performed and the results indicated a decrease in the quantity of glucose entering the PPP in resistant *** experiment suggested that the inhibition of major enzymes in the PPP consist of glucose-6-phosphate dehydrogenase deficiency(G6PD)in the oxidative arm and transketolase(TKT)in the non-oxidative *** addition,we observed that chronic treatment with sorafenib resulted in an increased oxidative stress in FLT3/ITD-positive leu-kemia cells,which was accompanied by decreased cell proliferation and an enhanc

背景与目的FMs样酪氨酸激酶3(FMs-like tyrosine kinase 3,FLT3)近膜结构域内的内部串联重复(internal tandem duplications,ITD)是急性髓系白血病(acute myeloid leukemia,AML)预后不良的指标。酪氨酸激酶抑制剂(如索拉非尼)是急性髓系白血病主要的靶向药物,但会产生耐药性,使其疗效受限。虽然目前已对FLT3抑制剂耐药的分子机制有所了解,但代谢物水平的细胞功能及与代谢通路的相互作用尚不清楚。本研究旨在阐明白血病细胞对FLT3抑制剂耐药后代谢组学的改变情况。方法我们建立了2株携带FLT3/ITD突变的索拉非尼耐药的细胞系,命名为鼠源BaF3/ITD-R和人源MV4-11-R细胞系。我们进行了非靶向代谢组学和稳定同位素标记代谢流分析,以确定与耐药相关的代谢改变。结果耐药细胞表现出完全改变的代谢图谱,其特征表现为葡萄糖需求增加,伴随进入磷酸戊糖途径(pentose phosphate pathway,PPP)的葡萄糖通量减少;氧化应激增加,同时谷胱甘肽合成增加。我们证明了耐药细胞中代谢改变得分最高的网络与核苷酸降解相关。我们还进行了稳定同位素示踪实验,结果提示在耐药细胞中进入PPP的葡萄糖通量减少。进一步研究表明,PPP中主要酶的抑制包括氧化途径的葡萄糖-6-磷酸脱氢酶(glucose-6-phosphate dehydrogenase,G6PD)缺陷和非氧化途径的转酮醇酶(transketolase,TKT)。此外,我们发现长期使用索拉非尼可导致FLT3/ITD阳性的白血病细胞氧化应激增加,同时伴有细胞增殖能力降低及抗氧化应答增强。结论我们通过比较代谢组学的相关数据鉴定出了独特的代谢和氧化还原适应特征,这可能促进了FLT3/ITD突变的白血病细胞发生索拉非尼耐药。
