Infant guinea pig retina model of glutamate toxicity and intervention of basic fibroblast growth factor

Impaired vision with oligemic ophthalmopathy is a result of excitotoxicity caused by excitatory amino acids, resulting in pathological changes, such as loss of retinal neurons and in particular retinal ganglionic cells. The present study utilized infant guinea pigs, aged 45-50 days, to establish injury models via intraperitoneal injection of fixed sodium glutamate doses. Results from hematoxylin- eosin staining revealed significantly reduced retinal ganglionic cell numbers and retinal damage at 10 days after 7 consecutive days of 3 g/kg sodium glutamate treatment; these animals served as the injury model group. In addition, models of moderate injury （glutamate 3 g/kg daily, for 7 consecutive days） were intraperitoneally pretreated with basic fibroblast growth factor （800 U/kg daily）. Immunohistochemistry results confirmed reduced anti-apoptotic gene bcl-2 expression in the ganglion cell layer of glutamate-injured guinea pigs. Expression of the pro-apoptotic gene caspase-3 was increased in the ganglion cell layer and inner plexiform layer. Somatostatin expression was primarily distributed in the ganglion cell layer and inner nuclear layer. Expression of the presynaptic element synaptophysin was weak. However, following basic fibroblast growth factor injection, expressions of the above-described bioactive molecules were reversed, which suggested that basic fibroblast growth factor exerted protective effects on sodium glutamate-induced retinal injury in infant guinea pigs by regulating expression of synaptophysin, somatostatin, Bcl-2, and caspase-3.

以市政污泥为原料,通过热解、活化及共沉淀工艺制备了铝铈负载污泥生物炭复合材料(Al/CeCSBC),运用多种分析技术表征了材料形貌、比表面积及结构特征,通过模拟废水的批量吸附实验,考察了pH、初始氟离子质量浓度及吸附时间对Al/Ce-CSBC吸附氟性能的影响,最后探究了其除氟机制。结果表明,活化和改性均提高污泥生物炭的比表面积,其中Al/Ce-CSBC具有丰富的狭缝形介孔,比表面积为176.36 m^(2)·g^(-1),pH^(PZC)为9.5。负载于材料表面的无定形铝/铈氧化物是主要吸附组分,且发挥了双金属协同作用,在pH=4.0~9.0内其可保持较高的吸附容量。吸附动力学符合伪二级模型,吸附等温线符合Freundlich模型。最大吸附容量可达到41.74 mg·g^(-1),显著优于其他常见生物炭材料。其吸附机理主要包括静电吸附、离子交换和表面络合。本文可为污泥的资源化利用提供重要的参考。

Tectonic modes of mantle convection and their implications for Earth's tectonic evolution based on three-dimensional numerical simulations

Five tectonic modes of mantle convection are obtained and analyzed with three-dimensional numerical models in a spherical shell domain. The five tectonic convective modes are non-plate mobile-lid, plate-like mobile-lid, episodic plate-like mobile-lid, episodic stagnant-lid, and stagnant-lid convective modes, respectively. The typical characteristics of these five tectonic modes and their numerical classification criteria based on plateness, mobility, and their standard deviations are presented and discussed. The results show that the yield stress of the lithosphere has profound effects on the tectonic convective *** the gradual increase of yield stress, the tectonic mode of mantle convection changes from one to another sequentially through the aforementioned five modes. Additionally, as the Rayleigh number increases, the range of yield stress for the platelike mobile-lid convective mode decreases, and the dimensionless transition stress between different tectonic modes ***, the dimensional transition stress between the non-plate mobile-lid convective mode and plate-like mobile-lid convective mode increases with the increase of Rayleigh number, but decreases between other tectonic modes. Furthermore, we find that the transition stress between different tectonic modes is inversely proportional to the internal heating rate, with the transition stress decreasing as the internal heating rate increases. The fitting analysis of the transition stress between tectonic modes shows that Earth's current plate tectonics correspond to a lithospheric yield stress of 150–250 MPa, which aligns with the strength of serpentinized mantle rock determined by experimental petrography. 目的:探讨槲皮素对转基因前列腺腺癌小鼠C2细胞系(TRAMP-C2)凋亡影响及其作用机制。方法:采用不同浓度的槲皮素处理体外培养的TRAMP-C2细胞,应用MTT法检测槲皮素对TRAMP-C2细胞增殖的影响;采用流式细胞仪检测TRAMP-C2细胞的凋亡率,并应用基因芯片技术筛选槲皮素作用前后TRAMP-C2细胞差异性表达基因。结果:槲皮素能抑制TRAMP-C2细胞生长,诱导TRAMP-C2细胞凋亡,呈剂量及时间依赖性;基因芯片分析发现,应用5×10-5mol/L浓度槲皮素处理48h后的TRAMP-C2细胞有69个基因表达下调,94个基因表达上调,其中血小板反应素表达明显上调,为对照组的36倍。结论:槲皮素能诱导TRAMP-C2细胞凋亡,具有直接抗肿瘤作用,其作用机制可能与上调TRAMP-C2细胞血小板反应素-1的表达有关。

Molecular dynamics simulations of helium transport through inorganic mineral nanopores

Helium transport through nanoscale inorganic mineral pores and pore throats is essential for its overall migration. To elucidate helium's transport dynamics within nanopores, we employed equilibrium and non-equilibrium molecular dynamics simulations to investigate helium's static self-diffusion and pressure-driven flow in quartz slit-shaped nanopores. We also introduced water and various gases, including hydrogen, methane, ethane, nitrogen, and carbon dioxide, into the nanopores to assess their influence on helium transport. Our findings indicate minimal helium adsorption on quartz pore surfaces. Under conditions where the pore size is less than 5 nm and the pressure under 10 MPa, environmental factors markedly influence helium diffusion. Large pore sizes, high temperatures, and low gas pressures enhance helium desorption and facilitate faster diffusion. We observed a positive correlation between helium flow velocity and factors such as pore size, pressure gradient, and surface smoothness of the pores. Notably, the presence of pore water and carrier gases in quartz nanopores, which diffuse more slowly than helium, tends to reduce helium surface adsorption and slow its diffusion. Among the carrier gases studied, nitrogen showed similar adsorption capacity, diffusivity, and stability to helium, while carbon dioxide displayed the highest adsorption capacity and the slowest diffusion rate, markedly differing from helium. Based on the simulation results, we concluded that water and carrier gases primarily function as transport mediums in helium migration, moving together with helium. Nitrogen, which shares similar properties with helium, effectively assists in this co-migration process. Conversely, carbon dioxide, due to its high adsorption capacity and slow diffusion, tends to be lost during co-migration. 为解决在修井打捞作业中常用的几类捞矛存在的不适用性等问题,研制了新型整体式液压可退捞矛。该捞矛依靠液缸推动卡瓦下行、扩张进行打捞,而释放丢手则通过提放连接工具的管柱实现。捞矛外筒及锚杆都无变径点,避免了应力集中。与捞矛外筒同等直径的引导槽备块对卡瓦片提供了足够的支撑强度,保证了工具的抗扭性能。卡瓦与引导槽内的倒角结构满足了卡瓦整体外张的角度,并确保了较好的咬合效果。经过地面试验及3井次的入井打捞作业,打捞成功率和丢手成功率均为100%。

几何误差测量是机床几何精度评定的关键一环。现有的几何误差测量方法在测量效率、测量空间上存在着不同程度的弊端。为此,提出一种基于激光跟踪仪的机床误差快速测量方法。首先,通过测量机床工作空间中不共面的4个位置的坐标,实现机床坐标系和跟踪仪坐标系的对准;其次,通过测量机床直线轴行程内若干位置的坐标,求解出机床各直线轴的3项位置误差和各直线轴间的垂直度误差;再次,通过测量机床工作空间体对角线上若干点的位置坐标,实现机床空间误差的测量;最后,在一台卧式加工中心上和XM60多光束激光干涉仪进行了对比试验,结果表明两仪器测量得到的误差曲线具有较高的相似性。该方法为机床误差测量提供了新手段和新思路。

Constraints on holographic QCD phase transitions from PTA observations

The underlying physics of QCD phase transition in the early Universe remains largely unknown due to its strong-coupling nature during the quark-gluon plasma/hadron gas transition, yet a holographic model has been proposed to quantitatively fit the lattice QCD data while with its duration of the first-order phase transition(FoPT) left undetermined. At specific baryon chemical potential, the first-order QCD phase transition agrees with the observational constraint of baryon asymmetry. It, therefore, provides a scenario for phase transition gravitational waves(GWs) within the standard model of particle physics. If these background GWs could contribute dominantly to the recently claimed common-spectrum red noise from pulsar timing array(PTA) observations, the duration of this FoPT can be well constrained, and the associated primordial black holes are still allowed by current observations.
