On the ionospheric response in the Taiwan region to Typhoon Meranti in September 2016

Previous studies have revealed that typhoons can affect the ionosphere. This paper uses GNSS observations provided by GNSS tracking stations of Taiwan region, ERA5 gravity wave dissipation products,and Himawari-8 band 07 brightness temperature observation data to analyze the impact of Typhoon Meranti on Taiwan region's ionosphere from September 13 to 15, 2016. In addition to the daily effects of traveling ionospheric disturbances(TIDs), TIDs of various intensities caused by Meranti-induced gravity waves were observed during this period, which is inseparable from the state of the typhoon and the topographic features during its progress. The ionospheric disturbance caused by the edge of a typhoon is usually larger than that at the typhoon eye. When the typhoon approached Taiwan region with high intensity on September 14, it was observed that the gravity waves caused by the typhoon had a wavelength of about 174-293 km, a period of 13.5-23.5 min, and a propagation speed of 142-302 m/s. When the vortex structure of a typhoon interacts with complex surfaces such as mountains, it is easier to excite more medium-scale gravity waves. The origin that excites gravity waves and causes ionospheric anomalies is different from the location of the typhoon eye and changes over time. The gravity waves caused by the typhoon lasted for three days in the central mountains of Taiwan region. Still, background wind conditions restricted the propagation of gravity waves from the mesosphere to the thermosphere and led to TID predominantly concentrated between 06:00 and 13:00 UT each day.

目的:采用高通量测序技术探索海珠益肝加味方对自身免疫性肝炎(autoimmune hepatitis,AIH)小鼠肝组织微小RNA(micro RNA,miRNA)表达谱的影响。方法:将24只小鼠随机分为空白组(NC组)、模型组(MODEL组)和海珠益肝加味方(TREAT组),每组8只。除NC组外,其余小鼠采用尾静脉注射刀豆蛋白(20 mg·kg^(-1))法制备AIH模型。TREAT组采用药物灌胃(13.4 g·kg^(-1)),其余2组采用蒸馏水灌胃,每日1次,连续3 d。HE染色观察小鼠肝组织病理形态,赖氏法测定血清谷草转氨酶(aspartate transaminase,AST)、谷丙转氨酶(alanine aminotransferase,ALT)含量。采用高通量测序技术检测小鼠肝组织miRNA表达谱,筛选差异表达miRNA和靶基因,并进行基因本体(Gene Ontology,GO)和京都基因与基因组百科全书(Kyoto Encyclopedia of Genes and Genomes,KEGG)分析。结果:HE染色显示,NC组小鼠肝细胞形态无明显异常,MODEL组小鼠肝细胞水肿伴炎症细胞浸润。TREAT组小鼠肝细胞轻微水肿,伴少量炎症细胞浸润。与NC组比较,MODEL组小鼠血清ALT、AST含量升高(P<0.01)。与MODEL组比较,TREAT组小鼠血清ALT、AST含量降低(P<0.01)。RNA-seq测序显示,与NC组比较,MODEL组小鼠肝组织差异表达miRNA共34个,其中18个上调,16个下调。与MODEL组比较,TREAT组小鼠肝组织差异表达miRNA共141个,其中13个上调,128个下调。GO功能主要富集于信号转导、细胞凋亡调控、细胞表面受体信号通路、钙离子跨膜运输等。KEGG通路主要富集于MAPK信号通路、ErbB信号通路、FoxO信号通路、Hippo信号通路等。结论:海珠益肝加味方可能通过调控肝组织miRNA表达谱,从而发挥治疗小鼠AIH的作用。

Genetic Engineered Ultrasound-Triggered Injectable Hydrogels for Promoting Bone Reconstruction

Genetic engineering technology can achieve specific gene therapy for a variety of diseases, but the current strategy still has some flaws, such as a complex system, single treatment, and large implantation trauma. Herein, the genetic engineering injectable hydrogels were constructed by ultrasonic technology for the first time to realize in vivo ultrasound-triggered in situ cross-linking and cell gene transfection, and finally complete in situ gene therapy to promote bone reconstruction. First, ultrasound-triggered calcium release was used to activate transglutaminase and catalyze the transamidation between fibrinogen. Simultaneously, liposome loaded with Zinc-finger E-box-binding homeobox 1 (ZEB1) gene plasmid (Lip-ZEB1) was combined to construct an ultrasound-triggered in situ cross-linked hydrogels that can deliver Lip-ZEB1. Second, ultrasound-triggered injectable hydrogel introduced ZEB1 gene plasmid into endothelial cell genome through Lip-ZEB1 sustained release, and then acted on the ZEB1/Notch signal pathway of cells, promoting angiogenesis and local bone reconstruction of osteoporosis through genetic engineering. Overall, this strategy provides an advanced gene delivery system through genetic engineered ultrasound-triggered injectable hydrogels.

Vesicular exocytosis is a key process involved in neurotransmitter release,whereas vesicle recycling is critical for the homeostasis of plasma membrane structures and the maintenance of ***^2+ regulates several fundamental cellular processes(Zhu et al.,2016,xiong and Zhu,2016).Especially,Ca^2+ influx plays a dual role of triggering vesicular exocytosis and modulating compensatory endocytosis in neurons and endocrine cells.

Methylated cytosine of CpG dinucleotides in vertebrates may be oxidized by Tet proteins,a process that can lead to DNA demethylation. The predominant oxidation product, 5-hydroxymethylcytosine(5hmC), has been implicated in embryogenesis, cell differentiation,and human diseases. Recently, the SRA domain of UHRF2(UHRF2-SRA) has been reportedto specifically recognize 5hmC, but how UHRF2 recognizes this modification is *** we report the structure of UHRF2-SRA in complex with a 5hmC-containing DNA. Thestructure reveals that the conformation of a phenylalanine allows the formation of an optimal5 hmC binding pocket, and a hydrogen bond between the hydroxyl group of 5hmCand UHRF2-SRA is critical for their preferential binding. Further structural and biochemical analyses unveiled the role of SRA domains as a versatile reader of modified DNA, and the knowledge should facilitate further understanding of the biological function of UHRF2 and the comprehension of DNA hydroxymethylation in general.

Nucleosome is the basic repeating unit of eukaryotic chromatin. Classical nucleosome contains one H3-H4 tetramer and two H2A–H2B dimmers wrapped by a certain length of DNA. Many non-allelic variants have been found for H3, but not for H4, suggesting that the H3 variants play very important roles in the functions of chromatin. Two ubiquitous variants of canonical histones H3.1 in mammals are CENP-A and H3.3. CENP-A is a centromere-specific histone H3 variant assembled onto centromeric chromatin during late M–early G1 phases of the cell cycle1. Deposition of CENP-A at the centromere requires histone chaperone HJURP. H3.3 is synthesized throughout the cell cycle and localized in gene bodies as well as in pericentric and telomeric heterochromatin. Two principal H3.3 deposition pathways are mediated by histone chaperones death domain–associated protein6（DAXX） and HIRA. DAXX cooperates with a SNF2-like chromatin remodeler, ATRX, to direct the deposition of H3.3 onto pericentric and telomeric heterochromatin, whereas HIRA mainly targets H3.3 to gene bodies. We solved the crystal structure of CENP-A–H4 with the chaperone HJURP（Hu, Liu et al. 2011） and H3.3–H4 in complex with its chaperone DAXX（Liu, xiong et al. 2012）. Basing on the structure information, our analysis reveal a novel site in the CENP-A which distinguishes it from histone H3 in its ability to bind HJURP. And by a combination of structural, biochemical and cell-based targeting analyses, we show that Ala87 and Gly90 are the principal determinants of human H3.3 specificity. These findings provide key information for specific recognition of H3 variants and mechanistic insights into the process of specific chromatin assembly.

大数据技术能够有效处理多来源、多类型、多尺度数据,在生态环境领域受到广泛关注,但中国仍处于起步阶段,探索大数据技术在生态环境领域的应用对推动大数据发展具有重要意义,对生态环境精准监管和综合决策具有显著参考价值。基于国内外最新研究,文章将生态环境大数据技术体系分为数据采集、数据处理和实践应用3个部分,数据采集包括地面监测、卫星遥感监测、地理信息、社会统计及网络抓取,数据处理包括存储管理、预处理、深入处理与整合挖掘4个流程,主要应用于生态环境的监测评价、模拟预测与优化管理3个方面。在监测评价方面,大数据技术有利于实时监控、长期跟踪、网格化评价生态环境质量,提高了生态环境监测评价的有效性,但多领域监测数据的耦合分析、智能纠错、评价方法优化能力有待提升;在模拟预测方面,大数据技术与专业模型整合增加了不同时空尺度下复杂环境要素的模拟精度和预测速度,实现动态预警和生态风险评估,但仍缺乏基于大数据技术的高精度存储、分析和集成模型,缺少更专业、更开放的大数据分析系统支撑模型库;在优化管理方面,促进污染的有效溯源、科学控制与全过程监管,但新型污染物和特征污染物时空分布特征尚不明确,系统性数学模型与风险评价管理体系尚不完善,给基于大数据平台的优化管理带来局限性。因此,应有效集成多领域生态环境监测数据,提升数据处理技术能力,推进大数据应用,以满足政府多源管控需求,提升政府科学决策效果。
