Efficient in vivo delivery of anti-inflammatory proteins to modulate the microenvironment of an injured spinal cord and promote neuroprotection and functional recovery is a great challenge.Nucleoside-modified messenger RNA(mRNA)has become a promising new modality that can be utilized for the safe and efficient delivery of therapeutic proteins.Here,we used lipid nanoparticle(LNP)-encapsulated human interleukin-10(hIL-10)-encoding nucleoside-modified mRNA to induce neuroprotection and functional recovery following rat spinal cord contusion injury.Intralesional administration of hIL-10 mRNA-LNP to rats led to a remarkable reduction of the microglia/macrophage reaction in the injured spinal segment and induced significant functional recovery compared to controls.Furthermore,hIL-10 mRNA treatment induced increased expression in tissue inhibitor of matrix metalloproteinase 1 and ciliary neurotrophic factor levels in the affected spinal segment indicating a time-delayed secondary effect of IL-105 d after injection.Our results suggest that treatment with nucleoside-modified mRNAs encoding neuroprotective factors is an effective strategy for spinal cord injury repair.

Scavenging and release of REE and HFSE by Na-metasomatism in magmatic-hydrothermal systems

Exploitable or potentially exploitable deposits of critical metals,such as rare-earth(REE)and high-field-strength elements(HFSE),are commonly associated with alkaline or peralkaline igneous rocks.However,the origin,transport and concentration of these metals in peralkaline systems remains poorly understood.This study presents the results of a mineralogical and geochemical investigation of the Na-metasomatism of alkali amphiboles and clinopyroxenes from a barren peralkaline granite pluton in NE China,to assess the remobilization and redistribution of REE and HFSE during magmatic-hydrothermal evolution.Alkali amphiboles and aegirine-augites from the peralkaline granites show evolutionary trends from sodic-calcic to sodic compositions,with increasing REE and HFSE concentrations as a function of increasing Na-index[Na^(#),defined as molar Na/(Na+Ca)ratios].The Na-amphiboles(i.e.,arfvedsonite)and aegirine-augites can be subsequently altered,or breakdown,to form hydrothermal aegirine during late-or post-magmatic alteration.Representative compositions analyzed by insitu LA-ICPMS show that the primary aegirine-augites have high and variable REE(2194-3627 ppm)and HFSE(4194-16,862 ppm)contents,suggesting that these critical metals can be scavenged by alkali amphiboles and aegirine-augites.Compared to the primary aegirine-augites,the presentative early replacement aegirine(Aeg-I,Na^(#)=0.91-0.94)has notably lower REE(1484-1972)and HFSE(4351-5621)contents.In contrast,the late hydrothermal aegirine(Aeg-II,Na^(#)=0.92-0.96)has significantly lower REE(317-456 ppm)and HFSE(6.44-72.2 ppm)contents.Given that the increasing Na^(#)from aegirine-augites to hydrothermal aegirines likely resulted from Na-metasomatism,a scavenging-release model can explain the remobilization of REE and HFSE in peralkaline granitic systems.The scavenging and release of REE and HFSE by Na-metasomatism provides key insights into the genesis of globally significant REE and HFSE deposits.The high Na-index of the hydrothermal aegirine might be useful as a geo

Chronic Hepatitis b in Indian Americans: Lack of Screening and Poor Linkage to Care

background: Chronic hepatitis b (CHb) is a major cause of liver-related morbidity and mortality in the United States (US) and globally. CHb disproportionately affects Asian Americans and many other immigrant minority populations, primarily owing to the high prevalence of CHb in their countries of origin. India is a country with a medium-to-high prevalence of hepatitis b (Hb) (>2%) and has over 40 million people infected with hepatitis b virus (HbV), with more than 115,000 deaths annually from HbV-related complications. Indian Americans are one of the largest immigrant populations in the US but remain underdiagnosed and poorly linked to clinical care. We, therefore, assessed the HbV prevalence and evaluated the linkage-to-care (LTC) among Indian Americans to develop strategic plans to reduce the impact of HbV in the US. Methods: between April 2022 and January 2024, serologic screening and surveys were provided to 328 Indian American adults (age 20 - 80) in New York City. All participants were tested for a triple panel consisting of hepatitis b surface antigen (HbsAg), hepatitis b surface antibody (anti-Hbs), and hepatitis b core IgG antibody (anti-Hbc). A survey was conducted on the subjects chronically infected with HbV regarding their histories of infection. Self-administered questionnaires were employed to evaluate demographic and epidemiologic characteristics. Results: Of 328 screened and evaluated (246 males and 82 females), 10 (3.0%) were HbV-infected, 222 (67.7%) were susceptible to HbV, and 96 (29.3%) were immune. The prevalence of chronic HbV varied between the age groups: 4.6% (age 20 - 40), 3.4% (age 41 - 60), and 1.7% (age 61 - 80). Of 10 chronically infected, only two subjects had been previously diagnosed but were not engaged in care. Conclusion: HbV disproportionately affects Asian Americans, primarily owing to immigration from parts of the world where the disease is endemic. Adding Value to Crop Production Systems by Integrating Forage Cover Crop Grazing

In addition to their value as cereal grains, wheat (Triticum aestivum L.) and triticale (× Triticosecale Wittmack) are important cool-season annual forages and cover crops. Yearling steer (bos taurus) performance was compared in the spring following autumn establishment as for age cover crops after soybean [Glycine max (L.) Merr.] grain harvest. Replicated pastures (0.4 ha) were no-till seeded in three consecutive years into soybean stubble in autumn, fertilized, and grazed the following spring near Ithaca, NE, USA. Each pasture (n = 3) was continuously stocked in spring with four yearling steers (380 ± 38 kg) for 17, 32, and 28 d in 2005, 2006, and 2007, respectively. In 2005, average daily gain (ADG) for steers grazing triticale exceeded the ADG for wheat by 0.31 kghd-1d-1. In 2006, wheat ADG exceeded that for triticale by 0.12 kghd-1d-1. In 2007, steers grazing wheat lost weight, while steers grazing triticale gained 0.20 kghd-1d-1. based on the 3-year average animal gains valued at $1.32 kg-1, mean net return ($ ha-1 yr-1) was $62.15 for triticale and $22.55 for wheat. Since these grazed cover crops provide ecosystem services in addition to forage, grazing could be viewed as a mechanism for recovering costs and adds additional value to the system. based on this 3-year grazing trial, triticale was superior to wheat and likely will provide the most stable beef yearling performance across years with variable weather for the western Cornbelt USA.

C-type lectins(CTLs)represent a large family of soluble and membrane-bound proteins which bind calcium dependently via carbohydrate recognition domains(CRDs)to glycan residues presented on the surface of a variety of pathogens.The deconvolution of a cell’s glycan code by CTLs underpins several important physiological processes in mammals such as pathogen neutralization and opsonization,leukocyte trafficking,and the inflammatory response.However,as our knowledge of CTLs has developed it has become apparent that the role of this innate immune family of proteins can be double-edged,where some pathogens have developed approaches to subvert and exploit CTL interactions to promote infection and sustain the pathological state.Equally,CTL interactions with host glycoproteins can contribute to inflammatory diseases such as arthritis and cancer whereby,in certain contexts,they exacerbate inflammation and drive malignant progression.This review discusses the‘dual agent’roles of some of the major mammalian CTLs in both resolving and promoting infection,inflammation and inflammatory disease and highlights opportunities and emerging approaches for their therapeutic modulation.

Extensive adhesion formation in a total knee replacement in the setting of a gastrointestinal stromal tumor:A case report

bACKGROUND Gastrointestinal stromal tumors(GISTs)are rare primary neoplasms of the gastrointestinal tract,accounting for 1%to 2%of all gastrointestinal neoplasms worldwide.GISTs are frequently discovered incidentally during workup for other diagnosis or intestinal obstruction,as they can present with few or no symptoms.Simultaneously,GISTs confer a high degree of malignant transformation,with a progression in about 10%to 30%of cases.CASE SUMMARY A 63-year-old healthy female presented to our institution with complaints of right knee pain and limited passive and active motion in the setting of a previous right total knee arthroplasty(TKA).One year after TKA,the patient was incidentally diagnosed with a GIST,which was successfully removed.After removal,the patient continued to have limited range of motion of the right knee and subsequently underwent revision TKA.Intraoperatively significant fibrotic adhesions were found encapsulating the femoral and tibial components.The patient’s pain improved postoperatively,however,she continued to have decreased range of motion with difficulty ambulating.CONCLUSION We propose that this case may demonstrate a proinflammatory milieu arising from a GIST,which had a direct influence on the outcome of recent total knee arthroplasty.This proposed mechanism between neoplastic cytokinetic activity and adhesion formation could have implications on preoperative and postoperative orthopedic management of total knee arthroplasty.

Landscape Genetics of Plants: Challenges and Opportunities

Dispersal is one of the most important but least understood processes in plant ecology and evolutionary biology.Dispersal of seeds maintains and establishes populations,and pollen and seed dispersal are responsible for gene flow within and among populations.Traditional views of dispersal and gene flow assume models that are governed solely by geographic distance and do not account for variation in dispersal vector behavior in response to heterogenous landscapes.Landscape genetics integrates population genetics with Geographic Information Systems(GIS)to evaluate the effects of landscape features on gene flow patterns(effective dispersal).Surprisingly,relatively few landscape genetic studies have been conducted on plants.Plants present advantages because their populations are stationary,allowing more reliable estimates of the effects of landscape features on effective dispersal rates.On the other hand,plant dispersal is intrinsically complex because it depends on the habitat preferences of the plant and its pollen and seed dispersal vectors.We discuss strategies to assess the separate contributions of pollen and seed movement to effective dispersal and to delineate the effects of plant habitat quality from those of landscape features that affect vector behavior.Preliminary analyses of seed dispersal for three species indicate that isolation by landscape resistance is a better predictor of the rates and patterns of dispersal than geographic distance.Rates of effective dispersal are lower in areas of high plant habitat quality,which may be due to the effects of the shape of the dispersal kernel or to movement behaviors of biotic vectors.Landscape genetic studies in plants have the potential to provide novel insights into the process of gene flow among populations and to improve our understanding of the behavior of biotic and abiotic dispersal vectors in response to heterogeneous landscapes.

The superτ-charm facility(STCF)is an electron–positron collider proposed by the Chinese particle physics community.It is designed to operate in a center-of-mass energy range from 2 to 7 GeV with a peak luminosity of 0.5×10^(35) cm^(–2)·s^(–1) or higher.The STCF will produce a data sample about a factor of 100 larger than that of the presentτ-charm factory—the bEPCII,providing a unique platform for exploring the asymmetry of matter-antimatter(charge-parity violation),in-depth studies of the internal structure of hadrons and the nature of non-perturbative strong interactions,as well as searching for exotic hadrons and physics beyond the Standard Model.The STCF project in China is under development with an extensive R&D program.This document presents the physics opportunities at the STCF,describes conceptual designs of the STCF detector system,and discusses future plans for detector R&D and physics case studies.

Influence of towing speed on effectiveness of rolling dynamic compaction

The influence of towing speed on the effectiveness of the 4-sided impact roller using earth pressure cells(EPCs)is investigated.Two field trials were undertaken;the first trial used three EPCs placed at varying depths between 0.5 m and 1.5 m with towing speeds of 9-12 km/h.The second used three EPCs placed at a uniform depth of 0.8 m,with towing speeds of 5-15 km/h.The findings from the two trials confirmed that towing speed influences the pressure imparted to the ground and hence compactive effort.This paper proposes that the energy imparted to the ground is best described in terms of work done,which is the sum of the change in both potential and kinetic energies.Current practice of using either kinetic energy or gravitational potential energy should be avoided as neither can accurately quantify rolling dynamic compaction(RDC)when towing speed is varied.

Evasion of apoptosis is a hallmark of cancer,attributed in part to overexpression of the antiapoptotic protein b-cell lymphoma 2(bcl-2).In a variety of cancer types,including lymphoma,bcl-2 is overexpressed.Therapeutic targeting of bcl-2 has demonstrated efficacy in the clinic and is the subject of extensive clinical testing in combination with chemotherapy.Therefore,the development of co-delivery systems for bcl-2 targeting agents,such as small interfering RNA(siRNA),and chemotherapeutics,such as doxorubicin(DOX),holds promise for enabling combination cancer therapies.Lipid nanoparticles(LNPs)are a clinically advanced nucleic acid delivery system with a compact structure suitable for siRNA encapsulation and delivery.Inspired by ongoing clinical trials of albumin-hitchhiking doxorubicin prodrugs,here we developed a DOX-siRNA co-delivery strategy via conjugation of doxorubicin to the surface of siRNAloaded LNPs.Our optimized LNPs enabled potent knockdown of bcl-2 and efficient delivery of DOX into the nucleus of burkitts'lymphoma(Raji)cells,leading to effective inhibition of tumor growth in a mouse model of lymphoma.based on these results,our LNPs may provide a platform for the co-delivery of various nucleic acids and DOX for the development of new combination cancer therapies.
