Metabolic pathways as possible therapeutic targets for progressive multiple sclerosis

Unlike relapsing remitting multiple sclerosis, there are very few therapeutic options for patients with progressive forms of multiple sclerosis. While immune mechanisms are key participants in the pathogenesis of relapsing remitting multiple sclerosis, the mechanisms underlying the development of progressive multiple sclerosis are less well understood. Putative mechanisms behind progressive multiple sclerosis have been put forth： insufficient energy production via mitochondrial dysfunction, activated microglia, iron accumulation, oxidative stress, activated astrocytes, Wallerian degeneration, apoptosis, etc. Furthermore, repair processes such as remyelination are incomplete. Experimental therapies that strive to improve metabolism within neurons and glia, e.g., oligodendrocytes, could act to counter inadequate energy supplies and/or support remyelination. Most experimental approaches have been examined as standalone interventions; however, it is apparent that the biochemical steps being targeted are part of larger pathways, which are further intertwined with other metabolic pathways. Thus, the potential benefits of a tested intervention, or of an established therapy, e.g., ocrelizumab, could be undermined by constraints on upstream and/or downstream steps. If correct, then this argues for a more comprehensive, multifaceted approach to therapy. Here we review experimental approaches to support neuronal and glial metabolism, and/or promote remyelination, which may have potential to lessen or delay progressive multiple sclerosis.

AIM To systematically review safety/efficacy of therapeutic endoscopic-retrograde-cholangiopancreatography(ERCP) performed during pregnancy, considering fetal viability, fetal teratogenicity, premature delivery, and future postpartum development of the *** systematic computerized literature search performed using PubMed with the key words "ERCP" and "pregnancy". Two clinicians independently reviewed the literature, and decided on which articles to incorporate in this review based on consensus and preassigned priorities. Large clinical trials, meta-analyses, systematic reviews, and controlled trials were assigned higher priority than review articles or small clinical series, and individual case reports were assigned lowest priority. Dr. Cappell has formal training and considerable experience in conducting systematic reviews, with 4 published systematic reviews in peer-reviewed journals indexed in PubM ed during the last 2 years, and with a PhD in neurophysiology that involved 5 years of training and research in biomedical *** Advances in imaging modalities, including abdominal ultrasound, MRCP, and endoscopic ultrasound, have generally obviated the need for diagnostic ERCP in nonpregnant and pregnant patients. Clinical experience with performing ERCP during pregnancy is burgeoning, with > 500 cases of therapeutic ERCP reported in the literature, aside from a national registry study of 58 patients. These studies show that therapeutic ERCP has a very high rate of technical success in clearing the bile duct of gallstones, and has a relatively low and acceptable rate of maternal and fetal complications. The great majority of births after therapeutic ERCP are full-term, have normal birth weights, and are healthy. Bluestem Gall Midge Annual Production Cycle and Effects on Grass seed Production

The bluestem gall midge (stenodiplosis wattsii Gagné) is native to the grasslands of North America. It feeds on the developing seeds of warm-season grasses during a portion of its lifecycle, but little is known of the biology and extent of gall midge infestations in native warm-season grasses in the UsA. We investigated the infestations of the bluestem gall midge in big bluestem (Andropogon gerardii Vitman), sand bluestem (Andropogon gerardii var paucipilus), little bluestem [schizachyrium scoparium (Michx.) Nash], and indiangrass [sorghastrum nutans (L.) Nash] commercial seed production fields during 2001 through 2004. Our objectives were to determine the annual production cycle of the bluestem gall midge, the range of the infestation in Nebraska, and estimate its impact on seed production. In eastern Nebraska, the midge goes through four generations with the fourth entering a larval diapause, overwintering in seeds and emerging in mid-June of the following year. The bluestem gall midge damaged from 31% of the little bluestem seed to 67% of the big bluestem seed, and, on average, reduced the production of viable seed by 54% across the four grass species in eastern Nebraska. Additionally, the warm-season prairie grasses were surveyed in nine other counties in Nebraska and the bluestem gall midge was found in each species surveyed from all nine counties. This study is the first to document the negative effects of the bluestem gall midge on the production of viable seed in sand bluestem and indiangrass.

The thermal and storage stability of bovine haemoglobin by ultraviolet-visible and circular dichroism spectroscopies

The effects of temperature,pH and long-term storage on the secondary structure and conformation changes of bovine haemoglobin（bHb） were studied using circular dichroism（CD） and ultraviolet-visible（UV-vis） *** network software was used to deconvolute the CD data to obtain the fractional content of the five secondary *** storage stability of bHb solutions in pH 6,7 and8 buffers was significantly higher at 4 ℃ than at 23 ℃ for the first 3 days.A complete denaturation of bHb was observed after 40 days irrespective of storage temperature or *** bHb solutions were also exposed to heating and cooling cycles between 25 and 65 ℃ and structural changes were followed by UVvis and CD *** experiments demonstrated that α-helix content of bHb decreased steadily with the increasing temperature above 35 ℃ at all pH *** loss in a-helicity and gain in random coil conformations was pH-dependent and the greatest under alkaline ***,there was minimal recovery of the secondary structure content upon cooling to 25 ℃.The use of bHb as a model drug is very common and this study elucidates the significance of storage and processing conditions on its stability.

An Awareness-Adoption Matrix for strategic Decision Making in Agricultural Development Projects: A Case study in Yunnan Province, China

significant achievements have been made in generating sustainable agricultural technologies in developing countries. Usually, these new technologies have been more effective in increasing production and productivity than existing technologies. However, many new technologies considered ‘effective’ have not been successful in alleviating the associated sustainability problems, due to poor adoption by targeted users. success of any novel technical intervention should be judged on the basis of how widespread adoption is in the target area by targeted users. In this context, a case study identified the factors affecting farmers’ adoption of improved technologies extended by an agricultural development project in a rural Village in Yunnan Province, China. Initial adoption/adaptation of project technologies was influenced by farmers’ awareness of the technology. Farmers were more aware of polythene mulch, contour cultivation, intercropping and tree planting technologies than others. This led to a comparatively high initial uptake of polythene mulch, contour cultivation, sweet chestnut, and intercropping technologies. Farmers had inadequate knowledge about some Project technologies, the adoption of which was particularly low. This reveals the need for increasing farmers’ awareness about the rationale for Project technologies to achieve greater adoption/adaptation of project technologies by farmers over wider areas. Farmers’ testing of the technology leads to better adoption/adaptation compared to mere awareness. However, considerable time and resources are required for testing/trying technologies. This justifies the usefulness of comparatively quick and less resource demanding options for awareness creation. Development of effective cropping technologies is important for sustainable agricultural development.
