A reversed-phase high performance liquid chromatography (HPLC) method was developed, validated, and used for the quantitative determination of gatifloxacin (GA) and ambroxol hydrochloride (AM), from its tablet dosage form. Chromatographic separation was performed on a HiQ Sil C18 column (250 mm×4.6 mm, 5 μm), with a mobile phase comprising of a mixture of 0.01 mol/L potassium dihydrogen orthophosphate buffer and acetonitrile (70∶30, v/v), and pH adjusted to 3 with orthophosphoric acid, at a flow rate of 1 mL/min, with detection at 247 nm. Separation was completed in less than 10 min. As per International Conference on Harmonisation (ICH) guidelines the method was validated for linearity, accuracy, precision, limit of quantitation, limit of detection, and robustness. Linearity of GA was found to be in the range of 10-60 μg/mL and that for AM was found to be 5-30 μg/mL. The correlation coefficients were 0.999 6 and 0.999 3 for GA and AM respectively. The results of the tablet analysis (n=5) were found to be 99.94% with ±0.25% standard deviation (SD) and 99.98% with±0.36% SD for GA and AM respectively. Percent recovery of GA was found to be 99.92%-100.02% and that of AM was 99.86%-100.16%. The assay experiment shows that the method is free from interference of excipients. This demonstrates that the developed HPLC method is simple, linear, precise, and accurate, and can be conveniently adopted for the routine quality control analysis of the tablet.

In the privacy preservation of association rules, sensitivity analysis should be reported after the quantification of items in terms of their occurrence. The traditional methodologies, used for preserving confidentiality of association rules, are based on the assumptions while safeguarding susceptible information rather than recognition of insightful items. Therefore, it is time to go one step ahead in order to remove such assumptions in the protection of responsive information especially in XML association rule mining. Thus, we focus on this central and highly researched area in terms of generating XML association rule mining without arguing on the disclosure risks involvement in such mining process. Hence, we described the identification of susceptible items in order to hide the confidential information through a supervised learning technique. These susceptible items show the high dependency on other items that are measured in terms of statistical significance with Bayesian Network. Thus, we proposed two methodologies based on items probabilistic occurrence and mode of items. Additionally, all this information is modeled and named PPDM (Privacy Preservation in Data Mining) model for XARs. Furthermore, the PPDM model is helpful for sharing markets information among competitors with a lower chance of generating monopoly. Finally, PPDM model introduces great accuracy in computing sensitivity of items and opens new dimensions to the academia for the standardization of such NP-hard problems.

Vasospasm as a Complication after Aneurysmal Rupture and Its Relation with Surgical Clipping and Endovascular Coiling among a Georgian Sample

Background: Potentially lethal, aneurysmal subarachnoid hemorrhage has a bad prognosis for many individuals. Over the past few decades, endovascular and surgical interventions have been developed, including surgical clipping, and endovascular coiling. Patients who have aSAH are also susceptible to delayed cerebral ischemia and cerebral vasospasm. The aim of this study is to compare the outcome of endovascular coiling with surgical clipping in patients with SAH, specifically in relation to prevalence of vasospasm, in the country of Georgia. Method: In this study, we present a retrospective review of the outcomes of 217 patients with acute subarachnoid hemorrhage who underwent endovascular coiling or surgical clipping. The data were gathered from patients who are admitted to New Vision University Hospital and Caucasus Medical Center in Tbilisi, Georgia, between 2017 and 2022. Results: Vasospasm was prevalent in 217 of the patients who had aneurysmal rupture when they first appeared. Endovascular coiling or surgical clipping was used to treat aneurysmal rupture. In our sample, 24.81 percent of patients who underwent coiling experienced vasospasm after 14 days, compared to 31.25 percent of patients who underwent clipping. After endovascular coiling and surgical clipping, the severity of vasospasm was only slightly different, according to Lindegaard ratios. Finally, 32 patients (23.35 percent) died after coiling whereas 55 patients (68.75 percent) died within three decades of clipping. Conclusion: After 5 years of data collection, this study has demonstrated the most favorable option for treatment is endovascular coiling. However, the treatment choice takes multiple factors into account, and clipping is not ideal for some ruptured aneurysms. Despite the fact that endovascular coiling is usually successful and minimally invasive, complications can occur and additional monitoring and potential surgical intervention are indicated.

The pivotal aim of this study is to evaluate the rock mass characterization and deformation modulus. It is vital for rock mass classification to investigate important parameters of discontinuities. Therefore, Rock Mass Rating (RMR) and Tunneling quality index (Q) classification systems are applied to analyze 22 segments along proposed tunnel routes for hydropower in Kandiah valley, Khyber Pakhtunkhwa, Pakistan. RMR revealed the range of fair to good quality rocks, whereas Q yielded poor to fair quality rocks for investigated segments of the rock mass. Besides, Em values were acquired by empirical equations and computer-aided program RocLab, and both methods presented almost similar variation trend of their results. Hence, the correlations of Em with Q and RMR were carried out with higher values of the regression coefficient. This study has scientific significance to initially understand the rock mass conditions of Kandiah valley.

Screening and Statistical Optimization of Physiochemical Parameters for the Production of Xylanases from Agro-Industrial Wastes

Xylanases are mostly produced through submerged fermentation;nonetheless solid-state fermentation has increased profound attention and consideration of scholars having high conversion level biomass to energy conservation. This study depicted the purification of xylanases and their possible utilization in industry. The present study was carried out to examine the culture influence of fungal strain Fomes fomentarius (F. fomentarius) using different agro-industrial residues (wheat straw, rice husk, sugarcane bagasse and siris pods). F. fomentarius showed maximum enzyme production after 72 h of fermentation, when grown on wheat straw in solid state fermentation process while maximum activity showed on pH 6.0 at 30°C. The other parameters optimized by statistical design (RSM) showed maximum xylanase activity (146 ± 8 IU/mL) at 65% moisture content, 4 mL inoculums size, 175 mg Ammonium sulphate, 200 mg Calcium carbonate and 1.4 grams of glucose. Xylanase was salted out at 60% ammonium sulphate concentration and enzyme was further purified by Sephadex G-100 gel filtration chromatography with 2.2 fold increase in activity. The purified xylanase from F. fomentarius had optimum pH 6.0 and 40°C. Xylanase showed higher specificity for oat spelt xylan with kinetic constants Km 1.25 mg/mL and Vmax 54 mM/min. Xylanases have an industrial important enzyme used extensively in food, feed and paper industry.

Objective: Artemisia annua is the chief source of artemisinin, a potent antimalarial agent, in which other bioactive phytochemicals are also present. Due to low levels of bioactive compounds including artemisinin and flavonoids, it is necessary to increase the level of the secondary metabolites by regulating the expression of rol genes in the ***: A hybrid variety of A. annua(Hyb1209 r, Shennong) developed by the Centre for Novel Agricultural Products, University of York, UK, was selected to produce transgenics of rolB and rolC genes. Genetic transformation was carried out via Agrobacterium tumefaciens GV3101 harboring rolB and rolC genes of Agrobacterium rhizogenes cloned separately. HPLC was used for the qualitative and quantitative analysis of flavonoids and artemisinin. Furthermore, thin layer chromatography(TLC) was also used to analyze artemisinin ***: Comparative analysis via HPLC revealed considerable enhancement in the phytochemical content of transgenic A. annua plants as compared to the wild type plant. Transgenics of rolB gene showed an average increase of 321% in rutin, 97.2% in caffeic acid, and 218.4% in myricetin, respectively. In the case of rolC gene transgenics, an average increase of 197.5% in rutin, 76.3% in caffeic acid, and 209.3%in myricetin was observed. Transgenics of rolB and rolC genes showed a 14.3%–28.6% and 2.8%–12.7% increase in artemisinin content respectively by HPLC analysis. TLC analysis showed that an average 142.2%and 110.2% enhancement in artemisinin for rolB and rolC transgenics respectively, compared with the wild type. An enhanced production of total flavonoids(average 30.2% and 25.5% increase in rolB and rolC transgenics, respectively) and total phenolics(average 34.3% and 25.8% increase in rolB and rolC transgenics, respectively) was observed as a result of transformation. This study illustrates the classification of the rock mass and evaluation of rock squeezing, rock burst potential, deformation modulus along the proposed tunnel alignment of small hydropower in Swat Valley, Khyber Pakhtunkhwa (KP), Pakistan. The field and laboratory studies were conducted to classify the rock mass by using geomechanical classification systems i.e. Rock Mass Rating (RMR), tunneling quality index (Q), Rock Mass Index (RMi). The empirical relations classified the ground as non-squeezing and minor to non-squeezing conditions, respectively. Whereas, other methods depict minor to medium bursting potential along chainage 1+000 to 4+000 m, while results along chainage 2+400 - 2+800 m present medium to high bursting potential. Furthermore, numerical analyses were carried out by RS3 for elastic and plastic conditions in order to assess the total displacement of each section in unsupported and supported conditions. The results gave maximum displacement along chainage 2+400 - 2+800 m (19.2 mm in unsupported and 16mm in supported condition) and minimum displacement along chainage 0+876 - 1+000 m (1.4 mm in unsupported and 1.3 mm in supported condition). Hence, the estimated support by empirical methods has been optimized by using numerical analyses for the stability of rock mass along the tunnel.
