Abnormal accumulation and metabolism of lipid droplets can lead to a variety of diseases.Polarity,a key parameter of the microenvironment,is closely associated with many diseases and dysfunctions in the body.It is important to elucidate the relationship between the physiological activity of lipid droplets(LDs)and the polarity of the microenvironment.In this work,based on push-pull mechanism,a fluorescent probe(E)-3-(5-(4-(diphenylamino)phenyl)thiophen-2-yl)-1-(2-hydroxyphenyl)prop-2-en-1-one(PPTH)with aggregation-induced emission(AIE)properties for the detection of polarity changes in cells was synthesized.PPTH not only visualize intracellular polarity fluctuation of iron death and inflammation but also distinguish between normal and fatty liver tissue.

The aerial parts of Mosla chinensis Maxim.and Mosla chinensis cv.'Jiangxiangru'(MCJ)are widely utilized in traditional Chinese medicine(TCM),known collectively as Xiang-ru.However,due to clinical effectiveness concerns and frequent misidentification,the original plants have increasingly been substituted by various species within the genera Elsholtzia and Mosla.The challenge in distinguishing between these genera arises from their similar morphological and metabolic profiles.To address this issue,our study introduced a rapid method for metabolic characterization,employing high-resolution mass spectrometry-based metabolomics.Through detailed biosynthetic and chemometric analyses,we pinpointed five phenolic compounds—salviaflaside,cynaroside,scutellarein-7-O-D-glucoside,rutin,and vicenin-2—among 203 identified compounds,as reliable chemical markers for distinguishing Xiang-ru from closely related Elsholtzia species.This methodology holds promise for broad application in the analysis of plant aerial parts,especially in verifying the authenticity of aromatic traditional medicinal plants.Our findings underscore the importance of non-volatile compounds as dependable chemical markers in the authentication process of aromatic traditional medicinal plants.

Pancreatic cancer is among the most malignant cancers,and thus early intervention is the key to better survival outcomes.However,no methods have been derived that can reliably identify early precursors of development into malignancy.Therefore,it is urgent to discover early molecular changes during pancreatic tumorigenesis.As aberrant glycosylation is closely associated with cancer progression,numerous efforts have been made to mine glycosylation changes as biomarkers for diagnosis;however,detailed glycoproteomic information,especially site-specific N-glycosylation changes in pancreatic cancer with and without drug treatment,needs to be further explored.Herein,we used comprehensive solid-phase chemoenzymatic glycoproteomics to analyze glycans,glycosites,and intact glycopeptides in pancreatic cancer cells and patient sera.The profiling of N-glycans in cancer cells revealed an increase in the secreted glycoproteins from the primary tumor of MIA PaCa-2 cells,whereas human sera,which contain many secreted glycoproteins,had significant changes of glycans at their specific glycosites.These results indicated the potential role for tumor-specific glycosylation as disease biomarkers.We also found that AMG-510,a small molecule inhibitor against Kirsten rat sarcoma viral oncogene homolog(KRAS)G12C mutation,profoundly reduced the glycosylation level in MIA PaCa-2 cells,suggesting that KRAS plays a role in the cellular glycosylation process,and thus glycosylation inhibition contributes to the anti-tumor effect of AMG-510.

Irrigation regimes modulate non-structural carbohydrate remobilization and improve grain filling in rice(Oryza sativa L.)by regulating starch metabolism

Recently developed ‘super’ rice cultivars with greater yield potentials often suffer from the problem of poor grain filling, especially in inferior spikelets. Here, we studied the activities of enzymes related to starch metabolism in rice stems and grains, and the microstructures related to carbohydrate accumulation and transportation to investigate the effects of different water regimes on grain filling. Two ‘super’ rice cultivars were grown under two irrigation regimes of well-watered(WW) and alternate wetting and moderate soil drying(AWMD). Compared with the WW treatment,the activities of ADP glucose pyrophosphorylase(AGPase), starch synthase(StSase) and starch branching enzyme(SBE), and the accumulation of non-structural carbohydrates(NSCs) in the stems before heading were significantly improved, and more starch granules were stored in the stems in the AWMD treatment. After heading, the activities of α-amylase, β-amylase, sucrose phosphate synthase(SPS) and sucrose synthase in the synthetic direction(SSs)were increased in the stems to promote the remobilization of NSCs for grain filling under AWMD. During grain filling, the enzymatic activities of sucrose synthase in the cleavage direction(SSc), AGPase, StSase and SBE in the inferior spikelets were increased, which promoted grain filling, especially for the inferior spikelets under AWMD.However, there were no significant differences in vascular microstructures. The grain yield and grain weight could be improved by 13.1 and 7.5%, respectively, by optimizing of the irrigation regime. We concluded that the low activities of key enzymes in carbon metabolism is the key limitation for the poor grain filling, as opposed to the vascular microstructures, and AWMD can increase the amount of NSC accumulation in the stems before heading, improve the utilization rate of NSCs after heading, and increase the grain filling, especially in the inferior spikelets, by altering the activities of key enzymes in carbon metabolism.

Avian infectious bronchitis(IB)is a highly contagious infectious disease caused by infectious bronchitis virus(IBV),which is prevalent in many countries worldwide and causes serious harm to the poultry industry.At present,many commercial IBV vaccines have been used for the prevention and control of IB;however,IB outbreaks occur frequently.In this study,two new strains of IBV,SX/2106 and SX/2204,were isolated from two flocks which were immunized with IBV H120 vaccine in central China.Phylogenetic and recombination analysis indicated that SX/2106,which was clustered into the GI-19 lineage,may be derived from recombination events of the GI-19 and GI-7 strains and the LDT3-A vaccine.Genetic analysis showed that SX/2204 belongs to the GVI-1 lineage,which may have originated from the recombination of the GI-13 and GVI-1 strains and the H120 vaccine.The virus cross-neutralization test showed that the antigenicity of SX/2106 and SX/2204 was different from H120.Animal experiments found that both SX/2106 and SX/2204 could replicate effectively in the lungs and kidneys of chickens and cause disease and death,and H120 immunization could not provide effective protection against the two IBV isolates.It is noteworthy that the pathogenicity of SX/2204 has significantly increased compared to the GVI-1 strains isolated previously,with a mortality rate up to 60%.Considering the continuous mutation and recombination of the IBV genome to produce new variant strains,it is important to continuously monitor epidemic strains and develop new vaccines for the prevention and control of IBV epidemics.

Device-to-device(D2D) communications allow direct connections among devices to lower transmission latency and network loads of the existing cellular mobile networks. To fully exploit the benefits of D2D communications, relay access is one of the critical challenges upon employing the optimal cooperative relay user equipments(UEs) for data forwarding. The existed relay access approaches usually neglect the inherent motivation that whether the relay is reluctant to forward the information of other UEs based on its status, such as limited battery, service cost and social ties. In this paper, we jointly investigate both the transmission delay and the reliability performance for designing a relay access scheme combined with the impacts of the physical layer and the social layer. Meanwhile, considering the user mobility and status, a dynamic relay access model is constructed with the utility function of the weighted average of latency and social trust level under the long-term constraints. To solve this problem, Lyapunov framework is proposed for converting it into a renewal-reward system towards infinite time horizon, and numerical results show the benefits of the proposed scheme compared with other schemes under dynamic scenarios, as it can lower the latency while guaranteeing the reliability of relay access.

中国农民价值观的迭代跃升,遵循新中国成立后的价值同质建构向改革开放后社会主义制度下的多元价值衍生、再向新时代多元基础上的价值共识诉求的转变趋势,并呈现出主体观向主体意识、利益观向公私协力、文明观向团体文明、生态观向尊重自然、道德观向现代情理的演化形态。以中国式现代化的五个基本特征为遵循,农民价值观迭代跃升的时代诉求聚焦于农民的主体观、利益观、文明观、生态观、道德观分别向自主自强、共赢共富、平等包容、和谐共生、向上向善的价值方向整体跃升。现阶段要从创新问题瞄准机制、遵循“指导-引领-人本”贯通原则、顺应“传承-借鉴-淬炼”递进理路、强化“政府-组织-农民”联动保障入手,进行系统整体推进,以实现全方位的价值跃升目标。

储气库建设对供气管网安全、平衡供气具有重要战略意义,在天然气调峰中起到举足轻重的作用。目前,储气库工艺存在压缩机组中体放空对空排放安全隐患和增压气润滑油超标、采出气轻烃超标、甘醇再生系统尾气对空排放、集输管线积盐腐蚀等问题。针对中体放空,设置放空引射装置,将排放物引至火炬集中排放,避免可燃气体站内聚集的隐患;针对润滑油含量超标(体积比为16×10^(−6)),设置高压除油器,降低增压气油含量,消除二次污染;通过对脱水装置吸收塔和再生装置优化,避免甘醇起泡失活;通过对甘醇再生气系统改造,实现再生气回收利用,降低环境污染,节约能耗。
