NF-κB信号通路在“反流性食管炎-食管癌”中的作用及中药干预研究进展

反流性食管炎是指胃内容物反流进入食管引起的食管黏膜损伤性炎症疾病,其发病率呈上升趋势,现已成为食管癌重要的癌前疾病。研究表明,持续的炎症反应刺激食管黏膜,引起食管上皮细胞异常增殖、食管黏膜组织损伤,最终导致异型增生甚至癌变的发生。核转录因子-κB(NF-κB)信号通路是最经典的炎症、癌症信号传导途径之一。研究发现,NF-κB信号通路的异常激活对反流性食管炎及食管癌的发生发展及预后至关重要,其广泛参与食管上皮细胞及肿瘤细胞的增殖、自噬、凋亡、炎症反应等过程,加速反流性食管炎向食管癌的转化,使其成为反流性食管炎及食管癌治疗的潜在靶点。目前,反流性食管炎及食管癌尚无特效治疗手段,并伴随较大的不良反应。因此,寻找一种有前途且安全的药物仍然是当务之急。近年来,中医药以NF-κB信号通路为切入点开展大量研究,结果表明NF-κB信号通路是中医药防治反流性食管炎及食管癌的重要潜在靶点,但目前尚缺乏全面系统的阐述。因此,该文总结近年来的相关研究,分析NF-κB信号通路与反流性食管炎、食管癌及“炎-癌转化”的关系,梳理中药调控NF-κB信号通路干预反流性食管炎及食管癌的研究文献进行综述,为反流性食管炎及食管癌的防治提供新思路。

中药调节微RNA干预食管癌的机制研究进展

食管癌(EC)是常见的消化系统恶性肿瘤,预后极差。微RNA(miRNA)是肿瘤发生、发展的重要调控因子,可参与肿瘤细胞增殖、侵袭、转移和凋亡等恶性生物学行为。中药具有疗效确切、作用广泛、副作用小的特点,故本文以miRNA为切入点,系统阐述中药调节miRNA干预EC的作用机制。结果发现,姜黄素、款冬花多糖、白术多糖、麦冬皂苷B等中药有效成分和豆根管食通口服液可上调miRNA-532-3p(miR-532-3p)、miR-551b-3p、miR-99a、miR-34a、miR-199a-3p、miR-377等miRNA的表达,白杨素、藤梨根提取物等中药有效成分/部位和柴胡疏肝散合旋覆代赭汤、加味橘皮竹茹汤等中药复方可下调miR-199a-3p、miR-451、miR-21等miRNA的表达,从而调控信号通路(磷脂酰肌醇3激酶/蛋白激酶B等)、下游蛋白(锌指和同源框蛋白1等)或酶类(胸腺激酶1等)的表达,抑制EC细胞的增殖、侵袭和转移,诱导其凋亡,最终达到延缓EC进展的目的。

The normality of symmetry property of Cayley graphs of valencies 3 and 4 on the alternating group A5 is studied. We prove that all but four such graphs are normal;that A5 is not 5-CI. A complete classification of all arc-transitive Cayley graphs on A5 of valencies 3 and 4 as well as some examples of trivalent and tetravalent GRRs of A5 is given.

胰腺癌(pancreatic cancer,PC)是临床上常见的消化系统恶性肿瘤。由于早期症状不典型,多数患者在确诊时已处于晚期,给患者及家属的身心造成极大痛苦。磷脂酰肌醇3-激酶(PI3K)/蛋白激酶B(Akt)信号是影响癌症的经典通路。PC状态下,PI3K/Akt信号通路调控失常,可推动肿瘤细胞周期过渡,抑制肿瘤细胞自噬、凋亡,影响PC细胞的增殖、侵袭与转移。目前,手术、放化疗及靶向治疗仍是PC的主要治疗手段,但存在耐药性强、不良反应大等局限性。近年来,中医药在抗肿瘤领域发展迅速,以疗效明显、不良反应少等优势备受关注。大量研究表明,中医药可以通过调控PI3K/Akt通路对PC细胞的增殖、自噬、细胞周期、凋亡、侵袭和转移及降低耐药性以延缓PC进展,但目前行业内仍缺少与上述内容相关的全面综述。因此,文章就PI3K/Akt通路及其与PC的关系,以及基于PI3K/Akt通路干预PC的中医药进展进行综述,以期为PC的治疗及未来药物开发提供有益参考。

Infectious disease departments in hospitals require pressure gradient to create unidirectional airflow to prevent the spread of contaminants,typically by creating active air infiltration through the difference between supply and exhaust air *** door gap is the channel of air flow between rooms,so its height has an important influence on the pressure difference and infiltration air volume of the *** is still a lack of research on setting reasonable ventilation strategies according to the different heights of door gaps at different positions in the *** this study,model of a set of isolation wards was established and analyzed using the multi-zone simulation software CONTAM,and the ventilation strategies with different heights of door gaps were applied to the actual infection diseases *** results show that in a building with ventilation system divided by functional area,the difference in the height of the door gaps requires different active infiltration air *** fluctuations in the medical and patient corridors are greater than in other *** significance of this study is to understand the active infiltration of air to guide the design and operation of ventilation systems in infectious disease hospitals or building remodeled to isolate close contacts of COVID-19 *** is also instructive for the design of pressure gradients in clean workshops,biological laboratories,and other similar buildings.

Early prediction of growth patterns after pediatric kidney transplantation based on height-related single-nucleotide polymorphisms

Background:Growth retardation is a common complication of chronic kidney disease in children,which can be partially relieved after renal *** study aimed to develop and validate a predictive model for growth patterns of children with end-stage renal disease(ESRD)after kidney transplantation using machine learning algorithms based on genomic and clinical ***:A retrospective cohort of 110 children who received kidney transplants between May 2013 and September 2021 at the First Affiliated Hospital of Zhengzhou University were recruited for whole-exome sequencing(WES),and another 39 children who underwent transplant from October 2021 to March 2022 were enrolled for external *** on previous studies,we comprehensively collected 729 height-related single-nucleotide polymorphisms(SNPs)in exon *** machine learning algorithms and 10-fold cross-validation analysis were employed for model ***:The 110 children were divided into two groups according to change in height-for-age *** univariate analysis,age and 19 SNPs were incorporated into the model and *** random forest model showed the best prediction efficacy with an accuracy of 0.8125 and an area under curve(AUC)of 0.924,and also performed well in the external validation cohort(accuracy,0.7949;AUC,0.796).Conclusions:A model with good performance for predicting post-transplant growth patterns in children based on SNPs and clinical variables was constructed and validated using machine learning *** model is expected to guide clinicians in the management of children after renal transplantation,including the use of growth hormone,glucocorticoid withdrawal,and nutritional supplementation,to alleviate growth retardation in children with ESRD.

The adenosine subfamily G protein-coupled receptors A_(2A)R and A_(2B)R have been identified as promising cancer immunotherapy *** of the A_(2A)R/A_(2B)R dual antagonists,AB928,has progressed to a phaseⅡclinical trial to treat rectal ***,the precise mechanism underlying its dual-antagonistic properties remains ***,we report crystal structures of the A_(2A)R complexed with AB928 and a selective A_(2A)R antagonist *** structures revealed a common binding mode on A_(2A)R,wherein the ligands established extensive interactions with residues from the orthosteric and secondary *** contrast,the cAMP assay and A_(2A)R and A_(2B)R molecular dynamics simulations indicated that the ligands adopted distinct binding modes on A_(2B)*** analysis of their chemical structures suggested that AB928 readily adapted to the A_(2B)R pocket,while 2-118 did not due to intrinsic *** disparity potentially accounted for the difference in inhibitory efficacy between A_(2B)R and A_(2A)*** study serves as a valuable structural template for the future development of selective or dual inhibitors targeting A_(2A)R/A_(2B)R for cancer therapy.
