Evaluation of the Safety of Three Phenolic Compounds from <i>Dipteryx alata</i>Vogel with Antiophidian Potential

Phenolic compounds from Dipteryx alata Vogel were assayed against the in vitro neurotoxic effect induced by Bothrops jararacussu (Bjssu) venom. Mutagenicity was assessed by the Ames test using Salmonella typhimurium strains TA98, TA97a, TA100, and TA102, in experiments with and without metabolic activation. Anti-bothropic activity was obtained by using mouse phrenic nerve-diaphragm (PND) preparation and myographic technique. Control experiments with physiological Tyrode solution were used for keeping the PND preparations alive (n = 4). Concentrations of phe-nolic compounds were as follow: protocatechuic and vanillic acids (200 μg/mL, n = 4), vanillin (50 μg/mL, n = 4). These compounds were used alone or pre-incubated with the venom (40 μg/mL), 30 min prior the addition to the organ bath (n = 4). Phenolic compounds significantly inhibited the neuromuscular blockade of Bjssu in the following order of potency: vanillic acid > protocatechuic = vanillin. Vanillic acid added 10 min after the Bjssu venom was also able to avoid the venomblockade evolution. The mutagenicity assay indicated that all phytochemicals were unable to in-crease the number of revertants, demonstrating the absence of mutagenic activity. This study demonstrated both the safety and therapeutical potential of the three phenolic compounds as novel complementary anti-bothropic agents.

目的:运用B超评估受体对无眼眼眶植入凝胶聚乙烯义眼台和多孔聚乙烯义眼台的反应。方法:兔36只行单侧眼内容物剜除并凝胶聚乙烯义眼台或多孔聚乙烯义眼台植入术,术后每周进行临床检查,术后30,60,90d行B超检查。结果:植入凝胶聚乙烯义眼台的兔除1只外,义眼台全都脱出,可能是凝胶义眼台吸水后体积增大造成的。B超检查显示随访期间凝胶义眼台内部无血管。植入多孔聚乙烯义眼台的兔有5只(27.8%),术后30d发生义眼台脱出。根据B超检查,多孔聚乙烯义眼台显示结构不规则有异质性,回声与血管组织类似。结论:如何确定植入兔无眼眼眶的凝胶聚乙烯义眼台的最佳大小,还需要进一步研究。我们的研究显示,B超能有效评估无眼球眼眶植入的义眼台,为进一步的活体研究提供了有用信息,有可能取代其他评估义眼台血管化的昂贵检查。

BACKGROUND Patients with cirrhosis are at risk of cirrhotic cardiomyopathy,with resulting cardiac dysfunction and exercise *** minute walking test(6MWT)assesses functional status and predicts morbidity and mortality in cardiopulmonary *** To determine if it associates with mortality by analyzing 6MWT performance in patients with liver *** A cohort of 106 cirrhotic patients was evaluated in the outpatient setting with echocardiogram and 6MWT and follow up for one year to document hepatic decompensation and *** distance in meters was recorded at the end of 6 min(6MWD).RESULTS This cohort had a mean age of 51 years and 56%male;patients were staged as Child A in 21.7%,B 66%and C 12.3%.Walk distance inversely correlated with Child scores,and was significantly reduced as Child stages *** who died(10.4%)showed shorter mean 6MWD(P=0.006).Low 6MWD was an independent predictor of mortality(P=0.01).CONCLUSION 6MWT is a noninvasive inexpensive test whose result is related to Child scores and *** is useful to identify patients with liver cirrhosis at high risk of mortality for closer monitoring and potential early intervention.

Background： leptin has a strong relation to important traits in animal production, such as carcass composition,feed intake, and reproduction. It is mainly produced by adipose cells and acts predominantly in the *** this study, circulating leptin and its gene expression in muscle were evaluated in two groups of young Nellore bulls with divergent feed efficiency. Individual dry matter intake（DMI） and average daily gain（ADG） of 98 Nellore bulls were evaluated in feedlot for 70 d to determinate the residual feed intake（RFI） and select 20 animals for the high feed efficient（LRFI） and 20 for the low feed efficient（HRFI） groups. Blood samples were collected on d 56 and at slaughter（80 d） to determine circulating plasma leptin. Samples of Longissimus dorsi were taken at slaughter for leptin gene expression ***： DMI and RFI were different between groups and LRFI animals showed less back fat and rump fat thickness,as well as less pelvic and kidney fat weight. Circulating leptin increased over time in all animals. Plasma leptin was greater in LRFI on 56 d and at slaughter（P = 0.0049）. Gene expression of leptin were greater in LRFI animals（P = 0.0022） in accordance with the plasma levels. The animals of the LRFI group were leaner, ate less, and had more circulating leptin and its gene ***： These findings demonstrated that leptin plays its physiological role in young Nellore bulls, probably controlling food intake because feed efficient animals have more leptin and lower residual feed intake.

Management of hepatitis C in patients with chronic kidney disease

Hepatitis C virus(HCV) infection is highly prevalent among chronic kidney disease(CKD) subjects under hemodialysis and in kidney transplantation(KT) recipients, being an important cause of morbidity and mortality in these patients. The vast majority of HCV chronic infections in the hemodialysis setting are currently attributable to nosocomial transmission. Acute and chronic hepatitis C exhibits distinct clinical and laboratorial features, which can impact on managementand treatment decisions. In hemodialysis subjects, acute infections are usually asymptomatic and anicteric; since spontaneous viral clearance is very uncommon in this context, acute infections should be treated as soon as possible. In KT recipients, the occurrence of acute hepatitis C can have a more severe course, with a rapid progression of liver fibrosis. In these patients, it is recommended to use pegylated interferon(PEG-IFN) in combination with ribavirin, with doses adjusted according to estimated glomerular filtration rate. There is no evidence suggesting that chronic hepatitis C exhibits a more aggressive course in CKD subjects under conservative management. In these subjects, indication of treatment with PEG-IFN plus ribavirin relies on the CKD stage, rate of progression of renal dysfunction and the possibility of a preemptive transplant. HCV infection has been associated with both liver disease-related deaths and cardiovascular mortality in hemodialysis patients. Among those individuals, low HCV viral loads and the phenomenon of intermittent HCV viremia are often observed, and sequential HCV RNA monitoring is needed. Despite the poor tolerability and suboptimal efficacy of antiviral therapy in CKD patients, many patients can achieve sustained virological response, which improve patient and graft outcomes. Hepatitis C eradication before KT theoretically improves survival and reduces the occurrence of chronic graft nephropathy, de novo glomerulonephritis and post-transplant diabetes mellitus.

Preliminary Developments of Miniplate-Type Fuel of U-2.5Zr-7.5Nb Alloy Dispersed and Cladded in Zircaloy

Nuclear fuel based on uranium metal alloys is utilized in research and test reactors. For the purpose of the reduction of fuel enrichment, high densities of uranium-235 in this kind of fuel are needed. This can be achieved when uranium alloys are used containing elements such as Zr, Mo and Nb. The construction of fuel element with high-uranium density requires materials with low cross sections for neutron absorption, stability under irradiation and absence of the chemical interactions between the fuel and cladding elements. In case of U-Zr-Nb alloys, Zry （zircaloy） cladding is a better option due to the fact that they have a higher chemical compatibility when compared with the use of aluminum alloys. This study aims to develop plate type nuclear fuel using the U-2.5Zr-7.5Nb alloy dispersed in Zry. Powders of this uranium based alloy and Zry were obtained by hydriding-dehydriding process. These powders were homogenized, compacted in pellet that was sandwiched in plates and frame of Zry. This assembly was hot rolled forming the dispersion fuel miniplate.

Influence of the Hydriding Cycles on U-IOMo Alloy Comminution Process

Hydriding-dehydriding process has been widely employed to comminute uranium alloys. Developments have been made to improve this process because it is a cheaper way to obtain powder of uranium alloys compared to the other methods, such as atomization. This work presented an innovative comminution process of U-10Mo alloy in laboratory scale where a Sievert type equipment was utilized and only hydriding cycles were employed without repetition of the dehydriding procedure. Experiments of comminution of the U-10Mo alloy were carried out by employing alloy samples which were submitted to different heat treatment conditions and, subsequently, to different hydriding cycles. The heat treatments considered were hot rolled at 800 ℃, homogenization at 900 ℃ or 1,000 ℃ and aging at 520℃. It was observed that the number of hydriding cycles and heat treatment conditions have influenced the particle size distributions. Samples that were hot-rolled, homogenized at 1,000 ℃ and aged, and comminuted with five cycles of hydriding have produced powders with particle size distribution more uniform with the major quantity of particles in the range from 50μm to 200 μm while the other conditions have obtained a great quantity above 200 μm.

Epidemiological Profile of Breast Neoplasia in the Municipality of Valença (RJ Brazil): Study from 2009 to 2017

Breast neoplasms are the main oncological pathology in Gynecology and Obstetrics, with increasing rates of incidence and mortality in the female population. According to the World Health Organization (WHO), more than 1 million new cases of breast cancer are diagnosed worldwide every year. This study aims to understand and analyze the epidemiological profile of patients diagnosed with breast cancer in the last nine years in the city of Valença—RJ, Brazil. A descriptive, documentary-retrospective analysis was carried out from January 2009 to December 2017, on 96 records of women who underwent breast biopsy at the Escola Escola de Valença (HEV), referred by the HEV Gynecology outpatient clinic. The results compared the variables age, race, parity, alcoholism, smoking, comorbidities, histological type, family history and screening performed with the scenario of breast cancer in the world, concluding that it is essential to identify patients at risk development of breast cancer to guide prevention, screening and early detection of pathology. The cases in the municipality are similar to other studies in relation to smoking, alcohol consumption, associated comorbidities, histological type and family history. However, the prevalence in younger people stands out, as well as the high percentage of women who have not had previous screening, which highlights the importance of the development and spread of women’s health in this city in the south of the state.

High Sugar-Fat Diet Induces Metabolic-Inflammatory Disorders Independent of Obesity Development

Background: The modern dietary habit, which is rich in refined carbohydrates and saturated fats, increases the risk of chronic diseases due to the proinflammatory effect of these nutrients. Aim: To evaluate the impact of high sugar-fat diet in the development of metabolic-inflammatory disorders in non-obese animals. Methods: Male Wistar rats were distributed into two groups according to the diet: control and high sugar-fat for 30 weeks. It was analyzed: dietary efficiency;chow, water and caloric intake;metabolic and hormonal profile in plasma and inflammatory cytokines in epididymal adipose tissue. Data were compared by Student’s t test or by Mann-Whitney U test with p Results: HSF presented lower chow intake, higher water consumption and dietary efficiency with no difference in the caloric intake. The final body weight (FBW) and weight gain (WG) were lower in the HSF group and there was no difference in the adiposity index (AI). HSF diet-induced hyperglycemia and hyperinsulinemia with no difference for Homeostatic Model Assessment for Insulin Resistance (HOMA-IR). Triglycerides, uric acid, adiponectin and leptin levels were higher in the HSF group. The HSF group showed increased interleukin-6 (IL-6) and tumoral necrosis factor-alpha (TNF-α) levels in epidydimal adipose tissue. The urinary protein-creatinine ratio and albuminuria were higher in the HSF group. Conclusion: HSF diet intake is directly involved in the development of metabolic-inflammatory disorders independent of obesity, dissociating the view that increased adiposity is the major risk factor for complications commonly found in obese individuals.
