An inherent acceleratory effect of insulin on small intestinal transit and its pharmacological characterization in normal mice

AIM： To study an inherent effect of insulin on small intestinal transit and to explore involvement of various systems/mechanisms in normal mice. METHODS： Insulin at the doses of 2 μU/kg, 2 mU/kg, 2 U/kg or vehicle was subcutaneously administered to four groups of overnight fasted normal male mice. Blood glucose （BG） levels were measured 2 min before insulin administration and 2 min before sacrificing the animals for the measurement of small intestinal transit （SIT）. Charcoal meal was administered （0.3 mL） intragastrically 20 min after insulin administration and animals were sacrificed after 20 min and SIT was determined. For exploration of the various mechanisms involved in insulin-induced effect on SIT, the dose of insulin which can produce a significant acceleration of SIT without altering BG levels was determined. The following drugs, atropine （1 mg/kg）, clonidine （0.1 mg/kg）, ondansetron （1 mg/kg）, naloxone （5 mg/kg）, verapamil （8 mg/kg） and glibenclamide （10 mg/kg）, were administered intravenously 10 min prior to the administration of insulin （2 μU/kg）. RESULTS： The lower doses of insulin （2 μU/kg and 2 mU/kg） produced a significant acceleration of SIT from 52.0% to 70.7% and 73.5% without lowering blood glucose levels （P〈0.01）, while the highest dose of insulin （2 U/kg） produced a fall in blood glucose levels which was also associated with significant acceleration of SIT （P〈0.01）. After pretreatment of insulin （2 μU/kg） group with atropine, insulin could reverse 50% of the inhibition produced by atropine. In clonidine-pretreated group, insulin administration could reverse only 37% of the inhibition produced by clonidine and inhibition of SIT was significant compared with vehicle ＋ insulintreated group, i.e. from 74.7% to 27.7% （P〈0.01）. To study the anti-nociceptive effect of domper-idone and cisapride in mice. METHODS: Initially, the effect of these drugs on motor activity was tested using rotarod. The anti-nociception was tested using chemical and mechanical assay. In the chemical assay, the number of abdominal constrictions either in the saline treated animals or in the domperidone/cisapride (1, 5, or 10 mg/kg either po or ip) treated mice, were recorded for a period of 30 min after acetic acid challenge (10 mLAg, of 0.6 % acetic acid ip). In the tail clip assay, the time taken by the mouse to make attempts to dislodge the bulldog clamp placed at the tail (reaction time) was recorded with a cut off time of 30 s. The role of opioid pathways was examined by pretreating the animals with naloxone (1 mg/kg, ip) 30 min prior to domperidone and cisapride. Characterization of hepatic progenitors from human fetal liver during second trimester

AIM： To enrich hepatic progenitors using epithelial cell adhesion molecule （EpCAM） as a marker from human fetal liver and investigate the expression of human leukocyte antigen （HLA） and their markers associated with hepatic progenitor cells. METHODS： EpCAM ＋ve cells were isolated using magnetic cell sorting （MACS） from human fetuses （n = 10） at 15-25 wk gestation. Expression of markers for hepatic progenitors such as albumin, alpha-fetoprotein （AFP）, CD29 （integrin ~1）, CD49f （integrin c^6） and CD90 （Thy 1） was studied by using flow cytometry, immunocytochemistry and RT-PCR; HLA class Ⅰ （A, B, C） and class Ⅱ （DR） expression was studied by flow cytometry only. RESULTS： FACS analysis indicated that EpCAM ＋ve cells were positive for CD29, CD49f, CD90, CD34, HLA class I, albumin and AFP but negative for HLA class Ⅱ （DR） and CD45. RT PCR showed that EpCAM ＋ve cells expressed liver epithelial markers （Ck18）, biliary specific marker （Ck19） and hepatic markers （albumin, AFP）. On immunocytochemical staining, EpCAM ＋ve cells were shown positive signals for Ck18 and albumin. CONCLUSION： Our study suggests that these EpCAM ＋ve cells can be used as hepatic progenitors for cell transplantation with a minimum risk of alloreactivity and these cells may serve as a potential source for enrichment of hepatic progenitor.

Objective:To evaluate the anti-bacterial and anti-oxidant activity of and rographolide(AND)and echiodinin(ECH)of Andrographis ***:In this study,an attempt has been made to demonstrate the anti-microbial and anti-oxidant activity of isolated AND and ECH by broth micro-dilution method and 2,2-dipheny l-2-picryl-hydrazyl(DPPH)assay,*** elucidation was determined by electro-spray ionization-MSD,NMR(1H and13C)and IR ***:AND was effective against most of the strains tested including Mycobacterium smegmatis,showing broad spectrum of growth inhibition activity with Minimum inhibitory concentration values againstStaphylococcus aureus(100μg/mL),Streptococcus thermophilus(350μg/mL)Bacillus subtilis(100μg/mL),Escherichia coli(50μg/mL),Mycobacterium smegmatis(200μg/mL),klebsiella pneumonia(100μg/mL),andPseudomonas aeruginosa(200μg/mL).ECH showed specific anti-bacterial activity against Staphylococcus aureus,Escherichia coli,Bacillus subtilis and Pseudomonas aeruginosaat a concentration higher than 225μg/*** AND and ECH were not effective against the two yeast strains,Candida albicansand Saccharomyces cerevisiae tested in this ***:This preliminary study showed promising anti-bacterial activity and moderate free radical scavenging activity of AND and ECH,and it may provide the scientific rationale for its popular folklore medicines.

康契普拉姆地区是印度最大的手工丝织品产区,但目前该地区仍主要采用传统的蚕丝染色方法,染色常与脱胶同时进行,染色后蚕丝的色牢度较差。为提高染色效率和改善蚕丝的染色性能,采用现代染色工艺流程对蚕丝进行染色。以色牢度为考察指标,对不同方法染色后蚕丝的染色性能进行测试分析的结果显示:与传统染色方法相比,采用现代染色工艺流程染色的蚕丝,其染色性能稳定,色牢度明显提高。

Virgin Coconut Meal (VCM) was used for the development of instant wheat sooji (semolina) halwa mix with better nutritional attributes. Central composite rotatable design (CCRD) with 2 independent variables (sugar and VCM) and 4 responses (lightness, redness, taste and overall acceptability) was used for the optimisation. VCM incorporated instant halwa mix prepared using optimised levels of ingredients contained moisture 0.95%;fat 26.2%;protein 7.65%;total ash 0.86%;fibre 1.02% and received overall acceptability score of 8.5 on a 9 point hedonic scale providing 523.86 k·cal/ 100g. The changes in quality of stored VCM incorporated instant halwa mix packed in polypropylene (PP, 75 μ) and laminates of metallised polyester (MP, 90 μ) were monitored in order to assess the shelf-life. Instant halwa mix remained stable and acceptable for one year in both the packaging materials under ambient temperature conditions (15℃ - 34℃). However, the rate of lipid peroxidation was found to be slightly higher in PP packed samples as compared to MP packed ones. Fatty acid composition of VCM incorporated instant halwa mix remained practically unchanged during storage. Oleic acid was the major fatty acid present in fat extracted from halwa mix followed by palmitic and lauric acids.

In order to improve the nutritional quality of biscuits, a multigrain premix (MGP) was developed by using whole barley, sorghum, chickpea, pea and defatted soya flour, each at 20% level. The developed MGP had 26.28% protein, 10.13% insoluble dietary fiber and 7.38% soluble dietary fiber. The experiment was designed to optimise the MGP and wheat flour concentration for the development of multigrain biscuits with high protein, dietary fibre and to maximize the acceptability by the application of central composite rotatable design (CCRD) of Response Surface Methodology (RSM). The levels of incorporation of MGP and wheat flour were taken as variables whereas protein, soluble, insoluble fibers, biscuit dough hardness, breaking strength and overall acceptability (OAA) as responses. The optimum level of MGP and wheat flour obtained using numerical optimization was found to be 40 g and 60 g respectively. The biscuits prepared using these had 16.61% protein, 2.57% soluble fibre, and 6.67% insoluble fibre which is significantly (p ≤ 0.05) higher than control biscuit.

印度泰米尔那德邦的扎丽纱手工生产中常采用上浆处理提高蚕丝纱线的可织性和降低断头率。白胶是常用的一种粘稠度高的上浆剂,而硬挺剂是新出现的一种天然胶,其粘稠度相对较低。分别测试分析利用白胶、硬挺剂上浆处理对已染色蚕丝纱线的质量、色泽、韧性和延展性的影响,结果显示:用白胶处理后紫罗兰色蚕丝纱线的质量增加率显著提高,色度L*值、a*值和b*值变化较大,色差值ΔE*>2,但蚕丝纱线的韧性、延展性未发生显著变化;而用硬挺剂处理,对紫罗兰色、黄色和紫色3种颜色蚕丝纱线的质量、色差值、韧性和延展性均无明显影响。依据试验结果,建议采用硬挺剂对染色蚕丝纱线进行上浆处理。
