Objective:To characterize the bacterial pathogens in patients having gram negative septicaemia. Results:The most prevalent isolates Escherichia coli（***） 58.6%,Klebsiella Spp.32.9%and Pseudomonas 8.6%,were resistant to most of the antimicrobials including cefazolin,ceftriaxone, cefuroxime,ampicillin and co-trimoxazole but sensitive to imipenem and *** and MBL production was seen 7.3%and 12.2%of *** isolates *** isoaltes were found to have blaCTX-M-15 and two of them also showed blaTEM-1 type ***, none of them showed blaSHV.Conjugation experiments using J-53 cells confirmed these resistant markers as plasmid ***:This work highlights the molecular epidemiology of escalating antimicrobial resistance and likely switch over of blaCTX-M-15 type extended spectrum beta-lactamases by blaTEM type ESBLs in ***,the antimicrobial resistance by horizontal gene transfer was predominant among Enterobacteraceae in the community setting.

Synthesis, characterization and electrochemical evaluation of anticorrosion property of a tetrapolymer for carbon steel in strong acid media

A novel tetrapolymer(tp) consisting of carboxylate, sulphonate, phosphonate and sulfur dioxide based comonomers was synthesized using Butler cyclopoymerization technique. The synthesized tetrapolymer was characterized using FTIR,1H-NMR,13CNMR and elemental analysis. The performance of the tetrapolymer as a corrosion inhibitor for St37 carbon steel in 15% HCl and 15% H2SO4 acid media was assessed using electrochemical impedance spectroscopy(EIS), linear polarization resistance(LPR), potentiodynamic polarization(PDP) and electrochemical frequency modulation(EFM) techniques. The influence of addition of a small amount of KI on the corrosion inhibition efficiency of tp was also assessed. Results obtained showed that the tetrapolymer moderately inhibited the corrosion of St37 steel in the acid media with protection efficiency of 79.5% and 61.1% at the optimum concentration of 1000 mg·L^-1 studied in HCl and H2SO4 media respectively. On addition of 5 mmol·L^-1 KI to the optimum tetrapolymer concentration, the protection efficiency was upgraded to 90.6% and 93.5% in HCl and H2SO4 environment, respectively. The enhanced performance of the polymer in the presence of KI is due to synergistic action deduced from synergism parameter(S1) which was found to be greater than *** tetrapolymer afforded the corrosion inhibition of St37 steel in the acid media by virtue of adsorption of the polymer molecules on the steel surface which was confirmed by ATR-FTIR analysis of the adsorbed film extracted from the steel surface. tp + KI formed complex with St37 steel surface in H2SO4 solution but not in HCl solution.

Summary：A multi-centred study was designed to collect dengue epidemiologic data from government and registered private hospitals/clinics and maintained archive of frozen specimens in bio-bank to be used for future dengue epidemic control program, and assess the epidemiology of dengue fever（DF） by evaluating biochemical and oxidative status of patients. ELISA Ig M antibodies test was done to confirm DF. From August 2010 to December 2011, 101 confirmed blood samples of DF patients referred to pathology lab of Jinnah Hospital Lahore were subjected to the epidemiologic assessment by evaluating the biochemical and physiological indices and alterations of circulating antioxidants. Clinical features of DF patients and effect of fever on blood components and serum proteins of liver were recorded. The hospital stay in DF, dengue hemorrhagic fever（DHF） and dengue shock syndrome（DSS） showed significant difference. Significant increases in serum alanine amino transferase（ALT）（P=0.000）, aspartate amino transferase（AST）（P=0.000）, alkaline phosphatase（ALP）（P=0.000）, malondialdehyde（MDA） along with significant decreases in total protein（tp）（P=0.000）, reduced glutathione（GSH）（P=0.000）, superoxide dismutase（SOD）, catalase（CAT）（P=0.000）, and sialic acid contents（P=0.016） were observed. A positive correlation existed between bound sialic acid levels, liver enzymes and circulating antioxidants（r=0.656, P=0.016）. In the present study, alterations of circulating antioxidants in DF suggest that DF might be a metabolic response to an acute, self-limiting tropical viral infection, and a consequence of the viral inflammatory process.

龙胆泻肝汤来源于?医方集解?,该方为泻肝胆实火,清三焦湿热之代表方剂。它由龙胆草、栀子、黄芩、柴胡、生地、木通、车前子、泽泻、当归、甘草组成。单味中药的现代研究结果表明该方诸药分别具有抗菌、消炎、解热、利胆、保肝、降压、祛血脂、利尿等作用,成方研究结果表明龙胆泻肝汤具有增加幼鼠胸腺重量,提高巨噬细胞吞噬功能,促进淋巴细胞转化等免疫增强作用。我们通过大量的文献调查,结合现代免疫学研究成果,认为“泻肝胆实火,清三焦湿热”的作用机理可能与干预免疫系统和肠道黏膜免疫系统等作用机理有关,但迄今为止,国内外未见这方面的研究报道。本实验目的是研究龙胆泻肝汤对肝胆系统功能活动及免疫系统和肠道黏膜免疫的影响,探讨其护肝、保肝、抗炎的作用机理。主要完成了龙胆泻肝汤对肝胆功能相关生理生化参数的影响、对肝损伤的保护作用和对免疫系统及肠道黏膜免疫系统的影响等三方面的研究。为了观察本方在实验条件下对肝胆功能的影响及是否有毒性副作用,我们使用生物化学方法检测反映肌肉组织、肝脏、肾脏等器官功能的天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总蛋白(tp)、白蛋白(ALB)、尿酸(UA)、肌酐(CRE)、肌酸激酶(CK)等指标。结果表明连续4周按小鼠体重0.5～2 g/Kg/日剂量古典配伍的龙胆泻肝汤,对小鼠的主要生物化学参数没有显著影响,说明古典配伍的龙胆泻肝汤在本实验条件下对小鼠肝功能、肾脏功能、肌肉组织等没有毒性副作用。为了研究龙胆泻肝汤的保肝作用,我们观察龙胆泻肝汤对四氯化碳(CCl)引起的小鼠实验性肝损伤的预防作用。结果表明龙胆泻肝汤可使实验性肝损伤小鼠血清中升高的乳酸脱氢酶(LDH)、丙氨酸氨基转移酶(ALT)值降低,且对前者有显著影响;龙胆泻肝汤显著改善或减少了CCl引起的动物肝细胞浆疏松化、肝细胞的空泡变性、脂肪变性和坏死,说明龙胆泻肝汤对小鼠实验性肝损伤有一定保护作用。其保肝作用的机理可能与消除四氯化碳进入体内生成的自由基对细胞膜脂质过氧化作用有关。为了探讨龙胆泻肝汤在临床广泛用于抗炎及治疗感染性病证的机理,我们研究了龙胆泻肝汤对小鼠脾脏T细胞亚群动态变化、免疫器官指数以及肠道黏液所含的分泌型IgA(S IgA)水平的变化等免疫系统的影响。为此,选用120只健康的ICR小鼠,随机分成5组,除空白组外所有小鼠腹腔注射0.025 mg/ mL脂多糖,0.2 mL/只;然后灌胃给与相应的药物,0.2 mL/d/只,连续用药20 d。于第15 d、20 d灌胃30 min后,从各组中随机抽出8只,雄雌各半,称量体重,断髓致死,采集样品观察。结果表明:龙胆泻肝汤按小鼠体重0.5 g/Kg/日的剂量给药,既能提高CD4 T淋巴细胞百分率,又能抑制CD8 T淋巴细胞百分率,从而使CD4/ CD8作用的比值显著上升;按小鼠体重0.5 g/Kg/日和2 g/Kg/日的剂量给药,3周后可显著提高小鼠脾指数;按体重0.5 g/Kg/日的3个剂量给药组,对小鼠肠道黏液中的分泌性SIgA均无显著作用,但随服药时间的延长有下降趋势。表明适当剂量的龙胆泻肝汤具有增强系统免疫的功能,其作用机理与促进脾脏内淋巴细胞增殖、分化有关。因龙胆泻肝汤属大寒沉降类中药,对肠道蠕动、黏液分泌有明显的抑制,因此,可能亦有轻度抑制肠道黏膜免疫的作用,但本方对黏膜淋巴细胞组织、黏膜下淋巴组织、肠固有膜淋巴细胞以及相关淋巴因子IFN-γ、IL-5、IFN-α和IL-2等的酶联免疫组织化学的实验研究,目前我们尚未完成。因此,要阐明本方对肠道黏膜免疫系统确切的影响,还有待进一步的研究探讨。

水产养殖业正处在高度集约化的迅速发展阶段,我们急需简单高效的方法来提高增养殖效率。饲料添加剂是一种重要的养殖手段,在养殖中通过饲料添加剂的使用来提高养殖效率对养殖业的发展具有重要意义。虾青素是一种酮类胡萝卜素,具有抗氧化,抗炎等优点。作为一种广泛应用的饲料添加剂,虾青素在提高水生动物的生长发育和繁殖性能方面具有巨大的潜力。本实验通过研究饲料中添加不同水平虾青素对吉富罗非鱼(GIFT,Oreochromis niloticus)生长性能、肝脏抗氧化、血液生理生化以及性腺的影响,来探究饲料中添加虾青素对吉富罗非鱼雌鱼发育和繁殖能力的影响。我们选取平均体重为(227±0.3)g的健康吉富罗非鱼120尾,随机分成5组,每组3个重复,每个重复放养24尾鱼。5组试验鱼分别饲喂添加0.00%(对照组)、0.05%、0.1%、0.15%、0.2%虾青素的实验饲料,并在循环水养殖系统中养殖52天。1.饲料中添加不同水平虾青素对雌性吉富罗非鱼生长性能,体指数和肝脏抗氧化的影响实验结果表明,饲料中添加虾青素显著影响FSW、SGR和FCR(P0.05);虾青素各添加组(0.05%、0.1%、0.15%和0.2%)VSI、HIS和GSI均高于对照组,但与对照组相比均无显著性差异(P>0.05)。饲料中添加虾青素对雌性吉富罗非鱼肝脏SOD、CAT和MDA均无显著影响(P>0.05)。其中,0.05%、0.1%和0.2%虾青素添加组MDA水平高于对照组,而0.15%虾青素添加组低于对照组;0.05%、0.15%和0.2%虾青素添加组SOD水平高于对照组,而0.1%虾青素添加组低于对照组;0.05%和0.15%虾青素添加组CAT水平高于对照组,而0.1%和0.2%虾青素添加组低于对照组。2.饲料中添加不同水平虾青素对雌性吉富罗非鱼血细胞和血液生化指标的影响实验结果表明;(1)饲料中添加虾青素对雌性吉富罗非鱼WBC有影响(P0.05)。其中,虾青素各添加组(0.05%、0.1%、0.15%和0.2%)RBC和HGB均高于对照组;0.1%和0.2%虾青素添加组HCT高于对照组,而0.05%和0.15%虾青素添加组低于对照组。(3)饲料中添加虾青素对雌性吉富罗非鱼血清ALT和AST无显著影响(P>0.05)。其中,0.05%和0.2%虾青素添加组ALT和AST水平高于对照组,而0.1%和0.15%虾青素添加组低于对照组。(4)饲料中添加虾青素能显著影响雌性吉富罗非鱼血清tp、TC和TG水平(P>0.05)。3.饲料中添加不同水平虾青素对雌性吉富罗非鱼血清性激素和性腺的影响饲料中添加虾青素可显著影响雌性吉富罗非鱼血清E2和FSH水平(P0.05)。其中,0.05%、0.1%和0.15%虾青素添加组LH和P水平高于对照组,而0.2%虾青素添加组低于对照组;0.15%虾青素添加组T水平高于对照组,而0.05%、0.1%和0.2%虾青素添加组低于对照组。通过对饲料中添加虾青素水平对性腺切片的影响,我们发现添加0.15%和0.2%的虾青素可以改善卵质量、成熟、发育。
