Changes in the fungal and bacterial biomass and community structure in litter after the volcanic eruptions of Mount Usu, northern Japan were investigated using a chronosequence approach, which is widely used for analyzing vegetation succession. The vegetation changed from bare ground(10 years after the eruptions) with little plant cover and poor soil to monotonic grassland dominated by Polygonum sachalinense with undeveloped soil(33 years) and then to deciduous broad-leaved forest dominated by Populus maximowiczii with diverse species composition and well-developed soil(100 years). At three chronosequential sites, we evaluated the compositions of phospholipid fatty acids(PLFAs), carbon(C) and nitrogen(N) contents and the isotope ratios of C(δ13C) and N(δ15N) in the litter of two dominant species, Polygonum sachalinense and Populus maximowiczii. The C/N ratio, δ13C and δ15N in the litter of these two species were higher in the forest than that in the bare ground and grassland. The PLFAs gradually increased from the bare ground to the forest, showing that microbial biomass increased with the development of the soil and/or vegetation. The fungi-to-bacteria ratio of PLFA was constant at 5.3 ± 1.4 in all three sites, suggesting that fungi were predominant. A canonical correspondence analysis suggested that the PLFA composition was related tothe successional ages and the developing soil properties(P < 0.05, ANOSIM). The chronosequential analysis effectively detected the successional changes in both microbial and plant communities.

Effect of high-pressure homogenization on stability of emulsions containing zein and pectin

The aim of this study was to investigate the effect of high-pressure homogenization on the droplet size and physical stability of different formulations of pectin–zein stabilized rice bran oil emulsions. The obtained emulsions, both before and after passing through highpressure homogenizer, were subjected to stability test under environmental stress conditions,that is, temperature cycling at 4 °C/40 °C for 6 cycles and centrifugal test at 3000 rpm for 10 min. Applying high-pressure homogenization after mechanical homogenization caused only a small additional decrease in emulsion droplet size. The droplet size of emulsions was influenced by the type of pectin used;emulsions using high methoxy pectin(HMP) were smaller than that using low methoxy pectin(LMP). This is due to a greater emulsifying property of HMP than LMP. The emulsions stabilized by HMP–zein showed good physical stability with lower percent creaming index than those using LMP, both before and after passing through high-pressure homogenizer. The stability of emulsions after passing through high-pressure homogenizer was slightly higher when using higher zein concentration, resulting from stronger pectin–zein complexes that could rearrange and adsorb onto the emulsion droplets.

We investigated the delivery of drugs to the posterior segment of the eye by non-invasive topical instillation using submicron-sized poly(D,L-lactide-co-glycolide)(PLGA) nanoparticles(NPs). Surface-modified PLGA NPs were developed to improve the drug delivery efficiency to the retina and were administered as topical eye drops to mice. Chitosan(CS) and glycol chitosan(GCS), which are mucoadhesive polymers, and polysorbate 80(P80) were used as surface modifiers, and have been reported to increase the association of NPs with cells.Coumarin-6 was used as a model drug and fluorescent marker, and after ocular administration of PLGA NP eye drops, the fluorescence intensity of coumarin-6 was observed in the retina. The fluorescence image analysis indicated that there are several possible routes to the retina and fates of PLGA NPs in ocular tissue, and that these pathways involved the corneal,non-corneal, or uveal routes. Delivery to the mouse retina segments after topical administration was increased by surface modification with CS, GCS, or P80. Surface-modified PLGA NPs are a promising method for retinal drug delivery via topical instillation.

AIM To examine whether nizatidine stimulatesduodenal HCO3- secretion in rats by inhibitingAChE activity.METHODS Under pentobarbital anesthesia,aproximal duodenal loop was perfused withsaline,and the HCO3 secretion was measuredat pH 7.0 using a pH-stat method and by adding10mM HCI.Nizatidine,neostigmine,carbacholor famotidine was administered i.***.as a singleinjection.RESULTS Intravenous administration ofnizatidine（3-30 mg/kg）dose-dependentlyincreased duodenal HCO3- secretion,and theeffect at 10mg/kg was equivalent to thatobtained by carbachol at 0.01 mg/kg.Thisnizatidine action was observed at the same doserange that inhibited acid secretion and enhancedgastric motility,mimicked by i.***.injection ofneostigmine（0.03 mg/kg）,and significantlyattenuated by bilateral vagotomy and prior s.***.administration of atropine but not byindomethacin,a cyclooxygenase inhibitor,orNG-nitro-L-arginine methyl ester,a NO synthaseinhibitor.The HCO3- secretory response toacetylcholine（0.001 mg/kg）was significantlypotentiated by the concurrent administration ofnizatidine（3mg/kg,i.v.）.The IC50of nizatidine for AChE of rat erythrocytes was1.4×10-6M,about 12 times higher than that ofneostigmine.Neither famotidine(>10-3M,30mg/kg,i.v.)nor cisapride(> 10-3M,3mg/kg,i.v.)had any influence on AChEactivity or duodenal HCO3- secretion.Duodenaldamage induced by acid perfusion（100 mM HCIfor 4 h）in the presence of indomethacin wassignificantly prevented by nizatidine andneostigmine,at the doses that increased theHCO3- secretion.CONCLUSION Nizatidine stimulates duodenalHCO3- secretion,in both vagal-dependent andatropine-sensitive manners,and the action isassociated with the anti-AChE activity of thisagent.

Microscopic Analyses of Insoluble Particles in an Ice Core of ürümqi Glacier No. 1: Quantification of Mineral and Organic Particles

Insoluble particle concentration in ice cores is commonly analyzed as a proxy for varia- tions in atmospheric mineral dust （aerosol concentration）. However, recent studies have revealed that the mineral dust is not only a constituent of the particles but that biogenic organic particles are also contained. We microscopically analyzed insoluble particles in a shallow ice core drilled on a mountain glacier, the Urumqi Glacier No. 1, in eastern Tienshan, China. We distinguished different morphological particles in the ice core and quantified them separately. Results showed that the insoluble particles in this ice core consisted mainly of mineral particles, amorphous organic particles, pollen, and micro- organisms. Mineral particles were the most dominant, accounting for approximately 67% of total par- ticles, and amorphous organic particles were the second most dominant, accounting for approximately 33% of the total. The annual variation in the particles for the last 11 years differed between mineral and amorphous organic particles. The results suggest that the total insoluble particle concentration in the ice core reflects not only the atmospheric mineral dust but also the organic particles blown from ground soil or produced by microbes on the glacial surface.

We produced 5-μJ, 6.5-fs visible pulses at a repetition rate of 1 kHz using filamentation in a gas cell filled with krypton followed by spectral selection and phase compensation by a combination of dielectric mirrors. The visible pulses have a smooth spectrum from 520 to 650 nm with a shot-to-shot stability in each spectral component of better than 2%(standard deviation). This pulse compression scheme is simple and robust, and can be easily integrated into intense ultrashort-pulse laser systems.

AIM To determine the possibility that diabetes mellitus promotes pancreatic ductal adenocarcinoma via glyceraldehyde(GA)-derived advanced glycation-end products(GA-AGEs).METHODS PANC-1,a human pancreatic cancer cell line,was treated with 1-4 mmol/L GA for 24 h. The cell viability and intracellular GA-AGEs were measured by WST-8 assay and slot blotting. Moreover,immunostaining of PANC-1 cells with an anti-GA-AGE antibody was performed. Western blotting(WB) was used to analyze the molecular weight of GA-AGEs. Heat shock proteins 90α,90β,70,27 and cleaved caspase-3 were analyzed by WB. In addition,PANC-1 cells were treated with GA-AGEs-bovine serum albumin(GA-AGEs-BSA),as a model of extracellular GA-AGEs,and proliferation of PANC-1 cells was measured.RESULTS In PANC-1 cells,GA induced the production of GA-AGEs and cell death in a dose-dependent manner. PANC-1 cell viability was approximately 40% with a 2 mmol/L GA treatment and decreased to almost 0% with a 4 mmol/L GA treatment(each significant difference was P < 0.01). Cells treated with 2 and 4 mmol/L GA produced 6.4 and 21.2 μg/mg protein of GA-AGEs,respectively(P <0.05 and P < 0.01). The dose-dependent production of some high-molecular-weight(HMW) complexes of HSP90β,HSP70,and HSP27 was observed following administration of GA. We considered HMW complexes to be dimers and trimers with GA-AGEs-mediated aggregation. Cleaved caspase-3 could not be detected with WB. Furthermore,10 and 20 μg/m L GA-AGEs-BSA was 27% and 34% greater than that of control cells,respectively(P < 0.05 and P < 0.01).CONCLUSION Although intracellular GA-AGEs induce pancreatic cancer cell death,their secretion and release may promote the proliferation of other pancreatic cancer cells.

In this paper, a grid-based distributed hydrological model BTOPMC （Block-wise use of TOPMODEL） is introduced, which was developed from the original TOPMODEL. In order to broaden the model＇s application to arid regions, improvement methodology is also implemented. The canopy interception and soil infiltration processes were incorporated into the original BTOPMC to model event-based runoff simulation in large arid regions. One designed infiltration model with application of time compression approximation method is emphasized and validated for improving model＇s performance for event hydrological simulations with a case study of Lushi River basin.

As an optimal surgical procedure to accurately evaluate lymph node(LN)metastasis during surgery with minimal surgical resection,we have been developing sentinel node(SN)biopsy for early gastric cancer since the 1990s.Twelve institutions from the Japanese Society of Sentinel Node Navigation Surgery(SNNS),including Keio University Hospital,conducted a multicenter prospective trial to validate the SN concept using the dual-tracer method with blue dye and a radioisotope.According to the results,397 patients were included in the final analysis,and the overall accuracy in detecting LN metastasis using SN biopsy was 99%(383 of 387).Based on the validation study,we are targeting cT1N0 with a primary tumor of≤4 cm in diameter as an indication for SN biopsy for gastric cancer.We are currently running a multicenter nonrandomized phaseⅢtrial to assess the safety and efficacy of SN navigation surgery.The Korean group has reported the result of a multicenter randomized phaseⅢtrial.Since meticulous gastric cancer in the remnant stomach was rescued by subsequent gastrectomy,the disease-specific survival was comparable between the two techniques,implying that SN navigation surgery can be an alternative to standard gastrectomy.With the development of SN biopsy procedure and treatment modalities,the application of SN biopsy will be expanded to achieve an individualized minimally invasive surgery.

Objective:Concurrent chemoradiotherapy using cisplatin was thought to be standard treatment for squamous cell carcinoma of cervix,but it had not been effective for adenocarcinoma.Concurrent chemoradiotherapy using irinotecan hydrochloride(CPT-11)had been effective for colorectal cancer,thus,we chose CPT-11 as a candidate for gynecologic adenocarcinoma.To evaluate the maximum tolerated dose(MTD)of weekly CPT-11 with external pelvic radiotherapy,a phase 1/2 study was conducted according to modified Fibonacci method.Methods:Eligible patients were advanced uterine cancer with measurable diseases[performance score(PS):0-2].Study period was from August 1 st,2002 to December 31 st,2008.The starting dose level(DL)of CPT-11 was 30 mg/m2(DL1)given weekly for 4 weeks.Subsequently,dose escalation was scheduled in 10 mg/m2 increments to 60 mg/m^2(DL4).The fixed radiotherapy consisted of whole pelvic 1.8 Gy/d,once a day in weekday for five weeks and it amounted to 45 Gy(25 fractions)in total.Results:Seventeen patients were enrolled.As for toxicities,one(1/17:5.9%)grade(G)4 neutropenia lasting 7 days had been seen in DL4.G2 diarrhea was identified in 35.3%(6/17)of the patients,and 11.8%(2/17)G3 diarrhea was observed in DL3 and DL4.Thus,the MTD of CPT-11 was defined as dose of 60 mg/m^2.The recommended dose was decided as 50 mg/m^2.The response rate was 88.2%[9 complete response(CR),3 partial response(PR),3 stable disease(SD),2 not evaluable(NE)].Disease control rate at 1 month after treatment completion was 100%but distant metastases were found in 24%(4/17)in longer outcome.Conclusions:MTD was 60 mg/m^2 and recommended dose was set as 50 mg/m2.This concurrent chemoradiation using weekly CPT-11 was feasible at 50 mg/m^2,and it might be effective even in adenocarcinoma of the uterus.
