AIM To assess in rodent and human adipocytes the antilipolytic capacity of hexaquis(benzylammonium) decavanadate(B6V10), previously shown to exert antidiabetic effects in rodent models, such as lowering free fatty acids(FFA) and glucose circulating *** Adipose tissue(AT) samples were obtained after informed consent from overweight women undergoing plastic surgery. Comparison of the effects of B6V10 and reference antilipolytic agents(insulin,benzylamine,vanadate) on the lipolytic activity was performed on adipocytes freshly isolated from rat, mouse and human AT. Glycerol release was measured using colorimetric assay as an index of lipolytic activity. The influence of B6V10 and reference agents on glucose transport into human fat cells was determined using the radiolabelled 2-deoxyglucose uptake *** In all the species studied, B6V10 exhibited a dosedependent inhibition of adipocyte lipolysis when triglyceride breakdown was moderately enhanced by β-adrenergic receptor stimulation. B6V10 exerted on human adipocyte a maximal lipolysis inhibition of glycerol release that was stronger than that elicited by insulin. However, B6V10 did not inhibit basal and maximally stimulated lipolysis. When incubated at dose ≥ 10 μmol/L, B6V10 stimulated by twofold the glucose uptake in human fat cells, but-similarly to benzylamine-without reaching the maximal effect of insulin, while it reproduced one-half of the insulin-stimulation of lipogenesis in mouse fat cells. COnCLUSIOn B6V10 exerts insulin-like actions in adipocytes, including lipolysis inhibition and glucose transport activation. B6V10 may be useful in limiting lipotoxicity related to obesity and insulin resistance.

Real-world evidence (RWE) is clinical evidence on a medical product’s safety and efficacy that is generated using real-world data (RWD) resulting from routine healthcare delivery. This study evaluates the clinical efficacy of amoxicillin + clavulanic acid in children with pharyngitis, acute otitis, or acute rhinosinusitis with suspected bacterial origin under normal office and home conditions. Methods: This was a real-life, prospective, observational, pharmacovigilance study. It included children of both sexes between 2 and 12 years old, with a diagnosis of Rhinopharyngitis (tonsillitis), Acute Otitis Media and Rhinosinusitis. The main effectiveness variable evaluated was reduction and time to resolution of symptoms. All patients received Amoxicillin/Clavulanic Acid suspension 600 mg/42.9 mg/5 mL at a dose of 90 mg/Kg/day in two doses, every 12 hours for 7 days. The evaluations were carried out at the beginning, at 72 hours (3rd day) and at 7 days. All patients underwent culture and antibiogram. Results: The majority of cultures were negative for pathogenic germs, suspecting unidentifiable germs, or viral etiology despite the rigorous selection of subjects following validated scores. The most frequently isolated germ was Staphylococcus aureus;growth of gram-negative bacteria was reported in 33.33% of the cultures. There was a significant improvement in symptoms in children with tonsillitis and rhinosinusitis from the first 72 hours of treatment, persisting until the 7 days. In the otitis media group, returning to normal by the tenth day. During the conduction of this investigation, no adverse effects associated with the prescribed therapy were reported.

The ability of sperm selection techniques to remove single- or double-strand DnA damage

A wide variety of techniques for the preparation of sperm are currently available, of which the most commonly employed are densitygradient centrifugation （DGC） and swim-up （SUP）. To date, these methods appear to be effective in selecting functional sperm for assisted reproduction techniques （ART）, but they may have negative effects on sperm DnA. In this study, the ability of these semen processing techniques to eliminate spermatozoa containing single- and double-strand DnA damage was assessed by the two-tailed comet assay and the sperm chromatin dispersion test in 157 semen samples from patients seeking assisted reproduction treatment. Our results indicated that SUP and DGC are equally efficient in eliminating spermatozoa containing double-strand DnA damage and sperm with highly damaged （degraded） DnA, as characterized by the presence of both single- and double-strand DnA breaks. However, DGC is more efficient than SUP in selecting spermatozoa that are free from single-strand DnA damage. Future studies should characterise the importance of the various types of DnA damage and examine the sperm processing protocols used in each laboratory to determine their ability to eliminate DnA damage and hence, prevent the potential transmission of genetic mutations via ART.

BACKGROUnD Antiviral treatment of patients with chronic hepatitis B(CHB)in the grey zone of treatment comands risk management in order to optimize the health *** this sense,the identification of HBV mutants related with an increased risk of hepatocellular carcinoma(HCC)could be useful to identify subpopulations with potential indication of antiviral *** To analyze the prevalence/persistence of hepatitis B virus(HBV)preS and basal core promoter(BCP)/precore/core variants associated to HCC development in CHB patients in the grey *** Work was designed as a longitudinal retrospective study,including 106 plasma samples from 31 patients with CHB in the grey zone of treatment:Hepatitis B e antigen negative,HBV-DnA levels between 12-20000 IU/mL,normal or discordant transaminase levels during follow up and mild/moderate necroinflammatory activity in liver biopsy or Fibroscan(up to 9.5 kPa).Serum HBVDnA was tested using the Abbott Real Time HBV Assay and the BCP/precore/core and the hepatitis B surface antigen(HBsAg)coding regions were analyzed in positive samples by PCR/bulk-sequencing to identify the HCCrelated HBV *** High-risk HCC related mutants were detected in 24(77%)patients:19(61%)in the BCP/precore/core,and 7(23%)in the HBsAg coding region(2 preS1 and 5 preS2 deletions).The prevalence of preS deletions was genotype-dependent:3/5(60%)patients with preS2 deletions and 1/2 with preS1 deletions were infected with the HBV-E *** HBV-E was the most prevalent in sub-Saharan patients,a correlation between preS deletions and ethnicity was also found:6/8(75%)sub-Saharan vs 1/19(5%)Caucasian patients had preS deletions(P=0.00016).Remarkably,this correlation was maintained in those patients infected with HBV-A,a minor genotype in sub-Saharan patients:2/2 patients infected with HBV-A from West Africa vs 0/6 of Caucasian origin had preS *** HCC related variants were the major strains and persisted over time(up to 48 mo).Pa

Evaluation of Sawdust and Rice Husks as Fillers for Phenolic Resin Based Wood-Polymer Composites

We produced Wood-Polymer Composites (WPCs) with phenolic resin (PR) filled with saw dust (SD) and rice husks (RH) in a PR:fillerratio of 60:40 wt.%. RH and SD were grinded and sieved into particles μm. The aim of this research work was to evaluate sawdust and rice husks as fillers for sustainable phenolic resin based WPCs. Therefore, we investigated the thermal stability of PR/RH and PR/SD WPCs then we studied and compared the tensile, flexural properties of PR/SD and PR/RH WPCs samples, as well as their dimensional stability after water absorption test. Furthermore, through ultraviolet light exposure, we evaluated the effects of photo-oxidation on the water stability and mechanical properties of PR/RH and PR/SD WPCs samples compared to unexposed ones. PR filled with SD presented better mechanical properties compared to PR/RH WPCs samples. However, PR/RH WPCs showed good mechanical properties, and better thermal resistance and better water repulsion capabilities compared to PR/SD WPCs samples. Although, long time UV exposure ended up lowering considerably the mechanical properties and water resistance of PR/SD and PR/RH WPCs, both RH and SD offer great added value as fillers for PR based WPCs;SD having better interactions with PR matrix compared to RH.

Development of Atmospheric Plasma Sprayed Dielectric Ceramic Coatings for High Efficiency Tubular Ozone Generators

Oxidative degradation of hazardous materials by ozone treatment like in sterilization of water, dump waste, pulp bleach and chemical processing, is superior to the traditional chlorine chemistry with respect to by-products and environmental protection. For an efficient and cost effective production of ozone for applications in drinking water and wastewater purification, a new concept of tubular composite material components has been developed. A borosilicate glass tube was coated with a layer system consisting of an intermetallic electrode and a dielectric oxide ceramic surface layer. Thermo-mechanical and dielectric properties are investigated with respect to the use of different thermal spray powders as well as the use of a high and a low energetic atmospheric spray gun. The materials and ozone production system of thermal sprayed ozonizer tubes are described and analyzed.

Molecular Diversity of Kenyan Lablab Bean (<i>Lablab purpureus</i>(L.) Sweet) Accessions Using Amplified Fragment Length Polymorphism Markers

Lablab purpureus (L.) Sweet is a multipurpose legume that combines use as human food and animal feed in addition to serving as a cover crop for soil conservation. In this work, molecular diversity in Lablab purpureus was assessed using amplified fragment length polymorphism markers on fifty Kenyan lablab accessions obtained from farmers’ fields and the Kenya national gene bank. One hundred and eighty polymorphic bands were revealed using fifteen selective primer pairs. The overall mean expected heterozygosity (He) for the five populations was 0.189. Estimates of components of molecular variance revealed that most of the genetic variation resided within populations (99%) and only 1% variance was among the populations, while Principal Coordinate Analysis showed an overlap between accessions from different geographic origins. The UPGMA cluster analysis generated from the distance matrix of the 50 assayed accessions, revealed low diversity among most of the accessions. The low diversity observed may be due to the narrow genetic base for breeding stocks, and extensive exchange of germplasm among smallholder farmers across the country. Results obtained from this study are discussed in light of the need to enhance the genetic management and improvement of this multipurpose crop species.

Occult hepatitis B virus(HBV) infection(OBI) is defined as the presence of HBV DnA in the liver(with or without detectable HBV DnA in serum) for individuals testing HBV surface antigen *** recently,the clinical effect of OBI was unclear on the progression of liver disease;on the development of hepatocellular carcinoma;and on the risk for reactivation or transmission of HBV *** studies suggest a high prevalence of OBI among patients with cryptogenic chronic liver disease,but its role in the progression to cirrhosis remains *** OBI has been well documented in human immunodeficiency virus(HIV) -positive patients,especially among those coinfected with hepatitis C virus,further studies are needed to determine its current clinical impact in HIV setting.

Efficacy and safety of bridging thrombolysis initiated before transfer in a drip-and-ship stroke service

Objective Data regarding the efficacy and safety of bridging thrombolysis(BT)initiated before transfer for evaluation of endovascular therapy is ***,therefore,analyse efficacy and safety of BT in patients treated within a drip-and-ship stroke *** Consecutive adult patients suffering from acute ischaemic stroke and large-vessel occlusions(LVO)transferred to our comprehensive stroke centre for evaluation of endovascular therapy in 2017–2020 were identified from a local prospective stroke database and categorised according to BT and *** was defined as intravenous thrombolysis initiated before *** was assessed before and after *** outcome before stroke and at 3 months using the modified Rankin scale(mRS)was *** outcome was defined as mRS 0–1 or return to prestroke *** safety analysis,intracranial haemorrhages and mortality at 3 months were *** analysis was limited to patients with anterior circulation *** Of n=714 patients,n=394(55.2%)received *** patients in the BT group with documented LVO before transfer recanalised without endovascular therapy(n=46,11.7%)than patients who did not receive BT before transfer(n=4,1.3%,pn multivariate analysis,BT was the strongest independent predictor of early recanalisation(adjusted OR 10.9,95%CI 3.8 to 31.1,pnded to be an independent predictor of an excellent outcome at 3 months(adjusted OR 1.38,95%CI 0.97 to 1.96,p=0.077).There were no differences in safety between the BT and no-BT *** BT initiated before transfer was a strong independent predictor of early recanalisation.
