Systematic mutational analysis of epitope-grafted ED3 immunogenicity reveals a DENV3-DENV4 bi-serospecific ED3 mutant

Objective:To identify the residue determinants of the serospecificity and sero-cross-reactivity of dengue virus(DENV)envelope protein domain 3(ED3),which contains two major putative epitopes of ***:We constructed ED3 from DENV3(3ED3)and DENV4(4ED3),and six epitope-grafted variants,where we transferred epitope 1(L304I,K305D,V309m,and S310A)and/or epitope 2(D383N,K384S,K387T,and N389H)of 4ED3 onto 3ED3 and *** albino mice aged 3-4 weeks were immunized against wildtype and epitope-grafted ED3 variants and anti-ED3 IgG antibody responses were determined using ***:mouse immunization using 3ED3 and 4ED3 generated serotype-specific antisera,as ***,most epitope-grafted ED3s produced antisera serospecific to the template ED3 with little or no cross-recognition of ED3 of the serotype from which the epitopes were *** indicated that a mere grafting of the epitope was not sufficient to transfer serospecificity,contrary to our ***,one epitope-grafted ED3 mutant,where epitope 1 of 3ED3 was grafted onto 4ED3(4ED3^(epi1)),generated antisera that was serospecific to both 4ED3 and ***:The 4ED3^(epi1)is a chimeric ED3 that produces antisera possessing serospecificity to both 3ED3 and 4ED3 onto a common 4ED3 *** 4ED3^(epi1),therefore,provides a unique tool for analyzing serospecificity and sero-cross-reactivity in *** believe that chimeric ED3 may provide a template for future recombinant ED3 possessing serospecificity of multiple DENVs onto a single scaffold and may pave a way developing tri-and/or tetravalent anti-DENV antisera.

Investigation of material composition,design,and performance of open-graded asphalt mixtures for semi-flexible pavement:A comprehensive experimental study

The primary goal of this study is the design and construction of semi-flexible pavement(SFP)mixture in accordance with the engineering and mechanical *** study involves the use of a range of gradation curves,air void contents,cellulose and synthesized fibers,and neat and modified asphalt binders to prepare the open-graded asphalt(OGA)*** analyze the characteristics of these mixtures,a variety of test,namely binder drainage,semi-circular bending(SCB),Cantabro,wheel tracking,indirect tensile strength(ITS),and permeability tests were ***,to analyze the prepared grouting material,flexural strength,compressive strength,and fluidity tests were *** the final stage,SFP was compared to HmA in terms of engineering characteristics and *** to the results,SFP was more resistant to skid,rutting,fire,and moisture damage,while HmA had a better performance in fracture tests,including SCB *** to the results of the mechanical performance tests conducted on OGA mixtures,the highest and lowest values for air void content to achieve the highest mechanical performance level were 30%-35%and 25%,***,based on the laboratory results,it was determined that the required void ratio for constructing OGA mixtures was 24%-26%based on the bitumen type and fibers amount in the ***,SFP mixture can be regarded as a viable alternative to common pavements thanks to its high resistance to rutting and moisture damage,long freezing-thawing fatigue life,and adequate fire and skid resistance.

Towards implementation of alloy-specific thermo-fluid modelling for laser powder-bed fusion of mg alloys

multi-physics thermo-fluid modeling has been extensively used as an approach to understand melt pool dynamics and defect formation as well as optimizing the process-related parameters of laser powder-bed fusion(L-PBF).However,its capabilities for being implemented as a reliable tool for material design,where minor changes in material-related parameters must be accurately captured,is still in *** the present research,first,a thermo-fluid computational fluid dynamics(CFD)model is developed and validated against experimental *** the predicted material properties of the pure mg and commercial ZK60 and WE43 mg alloys,parametric studies are done attempting to elucidate how the difference in some of the material properties,i.e.,saturated vapor pressure,viscosity,and solidification range,can influence the melt pool *** is found that a higher saturated vapor pressure,associated with the ZK60 alloy,leads to a deeper unstable keyhole,increasing the keyhole-induced porosity and evaporation mass *** viscosity and wider solidification range can increase the non-uniformity of temperature and velocity distribution on the keyhole walls,resulting in increased keyhole instability and formation of ***,the WE43 alloy showed the best behavior in terms of defect formation and evaporation mass loss,providing theoretical support to the extensive use of this alloy in *** summary,this study suggests an approach to investigate the effect of materials-related parameters on L-PBF melting and solidification,which can be extremely helpful for future design of new alloys suitable for L-PBF.

Although significant advances have been made in understanding the pathophysiology of psychiatric disorders(PDs),therapeutic advances have not been very *** psychotropic medications can reduce classical symptoms in patients with PDs,their long-term use has been reported to induce or exaggerate various pre-existing metabolic abnormalities including diabetes,obesity and non-alcoholic fatty liver disease(NAFLD).The mechanism(s)underlying these metabolic abnormalities is not clear;however,lipid/fatty acid accumulation due to enhanced de novo lipogenesis(DNL)has been shown to reduce membrane fluidity,increase oxidative stress and inflammation leading to the development of the aforementioned metabolic ***,emerging evidence suggest that DNL dysregulation and fatty acid accumulation could be the major mechanisms associated with the development of obesity,diabetes and NAFLD after long-term treatment with psychotropic medications in patients with *** support of this,several adjunctive drugs comprising of anti-oxidants and antiinflammatory agents,that are used in treating PDs in combination with psychotropic medications,have been shown to reduce insulin resistance and development of *** conclusion,the above evidence suggests that DNL could be a potential pathological factor associated with various metabolic abnormalities,and a new avenue for translational research and therapeutic drug designing in PDs.

Increasing demand for energy due to the populous Eastern Australia has driven oil and gas industries to find new sources of hydrocarbons as they are the primary energy *** study has been done on the Volador Formation in the Gippsland Basin by means of core-based petrophysical,sedimentological,and petrographic analyses as well as well log-based interpretation and capillary pressure *** wells from Kipper,Basker and Tuna fields with available dataset were investigated in this study:Kipper-1,Basker-1,Basker-2,Basker-5 and ***,the formation has good reservoir quality based on the high porosity and permeability values obtained through core and well log petrophysical *** formation made up of mostly moderate to coarse quartz grains that has experienced strong anti-compaction and is poorly *** and illite clays are seen dispersed in the rock formation,with the minority being mixed *** clays and diagenetic features including kaolinite cement and quartz overgrowth that can lead to porosity reduction only have insignificant impact on the overall reservoir *** addition,capillary pressure data shows that most samples are found in the transition to good reservoir zones(m this study have shown that the Volador Formation in the Gippsland Basin is worth for hydrocarbon exploration.

Time-dependent effects in transient liquid phase bonding of 304L and Cp-Ti using an Ag-Cu interlayer

One of the challenges for bimetal manufacturing is the joining ***,transient liquid phase(TLP)bonding was performed between 304L stainless steel and Cp-Ti using an Ag-Cu interlayer with a thickness of 75μm for bonding time of 20,40,60,and 90 *** bonding temperature of 860℃ was considered,which is under the β transus temperature of *** TLP bonding,various intermetallic compounds(ImCs),including Ti_(5)Cr_(7)Fe_(17),(Cr,Fe)_(2)Ti,Ti(Cu,Fe),Ti_(2)(Cu,Ag),and Ti_(2)Cu from 304L toward Cp-Ti formed in the ***,on the one side,with the increase in time,further diffusion of elements decreases the blocky ImCs such as Ti_(5)Cr_(7)Fe_(17),(Cr,Fe)_(2)Ti,Ti(Cu,Fe)in the 304L diffusion-affected zone(DAZ)and reaction zone,and on the other side,Ti_(2)(Cu,Ag)ImC transformed into fine morphology toward Cp-Ti *** microhardness test also demonstrated that the(Cr,Fe)_(2)Ti+Ti_(5)Cr_(7)Fe_(17) ImCs in the DAZ on the side of 304L have a hardness value of HV 564,making it the hardest *** maximum and minimum shear strength values are equal to 78.84 and 29.0 mPa,*** cleavage pattern dominated fracture surfaces due to the formation of brittle phases in dissimilar joints.

Engineered cardiac constructs(ECC)aid in the progression of regenerative medicine,disease modeling and targeted drug delivery to adjust and aim the release of remedial combination as well as decrease the side effects of *** this research,polycaprolactone/gold nanoparticles(PCL/GNPs)three-dimensional(3D)composite scaffolds were manufactured by 3D printing using the fused deposition modeling(FDm)method and then coated with gelatin/spironolactone(GEL/SPL).Scanning electron microscopy(SEm)and Fourier transform-infrared spectroscopy(FTIR–ATR)were applied to characterize the ***,drug release,biodegradation,behavior of the myoblasts(H9C2)cell line,and cytotoxicity of the 3D scaffolds were *** microstructural observation of the scaffolds reported interconnected pores with 150–300µm in *** 3D scaffolds were degraded significantly after 28 days of immersion in stimulated body fluid(SBF),with the maximum rate of GEL-coated 3D *** release from cross-linked GEL coating demonstrated the excess of drug release over time,and according to the control release systems,the drug delivery systems(DDS)went into balance after the 14th *** addition,cell culture study showed that with the addition of GNPs,the proliferation of(H9C2)was enhanced,and with GEL/SPL coating the cell attachment and viability were improved *** findings suggested that PCL/GNPs 3D scaffolds coated with GEL/SPL can be an appropriate choice for myocardial tissue engineering.
