Potassium transporter OsHAK9 regulates seed germination under salt stress by preventing gibberellin degradation through mediating OsGA2ox7 in rice

Soil salinity has a major impact on rice seed germination,severely limiting rice production.Herein,a rice germination defective mutant under salt stress(gdss)was identified by using chemical mutagenesis.The GDSS gene was detected via MutMap and shown to encode potassium transporter OsHAK9.Phenotypic analysis of complementation and mutant lines demonstrated that OsHAK9 was an essential regulator responsible for seed germination under salt stress.OsHAK9 is highly expressed in germinating seed embryos.Ion contents and non-invasive micro-test technology results showed that OsHAK9 restricted K^(+)efflux in salt-exposed germinating seeds for the balance of K^(+)/Na^(+).Disruption of OsHAK9 significantly reduced gibberellin 4(GA4)levels,and the germination defective phenotype of oshak9a was partly rescued by exogenous GA_(3)treatment under salt stress.RNA sequencing(RNA-seq)and real-time quantitative polymerase chain reaction analysis demonstrated that the disruption of OsHAK9 improved the GA-deactivated gene OsGA2ox7 expression in germinating seeds under salt stress,and the expression of OsGA2ox7 was significantly inhibited by salt stress.Null mutants of OsGA2ox7 created using clustered,regularly interspaced,short palindromic repeat(CRISPR)/CRISPR-associated nuclease 9 approach displayed a dramatically increased seed germination ability under salt stress.Overall,our results highlight that OsHAK9 regulates seed germination performance under salt stress involving preventing GA degradation by mediating OsGA2ox7,which provides a novel clue about the relationship between GA and OsHAKs in rice.

Smoothed-Particle Hydrodynamics Simulation of Ship Motion and Tank Sloshing under the Effect of Regular Waves

Predicting the response of liquefied natural gas(LNG)contained in vessels subjected to external waves is extremely important to ensure the safety of the transportation process.In this study,the coupled behavior due to ship motion and liquid tank sloshing has been simulated by the Smoothed-Particle Hydrodynamics(SPH)method.Firstly,the sloshing flow in a rectangular tank was simulated and the related loads were analyzed to verify and validate the accuracy of the present SPH solver.Then,a three-dimensional simplified LNG carrier model,including two prismatic liquid tanks and a wave tank,was introduced.Different conditions were examined corresponding to different wave lengths,wave heights,wave heading angles,and tank loading rates.Finally,the effects of liquid tank loading rate on LNG ship motions and sloshing loading were analyzed,thereby showing that the SPH method can effectively provide useful indications for the design of liquid cargo ships.

Cr-containing wastewater treatment based on Cr self-catalysis: a critical review

Chromium (Cr), as a transition metal material with multiple redox states, has exhibited the catalysis toward Fenton-like reactions over a wide pH range. Although it is not sensible to add Cr reagents as catalysts due to its toxicity, it is highly promising to remediate Cr-containing wastewater through Cr-initiated advanced oxidation processes (Cr-initiated AOPs), which are clean and low-cost. Moreover, the widely concerned Cr-complexes, considered as obstacles in the remediation process, can be effectively destroyed by AOPs. Cr self-catalysis is defined as Cr species is both substrate and catalyst. However, the full understanding of Cr self-catalysis, including the generation of intermediates Cr(IV)/Cr(V), the synergetic effects with co-existing ions, and the accumulation of toxic Cr(VI), remains a challenge for the practical application of Cr-initiated AOPs. In this review, relevant researches on Cr self-catalysis during Cr-initiated AOPs are summarized. Specifically, the Cr-Fenton-like reaction, Cr substituted materials, and Cr-sulfite reactions are explored as key mechanisms contributing to Cr self-catalysis. Moreover, Cr transformation processes, including synchronously Cr removal, Cr redox reactions, and Cr(VI) accumulation, in AOPs-based synergistic systems are systematically analyzed. Detailed approaches for the detection of active species in AOPs-based systems are also presented. The primary objective of this review is to explore the application of AOPs for Cr-containing wastewater remediation based on Cr self-catalysis, and provide fundamental insights and valuable information for future research on Cr-initiated AOPs.

AtVQ25 promotes salicylic acid-related leaf senescence by fine-tuning the self-repression of AtWRKY53

Most mechanistic details of chronologically ordered regulation of leaf senescence are unknown.Regulatory networks centered on AtWRKY53 are crucial for orchestrating and integrating various senescence-related signals. Notably, AtWRKY53binds to its own promoter and represses transcription of AtWRKY53, but the biological significance and mechanism underlying this selfrepression remain unclear. In this study, we identified the VQ motif-containing protein AtVQ25as a cooperator of AtWRKY53. The expression level of AtVQ25 peaked at mature stage and was specifically repressed after the onset of leaf senescence. AtVQ25-overexpressing plants and atvq25 mutants displayed precocious and delayed leaf senescence, respectively. Importantly, we identified AtWRKY53 as an interacting partner of AtVQ25. We determined that interaction between AtVQ25 and AtWRKY53 prevented AtWRKY53from binding to W-box elements on the AtWRKY53promoter and thus counteracted the selfrepression of AtWRKY53. In addition, our RNA-sequencing data revealed that the AtVQ25-AtWRKY53 module is related to the salicylic acid(SA) pathway. Precocious leaf senescence and SA-induced leaf senescence in AtVQ25-overexpressing lines were inhibited by an SA pathway mutant, atsid2, and Nah G transgenic plants; AtVQ25-overexpressing/atwrky53 plants were also insensitive to SA-induced leaf senescence. Collectively, we demonstrated that AtVQ25 directly attenuates the self-repression of AtWRKY53 during the onset of leaf senescence,which is substantially helpful for understanding the timing of leaf senescence onset modulated by AtWRKY53.

Modeling load distribution for rural photovoltaic grid areas using image recognition

Expanding photovoltaic(PV)resources in rural-grid areas is an essential means to augment the share of solar energy in the energy landscape,aligning with the“carbon peaking and carbon neutrality”objectives.However,rural power grids often lack digitalization;thus,the load distribution within these areas is not fully known.This hinders the calculation of the available PV capacity and deduction of node voltages.This study proposes a load-distribution modeling approach based on remote-sensing image recognition in pursuit of a scientific framework for developing distributed PV resources in rural grid areas.First,houses in remote-sensing images are accurately recognized using deep-learning techniques based on the YOLOv5 model.The distribution of the houses is then used to estimate the load distribution in the grid area.Next,equally spaced and clustered distribution models are used to adaptively determine the location of the nodes and load power in the distribution lines.Finally,by calculating the connectivity matrix of the nodes,a minimum spanning tree is extracted,the topology of the network is constructed,and the node parameters of the load-distribution model are calculated.The proposed scheme is implemented in a software package and its efficacy is demonstrated by analyzing typical remote-sensing images of rural grid areas.The results underscore the ability of the proposed approach to effectively discern the distribution-line structure and compute the node parameters,thereby offering vital support for determining PV access capability.

High-dimensional uncertainty quantification of projectile motion in the barrel of a truck-mounted howitzer based on probability density evolution method

This paper proposed an efficient research method for high-dimensional uncertainty quantification of projectile motion in the barrel of a truck-mounted howitzer.Firstly,the dynamic model of projectile motion is established considering the flexible deformation of the barrel and the interaction between the projectile and the barrel.Subsequently,the accuracy of the dynamic model is verified based on the external ballistic projectile attitude test platform.Furthermore,the probability density evolution method(PDEM)is developed to high-dimensional uncertainty quantification of projectile motion.The engineering example highlights the results of the proposed method are consistent with the results obtained by the Monte Carlo Simulation(MCS).Finally,the influence of parameter uncertainty on the projectile disturbance at muzzle under different working conditions is analyzed.The results show that the disturbance of the pitch angular,pitch angular velocity and pitch angular of velocity decreases with the increase of launching angle,and the random parameter ranges of both the projectile and coupling model have similar influence on the disturbance of projectile angular motion at muzzle.

Comparison between sepsis-induced coagulopathy and sepsis-associated coagulopathy criteria in identifying sepsis-associated disseminated intravascular coagulation

BACKGROUND:Disseminated intravascular coagulation(DIC)is associated with increased mortality in sepsis patients.In this study,we aimed to assess the clinical ability of sepsis-induced coagulopathy(SIC)and sepsis-associated coagulopathy(SAC)criteria in identifying overt-DIC and preDIC status in sepsis patients.METHODS:Data from 419 sepsis patients were retrospectively collected from July 2018 to December 2022.The performances of the SIC and SAC were assessed to identify overt-DIC on days 1,3,7,or 14.The SIC status or SIC score on day 1,the SAC status or SAC score on day 1,and the sum of the SIC or SAC scores on days 1 and 3 were compared in terms of their ability to identify pre-DIC.The SIC or SAC status on day 1 was evaluated as a pre-DIC indicator for anticoagulant initiation.RESULTS:On day 1,the incidences of coagulopathy according to overt-DIC,SIC and SAC criteria were 11.7%,22.0%and 31.5%,respectively.The specificity of SIC for identifying overt-DIC was significantly higher than that of the SAC criteria from day 1 to day 14(P3 had a significantly higher sensitivity(72.00%)and area under the curve(AUC)(0.69)in identifying pre-DIC than did the SIC or SAC status(sensitivity:SIC status 44.00%,SAC status 52.00%;AUC:SIC status 0.62,SAC status 0.61).The sum of the SIC scores on days 1 and 3 had a higher AUC value for identifying the pre-DIC state than that of SAC(0.79 vs.0.69,P<0.001).Favorable effects of anticoagulant therapy were observed in SIC(adjusted hazard ratio[HR]=0.216,95%confidence interval[95%CI]:0.060–0.783,P=0.018)and SAC(adjusted HR=0.146,95%CI:0.041–0.513,P=0.003).CONCLUSION:The SIC and SAC seem to be valuable for predicting overt-DIC.The sum of SIC scores on days 1 and 3 has the potential to help identify pre-DIC.

Assembly of highly efficient aqueous light-harvesting system from sequence-defined peptoids for cytosolic microRNA detection

Precisely controlled spatial distributions of artificial light-harvesting systems in aqueous media are of significant importance for mimicking natural light-harvesting systems;however,they are often restrained by the solubility and the aggregation-caused quenching effect of the hydrophobic chromophores.Herein,we report one highly efficient artificial light-harvesting system based on peptoid nanotubes that mimic the hierarchical cylindrical structure of natural systems.The high crystallinity of these nanotubes enabled the organization of arrays of donor chromophores with precisely controlled spatial distributions,favoring an efficient Förster resonance energy transfer(FRET)process in aqueous media.This FRET system exhibits an extremely high efficiency of 98.6%with a fluorescence quantum yield of 40%and an antenna effect of 29.9.We further demonstrated the use of this artificial light-harvesting system for quantifying miR-210 within cancer cells.The fluorescence intensity ratio of donor to acceptor is linearly related to the concentration of intercellular miR-210 in the range of 3.3–156 copies/cell.Such high sensitivity in intracellular detection of miR-210 using this artificial light-harvesting system offers a great opportunity and pathways for biological imaging and detection,and for the further creation of microRNA(miRNA)toolbox for quantitative epigenetics and personalized medicine.

Uncertainty quantification of mechanism motion based on coupled mechanism—motor dynamic model for ammunition delivery system

In this paper,a dynamic modeling method of motor driven electromechanical system is presented,and the uncertainty quantification of mechanism motion is investigated based on this method.The main contribution is to propose a novel mechanism-motor coupling dynamic modeling method,in which the relationship between mechanism motion and motor rotation is established according to the geometric coordination of the system.The advantages of this include establishing intuitive coupling between the mechanism and motor,facilitating the discussion for the influence of both mechanical and electrical parameters on the mechanism,and enabling dynamic simulation with controller to take the randomness of the electric load into account.Dynamic simulation considering feedback control of ammunition delivery system is carried out,and the feasibility of the model is verified experimentally.Based on probability density evolution theory,we comprehensively discuss the effects of system parameters on mechanism motion from the perspective of uncertainty quantization.Our work can not only provide guidance for engineering design of ammunition delivery mechanism,but also provide theoretical support for modeling and uncertainty quantification research of mechatronics system.

Dynamic Modeling of Key Operating Parameters for Supercritical Circulating Fluidized Bed Units based on Data-Knowledge-Driven Method

To address the pressing need for intelligent and efficient control of circulating fluidized bed(CFB)units,it is crucial to develop a dynamic model for the key operating parameters of supercritical circulating fluidized bed(SCFB)units.Therefore,data-knowledge-driven dynamic model of bed temperature,load,and main steam pressure of the SCFB unit has been proposed.Firstly,a knowledge-driven method is employed to develop a dynamic model for key operating parameters of SCFB units.The model parameters are determined based on the operating data of the unit and continuously optimized in real time.Then,Bidirectional Long Short-Term Memory combined with Convolutional Neural Network and Attention Mechanism is utilized to build the dynamic model of bed temperature,load,and main steam pressure.Finally,a collaboration and integration method based on the critic weight method and the variation coefficient method is proposed to establish data-knowledge-driven model of key operating parameters for SCFB units.The model displays great accuracy and fitting ability compared with other methods and effectively captures the dynamic characteristics,which can provide a research basis for the design of intelligent flexible control mode of SCFB unit.
