BACKGROUND Pyroptosis is an inflammatory form of programmed cell death,which has been shown to be related to the prognosis of many tumors.However,its role in gastric cancer(GC)is not fully understood.AIM To evaluate the expression of pyroptosis-related genes in GC and its correlation with prognosis.METHODS We constructed prognostic multigene markers of differentially expressed genes associated with pyroptosis by least absolute contraction and selection operator Cox regression.The risk model was analyzed by Kaplan-Meier curve,two-sided log-rank test and functional enrichment analysis.RESULTS Sixty-three pyroptosis-related genes were differentially expressed in tumor tissues and adjacent nontumor tissues.Based on these differentially expressed genes,5 gene signature were constructed and all GC patients were classified into two risk groups.Kaplan-Meier survival curve showed that the overall survival(OS)of patients in the high-risk group was significantly lower than that of the low-risk group.Multivariate Cox regression analyses showed that the risk score was an independent risk factor for OS.Receiver operating characteristic curve analysis confirmed the predictive ability of the model.External validation indicated increased OS in the low-risk group.The immune function and immune cell scores of the high-risk group were generally higher than those of the low-risk group.CONCLUSION Pyroptosis-related genes play a significant role in tumor immune microenvironment.This novel model,which contains 5 pyroptosis-related genes,is an independent predicting factor for OS in GC patients,and may help to evaluate the prognosis of GC.

Performance of Ti6Al4V fabricated by electron beam and laser hybrid preheating and selective melting strategy

Electron beam selective melting(EBM)and selective laser melting(SLM)are regarded as significant manufacturing processes for near-net-shaped Ti6Al4V components.Generally,in the conventional EBM process,preheating is necessitated to avoid"smoke"caused by the charging of electrons.In the conventional SLM process,laser as an energy source without the risk of"smoke"can be employed to melt metal powder at low temperatures.However,because of the low absorption rate of laser,the powder bed temperature cannot reach a high level.It is difficult to obtain as-built TiAl4V with favorable comprehensive properties via conventional EBM or SLM.Hence,two types of electron beam and laser hybrid preheating(EB-LHP)combined with selective melting strategies are proposed.Using laser to preheat powder allows EBM to be performed at a low powder bed temperature(EBM-LT),whereas using an electron beam to preheat powder allows SLM to be performed at a high powder bed temperature(SLM-HT).Ti6Al4V samples are fabricated using two different manufacturing strategies(i.e.,EBM-LT and SLM-HT)and two conventional processes,i.e.,EBM at a high powder bed temperature(EBM-HT)and SLM at a low powder bed temperature(SLM-LT).The temperature-dependent surface quality,microstructure,density,and mechanical properties of the as-built Ti6Al4V samples are characterized and compared.Results show that EBM-LT Ti6Al4V exhibits a higher ultimate tensile strength(981±43 MPa)and a lower elongation(12.2%±2.3%)than EBM-HT Ti6Al4V owing to the presence ofα′martensite.The SLM-HT Ti6Al4V possesses the highest ultimate tensile strength(1,059±62 MPa)and an elongation(14.8%±4.0%)comparable to that of the EBM-HT Ti6Al4V(16.6%±1.2%).

Developing mathematical models for high Knudsen number(Kn)flow for isotopic gas fractionation in tight sedimentary rocks is still challenging.In this study,carbon isotopic reversals(δ^(13)C_(1)>δ^(13)C_(2))were found for four Longmaxi shale samples based on gas degassing experiments.Gas in shale with higher gas content exhibits larger reversal.Then,a mathematical model was developed to simulate the carbon isotopic reversals of methane and ethane.This model is based on these hypotheses:(i)diffusion flow is dominating during gas transport process;(ii)diffusion coefficients are nonlinear depending on concentration gradient.Our model not only shows a good agreement with isotopic reversals,but also well predicts gas production rates by selecting appropriate exponents m and m^(*) of gas pressure gradient,where m is for ^(12)C and m^(*)is for ^(13)C.Moreover,the(m−m^(*))value has a positive correlation with fractionation level.(m1−m1^(*))of methane are much higher than that of ethane.Finally,the predicted carbon isotopic reversal magnitude(δ^(13)C_(1)−δ^(13)C_(2))exhibits a positive relationship with total gas content since gas in shale with higher gas content experiences a more extensive high Kn number diffusion flow.As a result,our model demonstrates an impressive agreement with the experimental carbon isotopic reversal data.

Synergistic effect of Zr and Mo on precipitation and high-temperature properties of Al-Si-Cu-Mg alloys

This study focuses on finding a solution to the sharp decline in mechanical properties of Al-Si-Cu-Mg alloys due to rapid coarsening of traditional intermediate phases at high temperature.A new type of modified al oy,to be used in automobile engines at high temperatures,was prepared by adding Zr and Mo into Al-Si-Cu-Mg alloy.The synergistic effects of Zr and Mo on the microstructure evolution and high-temperature mechanical properties were studied.Results show that the addition of Zr and Mo generates a series of intermetallic phases dispersed in the alloy.They can improve the strength of the alloy by hindering dislocation movement and crack propagation.In addition,some nano-strengthened phases show coherent interfaces with the matrix and improve grain refinement.The addition of Mo greatly improves the heat resistance of the alloy.The extremely low diffusivity of Mo enables it to improve the thermal stability of the intermetallic phases,inhibit precipitation during aging,reduce the size of the precipitates,and improve the heat resistance of the alloy.

Risk factors for brain injury in patients with exertional heatstroke:A 5-year experience

Purpose:Minimal data exist on brain injury in patients with exertional heatstroke(EHS)in developing country.In this study,we explored the risk factors for brain injury induced by EHS 90-day after onset.Methods:A retrospective cohort study of patients with EHS was conducted in the intensive care unit of the General Hospital of Southern Theater Command of PLA in China from April 2014 to June 2019.Patients were divided into non-brain injury(fully recovered)and brain injury groups(comprising deceased patients or those with neurological sequelae).The brain injury group was further subdivided into a death group and a sequela group for detailed analysis.General information,neurological performance and information on important organ injuries in the acute stage were recorded and analysed.Multivariable logistic regression was used to identify risk factors for brain injury after EHS and mortality risk factors for brain injury,and Kaplan-Meier survival curve was used to evaluate the effect of the neurological dysfunction on survival.Results:Out of the 147 EHS patients,117 were enrolled,of which 96(82.1%)recovered,13(11.1%)died,and 8(6.8%)experienced neurological sequelae.Statistically significant differences were found between non-brain injury and brain injury groups in age,hypotension,duration of consciousness disorders,time to drop core body temperature below 38.5℃,lymphocyte counts,platelet counts,procalcitonin,alanine aminotransferase,aspartate aminotransferase,creatinine,cystatin C,coagulation parameters,international normalized ratio,acute physiology and chronic health evaluation II scores,sequential organ failure assessment(SOFA)scores,and Glasgow coma scale scores(all p<0.05).Multivariate logistic regression showed that age(OR=1.090,95%CI:1.02-1.17,p=0.008),time to drop core temperature(OR=8.223,95%CI:2.30-29.40,p=0.001),and SOFA scores(OR=1.676,95%CI:1.29-2.18,p<0.001)are independent risk factors for brain injury induced by EHS.The Kaplan-Meier curves suggest significantly prolonged survival(p<0.001)in patients with

Development and validation of a machine learning-based early prediction model for massive intraoperative bleeding in patients with primary hepatic malignancies

BACKGROUND Surgical resection remains the primary treatment for hepatic malignancies,and intraoperative bleeding is associated with a significantly increased risk of death.Therefore,accurate prediction of intraoperative bleeding risk in patients with hepatic malignancies is essential to preventing bleeding in advance and providing safer and more effective treatment.AIM To develop a predictive model for intraoperative bleeding in primary hepatic malignancy patients for improving surgical planning and outcomes.METHODS The retrospective analysis enrolled patients diagnosed with primary hepatic malignancies who underwent surgery at the Hepatobiliary Surgery Department of the Fourth Hospital of Hebei Medical University between 2010 and 2020.Logistic regression analysis was performed to identify potential risk factors for intraoperative bleeding.A prediction model was developed using Python programming language,and its accuracy was evaluated using receiver operating characteristic(ROC)curve analysis.RESULTS Among 406 primary liver cancer patients,16.0%(65/406)suffered massive intraoperative bleeding.Logistic regression analysis identified four variables as associated with intraoperative bleeding in these patients:ascites[odds ratio(OR):22.839;Plirubin score(OR:2.361;Plity to predict intraoperative bleeding were 0.844 in the training set and 0.80 in the prediction set.CONCLUSION The developed and validated model predicts significant intraoperative blood loss in primary hepatic malignancies using four preoperative clinical factors by considering four preoperative clinical factors:ascites,history of alcohol consumption,TNM staging,and albumin-bilirubin score.Consequently,this model holds promise for enhancing individ

Chronic enteropathy associated with the SLCO2A1 gene(CEAS)is a complex gastroenterological condition characterized by multiple ulcers in the small intestine with chronic bleeding and protein loss.This review explores the potential mechanisms underlying the pathogenesis of CEAS,focusing on the role of SLCO2A1-encoded prostaglandin transporter OATP2A1 and its impact on prostaglandin E2(PGE2)levels.Studies have suggested that elevated PGE2 levels contribute to mucosal damage,inflammation,and disruption of the intestinal barrier.The effects of PGE2 on macrophage activation and Maxi-Cl channel functionality,as well as its interaction with nonsteroidal anti-inflammatory drugs play crucial roles in the progression of CEAS.Understanding the balance between its protective and pro-inflammatory effects and the complex interactions within the gastrointestinal tract can shed light on potential therapeutic targets for CEAS and guide the development of novel,targeted therapies.

We previously demonstrated that normal high-density lipoprotein(nHDL)can promote angiogenesis,whereas HDL from patients with coronary artery disease(d HDL)is dysfunctional and impairs angiogenesis.Autophagy plays a critical role in angiogenesis,and HDL regulates autophagy.However,it is unclear whether n HDL and d HDL regulate angiogenesis by affecting autophagy.Endothelial cells(ECs)were treated with n HDL and d HDL with or without an autophagy inhibitor.Autophagy,endothelial nitric oxide synthase(e NOS)expression,miRNA expression,nitric oxide(NO)production,superoxide anion(O2^(·-))generation,EC migration,and tube formation were evaluated.n HDL suppressed the expression of miR-181a-5p,which promotes autophagy and the expression of e NOS,resulting in NO production and the inhibition of O2^(·-)generation,and ultimately increasing in EC migration and tube formation.d HDL showed opposite effects compared to n HDL and ultimately inhibited EC migration and tube formation.We found that autophagy-related protein 5(ATG5)was a direct target of miR-181a-5p.ATG5 silencing or miR-181a-5p mimic inhibited n HDL-induced autophagy,e NOS expression,NO production,EC migration,tube formation,and enhanced O2^(·-)generation,whereas overexpression of ATG5 or miR-181a-5p inhibitor reversed the above effects of d HDL.ATG5 expression and angiogenesis were decreased in the ischemic lower limbs of hypercholesterolemic low-density lipoprotein receptor null(LDLr^(-/-))mice when compared to C57BL/6 mice.ATG5 overexpression improved angiogenesis in ischemic hypercholesterolemic LDLr^(-/-)mice.Taken together,nHDL was able to stimulate autophagy by suppressing miR-181a-5p,subsequently increasing e NOS expression,which generated NO and promoted angiogenesis.In contrast,d HDL inhibited angiogenesis,at least partially,by increasing miR-181a-5p expression,which decreased autophagy and e NOS expression,resulting in a decrease in NO production and an increase in O2^(·-)generation.Our findings reveal a novel mechanism by which HDL affects angiogenesis b

A class of Sturm-liouville problems with discontinuity is studied in this paper.The oscillation properties of eigenfunctions for Sturm-liouville problems with interface conditions are obtained.The main method used in this paper is based on Prufer transformation,which is different from the classical ones.Moreover,we give two examples to verify our main results.
