clobenzorex is an anorexigenic drug that is widely used in Mexicofor the treatment of obesity, since it helps to reduce body weight. This drug is available as immediate release capsules. To improve compliance to treatment, it was developed a new slow release formulation. In order to establish its usefulness, oral pharmacokinetics and weight reduction of slow release and immediate release formulations of clobenzorex in obese patients were compared. Sixty patients with a BMI higher than 27 kg/m2 were included in the study. Two groups of 30 patients were formed, one of them received 30 mg immediate release formulation b.i.d. and the other group received one 60 mg slow release formulation once a day, since under this scheme these formulations are prescribed. Blood samples were obtained at selected times during the first day and once weekly during 4 weeks. After the last dose, samples were obtained at selected times during 48 h. plasma levels were determined by HpLc-MS/MS and pharmacokinetic parameters were obtained. Reduction in cmax due to increased tmax, as well as, increased half-life were observed with the slow release formulation in comparison with immediate release formulation. Although lower plasma levels of clobenzorex were reached with the slow release formulation, reduction of body weight was similar with both products. Based on the results, it was concluded that slow release formulation of clobenzorex is an adequate formulation of clobenzorex, since pharmacokinetics and effects observed are compatible with a once a day administration.

Evaluation of the Influence of Hyoscine Butylbromide on the Oral Bioavailability of Lansoprazole in Healthy Adult Volunteers

The gastroesophageal reflux and/or peptic ulcer diseases are clinical conditions that occur usually accompanied of symptomatic pain. Lansoprazole, a proton pump inhibitor class drug is widely used in clinical practice for treatment of these diseases. However, its efficacy can be improved by combining with spasmolytic and/or visceral analgesic such as hyoscine butylbromide. Since hyoscine butylbromide is barely absorbed and exerts some local effects at gastrointestinal tract which may modify the absorption of lansoprazole, it is important to establish if there is a pharmacokinetic interaction after the oral concomitant administration of both drugs. For this objective, twenty-five subjects received under a crossover design an oral administration of lansoprazole (15 mg) plus placebo or a fixed-dose combination with hyoscine butiylbromide (15 mg + 10 mg, respectively). plasma samples were obtained at different times during 10 hours. Lansoprazole plasma concentrations were determined by a high performance liquid chromatography method coupled to tandem mass spectrometry. Fixed-dose combination was well tolerated. Lansoprazole pharmacokinetic parameters were: cmax 621.81 ± 212.79 and 450.38 ± 192.14 ng/mL;AUc0-t 1941.36 ± 845.57 and 1454.66 ± 757.28 ng·h/mL;tmax 2.83 ± 0.99 and 3.40 ± 1.82h;t1/2 1.35 ± 0.39 and 1.45 ± 0.51 h, for alone and combined fixed-dose formulation, respectively. pharmacokinetic parameters were compared by analysis of variance and ratios of AUc0-t, cmax and 90% confidence intervals obtained. Since confidence intervals exceed the 80% - 125% limits for these parameters, we conclude that there is a significantly pharmacokinetic interaction of lansoprazole when it is administered concomitantly with hyoscine butylbromide.

Epidemiology of hepatitis c virus infection

Globally, hepatitis c virus (HcV) has infected an estimated 130 million people, most of whom are chronically infected. HcV-infected people serve as a reservoir for transmission to others and are at risk for developing chronic liver disease, cirrhosis, and primary hepatocellular carcinoma (Hcc). It has been estimated that HcV accounts for 27% of cirrhosis and 25% of Hcc worldwide. HcV infection has likely been endemic in many populations for centuries. However, the wave of increased HcV-related morbidity and mortality that we are now facing is the result of an unprecedented increase in the spread of HcV during the 20th century. Two 20th century events appear to be responsible for this increase; the widespread availability of injectable therapies and the illicit use of injectable drugs.

The development of artificial intelligence(AI)and smart home technologies has driven the need for speech recognition-based *** demand stems from the quest for more intuitive and natural interaction between users and smart devices in their *** recognition allows users to control devices and perform everyday actions through spoken commands,eliminating the need for physical interfaces or touch screens and enabling specific tasks such as turning on or off the light,heating,or lowering the *** purpose of this study is to develop a speech-based classification model for recognizing human actions in the smart *** seeks to demonstrate the effectiveness and feasibility of using machine learning techniques in predicting categories,subcategories,and actions from sentences.A dataset labeled with relevant information about categories,subcategories,and actions related to human actions in the smart home is *** methodology uses machine learning techniques implemented in python,extracting features using countVectorizer to convert sentences into numerical *** results show that the classification model is able to accurately predict categories,subcategories,and actions based on sentences,with 82.99%accuracy for category,76.19%accuracy for subcategory,and 90.28%accuracy for *** study concludes that using machine learning techniques is effective for recognizing and classifying human actions in the smart home,supporting its feasibility in various scenarios and opening new possibilities for advanced natural language processing systems in the field of AI and smart homes.

Mirodenafil is a 5-phosphodiesterase inhibitor that is currently marketed in Korea for the treatment of erectile dysfunction;however, no information in other populations is available. It has been described that Mirodenafil is metabolized by cYp3A4, a metabolic pathway in which interethnic differences have been reported. The purpose of this study was to characterize the oral pharmacokinetics of Mirodenafil in Mexicans. Seventeen male healthy volunteers were enrolled in this study. After an overnight fast, volunteers received an oral 100 mg dose and blood samples were collected at selected times during 24 h. plasma was stored frozen and analyzed by an HpLc method. pharmacokinetic parameters obtained were: cmax 331.129 ± 32.689 ng/mL, tmax 1.574 ± 0.293 h, AUc24h 883.293 ± 104.088 ng·h/mL, AUc￥ 976.477 ± 108.812 ng·h/mL and t1/2 1.807 ± 0.171 h. parameter values observed in this study are similar to those reported in Koreans. Since efficacy and safety studies of Mirodenafil have been conducted in Koreans, it is expected that dosage regime to employ in Mexicans should be similar to the approved for Korean population.

Background:Despite efficacy in HcV eradication,direct-acting antiviral(DAA)therapy has raised controversies around their impact on hepatocellular carcinoma(Hcc)*** we reported the first Australian data on Hcc incidence in DAA-treated HcV patients with advanced fibrosis/***:We conducted a retrospective single center study of DAA-treated HcV patients with advanced fibrosis/cirrhosis from April 2015 to December *** with prior Hcc were included if they had complete response to Hcc ***:Among 138 patients who completed DAA therapy,133(96.4%)achieved sustained virologic response(median follow-up 23.8 months).Ten had prior Hcc and 5/10(50.0%)developed recurrence,while de novo Hcc developed in 7/128(5.5%).Median time from DAA to Hcc diagnosis was 34 weeks in recurrent Hcc *** novo 52 weeks(p=0.159).In patients with prior Hcc,those with recurrence(***)had shorter median time between last Hcc treatment and DAA(12 vs.164 weeks,pc response at 12 weeks(SVR12)(p=0.011),platelets(p=0.005),model for end-stage liver disease(MELD)score(p=0.029),alpha fetoprotein(AFp)(p=0.013),and prior Hcc(pciated with Hcc *** multivariate analysis,significant factors were prior Hcc(OR=4.80;95%cI:1.47–48.50;p=0.010),failed SVR12(OR=2.83;95%cI:1.71–16.30;p=0.016)and platelets(OR=0.97;95%cI:0.95–0.99;p=0.009).conclusions:Our study demonstrates a high incidence of recurrent Hcc in HcV patients with advanced fibrosis/cirrhosis treated with *** associated with Hcc development post-DAA were more advanced liver disease,failed SVR12 and prior Hcc,with higher rates of recurrence in those who started DAA earlier.

The gluten proteins of wheat grain are responsible for visco-elastic properties of flour,but they also trigger the immune-response of celiac *** this work,two low-gliadin RNA interference(RNAi)wheat lines that differ for the promoter driving the silencing(D-hordein andγ-gliadin promoters for D783 and D793 lines,respectively),were characterized at transcriptomic,and protein fraction levels in the *** of gliadins provides a readjustment in the grain protein fractions that also affects to the nongluten proteins(NGp),which were increased in both RNAi *** of wheat gluten by means of the R5 monoclonal antibody also showed a strong reduction in the content of gluten in both RNAi ***,fructans,an oligosaccharide linked with the development of non-celiac wheat sensitivity(NcWS)were also significantly decreased in RNAi *** down-regulation of gliadins fractions also impacts to other metabolic processes,particularly on carbohydrate metabolism,enzyme regulator activity and response to *** and transcription factors regulated by ABA were up-regulated,which could suggest the implication of this phytohormone on the stress response observed in the RNAi lines.

An Ontology Based cyclone Tracks classification Using SWRL Reasoning and SVM

Tropical cyclones(Tc)are often associated with severe weather conditions which cause great losses to lives and *** precise classification of cyclone tracks is significantly important in thefield of weather *** this paper we propose a novel hybrid model that integrates ontology and Support Vector Machine(SVM)to classify the tropical cyclone tracks into four types of classes namely straight,quasi-straight,curving and sinuous based on the track *** cyclone TRacks Ontology(TcTRO)described in this paper is a knowledge base which comprises of classes,objects and data properties that represent the interaction among the Tc characteristics.A set of SWRL(Semantic Web Rule Language)rules are directly inserted to the TcTRO ontology for reasoning and inferring new knowledge from ***,we propose a learning algorithm which utilizes the inferred knowledge for optimizing the feature *** to experiments on the IBTrAcS dataset,the proposed ontology based SVM classifier achieves an accuracy of 98.3%with reduced classification error rates.
