Genotoxicity of Water Extracts from Sewage Effluents in the Kanagawa Prefecture, Japan Using the Novel <i>umu</i>Tester Strain

The genotoxic activities of effluents from drainage water treatment plants were examined by using the novel umu tester strain NM8001, which lacks MutMst genes. To enhance the sensitivity of the LacZ assay, a BugBuster mix protein extraction reagent and TokyoGreen-β Gal for a fluorescence-galactosidase substrate were applied. Of the 24 sampling locations present in Kanagawa prefecture, Japan, water extracts from nine sampling points showed apparent genotoxic activities without metabolic activation. In contrast, water extracts from the upper sites of these water treatment plants did not show any significant genotoxic activities. The selected samples with genotoxic activity did not show significant mutagenicity toward Ames strains TA98 and TA100. Genotoxicity was also well correlated with the activity of a classical umu strain of TA1535/pSK1002;these findings indicate that the genotoxicity induced by oxidative damage was not a significant component of the genotoxicity.

Efficacy of the fat-dissociation method for nodal harvesting in gastric cancer

BACKGROUND There is an increased need for accurate staging for gastric cancer ***, it is necessary to carefully examine all dissected lymph nodes for precise staging. Recently, the fat-dissociation method has been developed as a quick and accurate method for harvesting dissected lymph nodes of colorectal cancer *** To investigate the usefulness of the fat-dissociation method for harvesting dissected lymph nodes of gastric cancer *** Fifty-six resected specimens from gastric cancer patients who underwent standard curative gastrectomy and lymph node dissection at our hospital were used. Group 2 lymph nodes were separated from each specimen, and the remaining adipose tissue containing the group 1 lymph nodes was used. Some resected specimens were subjected to the fat-dissociation method. One vial of Imofully~? was dissolved in 50 m L of saline and injected into the tissue. The tissue was incubated for 1 h and the dissolved fat was removed. Subsequently, the nodes were identified, picked up with scissors, and mapped. The number of nodes in each lymphatic compartment and duration of lymph node harvest and mapping were *** The fat-dissociation method was used for 24 samples, while the conventional dissection method was used for 32 samples. The total number of harvested lymph nodes was 45.9 in the fat dissociation group and 44.3 in the control group, and there was no significant difference between the two groups. There were also no significant differences in the number of lymph nodes between the two groups based on a comparison of the lymphatic compartments. However, the total median duration of the fat-dissociation method was 38.2 min, reflecting a reduced duration of approximately 60 min compared to the control *** Based on our results, the fat-dissociation method is effective in shortening the duration of lymph node harvest in gastric cancer surgery.

BACKGROUND Shear wave speed has been widely applied to quantify a degree of liver fibrosis. However, there is no standardized procedure, which makes it difficult to utilize the speed universally. AIM To provide procedural standardization of shear wave speed measurement. METHODS Point shear wave elastography (pSWE) was measured in 781 patients, and twodimensional shear wave elastography (2dSWE) was measured on the same day in 18 cases. Regions-of-interest were placed at 12 sites, and the median and robust coefficient-of-variation (CVR) were calculated. A residual sum-of-square (Σdi2) was computed for bootstrap values of 1000 iterations in 18 cases with each assumption of 1 to 12 measurements. The proportion of the Σdi2 (%Σdi2) was calculated as the ratio of Σdi2 to pSWE after converting it based on the correlation between pSWE and 2dSWE. RESULTS The CVR showed a significantly broader distribution in the left lobe (P < 0.0001),and the smallest CVR in the right anterior segment that covered 95% cases was 40.4%. pSWE was significantly higher in the left lobe than in the right lobe (1.63 ± 0.78 m/s vs 1.61 ± 0.78 m/s, P = 0.0004), and the difference between the lobes became further discrete when the subjects were limited to the cases with a CVR less than 40.4% in any segment (1.76 ± 0.80 m/s vs 1.70 ± 0.82 m/s, P < 0.0001). The highest values of the CVR in every 0.1 m/s interval were plotted in convex upward along pSWE and peaked at 1.93 m/s. pSWE and 2dSWE were significantly correlated (P < 0.0001, r = 0.95). In 216000 resamples from 18 cases, the %Σdi2 of 12 sites was 8.0% and gradually increased as the acquisition sites decreased to reach a significant difference with a %Σdi2 of 7 sites (P = 0.027). CONCLUSION These data suggest that shear wave speed should be measured at 8 or more sites of spreading in both lobes.

＜正＞ Recently, GPS （Global Positioning System） has been spreading rapidly. Especially, SPS （Standard Positioning Service） was improved in 2000 by releasing SA （Selective Availability）, and now it is paid attention from its confidence and usefulness. SPS is expected to be used various fields. First, we discovered that the position obtained by SPS shifts from true position. Then, we proposed the shift error reduction method using only satellites information （angle of elevation, angle of direction） from a GPS receiver. Next, we compared the result of simulation with that of positioning experiments with the index of an angle and a distance. As a result, we were able to forecast the direction of the shift and the amount of the shift and also were able to improve SPS precision.
