Phytase, also known as phytate-degrading enzyme, catalyzes the hydrolysis of phytate (inositol hexakisphosphate) with sequential release of phosphate and lower inositol phosphate. We report here a new plasmid construct designated as pMSuia from pBAD-TOPO that harbors a 1.1 kb phytase gene (phyMS) from Mycobacterium smegmatis, and expression as well as characterization of the purified recombinant M. smegmatis phytase. DNA sequencing analysis and multiple alignment exercise indicated that the M. smegmatis phytase is different from both known acid and alkaline phytases. The active ~45 kDa recombinant enzyme was expressed and confirmed by enzyme assay and Western blot analyses. Ni-NTA affinity purified recombinant M. smegmatis phytase exhibited specific activity of 233.51 U/mg, optimal pH of 3 and 7 and optimal temperature of 60°C. The purified enzyme retains almost 30% of the initial activity after incubation at 90°C for 60 min. The enzyme showed broad substrate specificity with Km and Vmax of the recombinant enzyme for sodium phytate substrate of 0.105 ± 0.016 mM and 26.93 ± 1.21 mM min-1, respectively.

Structural and Optical Properties of Zinc Borotellurite Glass Co-Doped with Lanthanum and Silver Oxide

A series of zinc borotellurite glass co-doped with lanthanum and silver oxide with the chemical formula of [{[(TeO2)0.7(B2O3)0.3]0.7(ZnO)0.3}0.96(La2O3)0.04]1-x(Ag2O)x where the molar frac-tion of silver oxide, x = 0.02, 0.04, 0.06, 0.08 and 0.10 had been successfully prepared via the conventional melt-quenching technique. The structural properties of the glasses were unveiled through X-ray Diffraction (XRD) and Fourier Transform Infra-Red (FTIR) spectroscopy while optical properties of the glasses were investigated with Ultra Violet Visible (UV-Vis) spectropho-toscopy. The short range periodic atomic arrangement in the glass matrix that implies the amorphous nature of the glass was confirmed with the presence of a broad hump in the XRD pattern. On the other hand, the three absorption bands observable in the FTIR spectra had proven the existence of BO4, BO3 as well as TeO4 units in the glass network. The absorbance values retrieved from UV-Vis spectroscopy were utilized to calculate the indirect energy band gap and Urbach energy values of the fabricated glass. By employing the equations proposed by Mott and Davis, the obtained indirect energy band gap have val-ues ranging from 2.16 to 4.16 *** decreasing trend in indirect energy band gap and increasing Urbach energy values were related to the increasing num-ber of nonbridging oxygen (NBO) in the glass that is created from the breaking of Te-O-Te or B-O-B bonds after lanthanum as well as silver oxide are incor-porated into the zinc borotellurite glass network.

The objective of this study is to verify potential of various types of microorganisms during spray drying and non-refrigerated storage that can be enhanced substantially by selecting suitable protective colloids. Four selected probiotics tested are Lactbacillus plantarum B13 and B18, which are the bacteria probiotics and Kluyveromyces lactis and Saccharomyces blouradii, non-bacteria probiotics. Two levels of experiment occur starting with formulation study of encapsulation agent followed by the viability study of different probiotics after spray dry and two weeks nonrefrigerated storage. The formulation of 30% of gum Arabic, 15% of gelatin and 45% of coconut oil can homogenize well at least for two hours and can produce acceptable dried product (below 4% of moisture content) at low outlet temperature (70℃ -75℃). K. lactis, S. blouradii gives 2.57% and 2.4% of viability percentage after spray drying process and 25.84% and 2.04% after two weeks nonrefrigerated storage respectively. The colonies of non-probiotics bacteria after both conditions are between 1010 and 106 cfu/mL which is among the accepted level for industrial application. However, the survival of probiotics in a spray-dried form during non-refrigerated storage is higher at low of moisture content compared to others.

Adult mosquito sampling techniques are essential for monitoring transmission of malaria and other mosquito borne infections. Preference for any sampling technique depends on both its field efficiency and the characteristics of local vector populations. Surveys on adult mosquitoes using Human Landing Catch (HLC) and CO2-baited CDC light trap (CDC-LT) techniques were conducted in several knowlesi malaria endemic areas between the months of March to December 2012 in several states of Peninsula Malaysia. These two techniques were relatively compared to determine the preferences of anopheline mosquitoes towards CO2-baited CDC-LT technique using HLC technique as the reference method. Cx. gelidus, An. maculates and An. introlatus were the main three species collected by HLC technique, whereas the species collected by CO2-baited CDC-LT technique were mostly An. cracens, Ar. durhami and Coquillettidia species. Most of the Anopheles species were collected almost exclusively by the human collectors except for An. cracens and An. introlatus which were collected using both techniques. Anopheles cracens was the most dominant species captured using CO2-baited CDC-LT technique. This is the first report showing An. cracens was caught using CO2-baited CDC-LT technique in Malaysia.

The diagnosis of breast cancer can cause a significant anxiety in someone’s life. The care of the breast cancer patient requires in many centres. The care of breast cancer patient is undertaken as a joint venture between the surgeon, medical oncologist, radiotherapist, pathologist and allied health professionals such as the clinical nurse specialist as well as counsellors. This is a case of a 39-years-old Malay woman presented to HUSM with a history of ulcerating breast lump with bloody pus discharge on the right breast. An initial mammogram study showed lesion suspicious of malignancy, and histopathological examination was consistent with invasive ductal carcinoma. She received neoadjuvant chemotherapy before proceeding to right modified radical mastectomy with axillary clearance and immediate chest wall reconstruction. Detailed discussion on the diagnosis, treatment, surgical intervention and progress of the patient with reference to available literature reviews are presented.

The formulation of omega-3 emulsion using rubber (Hevea Brasiliensis) seed oil based on the best performance of the emulsion in terms of higher viscosity, smaller droplet size, lower moisture content and slightly acidic pH value supported by degree of creaming in varying the type and composition of emulsifier used. Rubber seed oil contains significant value of alpha-linolenic acid which plays an important role in maintaining human health. Therefore, formulation of rubber seed oil emulsion is important to become a new source of omega-3 emulsion instead of fish oil. Rubber seed oil was mixed with distilled water and nonionic emulsifier which were lecithin and span 80 by homogenizer. From the analysis conducted, the best formulation was the emulsion with 50% of distilled water, 6% of lecithin and 47% of rubber seed oil.

A sensitive tapered optical fiber sensor incorporating graphene oxide(GO)and polyvinyl alcohol(PVA)composite film for the rapid measurement of changes in relative humidity was proposed and experimentally *** sensing principle was based on the intensity modulation of the transmitted light induced by the refractive index changes of the sensitive *** sensing region was obtained by tapering a section of single-mode optical fiber(SMF)from its original 125µm diameter down to 9.03µ*** tapered structure was then modified through deposition of GO/PVA nanocomposites by using the dip-coating *** field emission scanning electron microscope(FESEM)and Raman spectroscopy were used to characterize the structure of the composite *** evidenced by a Fourier transform infrared spectroscopy(FTIR)analysis,the presence of oxygen functional groups(such as−OH and COOH)on the GO structure enabled the attachment of PVA molecules through hydrogen bonding and strong adhesion between GO/PVA *** performance of the sensor was tested over a wide range(20%RH to 99.9%RH)of relative *** sensor showed a good response with its signal increasing linearly with the surrounding *** tapered optical fiber sensor with the coating of GO/0.3g PVA achieved the highest sensitivity[0.5290RH(%)].The stability,repeatability,reversibility,as well as response time of the designated sensor were also measured and analyzed.

Consumers’ emotion has become imperative in product design. In affective design field, Kansei Engineering (KE) has been recognized as a technology that enables discovery of consumer’s emotion and formulation of guide to design products that win consumers in the competitive market. Albeit powerful technology, there is no rule of thumb in its analysis and interpretation process. KE expertise is required to determine sets of related Kansei and the significant concept of emotion. Many research endeavours become handicapped with the limited number of available and accessible KE experts. This work is performed to simulate the role of experts with the use of Natphoric algorithm and thus provides solution to the complexity and flexibility in KE. The algorithm is designed to learn the process by implementing training datasets taken from previous KE research works. A framework for automated KE is then designed to realize the development of automated KE system.
