The cloning of non-structural-1 (NS1) gene of H9N2 subtype of avian influenza virus in pGEX-4T-1 and pMAL-c2X plasmids and expression in Escherichia coliDH5αstrain

Avian influenza is a viral contagious disease that affects poultry industry and human health. Vaccination has been considered as a preventive tool in the eradication of AI, but it causes some limitations including trade embargoes and interfering with serologic surveillance in differentiation between infected and vaccinated animals (DIVA strategy). Several distinct DIVA strategies have been presented to conquer these limitations. In this study, the open reading frame of NS1 gene of a H9N2 subtype of AI virus was amplified by polymerase chain reaction. After extraction and purification of NS1 gene from agarose gel, it was inserted into two different pGEX-4T-1 and pMAL-c2X plasmids and transferred in DH5α strain of Escherichia coli by using electroporation procedure. The E. coli colonies possessing recombinant NS1 gene were screened using PCR, restriction mapping and sequencing analysis. The expressed rNS1 protein was purified using affinity chromatography based on MBP (pMAL- c2X) and GST (pGEX-4T-1). The MBP-NS1 and GST- NS1 proteins on SDS-PAGE had bands with molecular weight of 68 and 52 kDa respectively. Western blotting with MBP-NS1 protein showed positive reaction using antisera obtained from chickens challenged with a H9N2 subtype strain. But, the most sera prepared from H9N2 vaccinated chickens were negative in WB. These findings indicated that the MBP-rNS1 protein of 26 kDa expressed by pMAL-c2X plasmid can be used in a DIVA for differentiation of AI infected and vaccinated chickens.

Preliminary Study on the Distribution of Irrawaddy Dolphin, Orcaella brevirostris, in Banten Bay

Irrawaddy dolphins (Orcaella brevirostris) are aquatic mammals with critically endangered status able to live in different water salinities habitats, like estuary, rivers and lakes. Banten Bay features estuarine area where rapid economic development, contributed from industry, ports, mining and fisheries, takes place, and thus the presence of Irrawaddy dolphins in the area faces various environmental threats. This research aimed to study the distribution of Irrawaddy dolphins in Banten Bay and explore environmental factors threatening its life. Dolphins observation was conducted from January 2013-February 2014, using direct visual effort by team of observer and indirect effort by local fishermen. Direct visual effort was performed on boat by three observers, with maximum vessel speed 15 km/hour and total length of transect was 404 Km. Indirect visual effort comprised of regular assessment by 32 local fishermen when they conducted capture fishing at two different areas (South and North of Banten Bay). Sightings data from direct and indirect visual observation compiled with respondents opinion were mapped using Arc View®?GIS 3.3, resulting in distribution map of Irrawaddy dolphin in Banten Bay. Irrawaddy dolphin was most frequently observed in the northeast, south and central areas of the bay. Environmental threats alarming the lives of Irrawaddy dolphin in the Banten Bay may include noise pollution, boat accident and entanglement.

Cloud computing is a kind of computing that depends on shared figuring assets instead of having nearby servers or individual gadgets to deal with applications. Technology is moving to the cloud more and more. It’s not just a trend, the shift away from ancient package models to package as service has steadily gained momentum over the last ten years. Looking forward, the following decade of cloud computing guarantees significantly more approaches to work from anyplace, utilizing cell phones. Cloud computing focused on better performances, better scalability and resource consumption but it also has some security issue with the data stored in it. The proposed algorithm intents to come with some solutions that will reduce the security threats and ensure far better security to the data stored in cloud.

Effect of Gold Nanoparticle on Percentage Depth Dose of 18 MV X-Ray in MAGICA Polymer Gel Dosimeter

Radiation-sensitive polymer gels are among the most promising three-dimensional dose verification tools and tissue-like phantom developed to date. This study is an investigating of percentage depth dose enhancement within the gel medium with the use of conformal distribution gold nanoparticle as contrast agents by high atomic number material. In this work, the normoxic polymer gel dosimeter MAGICA tissue-equivalence was first theoretically verified using MCNPX Monte Carlo code and experimentally by percentage depth dose curves within the gel medium. Then gold nanoparticles (GNPs) of 50 nm diameter with different concentrations of 0.1 mM, 0.2 mM, and 0.4 mM were embedded in MAGICA gel and irradiated by 18 MV photon beam. Experimental results have shown dose increase of 10%, 2% and 4% in 0.1 mM, 0.2 mM and 0.4 mM concentrations, respectively. Simulation results had good agreement in the optimum concentration of 0.1 mM. The largest error between experimental and simulation results was equal to 9.28% stood for 0.4 mM concentration. The results showed that the optimum concentration of gold nanoparticles to achieve maximum absorbed dose in both experimental and simulation was 0.1 mM and so it can be used for clinical studies.

Application of SimaPro7 on Al-Hilla City Sewerage Network, Iraq

SimaPro is a software package designed to make a valuable contribution for analyzing the environmental impact on products during their whole life cycle. A huge amount of knowledge about the environment is built into the program and database, enabling to analyze a product with a minimum of specialized knowledge. In this study, SimaPro was used to analyze and evaluate the impacts that resulted from sewerage network in Al-Hilla city, Iraq. The results of Life Cycle Assessment (LCA) showed that sewerage network had an impact and damage on the environment by 291 points for every cubic meter of collect wastewater. The most potential environmental impacts were global warming, respiratory inorganics and non-renewable energy, contributing to the sewerage network. The study also showed that most of the effects were as a result of energy consumption in the pumping of wastewater, pipes used and diesel using for network operation.

Potential of Fish Wastes as Feedstock for Biodiesel

Fish wastes are the discarded parts include the internal organs, viscera, bones, trimmings, tails, fins and skin of fishes. These discarded portions while disposing of cause major environmental damage. Usually, the discarded parts of fishes are ground into fishmeal for livestock and aquaculture feed. This study was undertaken to explore biodiesel production based on the fatty acids composition. The fish waste sample was collected from Kota Kinabalu, Sabah fish market. The sample was drained for excess water and oven-dried at 55°C - 60°C for complete dryness. Crude oils were extracted in petroleum ether in Soxhlet extraction method. Methylation of the extracted crude fish oil was carried out to yield fatty acid methyl esters (FAME). The FAME was analyzed by GCMS system and the reference to NIST library was used to identify the fatty acids present in the FAME. A total of 21 fatty acids were identified that composed of 53.53% saturated fatty acids (SFA), 22.1% monounsaturated fatty acid (MUFA) and 24.37% polyunsaturated fatty acids (PUFA). The important fatty acids [myristic acid (C14:0), palmitic acid (C16:0), palmitoleic acid (C16:1), oleic acid (C18:1), linoleic acid (C18:2), linolenic acid (C18:3), docosapentaenoic acid (C22:5) and docosahexaenoic acid (C22:6)] found in fish oil indicated the potentiality of biodiesel production if fish waste was stocked. The highest percentage of SFA causes higher viscosity, cetane number and density and hence these properties of biodiesel produced from the fish waste are expected to be high. Therefore, the fish waste has high potential of fatty acid in FAME to produce biodiesel through transesterification process.

Addiction Treatment Aftercare Outcome Study

Aftercare is crucial once an individual has completed drug or alcohol treatment and is in recovery. There is a continuity of care that should be followed once initial treatment is completed. This usually involves a lower level of treatment such as outpatient care and a sober living environment. In order to assess the efficacy and benefit of our addiction treatment program, we investigate a set of patients in which addiction treatment outcome and rehabilitation is determined for patients who have completed treatment and followed up. We determine abstinence rates and identify predictors of treatment outcome.

Protective Effect of Curcumin on Anxiety, Learning Behavior, Neuromuscular Activities, Brain Neurotransmitters and Oxidative Stress Enzymes in Cadmium Intoxicated Mice

Cadmium (Cd) exposure can induce acute lethal health-related threats to humans since it has an exceptional ability to accumulate in living organisms and cause toxicological effects. Curcumin (Cur) on the other hand has a wide variety of biological activities and several animal studies have suggested for a potential therapeutic or preventive effects against several ailments and infections. To study the effect of Cur on the toxicity of Cd, sixty Swiss-Webster strain male mice were divided into 6 groups of ten each at random. Group-1 served as the na?ve control and received no treatment. Group-2, 3 and 4 were the experimental controls and were administered once a day with a single oral dose of 50% dimethyl sulphoxide (DMSO), Cur (300 mg/kg) or Cd (100 mg/kg) respectively, for 2 weeks. Group-5 and 6 received Cur and Cd in combination once a day orally for 2 weeks except that Cur in a dose of 150 and 300 mg/kg to group 5 and 6 respectively, was administered one hour before Cd (100 mg/kg) administration to both groups. After treatment period, the animals were subjected to behavioral tests and thereafter, the animals were sacrificed for the estimation of neurotransmitters like serotonin (5-HT), dopamine (DA) and it’s metabolite 3,4-dihydroxyphenylacetic acid (DOPAC) as well as oxidative stress enzymes like lipid peroxides in the form of thiobarbituric acid–reactive substances (TBARS) and total glutathione (GSH) in the forebrain tissue. Cd reduced significantly the body weight gain, the locomotor activity, anxiety behavior in the plus maze and the learning capability (cognitive effect) in the shuttle-box test. Biochemical analysis further revealed that Cd exposure significantly altered the brain neurotransmitters and the oxidative stress enzymes. However, administration of Cur along with Cd had an ameliorating effect on all the behavioral and biochemical parameters studied herein and reduced the toxicity of Cd significantly and dose-dependently. Mineral Chemistry and Geothermometry of Amphibole and Plagioclase in the Metabasites, Located at the Tanbour Metamorphic Complex in Southern Iran

Metabasite refers to metamorphosed basalts and other mafic igneous rocks (rich in iron and magnesium). When a mafic igneous rock is subjected to new pressure and temperature conditions during metamorphism, these chemical components will rearrange themselves to form new minerals. Metabasites can be found in many metamorphic belts including Sanandaj-Sirjan metamorphic belt of Iran. The study area is a Tanbour metamorphic complex in Eastern of Sirjan city, which is geologically located at the Sanandaj-Sirjan metamorphic belt in Southern Iran. Metabasite in this complex consists of greenschist, epidote amphibolite and amphibolite. Amphibole and plagioclase are the main minerals in the greenschist and amphibolite, and the a secondary mineral in some micaschist seen in the study area. The electron microprobe analysis was done on this mineralization in greenschist, epidote amphibolite and amphibolite, which showed that the amphiboles in greenschist was a member of the calcic group and Actinolite type, and the amphiboles in epidote amphibolite was a member of the calcic group and these amphiboles were tschermakite up to Ferro-Tschermakite + Ferro-Hornblende type. The amphibole in amphibolite is a member of the calcic group and this amphibole is Magnesio-Hornblende type. The plagioclases in the greenschist is pure albite (An 3.29 - 3.6), and in the epidote amphibolite is oligoclase (An 19.5 - 24.2), while in the amphibolites is oligoclase (An 16.9 - 26.6). The estimated P–T conditions are in favor of their metamorphism under epidote amphibolite (550°C and 8 kbar) and amphibolite (611°C - 652° Cand 10.5 kbar) facies.
