Antihyperglycemic effect of Juniperus phoenicea *** alloxan-induced diabetic rats and diterpenoids isolated from the fruits

Objective:To explore the traditional use of Juniperus phoenicea L.(***)growing in Egypt as antidiabetic ***:The antihyperglycemic activities of the crude 80%ethanol and successive extracts of leaves and fruits of the plant were investigated in alloxan-induced diabetic rats after collecting blood samples through retro-orbital puncture *** a consequence of the biological results,phytochemical investigation of the chloroform fraction of fruits was carried out by column chromatography and thin layer ***:Results revealed the reduction in blood glucose levels in rats,which were significantly different from control at 4 and 8 weeks(P<0.01).The highest antihyperglycemic activity was exhibited by the crude extracts of fruits and leaves of which the potency was 83.6%and 81.9%,respectively,after 8 weeks,comparing to metformin drug(100%potency).Chloroform fractions of leaves and fruits were the most potent fractions(potencies were 70.3%,71.4%),respectively,along with ethyl acetate fraction of fruits(71.4%).Phytochemical investigating of the chloroform fraction of fruits resulted in the isolation and identification of 5 abietane ***,7-dehydroabietanone,sugiol,6-α-hydroxy-7-oxoferruginol,totarolone and a labdane diterpenoid,varodiol were isolated for the first time from the fruits of *** growing in *** identification of these compounds was based on spectroscopic analysis:1hydrogen-nuclear magnetic resonance and electron impact mass spectrometry,comparing the results with the ***:It has become clear that leaves and fruits of the Egyptian *** provide effective antihyperglycemic action in diabetic rats as was reported in folk *** high contents of terpenoids in the non-polar fractions may attribute to the antidiabetic effect of the plant.

BACKGROUND Hepatocellular carcinoma(HCC)is the fifth most common *** expression of microRNAs(miRNAs)-326,miRNA-424,and miRNA-511 has been associated with the diagnosis and prognosis of HCC in different ***,limited information is available regarding their expression in Egyptian HCC *** To assess the role of circulating miRNAs-326,miRNA-424,and miRNA-511 in Egyptian HCC *** This prospective observational study included 70 HCC patients and 25 healthy *** circulating levels of these three miRNAs were evaluated by real-time *** operating characteristic curve analysis was used to test the diagnostic accuracy of micro RNA expression *** All miRNAs were differentially expressed in HCC patients;miRNAs326 and miRNA-424 were upregulated,while miRNA-511 was *** miRNA-326 and miRNA-424 showed sensitivity and specificity of 97%,71.4%,and 52%,60%,respectively,to differentiate HCC from ***,miRNA-326 was associated with survival and could differentiate between Child grades(A vs B);miRNA-424 significantly differentiated early vs intermediate stages of HCC;while miRNA-511 was significantly correlated with response to modified Response Evaluation Criteria in Solid Tumors(m RECIST).CONCLUSION We conclude that miRNA-326,miRNA-424,and miRNA-511 have diagnostic and prognostic roles in Egyptian patients with hepatitis C virus-related HCC and should be considered for better disease management.

Ibandronate sodium monohydrate is a highly polar aliphatic compound that belongs to the bisphosphonate class,a widely used bone resorption *** aliphatic nature of ibandronate precludes direct photometric detection and its high polar-ity urged analysts to use ion-pairing agents in the mobile phase to promote retention in reversed-phase *** this work,a reversed-phase method for determination of ibandronate sodium monohydrate in pharmaceutical tablets is introduced by employing an online postcolumn ligand exchange reaction to enable photometric *** method offers for the first time the ability to separate the drug from its well-known impurities on conventional reversed-phase *** detection reac-tion depends on the ability of ibandronate and its degradation products to displace salicylate in the iron(III)salicylate complex,forming various colorless iron(III)complexes and showing a negative chromatographic signal for ibandronate and its degra-dants atλ_(max)=525 *** chromatographic separation was achieved using a Hypersil BDS C_(8) column(5μm particle size,150 mm×4.6 mm i.d.)and a mobile phase consisting of a mixture of aqueous formate buffer pH 3(pH 3,5 mM)and methanol in a ratio of 80:20%(v/v),delivered at a flow rate of 0.8 mL/*** postcolumn iron(III)salicylate reagent was prepared at 250μM in aqueous formate buffer,delivered using a syringe pump at a flow rate of 0.5 mL/*** proposed method was validated as per the guidelines of the International Conference on Harmonization Q2(R1)and was found linear over the range 40-120μg/mL(r=0.9995)with a limit of detection of 3.19μg/*** to currently reported methods,the proposed method is considerably simpler,faster,cheaper,and adequately sensitive and makes use of the most popular LC detectors.

Fetal Cerebellar Vermis Assessment by MRI. What Does It Add?

Background: Posterior fossa anomalies (PFAs) are considered one of the commonest brain anomalies identified by the fetal imaging techniques. The current study aims to estimate the significance of cerebellar vermis assessment by fetal MRI in fetal brain malformations. Patients and Methods: The study was conducted at a tertiary University hospital in Upper Egypt. Data were gathered prospectively from sixty pregnant women with sonographically diagnosed fetal brain malformations referred for Magnetic Resonance Imaging (MRI) between April 2015 and May 2019 followed by postpartum neonatal MRI brain examination for non-terminated and live birth cases. Results: Of the sixty fetuses with brain malformations, additional cerebellar vermis findings were seen on MRI in 10 cases (16.6%). Fetal MRI sensitivity, specificity, positive and negative predictive values were 100% that were in correlation with those of prenatal ultrasound turned out to be significantly higher. Conclusion: This study recommends fetal MRI for cerebellar vermis assessment in fetal brain malformations to guide the clinical management.

Inter-Specific Biochemical Diversity between <i>Echis pyramidum</i>and <i>Eryx colubrinus</i>Inhabiting El-Faiyum, Egypt

Discontinuous polyacrylamide gel electrophoreses for lactate dehydrogenase (Ldh) and alfa-esterase (α-Est) isoenzymes were conducted for biochemical differentiation between Echis pyramidum and Eryx colubrinus inhabiting El-Faiyum, Egypt. Total lipids and total protein of liver and muscle tissues in both species were also analyzed. Three Ldh isoforms were recorded in both species and the activity and relative front (RF) of Ldh-1 seemed to be higher in E. pyramidum than in E. colubrinus. This high activity could be supported by the significant increase in the total lipids and total protein in liver and muscle tissues of this species. It is thus possibly reasonable to suppose that E. pyramidum is more active, energetic and adaptable in its habitat than E. colubrinus. α-Est showed five isozymic forms fractions in E. pyramidum, while it showed only four isoforms in E. colubrinus. α-Est-1 was the first clear, dense and thick isoform in E. pyramidum, but it was completely absent in E. colubrinus. High activity of the esterase isoform;α-Est-1 only in the heart tissue of E. pyramidum may reflect the high ability of E. pyramidum to be more resistant to the accumulated toxic residues in its body tissues than E. colubrinus.

AIM: To investigate the association of the functional monocyte chemotactic protein-1(MCP-1) promoter polymorphism(A-2518G) with spontaneous bacterial peritonitis(SBP).METHODS: Fifty patients with post-hepatitis C liver cirrhosis and ascites were categorized into two groups; group Ⅰ included 25 patients with SBP and group Ⅱ included 25 patients free from SBP. In addition, a group of 20 healthy volunteers were included. We assessed the MCP-1 gene polymorphism and gene expression as well as interleukin(IL)-10 levels in both blood and ascitic fluid. RESULTS: A significant MCP-1 gene polymorphism was detected in groups Ⅰ and Ⅱ(P = 0.001 and 0.02 respectively). Group Ⅰ was associated with a significantly higher frequency of AG genotype [control 8(40%) vs SBP 19(76.0%), P < 0.001], and group Ⅱ was associated with a significantly higher frequency of GG genotype when compared to healthy volunteers [control 1(5%) vs cirrhotic 16(64%), P < 0.001]. Accordingly, the frequency of G allele was significantly higher in both groups(Ⅰ and Ⅱ) [control 10(25%) vs SBP 27(54%), P < 0.001 and vs cirrhotic 37(74.0%), P < 0.001, respectively]. The total blood and ascetic fluid levels of IL-10 and MCP-1 gene expression were significantly higher in group Ⅰ than in group Ⅱ. Group Ⅰ showed significant reductions in the levels of MCP-1 gene expression and IL-10 in the whole blood and ascetic fluid after therapy. CONCLUSION: MCP-1 GG genotype and G allele may predispose HCV infected patients to a more progressive disease course, while AG genotype may increase the susceptibility to SBP. Patients carrying these genotypes should be under supervision to prevent or restrict further complications.
