Unveiling an ancient whole-genome duplication event in Stentor,the model unicellular eukaryotes

Whole-genome duplication(WGD) events are widespread across eukaryotes and have played a significant role in moulding the genetic architectures of diverse organisms. In the present study, the newly sequenced genome of a giant ciliated protist, Stentor roeselii, provides an opportunity for the analysis of the collinearity and retention of reciprocal best-hit genes between two Stentor species. As a main result, we have unveiled a previously undetected ancient WGD event shaping the genome of its congener, Stentor coeruleus, a model protist used in cytological and evolutionary studies. Genomes of two congeners, S. coeruleus and S. roeselii, are compared and analyzed, revealing that:(i)the former exhibits a much higher retention rate of colinear-gene pairs(28%) than does S. roeselii, and in S. coeruleus, 75% of genes that have a RBH hit in S. roeselii, have paralogs with high amino-acid identity, consistent with a WGD event in the lineage leading to S. coeruleus;(ii) the S. roeselii genome possesses extremely short intergenic regions, implying that the lengths of intergenic regions are under strong selection;(iii) the unique characteristics of introns may have been shaped in the common ancestor of heterotrichs;(iv) gene families that play a role in activities of multiple protein kinases and voltage-gated ion channels expanded rapidly in the ancestor of both taxa, possibly relating to the remarkable regenerative ability in Stentor. This study offers new insights into the evolutionary dynamics of ciliate genomes,with implications for understanding of the processes underlying the evolution of genomic complexity.

Distribution and Hydrogeochemical Characteristic of High Iodine Groundwater in Oasis Zone in the Tarim Basin in Xinjiang, China

Groundwater is the main water supply source in the Tarim Basin in China. Endemic disease caused by high iodine(I) groundwater in the Tarim Basin was reported previously. Therefore, it is crucial to systematically identify the distribution and genesis of groundwater I. Based on hydrochemical analysis of 717 groundwater samples collected in 2015 – 2018, spatial distribution and hydrogeochemistry characteristic of high I groundwater in different aquifers were analyzed. Results showed that groundwater I ranged between 100.00 μg/L) accounted for 7.25% of the total samples. Horizontally,groundwater I significantly increased from recharge zone(RZ) to transition zone(TZ) and to evaporation zone(EZ). Vertically, groundwater in shallow confined aquifer(SCA) had the greatest I concentration, followed by single-structure phreatic aquifer(SSPA), phreatic aquifer in confined groundwater area(PACGA), while groundwater in deep confined aquifer(DCA) generally had low I concentration. Groundwater I enrichment in SSPA was mainly affected by organic matter(OM) decomposition and that in SCA was mainly affected by evaporite mineral dissolution, OM decomposition under alkaline environment. While I enrichment in groundwater of PACGA was restrained under neutral environment. Lacustrine sedimentary environment was crucial for I enrichment in groundwater. Besides, fine-grained lithology of aquifer, smooth topographic slope, shallow buried depth of groundwater, weak alkaline and reducing environment, reductive dissolution of iron oxide/hydroxide minerals and OM decomposition were advantageous to I enrichment in groundwater.

NK细胞联合西妥昔单抗对大肠癌细胞ADCC作用的研究

目的比较3株肠癌细胞株(LOVO、SW480、SW620)与自然杀伤(natural killer,NK)细胞、西妥昔单抗(Cetuximab)混合作用之后,NK细胞抗体依赖的细胞介导的细胞毒作用(antibody dependent cell cytotoxicity,ADCC),探讨Cetuximab抗肿瘤过程中ADCC的作用。方法免疫组织化学方法检测3种不同肠癌细胞系(LOVO、SW480、SW620)的EGFR表达水平;MTT法检测6种不同浓度Cetuximab作用48h后肿瘤细胞的生长抑制情况;NK细胞与Cetuximab包被的肿瘤细胞共孵育,LDH法检测NK细胞的ADCC功能变化。结果免疫组织化学结果显示,3株细胞EGFR表达:LOVO(),SW480(+),SW620(-);Cetuximab对LOVO细胞和SW480细胞的生长抑制呈剂量依赖,对SW620细胞无抑制作用;LOVO细胞和SW480细胞经Cetuximab处理后,NK细胞的ADCC作用均增强且有统计学意义(P<0.05)。结论单独的Cetuximab可抑制EGFR阳性的LOVO细胞和SW480细胞,而对EGFR阴性的SW620细胞生长抑制作用不明显,所选的2株EGFR阳性肿瘤细胞都可被Cetuximab所介导ADCC作用所抑制,高EGFR表达的肿瘤细胞更加敏感。

目的对比分析腹腔镜术后早期肠内营养和开放手术后肠外营养对机体恢复情况和免疫功能的影响。方法将60例胃癌患者随机平均分为两组,观察组行腹腔镜远端胃大部切除术和早期肠内营养,对照组行开放手术和肠外营养。比较分析两组患者术后营养支持期间的肠胃功能恢复情况、营养状况、免疫功能和住院天数。测定术前和术后1天、7天的肝功能、电解质、营养指标(体重、血清总蛋白、血清清蛋白、前清蛋白)和免疫指标(外周血CD4、CD8、CD4/CD8)。结果观察组患者术后排气时间短于对照组(P<0.05);术后第7天肝功能损害比对照组轻,而血清前清蛋白、CD4、CD4/CD8均高于对照组(P<0.05);术后平均住院时间少于对照组。结论腹腔镜手术和早期肠内营养有利于胃癌患者术后的肠胃功能恢复,对改善机体营养状况和促进免疫功能恢复具有积极意义,能缩短术后住院天数。

Solar-driven overall conversion of CO_(2)and H_(2)O into fuels and chemicals shows an ultimate strategy for carbon neutrality yet remains a huge ***,an integrated photocatalytic redox architecture of Zn NPs/GaN Nanowires(NWs)/Si is explored for light-driven overall conversion of CO_(2)and H_(2)O into CH_(4)and H_(2)O_(2)simultaneously without any external sacrificial agents and *** as-designed architecture affords a benchmark CH_(4)activity of 189 mmol gcat^(-1)h^(-1)with a high selectivity of 93.6%,in the synchronized formation of H_(2)O_(2)at a considerable rate of 25 m g^(-1)h^(-1).Moreover,a considerable turnover number of 27,280 mol CH_(4)per mol Zn was achieved over a long-term operation of 80 *** operando spectroscopic characterizations,isotope experiments,and density functional theory calculations,it is unraveled that Zn sites are synergetic with GaN to drive CO_(2)-to-CH_(4)conversion with a lowered energy barrier of 0.27 eV while inhibiting hydrogen evolution reaction with a relatively high energy barrier of 0.93 ***,owing to the unique surface properties of GaN,water is split into*OH and*H,followed by the formation of H_(2)O_(2)because of the alleviated adsorption strength of*OH by Zn ***,the hierarchical architecture enables the achievement of high activity and high selectivity of CH_(4)from CO_(2)reduction in distilled water along with the generation of H_(2)O_(2).This work provides an integrated photocatalytic redox architecture for the synchronized production of CH_(4)and H_(2)O_(2)with the only inputs of CO_(2),distilled water,and light.

EZH2-dependent myelination following sciatic nerve injury

T cell interactions with microglia in immune-inflammatory processes of ischemic stroke

Gastric Carcinoma(GC)is a highly fatal malignant tumor with a poor *** elevated mortality rates are primarily due to its proclivity for late-stage *** the metabolic interactions between tumor microenvironment and the systemic bloodstream could help to clearly understand the mechanisms and identify precise biomarkers of tumor growth,proliferation,and *** this study,an integrative approach that combines plasma metabolomics with mass spectrometry imaging of tumor tissue was developed to investigate the global metabolic landscape of GC tumorigenesis and *** results showed that the oxidized glutathione to glutathione ratio(GSSH/GSH)became increased in non-distal metastatic GC(M0),which means an accumulation of oxidative stress in tumor ***,it was found that the peroxidation of polyunsaturated fatty acids,such as 9,10-EpOMe,9-HOTrE,etc.,were accelerated in both plasma and tumor tissues of distal metastatic GC(M1).These changes were further confirmed the potential effect of CYP2E1 and GGT1 in metastatic potential of GC by mass spectrometry imaging(MSI)and immunohistochemistry(IHC).Collectively,our findings reveal the integrated multidimensional metabolomics approach is a clinical useful method to unravel the bloodtumor metabolic crosstalk,illuminate reprogrammed metabolic networks,and provide reliable circulating biomarkers.
