Nervous system disorders are prevalent health issues that will only continue to increase in frequency as the population ***-back axonopathy is a hallmark of many neurologic diseases and leads to axonal disconnection from their targets,which in turn leads to functional *** the course of many of neurologic diseases,axons can regenerate or sprout in an attempt to reconnect with the target and restore synapse *** amyotrophic lateral sclerosis(ALS),distal motor axons retract from neuromuscular junctions early in the disease-course before significant motor neuron *** is evidence of compensatory motor axon sprouting and reinnervation of neuromuscular junctions in ALS that is usually quickly overtaken by the disease *** drugs that enhance compensatory sprouting and encourage reinnervation may slow symptom progression and retain muscle function for a longer period of time in ALS and in other diseases that exhibit dying-back *** remain many outstanding questions as to the impact of distinct disease-causing mutations on axonal outgrowth and regeneration,especially in regards to motor neurons derived from patient induced pluripotent stem *** microfluidic chambers are powerful tools for studying the distal axons of human induced pluripotent stem cells-derived motor neurons,and have recently been used to demonstrate striking regeneration defects in human motor neurons harboring ALS disease-causing *** the human neuromuscular circuit with human induced pluripotent stem cells-derived motor neurons will be critical for developing drugs that enhance axonal regeneration,sprouting,and reinnervation of neuromuscular *** this review we will discuss compensatory axonal sprouting as a potential therapeutic target for ALS,and the use of compartmentalized microfluidic devices to find drugs that enhance regeneration and axonal sprouting of motor axons.

Phishing,an Internet fraudwhere individuals are deceived into revealing critical personal and account information,poses a significant risk to both consumers and web-based *** indicates a persistent rise in phishing ***,these fraudulent schemes are progressively becoming more intricate,thereby rendering them more challenging to ***,it is imperative to utilize sophisticated algorithms to address this *** learning is a highly effective approach for identifying and uncovering these harmful *** learning(ML)approaches can identify common characteristics in most phishing *** this paper,we propose an ensemble approach and compare it with six machine learning techniques to determine the type of website and whether it is normal or not based on two phishing *** that,we used the normalization technique on the dataset to transform the range of all the features into the same *** findings of this paper for all algorithms are as follows in the first dataset based on accuracy,precision,recall,and F1-score,respectively:Decision Tree(DT)(0.964,0.961,0.976,0.968),Random Forest(RF)(0.970,0.964,0.984,0.974),Gradient Boosting(GB)(0.960,0.959,0.971,0.965),XGBoost(XGB)(0.973,0.976,0.976,0.976),AdaBoost(0.934,0.934,0.950,0.942),Multi Layer Perceptron(MLP)(0.970,0.971,0.976,0.974)and Voting(0.978,0.975,0.987,0.981).So,the Voting classifier gave the best *** in the second dataset,all the algorithms gave the same results in four evaluation metrics,which indicates that each of them can effectively accomplish the prediction ***,this approach outperformed the previous work in detecting phishing websites with high accuracy,a lower false negative rate,a shorter prediction time,and a lower false positive rate.

背景在中国,子宫内膜癌是女性生殖道最常见的浸润性恶性肿瘤,其发病率随着生活方式的改变而持续上升。大多数EC是散发性的,只有10%是家族性的。EC是一组异质性肿瘤,不同亚型的患者对治疗反应的结果不同。大多数确诊患者为绝经后妇女,少数患者在育龄期确诊。然而,这些育龄期患者往往表现出强烈的生育意愿。多囊卵巢综合征、肥胖、胰岛素抵抗、糖尿病、高血压、高龄、种族、未生育、初潮早、绝经晚、月经周期不规则、不孕、无排卵或稀疏排卵、闭经、使用他莫昔芬和遗传易感性都是EC患者的危险因素。EC的主要治疗方式包括手术、放疗和化疗。它通常通过筋膜外全子宫+双侧输卵管切除术(剖腹手术或腹腔镜检查)进行治疗,伴或不伴双侧卵巢切除术,伴或不伴盆腔和/或腹主动脉旁淋巴结切除术。然而,这会导致生殖功能的永久性损害。保守治疗已被建议作为延迟或预防手术的一种手段。对于Ⅰ级、IA期、局限于子宫内膜的年轻女性的早期子宫内膜癌,经全面检查并慎重考虑后决定保留生育能力,是可行且有效的方案。因此,探索精确的诊断和预后生物标志物以及EC的创新治疗方案至关重要。目的目前,国内外关于子宫内膜癌保守治疗成功的报道很多,但关于保守治疗失败的报道相对较少。本研究报道了保守治疗失败的子宫内膜癌的分子分型,并评估其临床和病理特征。因此,本研究旨在研究的是育龄期妇女保守治疗失败的早期子宫内膜癌(EEC)患者的分子类别。方法2011年1月至2021年10月近10年在中国长春市吉林大学第二医院病历系统中40岁以下进行常规激素治疗的子宫内膜癌育龄期妇女共186例。我们的回顾性研究共纳入了15例保守治疗失败的EC患者,并收集了2020年9月至2021年3月的病历资料。因此,我们评估了保守治疗的结果和预后。此外,我们将术前刮宫活检结果与组织病理学结果进行了比较。进而,我们使用下一代测序(NGS)确定了所有患者的分子分型,以发现哪些分子类型的子宫内膜癌(EC)不适合接受保守治疗的生育管理。结果符合G1级EC诊断标准的15例患者的中位年龄为33.67岁,年龄范围为26-40岁。月经初潮的中位年龄为13.56岁,年龄范围为11-16岁。中期随访时间为39个月,从13个月到120个月不等。这些患者中,3例为单身且无妊娠史;其他12人已婚,但其中10人主要是不孕。总共有3名患者完全缓解后复发,1名患者显示出疾病进展并进行了子宫切除术。此外,11名进行子宫切除患者疾病稳定,孕激素无反应。其中,宫颈内上皮样瘤变1例,PCOS 2例,有肝病1例,高血压1例,肥胖8例。总体而言,15例EC患者在保守治疗失败后接受了手术干预,包括完全缓解(CR)后复发,疾病稳定(SD)和疾病进展(PD)。此外,在手术时15例患者中有12例保留卵巢,其中8例行术中卵巢活检,结果均为阴性。在诊断性刮宫活检中,所有患者病理报告均为G1 EC。手术病理与刮宫活检的结果不一致,15例患者中有6例显示出不同的病理类型。此外,分子分型结果显示CNL 13例,MSI-h 2例。然而,所有的患者在保守治疗后仍然无法生育。结论分子分型可能为评估预后提供可靠的附加信息,并有助于EEC患者治疗策略的选择。EC女性的保守治疗是安全有效的,但需要谨慎选择患者。然而,EEC患者保守治疗后妊娠失败的问题仍未解决。此外,不推荐对合并MSI-h的EEC患者进行保守治疗。然而,对于患有ChL的EEC患者可以尝试这样做,但需要定期随访以监测EC复发并防止疾病恶化。

An Improved Method for Preparation of Uniform and Functional Mitochondria from Fresh Liver

Background and Aims:As the major energy source for mammalian cells,mitochondria have been the subject of numerous ***,the isolation and purification of healthy mitochondria,especially from fresh tissue,remains *** most popular methods and kits involve various centrifugation steps which require substantial time and equipment but do not consistently provide pure preparations of functional *** aim of this study was to determine whether methods could be devised to improve the purity and yield of functional mitochondria from fresh ***:Fresh mouse liver was homogenized,and cells *** size analysis,quantitation of mitochondrial DNA,mitochondrial oxygen consumption,and purity of mitochondria(by electron microscopy)were measured in samples after various purification steps and significant differences ***:A two-step procedure consisting of centrifugation followed by filtration through 1.2μand 0.8μfilters resulted in uniform mitochondrial preparations with diameters between 520-540 nm,and approximately 5-times more pure *** mitochondria thus obtained had oxygen consumption and sensitivities to mitochondrial inhibitors that were indistinguishable from those purified by centrifugation *** microscopy confirmed the presence of more uniform and 4-5 times greater concentrations of mitochondria compared to centrifugation ***:A two-step procedure consisting of sequential centrifugation followed by filtration is a rapid method for the production of highly purified,uniform and functional mitochondria.

聚氨酯是一种聚合物树脂材料,将聚氨酯胶粘剂与沙子混合而制成的聚氨酯砂浆作为修补材料受到越来越多的关注。在这项研究中,通过测试聚氨酯砂浆和聚氨酯橡胶混合砂浆(聚氨酯与沙子和橡胶颗粒混合)的抗压、抗折强度,以此来评估橡胶颗粒对其材料性能的影响。同时以聚氨酯砂浆和聚氨酯橡胶混合砂浆作为粘结材料,进行了水泥砂浆界面粘结试验以及界面拉伸强度试验。本研究使用的材料设计如下。聚氨酯胶粘剂由A和B两种组分组成。A组分(多元醇)是一种含有羟基的化合物,B组分(MDI)是含有游离异氰酸酯基的化合物。试验变量分别选择A:B为5:1和6:1的两种比例,橡胶颗粒的粒径采用16目和60目这两种尺寸,以0,5%,10%,15%的比率等体积取代砂来形成试验配合比。此外,还制作了混合比为1:3:0.5的普通水泥砂浆。论文中的试验包括了54个配合比,共计420件试件。主要研究成果如下:1)聚氨酯砂浆在3h时的抗折强度平均在5MPa,抗压强度在11MPa左右。这表明聚氨酯砂浆可用作修补材料。与聚氨酯砂浆相同,聚氨酯橡胶混合砂浆的抗压强度和抗折强度在3h和6h时都有小幅下降,而养护龄期在24h时则略有增加,且测得掺入橡胶粒径为16目时试件的强度大于掺入60目橡胶颗粒的试件强度。同时,当聚氨酯胶粘剂中A和B组分的比例为6:1时,聚氨酯橡胶混合砂浆在3h,6h,1d,3d和7d的抗压强度和抗折强度均高于A和B组分的比例为5:1的情况。2)对组合粘结试件,当养护龄期为7d时,不含橡胶颗粒的聚氨酯试件(PU比为6:1,PC2-0)在7d最高强度为12 MPa;但是,随着橡胶含量的增加和橡胶粒径的减小,界面粘结强度不断降低。比较了由两种不同比例的A和B组分制成的聚氨酯粘结砂浆,以6:1的比例制备的测试件的结合强度更大。3)拉伸粘结强度试验的结果表明,粘结强度优选与非橡胶的空白聚氨酯砂浆样品相比;随着橡胶颗粒含量的增加,试验片的粘结破坏负荷与聚氨酯橡胶混合砂浆样品中的60目颗粒尺寸相比,16目橡胶颗粒的粘结强度降低的结果更大。在该测试中,样品PC2b-2的最高拉伸粘合试验为1.411 MPa。当PU的比例为6:1时,橡胶含量为5%和10%,粒径为60目时,仍显示出较高的粘合强度,可以满足维修项目的要求。论文创新包括首次把橡胶颗粒掺入聚氨酯砂浆试件,并测试了其拉伸粘结强度。聚氨酯混合砂浆与普通水泥砂浆组合试件的界面粘结强度试验结果也是首次报告。

Computational fuid dynamics(CFD)simulations of a single staged injection of h_(2) through a central wedge shaped strut and a multi staged injection through wall injectors are carried out by using Ansys CFX-12 *** terahedral grids for narow channel and quarter geometries of the combustor are generated by using ICEM *** three dimensional(3D)Reynods averaged Navier-stokes(RANS)simulations are carried out in the case of no h_(2) injection and compared with the simulations of single staged pilot and/or main h2 injections and multistage *** stuess transport(SST)based on k-ω turbulent model is *** field visualization(omplex shock waves interactions)and static pressure distribution along the wall of the combustor are pradicted and compared with the experimental schlieren images and measured wall static pressures for validation.A good agreement is found between the CFD predicted results and the measured *** narow and quarter geometries of the combustor give similar results with very small ***-staged injections of h_(2) enhance the turbulent h_(2)/air mixing by fomming vortices and additional shock waves(bow shocks).
