Effects of L-arginine and N~ω-nitro-L-arginine methylester on learning and memory and α7 nAChR expression in the prefrontal cortex and hippocampus of rats

Nitric oxide （NO） is a novel type of neurotransmitter that is closely associated with synaptic plasticity, learning and memory. In the present study, we assessed the effects of Larginine and NnitroL arginine methylester （LNAME, a nitric oxide synthase inhibitor） on learning and memory. Rats were assigned to three groups receiving intracerebroventricular injections of LArg （the NO precursor）, LNAME, or 0.9% NaCI （control）, once daily for seven con secutive days. Twelve hours after the last injection, they underwent an electric shockpaired Y maze test. Twentyfour hours later, the rats＇ memory of the safe illuminated arm was tested. After that, the levels of NO and a7 nicotinic acetylcholine receptor （a7 nAChR） in the prefrontal cortex and hippocampus were assessed using an NO assay kit, and immunohistochemistry and Western blots, respectively. We found that, compared to controls, LArgtreated rats received fewer foot shocks and made fewer errors to reach the learning criterion, and made fewer errors during the memorytesting session. In contrast, LNAMEtreated rats received more foot shocks and made more errors than controls to reach the learning criterion, and made more errors during the memorytesting session. In parallel, NO content in the prefrontal cortex and hippocampus was higher in LArgtreated rats and lower inLNAME rats, compared to controls. Similarly, （]7 nAChR immunoreactivity and protein expression in the prefrontal cortex and hippocampus were higher in LArgtreated rats and lower in LNAME rats, compared to controls. These results suggest that the modulation of NO content in the brain correlates with a7 nAChR distribution and expression in the prefrontal cortex and hippocampus, as well as with learning and memory performance in the Ymaze.

Synthesis and antitumor activity of nitric oxide releasing derivatives of AT1 antagonist

A series of novel nitric oxide-donating derivatives （7a-e, 8a-e） were synthesized by coupling furoxan and nitric oxide with irbesartan analogue and their cytotoxicity against BEL7402 cells in vitro were evaluated by MTT method. It was found that 8c exhibits the most cytotoxic activities with ICso value of 12.5 μmol/L. The hybrids of AT1 antagonist and nitric oxide donor appear to have beneficial effects on antitumor.

Endoscopic diagnosis of gastrointestinal graft-versus-host disease

AIM:To evaluate the diagnostic value of endoscopy in patients with gastrointestinal graft-versus-host disease (GI GVHD). METHODS:We identified 8 patients with GI GVHD following allogeneic hematopoietic stem cell trans-plantation (HSCT). GVHD was defined histologically as the presence of gland apoptosis, not explained by other inflammatory or infectious etiologies. RESULTS:The symptoms of GI GVHD included anorexia, nausea, vomiting, watery diarrhea, abdominal pain, GI bleeding, etc. Upper endoscopic appearance varied from subtle mucosal edema, hyperemia, erythema to obvious erosion. Colonoscopic examination showed diffuse edema, hyperemia, patchy erosion, scattered ulcer, sloughing and active bleeding. Histological changes in GI GVHD included apoptosis of crypt epithelial cells, dropout of crypts, and lymphocytic infiltration in epithelium and lamina propria. The involvement of stomach and rectocolon varied from diffuse to focal. CONCLUSION:Endoscopy may play a significant role in early diagnosis of GI GVHD patients following allogeneic HSCT, and histologic examination of gastrointestinal biopsies is needed to confirm the final diagnosis.

Direct fabrication of conductive LB films composed of N-docosylpyridinium-TCNQ

Langmuir-Blodgett (LB) films of N-docosylpyridinium-TCNQ (NDP-TCNQ) were prepared in air. The films deposited at the room temperature showed in-plane conductivity of 4×10-3 ***-1 and its absorption spectra in UV and IR regions resembled those for the films composed of mixed-valence TCNQ salts of NDP-(TCNQ)2 rather than for the NDP-TCNQ LB films preparaed under protection of nitrogen. In the case that the films left in a warm environment after each dipping cycle, the absorption in the corresponding region changed. Spectral analysis and XPS study revealed that more TCNQ molecules were produced. The oxidation of TCNQ- in air was considered to be the origin of neutral TCNQ formed.

Main controlling factor and mechanism of gas-in-place content of the Lower Cambrian shale from different sedimentary facies in the western Hubei area, South China

The Lower Cambrian shale gas in the western Hubei area,South China has a great resource prospect,but the gas-in-place(GIP)content in different sedimentary facies varies widely,and the relevant mechanism has been not well *** the present study,two sets of the Lower Cambrian shale samples from the Wells YD4 and YD5 in the western Hubei area,representing the deep-water shelf facies and shallowwater platform facies,respectively,were investigated on the differences of pore types,pore structure and methane adsorption capacity between them,and the main controlling factor and mechanism of their methane adsorption capacities and GIP contents were *** results show that the organic matter(OM)pores in the YD4 shale samples are dominant,while the inorganic mineral(IM)pores in the YD5 shale samples are primary,with underdeveloped OM *** pore specific surface area(SSA)and pore volume(PV)of the YD4 shale samples are mainly from micropores and mesopores,respectively,while those of the YD5 shale samples are mainly from micropores and macropores,*** methane adsorption capacity of the YD4 shale samples is significantly higher than that of the YD5 shale samples,with a maximum absolute adsorption capacity of 3.13 cm^(3)/g and 1.31 cm^(3)/g in average,*** with the shallow-water platform shale,the deep-water shelf shale has a higher TOC content,a better kerogen type and more developed OM pores,which is the main mechanism for its higher adsorption *** GIP content models based on two samples with a similar TOC content selected respectively from the Wells YD4 and YD5 further indicate that the GIP content of the deep-water shelf shale is mainly 34 m^(3)/t within a depth range of 1000—4000 m,with shale gas exploration and development potential,while the shallow-water platform shale has normally a GIP content of<1 m^(3)/t,with little shale gas *** the geological and geochemical conditions of shale gas formation a

针对现有电液伺服系统的非线性、参数不确定导致轨迹跟踪精度低的问题,提出了一种非线性自适应输出反馈控制器。分析了液压系统的非线性模型,为控制器设计提供理论基础。非线性自适应输出反馈控制器包含自适应线性扩展状态观测器(Linear Extended State Observer, LESO)和非线性扰动观测器(Nonlinear Disturbance Observer, NDO),用来估计系统中不可策略状态以及惯性载荷动力学中的失配扰动。实验结果表明,所提出的非线性自适应输出反馈鲁棒控制器可有效解决PI控制器存在的谐振问题。

Rational design of superior catalysts from topological semimetals with nontrivial energy window
