Hypoglycemic effect of Octomeles sumatrana aqueous extract in streptozotocin—induced diabetic rats and its molecular mechanisms

Objective:To investigate the hypoglycemic effect of the aqueous extract of Octomeles sumatrana (***)(OS) in streptozotocin-induced diabetic rats(STZ) and its molecular mechanisms. Methods:Diabetes was induced by intraperitoneal(i.p.) injection of streptozotocin(55 mg/kg) in to male Sprague-Dawley *** were divided into six different groups;normal control rats were not induced with STZ and served as reference,STZ diabetic control rats were given normal *** groups were treated with OS aqueous extract at 0.2,0.3 and 0.5 g/kg,orally twice daily continuously for 21 *** fifth group was treated with glibenclamide(6 mg/kg) in aqueous solution orally continuously for 21 *** completion of the treatment period,biochemical parameters and expression levels of glucose transporter 2(Slc2a2),glucose-6-phosphatase (GoPase) and phosphoenolpyruvate carboxykinase(PCK1) were determined in liver by quantitative real time ***:Administration of OS at different doses to STZ induced diabetic rats, resulted in significant decrease(P<0.05) in blood glucose level in a dose dependent manner by 36%,48%,and 64%at doses of 0.2,0.3 and 0.5 g/kg,respectively,in comparison to the STZ control *** with OS elicited an increase in the expression level of Slc2a2 gene but reduced the expression of G6Pase and PCK1 ***,OS treated rats,showed significantly lower levels of serum alanine transaminase(ALT),aspartate aminotransferase(AST) and urea levels compared to STZ untreated *** extract at different doses elicited signs of recovery in body weight gain when compared to STZ diabetic controls although food and water consumption were significantly lower in treated groups compared to STZ diabetic control ***:*** aqueous extract is beneficial for improvement of hyperglycemia by increasing gene expression of liver Slc2a2 and reducing expression of G6Pase and PCK1 genes in streptozotocin-induced diabetic rats.

Determination of Heavy Metals and Polycyclic Aromatic Hydrocarbons (PAH) Contents Using the Lichen<i>Dirinaria picta </i>in Universiti Kebangsaan Malaysia

A biomonitoring study on lichens was conducted to assess the levels of atmospheric heavy metal and polycyclic aromatic hydrocarbon (PAH) at ten sites at the Universiti Kebangsaan Malaysia Campus, Bangi, Selangor, Malaysia. Dirinaria picta, epiphytic lichen which commonly found at lowland areas was used as the bioindicator in this study where samples were collected from the bark of palm trees at sites ranging from areas with high volume of traffic to isolated areas in the campus. Eight metals (Cr, Fe, Co, Ni, Cu, Zn , Al, Pb), and ten PAH;naphthalene (PAH1), acenaphtylene (PAH2), acenaphthene (PAH3), fluorine (PAH4), anthracene (PAH5), phenanthrene (PAH6), fluoranthene (PAH7), pyrene (PAH8), benzo (b) anthracene (PAH9) and chrysene (PAH10) were analyzed in the naturally growing thallus of Dirinaria picta. The average concentration of the metals was observed to be in the range of 230.6 ppm (Fe) to 0.3 ppm (Co) and for PAH, it was observed in the range of 612 ppm (PAH1) to 79.5 ppm (PAH10). The One-way ANOVA analysis for both the heavy metals and the PAHs showed Ho was rejected. There are significant differences in heavy metals and PAHs concentrations between sites. The selectivity sequence of heavy metals were Fe > Cr > Ni > Al > Zn > Pb > Cu. Meanwhile for PAH, the sequence is PAH1 > PAH3 > PAH2 > PAH6 > PAH5 > PAH7 > PAH4 > PAH8 > PAH9 > PAH10.

Adult mosquito sampling techniques are essential for monitoring transmission of malaria and other mosquito borne infections. Preference for any sampling technique depends on both its field efficiency and the characteristics of local vector populations. Surveys on adult mosquitoes using Human Landing Catch (HLC) and CO2-baited CDC light trap (CDC-LT) techniques were conducted in several knowlesi malaria endemic areas between the months of March to December 2012 in several states of Peninsula Malaysia. These two techniques were relatively compared to determine the preferences of anopheline mosquitoes towards CO2-baited CDC-LT technique using HLC technique as the reference method. Cx. gelidus, An. maculates and An. introlatus were the main three species collected by HLC technique, whereas the species collected by CO2-baited CDC-LT technique were mostly An. cracens, Ar. durhami and Coquillettidia species. Most of the Anopheles species were collected almost exclusively by the human collectors except for An. cracens and An. introlatus which were collected using both techniques. Anopheles cracens was the most dominant species captured using CO2-baited CDC-LT technique. This is the first report showing An. cracens was caught using CO2-baited CDC-LT technique in Malaysia.

Production of Endopolysaccharides from Malaysia’s Local Mushrooms in Air-Lift Bioreactor

Four local mushroom species, viz. Auricularis polytricha, Lentinus edodes, Agrocybe sp and Pleurotus flabellatus were grown under submerged culture and screened for endopolysaccharides. The fermentation was done in 250 ml working volume Erlenmeyer flask and the fermentation curves for all species were established. Pleurotus flabellatus has the highest rate of biomass production at the rate of 0.180 g/L/day, at 10 days hence chosen for further investigation. Two additional media, viz. Mushroom Complete Media (MCM) and Yeast Malt (YM) were selected to be compared with potato extract(PE) media used initially. MCM media produced the highest biomass productivity at the rate of 0.311 g/L/day. Pleurotus flabellatus biomass was extracted using modified Mizuno method and the endopolysaccharide obtained was tested for β-glucan. The yield of β-glucan was 7.70 ± 1.11 g/100g. The polysaccharides were purified using column chromatography to yield four fractions. The fourth fraction F4, gave the highest molecular weight at 3.058 × 106 Dalton (11.8%) and 1.282 × 104 Dalton (88.2%). The mushroom, P. flabbelatus was cultured using air-lift bioreactor, and the highest productivity was obtained at air-flowrate 2 L/min, yielding 2.25 g/L/day. The yield of biomass against substrate used (glucose consumption) Yb/s was 0.78 g/g.
