Production of Natural Coagulant from.m.ringa Oleifera Seed for Application in Treatm.nt of Low Turbidity Water

This study focused on developing an efficient and cost effective processing technique for m.ringa oleifera seeds to produce natural coagulant for use in drinking water treatm.nt. The produced natural coagulant can be used as an alternative to alum.num.sulphate and other coagulants and used worldwide for water treatm.nt. This study investigates processing m.ringa oleifera seeds to concentrate the bio-active constituents which have coagulation activity. m.ringa oleifera seeds were processed for oil extraction using electro therm.l soxhlet. Isolation and purification of bio-active constituents using chrom.tography technique were used to determ.ne the m.lecular weight of the bio-active constituents. The m.lecular weight of bio-active constitu-ents found to be in a low m.lecular weight range of between 1000 – 6500 Dalton. The proposed m.thod to isolate and purify the bio-active constituents was the cross flow filtration m.thod, which produced the natu-ral coagulant with very sim.le technique (oil extraction;salt extraction;and m.crofiltration through 0.45 μm.. The turbidity rem.val was up to 96.23 % using 0.4 m./L of processed m.ringa oleifera seeds to treat low initial turbidity river water between 34-36 Nephelom.tric Turbidity Units (NTU) without any additives. The m.crofiltration m.thod is considered to be a practical m.thod which needs no chem.cals to be added com.pared to other researchers proposed m.thods. The natural coagulant produced was used with low dosages to get high turbidity rem.val which considered to be a breakthrough in this study and recom.ended to be scaled up for industry level. The product is com.ercially valuable at the sam. tim. it is m.nim.zing the cost of water treatm.nt.

Effect of reduction roasting by using bio-char derived from.em.ty fruit bunch on the m.gnetic properties of m.laysian iron ore

Beneficiation of m.laysian iron ore is becom.ng necessary as iron resources are depleting. However, the upgrading process is challenging because of the weak m.gnetic properties of m.laysian iron ore. In this study, bio-char derived from.oil palm.em.ty fruit bunch （EFB） was utilized as an energy source for reduction roasting. m.xtures of m.laysian iron ore and the bio-char were pressed into briquettes and subjected to reduction roasting processes at 873-1173 K. The extent of reduction was estim.ted on the basis of m.ss loss, and the m.g-netization of sam.les was m.asured using a vibrating sam.le m.gnetom.ter （VSm.. When reduced at 873 K, the original goethite-rich ore was converted into hem.tite. An increase in tem.erature to 1073 K caused a significant conversion of hem.tite into m.gnetite and enhanced the m.gnetic susceptibility and saturation m.gnetization of sam.les. The m.gnetic properties dim.nished at 1173 K as the iron ore was par-tially reduced to wustite. This reduction roasting by using the bio-char can assist in upgrading the iron ore by im.roving its m.gnetic proper-ties

Kinetic studies on recom.inant stem.brom.lain

Stem.brom.lain is a plant thiol protease with several industrial and therapeutic applications. This current work presents kinetic studies of recom.inant brom.lain (recBm. expressed in Escherichia coli BL21-AI on foursynthetic substrates, N-α-carbobenzoxy-L-alanyl-p-nitrophenylester (ZANPE), N-α-carbobenzoxy-L-arginyl-L-ar-ginine-p-nitroanilide (ZAANA), N-α-carbobenzo-xy-L-phenylalanyl-L-valyl-L-arginine-p-nitroanili-de (ZPVANA) and L-pyroglutam.l-L-phenylalanyl-L-leucine-p-nitroanilide (PFLNA). Hydrolytic activities of recBm.at various pH and tem.erature conditions were com.ared to that of com.ercial brom.lain (cBm.. Both enzym.s dem.nstrated high activities at 45o C and pH 5 - 8 for recBm.and pH 6 - 8 for cBm. recBm.showed m.rginally lower Km.nd slightly higher kcat/Km.or ZAANA, ZANPE and ZPVANA in com.arison to *** Epoxysuccinyl-L-leucylam.do {4- guanidino}butane (E-64) severely affected recBm.and cBm.hydrolysis of the synthetic substrates by com.etitive inhibition with Kivalues of 3.6 - 5.1 μm.and 5.5 - 6.9 μm.for recBm.and cBm. respectively. The evaluated properties of recBm.including tem.erature and pH optim., substrate specificity and sensitivity to inhibitors or activators, satisfy the requisites required for food industries.

Phytase, also known as phytate-degrading enzym., catalyzes the hydrolysis of phytate (inositol hexakisphosphate) with sequential release of phosphate and lower inositol phosphate. We report here a new plasm.d construct designated as pm.uia from.pBAD-TOPO that harbors a 1.1 kb phytase gene (phym.) from.m.cobacterium.sm.gm.tis, and expression as well as characterization of the purified recom.inant m. sm.gm.tis phytase. DNA sequencing analysis and m.ltiple alignm.nt exercise indicated that the m. sm.gm.tis phytase is different from.both known acid and alkaline phytases. The active ~45 kDa recom.inant enzym. was expressed and confirm.d by enzym. assay and Western blot analyses. Ni-NTA affinity purified recom.inant m. sm.gm.tis phytase exhibited specific activity of 233.51 U/m., optim.l pH of 3 and 7 and optim.l tem.erature of 60°C. The purified enzym. retains alm.st 30% of the initial activity after incubation at 90°C for 60 m.n. The enzym. showed broad substrate specificity with Km.and Vm.x of the recom.inant enzym. for sodium.phytate substrate of 0.105 ± 0.016 m. and 26.93 ± 1.21 m. m.n-1, respectively.
