Impact of source tissue and ex vivo expansion on the characterization of goat mesenchymal stem cells

Background： There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells （MSCs） have been isolated and characterized from various species, but are poorly characterized in goats. Results： Goat MSCs isolated from bone marrow （BM-MSCs） and adipose tissue （ASCs） have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency （CFE）, yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions： Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

BACKGROUND Cervical cancer is a formidable global health issue,particularly affecting women in lower-middle-income countries with little or no access to preventative vaccines,screening programs,and treatment *** case report presents a unique case of a large cervical cancer achieving complete response(CR)with concurrent chemoradiotherapy(CCRT),highlighting the effectiveness of this treatment approach even in advanced stages and underscoring the importance of adaptive radiotherapy(RT)in optimizing patient *** SUMMARY We present the case of a 53-year-old woman who presented with four years of abnormal vaginal bleeding and was found to have p16-positive,moderately differentiated cervical squamous cell *** tumor measured 14 cm×12 cm×8 cm,the largest size reported in the literature to achieve CR with *** this monumental feat,the patient remained disease-free and is currently on follow-up for 2 years;however,she continued to suffer from substantial morbidity caused by a vesicovaginal fistula and hydronephrosis,underscoring the continuing impact of cervical cancer on quality of *** In this case report,we highlight the effectiveness of CCRT in achieving CR,even in cases of bulky cervical cancer,with adaptive RT offering a customized strategy to improve patient *** also emphasize the necessity for multidisciplinary team discussions and highlight the need for strategies to mitigate treatment-related toxicities and long-term complications.

Allantoin attenuates deficits of behavioural and motor nerve conduction in an animal model of cisplatin-induced neurotoxicity in rats

Background: The aim of the present study was to evaluate the neuroprotective effect of allantoin in cisplatin‐induced toxicity in rats. Methods: Adult male Wistar rats weighing 160‐200 g were used. Neuropathy was induced by injecting cisplatin (2 mg/kg, ip, twice a week for 6 weeks) and the rats were concurrently treated with allantoin (200 and 400 mg/kg, po) for 8 weeks. At the end of the study, body weight and hemogram were measured. Behavioural tests were performed, including tests for cold and hot hyperalgesia, motor co‐ordination, locomotor activity, mechano‐tactile allodynia and mechanical hyperalgesia. The rats were then sacrificed and sciatic nerve conduction velocity was determined. The antioxidant enzyme and nitric oxide levels in sciatic nerve homogenates were measured. Results: In this study, allantoin restored the motor nerve conduction velocity deficits induced by cisplatin, and the allantoin‐treated rats showed improvement in cold and thermal hyperalgesia, mechano‐tactile allodynia, and mechanical hyperalgesia. Allantoin treatment also improved the rats’ hematological status, increasing haemoglobin, platelet and RBC counts compared to the cisplatin‐treated group. Allantoin treatment also mitigated the functional abnormalities seen in the cisplatin neuropathy group, protecting neurons from the neurotoxic effects of cisplatin. Conclusion: Allantoin shows promise for use as an adjuvant drug in cancer treatment to protect against cisplatin‐induced neuropathy.

Multi-objective energy management of island microgrids with D-FACTS devices considering clean energy, storage systems and electric vehicles

The development of microgrids is progressing due to intelligent load demands,clean energy,batteries and electric *** presence of such systems in microgrids causes power balance inconsistency,leading to increased power losses and deviation in *** this pa-per,a mixed-integer non-linear programming model is proposed for modelling island microgrid energy management considering smart loads,clean energy resources,electric vehicles and ***,a flexible distributed AC transmission system device is proposed to prevent voltage deviation and reduce power losses.A scenario-based multi-objective function has been proposed to decrease energy losses and voltage deviations and energy outages of clean energy resources,reduce emissions from fossil-fired distributed generation and finally decrease load outages to reduce the vulnerability of the islanded *** the proposed mixed-integer non-linear model and the high number of variables and constraints,a modified evolutionary algorithm based on particle swarm optimization has been proposed to solve the proposed model,which can be more efficient than other algorithms to achieve global optimal *** model presented is implemented on a 33-node island microgrid and the results illustrate that the proposed algorithm and model are effective in reducing energy losses and voltage deviation,as well as reducing the vulnerability of the *** simulation results demonstrate that the proposed approach can lead to significant improvements in the performance of the ***,the ap-proach can result in a 27%reduction in losses,a 6%reduction in pollution and a 31%improvement in ***,the approach allows maximum utilization of renewable energy sources,making it a promising solution for sustainable energy management.

Coconut wood（Cocos nucifera） is used as alternative and complementary raw material for making house and furniture. Decorative style and material strength of coconut wood started gaining attention as an alternative material in the furniture industry. The needs of furniture industries to determine the quality of a good coconut wood to make good product quality, requiring control during the selection process until the end of the process material to be a ready-made product. Determining the level of quality（grading） visually for coconut wood needs to be created automatically, so that it can be used for the determination of material suitable for use as furniture or construction materials for buildings and reducing dependence on human grader. This research presents our experimental work on coconut wood quality classification using k-Nearest Neighbors（k-NN）, Discriminant Analysis（DA）, Na?ve Bayes（NB） and Decision Tree（DT）. The Gray-Level Co-occurrence Matrix（GLCM） is used to extract the texture features of coconut wood images. Minimum Redundancy Maximum Relevance（m RMR） feature selection is used to improve accuracy of classifier. Experiment result shows that Decision Tree（DT） based on GLCM + m RMR gives the best accuracy rate at 84.12%, which is slightly better than 81.80% of KNN2 based on GLCM.

Naturally-occurring and artificially-induced polyploids have been documented in various fish species but to date no comparison has been reported of the impacts of ploidy on fish biomarker responses to organic *** study describes effects of ploidy,gender,and dose on biliary fluorescent aromatic compound（FAC） concentrations,hepatic ethoxyresorufin-O-deethylase（EROD） and glutathione S-transferase（GST） activities in one of the most commonly cultured warm-water species,the African catfish Clarias *** matured male and female diploid and triploid fish were intraperitoneally（i.p.）injected with 0,5 or 25 mg/kg benzo[a]pyrene（BaP） and liver and gallbladder were sampled48 hr *** significant differences were found between ploidies in bile concentrations of7,8 dihydrodiolbenzo[a]pyrene（7,8D BaP）,1-hydroxybenzo[a]pyrene（1-OH BaP） or3-hydroxybenzo[a]pyrene（3-OH BaP）.However,concentrations of the biliary FACs did differ between males and females at different dose of injection with generally higher concentrations in females at the low dose of BaP and higher concentrations in males at the higher BaP *** EROD activity did not exhibit gender-dependent difference,whereas it was significantly higher in triploids than *** activities were not significantly influenced by any of the tested *** work advanced our understanding of the role of ploidy,gender,and dose in biotransformation of pollutants in fish.

Tripartite Entanglement Measures of Generalized GHZ State in Uniform Acceleration

Using the single-mode approximation,we study entanglement measures including two independent quantities;i.e.,negativity and von Neumann entropy for a tripartite generalized Greenberger-Horne-Zeilinger(GHZ)state in noninertial *** on the calculated negativity,we study the whole entanglement measures named as the algebraic average π3-tangle and geometric average Π*** find that the difference between them is very small or disappears with the increase of the number of accelerated *** entanglement properties are discussed from one accelerated observer and others remaining stationary to all three accelerated *** results show that there will always exist entanglement,even if acceleration r arrives to *** degree of entanglement for all 1-1 tangles are *** equal to zero,but 1-2 tangles always decrease with the acceleration parameter *** notice that the von Neumann entropy increases with the number of the accelerated observers and SκΙζΙ(κ,ζ∈(A,B,C)) first increases and then decreases with the acceleration parameter *** implies that the subsystem ρκΙζΙ is first more disorder and then the disorder will be reduced as the acceleration parameter r ***,it is found that the von Neumann entropies SABCI,SABICI and SAIBICI always decrease with the controllable angle θ,while the entropies of the bipartite subsystems S2-2non(two accelerated qubits),S2-1non(one accelerated qubit) and S2-0non(without accelerated qubit) first increase with the angle θ and then decrease with it.

Recently,a trust system was introduced to enhance security and cooperation between nodes in wireless sensor networks(WSN).In routing,the trust system includes or avoids nodes related to the estimated trust values in the routing *** article introduces Enhanced Metaheuristics with Trust Aware Secure Route Selection Protocol(EMTA-SRSP)for *** presented EMTA-SRSP technique majorly involves the optimal selection of routes in *** accomplish this,the EMTA-SRSP technique involves the design of an oppositional Aquila optimization algorithm to choose safe routes for data *** the clustering process,the nodes with maximum residual energy will be considered cluster heads(CHs).In addition,the OAOA technique gets executed to choose optimal routes based on objective functions with multiple parameters such as energy,distance,and trust *** experimental validation of the EMTA-SRSP technique is tested,and the results exhibited a better performance of the EMTA-SRSP technique over other approaches.
