Albumin-bound kynurenic acid is an appropriate endogenous biomarker for assessment of the renal tubular OATs-MRP4 channel

Renal tubular secretion mediated by organic anion transporters(OATs)and the multidrug resistanceassociated protein 4(MRP4)is an important means of drug and toxin excretion.Unfortunately,there are no biomarkers to evaluate their function.The aim of this study was to identify and characterize an endogenous biomarker of the renal tubular OATs-MRP4 channel.Twenty-six uremic toxins were selected as candidate compounds,of which kynurenic acid was identified as a potential biomarker by assessing the protein-binding ratio and the uptake in OAT1-,OAT3-,and MRP4-overexpressing cell lines.OAT1/3 and MRP4 mediated the transcellular vectorial transport of kynurenic acid in vitro.Serum kynurenic acid concentration was dramatically increased in rats treated with a rat OAT1/3(rOAT1/3)inhibitor and in rOAT1/3 double knockout(rOAT1/3^(-/-))rats,and the renal concentrations were markedly elevated by the rat MRP4(rMRP4)inhibitor.Kynurenic acid was not filtered at the glomerulus(99%of albumin binding),and was specifically secreted in renal tubules through the OAT1/3-MRP4 channel with an appropriate affinity(Km)(496.7 mM and 382.2 mM for OAT1 and OAT3,respectively)and renal clearance half-life(t1/2)in vivo(3.7±0.7 h).There is a strong correlation in area under the plasma drug concentration-time curve(AUC0et)between cefmetazole and kynurenic acid,but not with creatinine,after inhibition of rOATs.In addition,the phase of increased kynurenic acid level is earlier than that of creatinine in acute kidney injury process.These results suggest that albumin-bound kynurenic acid is an appropriate endogenous biomarker for adjusting the dosage of drugs secreted by this channel or predicting kidney injury.

The utility of endogenous glycochenodeoxycholate-3-sulfate and 4β-hydroxycholesterol to evaluate the hepatic disposition of atorvastatin in rats

The liver is an important organ for drugs disposition,and thus how to accurately evaluate hepatic clearance is essential for proper drug dosing.However,there are many limitations in drug dosage adjustment based on liver function and pharmacogenomic testing.In this study,we evaluated the ability of endogenous glycochenodeoxycholate-3-sulfate(GCDCA-S)and 4β-hydroxycholesterol(4β-HC)plasma levels to evaluate organic anion-transporting polypeptide(Oatps)-mediated hepatic uptake and Cyp3 a-meidated metabolism of atorvastatin(ATV)in rats.The concentration of ATV and its metabolites,2-OH ATV and 4-OH ATV,was markedly increased after a single injection of rifampicin(RIF),an inhibitor of Oatps.Concurrently,plasma GCDCA-S levels were also elevated.After a single injection of the Cyp3 a inhibitor ketoconazole(KTZ),plasma ATV concentrations were significantly increased and 2-OH ATV concentrations were decreased,consistent with the metabolism of ATV by Cyp3 a.However,plasma 4β-HC was not affected by KTZ treatment despite it being a Cyp3 a metabolite of cholesterol.After repeated oral administration of RIF,plasma concentrations of ATV,2-OH ATV and 4-OH ATV were markedly increased and the hepatic uptake ratio of ATV and GCDCA-S was decreased.KTZ did not affect plasma concentrations of ATV,2-OH ATV and 4-OH ATV,but significantly decreased the metabolic ratio of total and 4-OH ATV.However,the plasma level and hepatic metabolism of 4β-HC were not changed by KTZ.The inhibition of hepatic uptake of GCDCA-S by RIF was fully reversed after a 7-d washout of RIF.Plasma concentration and hepatic uptake ratio of GCDCA-S were correlated with the plasma level and hepatic uptake of ATV in rats with ANIT-induced liver injury,respectively.These results demonstrate that plasma GCDCA-S is a sensitive probe for the assessment of Oatps-mediated hepatic uptake of ATV.However,Cyp3 a-mediated metabolism of ATV was not predicted by plasma 4β-HC levels in rats.

目的探讨术后实施健康教育对子宫内膜异位症患者用药依从性及生活质量的影响,帮助患者制定针对性的术后护理措施,改善其遵医行为,并有效地促进康复进程。方法采用复合逻辑检索方式,以“子宫内膜异位症”、“内异症”、“健康教育”、“生活质量”、“Endometriosis”、“Health Education”等为检索词,对Pubmed、Medline、EMbase、知网数据库、万方数据库和维普等数据库进行检索,并筛选子宫内膜异位症患者术后实施健康教育的随机对照研究文献。Review Manager 5.3软件进行Meta分析,以P<0.05为合并统计量有统计学意义。结果患者健康知识知晓度评分的合并结果显示,Chi^(2)=3.95,I^(2)=49%,Z=14.27(P<0.001)。生理维度生活质量的合并结果显示,Tau^(2)=0.21,Chi^(2)=13.88,df=3,I^(2)=78%,提示合并的文献存在异质性,采用随机效应模型分析,结果显示,Z=3.53(P<0.001);患者心理维度生活质量的合并结果显示,Tau^(2)=0.00,Chi^(2)=1.51,df=3,I^(2)=0%,提示合并的文献无明显的异质性。Z=7.48(P<0.001)。患者用药依从性合并结果显示,Z=4.00(P<0.001),差异有统计学意义。结论子宫内膜异位症患者术后实施健康教育,有助于提升患者对疾病认知,改善术后用药依从性,并进一步提高患者术后生活质量。

将病毒注入蛋胚培养疫苗时,针头冲击使注射孔周围产生裂纹导致培养失败。为解决目前人工手持照蛋器在暗室通过肉眼检测蛋胚裂纹效率低、误判率高这一问题,提出一种结合多光谱通道注意(MSCA)机制与并行堆叠拓扑(PST)模块的带孔蛋胚裂纹检测方法。首先搭建带孔蛋胚裂纹检测黑箱,采集病毒注射后带孔蛋胚的线形、网状、星形裂纹和带孔完好蛋胚图像并建立数据集;接着以ResNet-50为骨干模型,将其后4层的第一个残差模块替换为PST模块,以增加模型初期图像表达能力;最后在每个PST模块与残差模块后引入MSCA机制,MSCA机制通过二维离散余弦变换(2DDCT)压缩数据得到各通道频率分量,采用神经结构搜索(NAS)方式得到最佳频率分量,即对权重重新分配,增加裂纹特征权重比例,以确保模型快速、精准识别带孔蛋胚裂纹。结果表明,改进的网络模型对带孔蛋胚裂纹检测时间为0.42 s/枚,检测精度为96.43%,检测效率高于人工作业。与原始ResNet-50相比,检测精度提高3.66%,优于其他经典卷积网络模型,改进模型可用于疫苗培育前带孔蛋胚裂纹自动检测。

Objectives:The secondary products of lipid oxidation are one of the main factors inducing protein oxidation.The effects of oxidation treatment with malondialdehyde(MDA)on the immunoreactivity of amandin and its digestion were studied.Materials and Methods:The rabbit IgG binding ability of amandin was analyzed by western blotting,and the changes in amandin oxidation and immunoreactivity during digestion of amandin with different degrees of oxidation were investigated in combination with an almond allergen enzyme-linked immunosorbent assay kit.Alteration of linear epitopes of amandin by oxidation was investigated by liquid chromatography-tandemmass spectrometry(LC-MS/MS).Results:The results showed that the immunoreactivity of amandin was significantly reduced after 1 mmol/L MDA and 100 mmol/L MDA treat-ment.However,the 1 mmol/L MDA treatment was owing to cleavage of linear epitope peptide in amandin and oxidation of the active amino acid.The 100 mmol/L MDA treatment was due to aggregation of amandin and significant decrease in its solubility.Oxidation also reduced di-gestibility of amandin and significantly affected immunoreactivity during digestion.LC-MS/MS also identified four oxidation-prone methionine sites(aa 264-274,298-308,220-240,and 275-297)in gamma conglutinin 1.Conclusions:MDA treatment reduced the immunoreactivity of amandin.MDA treatment also led to protein aggregation,which slowed down the digestion of amandin and altered the immunoreactivity of amandin during digestion.

Clinical characteristics of 310 SARS-CoV-2 Omicron variant patients and comparison with Delta and Beta variant patients in China

Although the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)Omicron variant has spread,data on the clinical characteristics of infected patients are limited.In this study,the demographic,clinical characteristics,and laboratory data of 310 SARS-CoV-2 Omicron variant patients treated at Haihe Hospital of Tianjin were collected and analyzed.Information on these patients was compared to 96 patients with the Delta variant of concern(VOC)and 326 patients with the Beta VOC during the previous coronavirus disease 2019(COVID-19)outbreak in Harbin.Of the 310 patients infected with the Omicron variant,the median age was 35 years.Most patients were clinically classified as mild(57.74%),and the most common symptoms were cough(48.71%),fever(39.35%),and sore throat(38.26%).The results for different vaccination groups in the Omicron group showed that the median of“SARS-CoV-2 specific IgG”after 2 or 3 doses of vaccination was higher than the unvaccinated group(all Ps<0.05).Older age was associated with a higher proportion of moderate cases and lower asymptomatic and mild cases based on clinical classifications.Compared to the Delta and Beta groups,the median age of the Omicron group was younger.The total number of asymptomatic patients and mild patients in the Omicron virus group was higher than the Delta and Beta groups(60.97%vs.54.17%vs.47.55%).This study presented the clinical characteristics of the first group of patients infected with the Omicron variant in Tianjin,China,and compared their clinical features with patients infected by the Delta and Beta variants,which would increase our understanding of the characteristics of SARS-CoV-2 Omicron variant.

Aqueous Zn-ion batteries(AZIBs)are the potential options for the next-generation energy storage scenarios due to the cost effectiveness and intrinsic safety.Nevertheless,the industrial application of AZIBs is still impeded by a series of parasitic reactions and dendrites at zinc anodes.In this study,taurine(TAU)is used in electrolyte to simultaneously optimize the coordination condition of the ZnSO4electrolyte and interfacial chemistry at the anode.TAU can preferentially adsorb with the zinc metal and induce an in situ stable and protective interface on the anode,which would avoid the connection between H_(2)O and the zinc metal and promote the even deposition of Zn^(2+).The resulting Zn//Zn batteries achieve more than 3000 hours long cyclic lifespan under 1 mA cm^(-2)and an impressive cumulative capacity at 5 mA cm^(-2).Moreover,Zn//Cu batteries can realize a reversible plating/stripping process over 2,400cycles,with a desirable coulombic efficiency of 99.75%(1 mA cm^(-2)).Additionally,the additive endows Zn//NH_(4)V_(4)O_(10)batteries with more stable cyclic performance and ultrafast rate capability.These capabilities can promote the industrial application of AZIBs.

Recently,sodium-ion batteries gradually become the promising alternative to lithium-ion batteries because of cost considerations.In this work,a kind of Bi2MoO6 nanosheets@N,S codoped graphene composite is designed and fabricated for sodium storage applications.Detailed characterizations are employed to investigate its morphology,structure and chemical compositions.When evaluated as an anode material for sodium-ion batteries,the as-prepared composite is able to display a specific capacity of 254 mA·h/g after 50 cycles at a current density of 0.2 A/g,and 186 mA·h/g at 1.6 A/g during the rate capability test.As a result,the further morphology and structure optimization is still required for high performance sodium-ion batteries.
