High-throughput“read-on-ski”automated imaging and label-free detection system for toxicity screening of compounds using personalised human kidney organoids

Organoid models are used to study kidney physiology,such as the assessment of nephrotoxicity and underlying disease processes.Personalized human pluripotent stem cell-derived kidney organoids are ideal models for compound toxicity studies,but there is a need to accelerate basic and translational research in the field.Here,we developed an automated continuous imaging setup with the“read-on-ski”law of control to maximize temporal resolution with minimum culture plate vibration.High-accuracy performance was achieved:organoid screening and imaging were performed at a spatial resolution of 1.1µm for the entire multi-well plate under 3 min.We used the in-house developed multi-well spinning device and cisplatin-induced nephrotoxicity model to evaluate the toxicity in kidney organoids using this system.The acquired images were processed via machine learning-based classification and segmentation algorithms,and the toxicity in kidney organoids was determined with 95%accuracy.The results obtained by the automated“read-on-ski”imaging device,combined with label-free and non-invasive algorithms for detection,were verified using conventional biological procedures.Taking advantage of the close-to-in vivo-kidney organoid model,this new development opens the door for further application of scaled-up screening using organoids in basic research and drug discovery.

Dry eye is a common ocular disease that results in discomfort and impaired vision,impacting an individual’s quality of life.A great number of drugs administered in eye drops to treat dry eye are poorly soluble in water and are rapidly eliminated from the ocular surface,which limits their therapeutic effects.Therefore,it is imperative to design a novel drug delivery system that not only improves the water solubility of the drug but also prolongs its retention time on the ocular surface.Herein,we develop a copolymer from mono-functional POSS,PEG,and PPG(MPOSS-PEG-PPG,MPEP)that exhibits temperature-sensitive sol-gel transition behavior.This thermo-responsive hydrogel improves the water solubility of FK506 and simultaneously provides a mucoadhesive,long-acting ocular delivery system.In addition,the FK506-loaded POSS hydrogel possesses good biocompatibility and significantly improves adhesion to the ocular surface.In comparison with other FK506 formulations and the PEG-PPG-FK506(F127-FK506)hydrogel,this novel MPOSS-PEG-PPG-FK506(MPEP-FK506)hydrogel is a more effective treatment of dry eye in the murine dry eye model.Therefore,delivery of FK506 in this POSS hydrogel has the potential to prolong drug retention time on the ocular surface,which will improve its therapeutic efficacy in the management of dry eye.

Accelerated Primal-Dual Projection Neurodynamic Approach With Time Scaling for Linear and Set Constrained Convex Optimization Problems

The Nesterov accelerated dynamical approach serves as an essential tool for addressing convex optimization problems with accelerated convergence rates.Most previous studies in this field have primarily concentrated on unconstrained smooth con-vex optimization problems.In this paper,on the basis of primal-dual dynamical approach,Nesterov accelerated dynamical approach,projection operator and directional gradient,we present two accelerated primal-dual projection neurodynamic approaches with time scaling to address convex optimization problems with smooth and nonsmooth objective functions subject to linear and set constraints,which consist of a second-order ODE(ordinary differential equation)or differential conclusion system for the primal variables and a first-order ODE for the dual vari-ables.By satisfying specific conditions for time scaling,we demonstrate that the proposed approaches have a faster conver-gence rate.This only requires assuming convexity of the objective function.We validate the effectiveness of our proposed two accel-erated primal-dual projection neurodynamic approaches through numerical experiments.

Experimental study on reactions between alkaline basaltic melt and orthopyroxenes: constraints on the evolution of lithospheric mantle in the North China Craton

The experimental results of the reactions between an alkaline basaltic melt and mantle orthopyroxenes under high-temperature and high-pressure conditions of 1300–1400℃ and 2.0–3.0 GPa using a six-anvil apparatus are reported in this paper.The reactions are proposed to simulate the interactions between melts from the asthenospheric mantle and the lithospheric mantle.The starting melt in the experiments was made from the alkaline basalt occurring in Fuxin,Liaoning Province,and the orthopyroxenes were separated from the mantle xenoliths in Damaping,Hebei Province.The results show that clinopyroxenes were formed in all the reactions between the alkaline basaltic melt and orthopyroxenes under the studied P–T conditions.The formation of clinopyroxene in the reaction zone is mainly controlled by dissolution–crystallization,and the chemical compositions of the reacted melt are primarily infl uenced by the diff usion eff ect.Temperature is the most important parameter controlling the reactions between the melt and orthopyroxenes,which has a direct impact on the melting of orthopyroxenes and the diff usion of chemical components in the melt.Temperature also directly controls the chemical compositions of the newly formed clinopyroxenes in the reaction zone and the reacted melt.The formation of clinopyroxenes from the reactions between the alkaline basaltic melt and orthopyroxenes can result in an increase of CaO and Al_(2)O_(3) contents in the rocks containing this mineral.Therefore,the reactions between the alkaline basaltic melt from the asthenospheric mantle and orthopyroxenes from the lithospheric mantle can lead to the evolution of lithospheric mantle in the North China Craton from refractory to fertile with relatively high CaO and Al 2 O 3 contents.In addition,the reacted melts in some runs were transformed from the starting alkaline basaltic into tholeiitic after reactions,indicating that tholeiitic magma could be generated from alkaline basaltic one via reactions between the latter and orthopyroxene.

Chaotic CS Encryption:An Efficient Image Encryption Algorithm Based on Chebyshev Chaotic System and Compressive Sensing

Images are the most important carrier of human information. Moreover, how to safely transmit digital imagesthrough public channels has become an urgent problem. In this paper, we propose a novel image encryptionalgorithm, called chaotic compressive sensing (CS) encryption (CCSE), which can not only improve the efficiencyof image transmission but also introduce the high security of the chaotic system. Specifically, the proposed CCSEcan fully leverage the advantages of the Chebyshev chaotic system and CS, enabling it to withstand various attacks,such as differential attacks, and exhibit robustness. First, we use a sparse trans-form to sparse the plaintext imageand then use theArnold transformto perturb the image pixels. After that,we elaborate aChebyshev Toeplitz chaoticsensing matrix for CCSE. By using this Toeplitz matrix, the perturbed image is compressed and sampled to reducethe transmission bandwidth and the amount of data. Finally, a bilateral diffusion operator and a chaotic encryptionoperator are used to perturb and expand the image pixels to change the pixel position and value of the compressedimage, and ultimately obtain an encrypted image. Experimental results show that our method can be resistant tovarious attacks, such as the statistical attack and noise attack, and can outperform its current competitors.

The variation in basal channels and basal melt rates of Pine Island Ice Shelf

In recent years,there has been a significant acceleration in the thinning,calving and retreat of the Pine Island Ice Shelf(PIIS).The basal channels,results of enhanced basal melting,have the potential to significantly impact the stability of the PIIS.In this study,we used a variety of remote sensing data,including Landsat,REMA DEM,ICESat-1 and ICESat-2 satellite altimetry observations,and Ice Bridge airborne measurements,to study the spatiotemporal changes in the basal channels from 2003 to 2020 and basal melt rate from 2010 to 2017 of the PIIS under the Eulerian framework.We found that the basal channels are highly developed in the PIIS,with a total length exceeding 450 km.Most of the basal channels are ocean-sourced or groundingline-sourced basal channels,caused by the rapid melting under the ice shelf or near the groundingline.A raised seabed prevented warm water intrusion into the eastern branch of the PIIS,resulting in a lower basal melt rate in that area.In contrast,a deepsea trough facilitates warm seawater into the mainstream and the western branch of the PIIS,resulting in a higher basal melt rate in the main-stream,and the surface elevation changes above the basal channels of the mainstream and western branch are more significant.The El Ni?o event in 2015–2016 possibly slowed down the basal melting of the PIIS by modulating wind field,surface sea temperature and depth seawater temperature.Ocean and atmospheric changes were driven by El Ni?o,which can further explain and confirm the changes in the basal melting of the PIIS.

Spatial patterns of zooplankton abundance,biovolume,and size structure in response to environmental variables:a case study in the Yellow Sea and East China Sea

The Yellow Sea(YS)and East China Sea(ECS)are highly dynamic marginal seas of the northwestern Pacific Ocean.To gain an in-depth understanding of zooplankton community structure,zooplankton abundance,biovolume,and size structure in summer 2017 in the YS and ECS were assessed using ZooScan imaging analysis.Zooplankton abundance and biovolume ranged 2.94–1187.14 inds./m^(3)and 3.13–3438.51 mm^(3)/m^(3),respectively.Based on the biovolume data of the categorized size classes of 26 identified taxonomic groups,the zooplankton community was classified into five groups,and each group was coupled with distinctive oceanographic features.Under the influence of the Yellow Sea Cold Water Mass,the Yellow Sea offshore group featured the lowest bottom temperature(10.84±3.42℃)and the most abundant Calanoids(mainly in the 2–3 mm size class).In the Yellow Sea inshore group,Hydrozoans showed the largest biovolume and dominated in the 3–4-mm and>5-mm size classes.The East China Sea offshore group,which was affected by the Kuroshio Branch Current,featured high temperature and salinity,and the lowest bottom dissolved oxygen(2.58±0.5 mg/L).The lowest values of zooplankton abundance and biovolume in the East China Sea offshore group might be attributed to the bottom dissolved oxygen contents.The East China Sea inshore group,which was mainly influenced by the Zhejiang-Fujian Coastal Current and Changjiang Diluted Water,was characterized by high chlorophyll a and the largest biovolume of carnivorous Siphonophores(280.82±303.37 mm^(3)/m^(3)).The Changjiang River estuary offshore group showed the most abundant Cyclopoids,which might be associated with the less turbid water mass in this region.Seawater temperature was considered the most important factor in shaping the size compositions of Calanoids in different groups.

Rock mass quality prediction on tunnel faces with incomplete multi-source dataset via tree-augmented naive Bayesian network

Rock mass quality serves as a vital index for predicting the stability and safety status of rock tunnel faces.In tunneling practice,the rock mass quality is often assessed via a combination of qualitative and quantitative parameters.However,due to the harsh on-site construction conditions,it is rather difficult to obtain some of the evaluation parameters which are essential for the rock mass quality prediction.In this study,a novel improved Swin Transformer is proposed to detect,segment,and quantify rock mass characteristic parameters such as water leakage,fractures,weak interlayers.The site experiment results demonstrate that the improved Swin Transformer achieves optimal segmentation results and achieving accuracies of 92%,81%,and 86%for water leakage,fractures,and weak interlayers,respectively.A multisource rock tunnel face characteristic(RTFC)dataset includes 11 parameters for predicting rock mass quality is established.Considering the limitations in predictive performance of incomplete evaluation parameters exist in this dataset,a novel tree-augmented naive Bayesian network(BN)is proposed to address the challenge of the incomplete dataset and achieved a prediction accuracy of 88%.In comparison with other commonly used Machine Learning models the proposed BN-based approach proved an improved performance on predicting the rock mass quality with the incomplete dataset.By utilizing the established BN,a further sensitivity analysis is conducted to quantitatively evaluate the importance of the various parameters,results indicate that the rock strength and fractures parameter exert the most significant influence on rock mass quality.

Current electrochemical energy storage technology has evolved a variety of rechargeable battery systems.Recently,the resource shortage of raw materials in commercially available lithium-ion batteries has attracted widespread attention.The requirements to meet resourcefulness,sustainability,safety,and high energy density have motivated the development of rechargeable magnesium-ion batteries(RMBs).Although RMBs have made significant progress so far,there are still many obstacles to practical orientation.We systematically summarize the significant progress and the latest research on RMBs,including Mg^(2+)-conducting electrolytes,Mg^(2+)-storage cathodes,and Mg-based anodes.In this review,we mainly introduce the properties and features of various Mg^(2+)-conductive electrolytes,the mainstream cathode materials,and their respective Mg^(2+)-storage mechanisms,as well as the Mg metal(or alloy)anodes and the corresponding modification strategies.Finally,the future directions in various key components for RMBs are highlighted.
