Isolation of Functional RNA from Heavily Infested, Wilted and Necrotic Leaf Tissues of Tea with High Polyphenol content

A procedure is described to isolate intact RNA from tissues not previously undertaken-highly infested and wilted apical buds and leaves of tea [camellia sinensis （L.） O. Ktze.]. The protocol uses a final concentration of 450 mM β mercaptoethanol （βME） and 10% Polyvinylpyrrolidone （PVP） to circumvent problems associated with large amounts of polyphenols, polysaccharides, pigments and other secondary metabolites not easily removed by conventional procedures. Furthermore, the proposed protocol is applicable to normal tissues and other plant tissues with similar stresses, containing compounds that interfere with RNA extractions. Total RNA could be used for downstream applications such as mRNA isolation, reverse transcription, quantitative Polymerase chain Reaction （PcR）, cDNA library construction and Rapid amplification of cDNA ends （RAcE）.

A rapid, sensitive, robust, rugged and linear HPLc method is developed using QbD approach and validated as per IcH for the estimation of amlodipine impurities in tablet dosage form. Phosphate buffer with triethyl amine adjusted to pH to 2.8 is used as the mobile phase and 3μ particle size c18 column of 150 mm length and 4.6 mm internal diameter is used. Using photo diode array (PDA) detector, the compounds are monitored at 340 nm. All impurities are well separated and flow Gradient has been optimized to obtain the acceptable resolution between impurities and amlodipine. Diluent was chosen, based on the impurity peak shapes and recoveries. Test concentration and injection volume have been optimized to obtain limit of quantification (LOQ) values below the reporting threshold.

Objective:To investigate the antioxidant property of purified bioactive compound from sponge associated bacteria Bacillus ***:The potential active compound was subjected for assay of total antioxidant activity,α,α-diphenyl-α-picrylhydrazyl(DPPH)radical scavenging activity,nitric oxide radical scavenging activity,hydrogen peroxide(H_(2)O_(2))scavenging activity and total reducing ***,the 16S rRNA gene sequence was carried out to identify the sponge symbiotic ***:The results showed linear increase of total antioxidant activity,DPPH radical scavenging activity,nitric oxide radical scavenging activity,H_(2)O_(2)scavenging activity and total reducing ***_(50)value of the active compound for DPPH activity,H_(2)O_(2)scavenging activity,nitric oxide scavenging activity was recorded as 15.025μg/mL,23.73μg/mL,and 41.70μg/mL *** potential strain was identified as Bacillus species from GenBank database(GenBank Accession number JX985748).conclusions:The present study has reported the antioxidant property of purified bioactive compound from sponge associated Bacillus *** report will be helpful to pharmaceutical and antioxidant researchers for further studies.

Qualitative and Quantitative changes in Biochemical constituents in the Rice Plant Treated with Different Xenobiotics

Experiments were conducted at Entomology division of Directorate of Rice research to study the qualitative and quantitative changes in biochemical constituents in the rice plant treated with different xenobiotics, viz. insecticides like deltamethrin, imidacloprid, herbicide like 2,4-Dichlorophenoxyacetic acid, fungicide like carbendazim, green manure calotropis gigantea leaves, botanicals like Neem Seed Powder and fertilizer urea along with control plants of TN1, PTB33 and Swama varieties comprising of total 15 treatments. Application of butachlor, carbendazim, deltamethrin and excessive nitrogen fertilization resulted in increased population build up of brown plant hopper （BPH）. Significantly, lower honeydew excretion （13 mme area） in the BPH nymphs was observed on resistant PTB33 variety compared to susceptible TN1 and Swarna varieties （116 and 110 mm2 respectively）. In susceptible TN1 variety, the sucrose content was significantly higher （9.35 mg/g tissue） compared to BPH resistant PTB33 （5.32 mg/g tissue）. The BPH resistant PTB33 variety exhibited significantly lower free amino acid content （1.23 mg/g tissue） as compared to TN1 （2.51 mg/g）. The c/N ratio was comparatively high in the resistant PTB33 alone （4.32）, but was low in PTB33 applied with 2,4-D ＋ carbendazim （2.73）, 2,4-D ＋ imidacloprid ~ carbendazim （2.3）, excess urea fertilizer （2.18）. The total phenol contents measured in resistant PTB33 （7.5 mg/g dry wt）, susceptible TN1 （5.865 mg/g dry wt） and the popular variety Swarna （5.57 mg/g dry wt） were almost same and no statistical difference was observed. Further, treatment of these varieties with different xenobiotics and botanicals also did not significantly alter their total phenol content. However, the phenol content of the resistant PTB 33 alone and treated with xenobiotics was higher compared to the susceptible TN 1 and Swarna varieties alone and treated with xenobiotics. An Ordering Policy for Deteriorating Items with Time-Dependent Quadratic Demand and Salvage Value under Permissible Delay in Payment

The article deals with an economic order quantity (EOQ) inventory model for deteriorating items in which the supplier provides the purchaser a permissible delay in payment. This is so when deterioration of units in the inventory is subject to constant deterioration rate, demand rate is quadratic function of time and salvage value is associated with the deteriorated units. Shortages in the system are not allowed to occur. A mathematical formulation is developed when the supplier offers a permissible delay period to the customers under two circumstances: 1) when delay period is less than the cycle of time;and 2) when delay period is greater than the cycle of time. The method is suitable for the items like state-of-the-art aircrafts, super computers, laptops, android mobiles, seasonal items and machines and their spare parts. A solution procedure algorithm is given for finding the optimal order quantity which minimizes the total cost of an inventory system. The article includes numerical examples to support the effectiveness of the developed model. Finally, sensitivity analysis on some parameters on optimal solution is provided.

Identification and characterization of Heparan Sulphate Binding Proteins of <i>Entamoeba histolytica</i>

A large number of microbial pathogens bind to heparan sulphate on eukaryotic cell surfaces. Heparan Sulphate Binding Proteins (HSBPs) from Entamoeba histolytica culture lysates were obtained by sequential ammonium sulphate precipitation and Protein purify. SDS-PAGE and immunoblotting experiments indicated the presence of two major extracellular proteins in E. histolytica (51.2 kDa and 61.0 kDa). characterization of HSBPs by 2D Gel electrophoresis of 40% (NH4)2SO4 precipitated lysate of E. histolytica revealed that the isoelectric point of 51.2 kDa HSBP was at pH3.0. The protein of 61.0 kDa HSBP showed three spots in 40% (NH4)2SO4 lysate which had isoelectric point between pH 4.0 - 7.0. While in 80% (NH4)2SO4 precipitated lysate, 51.2 kDa HSBP showed only one spot which had isoelectric point at pH 3. However, 61.0 kDa HSBP revealed 2 spots which had isoelectric point between pH 4 and 5. The result showed that this parasite has proteins which interact with heparan sulphate whose molecular formula is c14H23NO21S-23. These proteins may have a role in binding of parasite to heparan sulphate on host cells. Further characterization by MALDI-TOF analysis of HSBPs from E. histolytica demonstrated HSBPs to be novel protein in this parasite which has been isolated, purified and characterized first time by our group in the present study.

Eight insecticidal crystal proteins of Bacillus thuringiensis, crylAa, crylAb, crylAc, crylB, cry2Aa, crylc, crylDa and cry 1Ea were assessed for toxicity against 1 st instar larvae of rice leaf folder, cnaphalocrocis medinalis （Guenee） at 48 HAT and 72 HAT. Bioassay results depicted crylAa was the most toxic （Lcso 2.35 ppm） followed by crylBa （Lcso 8,50 ppm） and crylAb （Lcso 8.73 ppm） at 48 HAT, whereas, at 72 HAT crylAb proved to be highly toxic （Lc50 0.50 ppm） followed by crylAa （Lcso 4.07 ppm）, crylAc （Lcso 4,84 ppm） and crylBa （Lcso 6.42 ppm）. Toxins cry2Aa, crylca, crylDa and crylEa did not resulted in any mortality at 48 HAT and 72 HAT, respectively. Baseline estimates for crylAb against 1st instar larvae of c. medinalis sampled from seven geographical locations revealed variation in Lc50＇s from 0.37 ppm to Lc50 16.25 ppm at 48 HAT and Lc50 0.50 ppm to Lc50 6.49 ppm 72 HAT, respectively with relative resistance ratios of 44-fold and 13-fold at 48 HAT and 72 HAT over the susceptible population.

Effect of Slow cooling in Reducing Pore Size in a Sintered Powder Metallurgical 6061Aluminium Alloy

The usage of powder metallurgy aluminium compacts in lieu of ferrous components in automotives helps to lower vehicle weight. The major drawback in the commercially available press sintered aluminium alloy is porosity which is mainly dependent on the powder metallurgical process parameters such as compaction pressure, sintering temperature and cooling rate after sintering. In this paper the effect of particle size and furnace controlled cooling after sintering on porosity level and micro hardness of an elemental 6061 aluminium alloy has been investigated. Aluminium particle sizes of 20 μm and 150 μm were used. The elemental 6061 aluminium alloy powders are warm compacted at 175 MPa. After sintering for about one hour at 600°c, the aluminium compacts were furnace cooled at the rate of 1°c /min to different temperatures of 500°c, 400°c, 300°c and 200?c. When the cooling temperature after sintering inside the furnace is effected at various temperatures from 600°c to 200°c, for a precipitate hardened aluminium compacts with aluminium particle size of 20 μm, the porosity level reduced by 26% and that for aluminium particle size of 150μm, the porosity level reduced by 23%. Marked improvement in micro hardness value is also observed correspondingly.

Integrative Modeling of Oxidative Stress and c1 Metabolism Reveals Upregulation of Formaldehyde and Downregulation of Glutathione

This research provides, to the authors’ knowledge, the first integrative model of oxidative stress and c1 metabolism in plants. Increased oxidative stress can cause irreversible damage to photosynthetic components and is harmful to plants. Perturbations at the genetic level may increase oxidative stress and upregulate antioxidant systems in plants. One of the key mechanisms involved in oxidative stress regulation is the ascorbate-glutathione cycle which operates in chloroplasts as well as the mitochondria and is responsible for removal of reactive oxygen species (ROS) generated during photosynthetic operations and respiration. In this research, the complexity of molecular pathway systems of oxidative stress is modeled and then integrated with a previously developed in silico model of c1 metabolism system. This molecular systems integration provides two important results: 1) demonstration of the scalability of the cytoSolve®?collaboratory™, a computational systems biology platform that allows for modular integration of molecular pathway models, by coupling the in silico model of oxidative stress with the in silico model of c1 metabolism, and 2) derivation of new insights on the effects of oxidative stress on c1 metabolism relative to formaldehyde (HcHO), a toxic molecule, and glutathione (GSH), an important indicator of oxidative homeostasis in living systems. Previous in silico modeling of c1 metabolism, without oxidative stress, observed complete removal of formaldehyde via formaldehyde detoxification pathway and no change in glutathione concentrations. The results from this research of integrative oxidative stress with c1 metabolism, however, demonstrate significant upregulation of formaldehyde concentrations, with concomitant downregulation and depletion of glutathione.
