Effects of phytoestrogen genistein on myocardial ischemia/reperfusion injury and apoptosis in rabbits

AIM: To study the effect of genistein (GST) on rabbit heart ischemia/reperfusion (I/R) injury. METHODS: Rabbit heart I/R injury was induced by occluding the left anterior descending coronary artery for 45 min and reperfusing for 180 min. GST (1.0 mg/kg) was intravenously injected 5 min before heart ischemia. Hemodynamic data, infarct size, and cardiomyocytic apoptosis were measured. The pathologic changes of I/R myocardium were observed. RESULTS: During the I/R, heart rate, mean arterial blood pressure, myocardial oxygen consumption, left ventricu- lar (LV) –dp/dtmax and +dp/dtmax were decreased progressively. The infarct size was occupied 60.23 %±3.97 % (% of area at risk) in vehicle +I/R group while GST reduced the infarct size to 39.62 %±4.30 % (P<0.01). DNA ladder patter in myocardium was revealed by agarose gel electrophoresis in vehicle +I/R group while was not found in GST+I/R group. Apoptotic cardiomyocytes were sparse within ischemic myocardium at risk in GST+I/R group as compared with that in vehicle +I/R group (TUNEL stain). Apoptosis rate in ischemic myocardium from vehicle +I/R and GST+I/R groups detected by flow cytometry were 15.33 %±1.31 % and 3.88 %±0.33 %, respectively. Fas and Bax protein expressions in ischemic myocardium of vehicle +I/R group were higher than that in GST+I/R group (P<0.01). Bcl-2/Bax ratio in vehicle +I/R group was lower than that in nonischemic myocar- dium (P<0.01), while in GST+I/R group, the Bcl-2/Bax ratio was higher than that in vehicle +I/R group (P<0.01). CONCLUSION: GST reduced infarct size and apoptosis of myocytes in I/R rabbit heart.

A lamellar-structure TC21 titanium alloy was hot-rolled and subsequently annealed at 820,880 and 940℃ for 1 and 6 h,and the effects of annealing parameters on static globularization and morphology evolution of bothαandβphases were *** results show thatαglobularization process is sluggish due to the limited boundary splitting at 820℃.With increasing temperature to 880℃,the accelerated boundary splitting and termination migration promote theα*** 820 and 880℃,the static recovery(SRV)and recrystallization(SRX)induce the grain refinement of interlamellarβ***,the excessively high temperature of 940℃ results in the coarsening ofαgrains due to the assistance of Ostwald ripening,and produces coarseβgrains mainly due to the absence of SRX in interlamellarβ***,880℃ is an appropriate annealing temperature to produce a homogeneous microstructure in which globularizedαand refinedβgrains distribute homogeneously.

Objective To examine the effects of urotensin Ⅱ （UⅡ） on the discharges of neurons in CA1 area of hippocampal slices by using extracellular recording technique. Results ①In response to the application of UⅡ （0.3, 3.0, 30.0,300.0 nmol/L, n = 77） into the perfusate for 2 min, the spontaneous discharge rates （SDR） of 63/77 （81.8% ） neurons were significantly decreased in a dose-dependent manner. ②Pretreatment with bicuculline（BIC, 100 μmol/L）, a specific GABAs receptor antagonist, led to a marked increase in the SDR of 6/7 （85.71%） neurons in an epileptiform pattern. The increased discharges were not significantly changed after UⅡ （30.0 nmol/L） was applied into the perfusate for 2 min. ③Pretreatment with picrotoxin （ PIC, 50 μmol/L）, a selective blocker of Cl^- channel, led to an increase in the SDR of all 8/8（ 100% ） neurons. The increased discharges were not influenced by the UⅡ （30.0 nmol/L） applied. ④Application of nitric oxide synthase （NOS） inhibitor N^G nitro-L-arginine methyl ester （L-NAME, 50μmol/L） into the perfusate for 2 min also significantly augmented the SDR of 14/16 （87.5 % ） neurons , then UⅡ （30.0 nmol/L） applied into the perfusate reduced the increased the SDR of all 14/14 （ 100% ） neurons. Conclusion These results suggest that UⅡ may decrease neuronal activity by potentiating GABAA receptor-mediated CI current in hippocampal CA1 neurons, and involved with the mediation of nitric oxide.

Although solar steam generation strategy is efficient in desalinating seawater,it is still challenging to achieve continuous solar-thermal desalination of seawater and catalytic degradation of organic ***,dynamic regulations of hydrogen bonding networks and solvation structures are realized by designing an asymmetric bilayer membrane consisting of a bacterial cellulose/carbon nanotube/Co_(2)(OH)_(2)CO_(3)nanorod top layer and a bacterial cellulose/Co_(2)(OH)_(2)CO_(3)nanorod(BCH)bottom ***,the hydrogen bonding networks inside the membrane can be tuned by the rich surface–OH groups of the bacterial cellulose and Co_(2)(OH)_(2)CO_(3)as well as the ions and radicals in situ generated during the catalysis ***,both SO_(4)^(2−)and HSO_(5)−can regulate the solvation structure of Na^(+)and be adsorbed more preferentially on the evaporation surface than Cl^(−),thus hindering the de-solvation of the solvated Na^(+)and subsequent nucleation/growth of ***,the heat generated by the solar-thermal energy conversion can accelerate the reaction kinetics and enhance the catalytic degradation *** work provides a flow-bed water purification system with an asymmetric solar-thermal and catalytic membrane for synergistic solar thermal desalination of seawater/brine and catalytic degradation of organic pollutants.

AIM: To study the effect of capsaicin on carotid sinus baroreceptor activity (CBA). METHODS: The functional curve of carotid baroreceptor (FCCB) was constructed and the functional parameters of carotid sinus baroreceptor were measured by recording sinus nerve afferent discharge in anesthetized rats with perfused isolated carotid sinus. RESULTS: Low-concentration of capsaicin (0.2 μmol/L) had no significant effect on CBA, while perfusion of the isolated carotid sinus with middle-concentration of capsaicin (1 μmol/L) could shift FCCB to the left and upward, with peak slope (PS) increased from (2.47 %±0.14 %)/mmHg to (2.88 %±0.10 %)/mmHg (P<0.05) and peak integral value of carotid sinus nerve discharge (PIV) enhanced from 211 %±5 % to 238 %±6 % (P<0.01). The threshold pressure (TP) and saturation pressure (SP) were significantly decreased from 68.0±1.1 to 62.7±1.0 mmHg (P<0.01) and from 171.0±1.6 to 165.0±0.6 mmHg (P<0.01). Through the long-term plot study on the litter and its decomposition in the evergreen broad-leaved forest ecosystem in Hangzhou for more than two years,it was resulted that the annual litter production was 5.85 t ha^-1,most of which was the fallen leave (79.5 percent) and the withered branches and fruits were far less (7.1 and 13.4 percents respectively).The dynamics of the fallen litter was shown as a curve of two-peak pattern which appeared in April and September each *** half-life of the litter was 1.59 *** decay rate of the litter attenuted as an exponential *** annual amount of the nutrient returned to the ground through the litter was as large as 223.69kg ha^-*** total current amount of the litter on the ground was 7.47t ha^-*** decay rate in the first half of a year was 45.18 *** ecosystem remained in the stage of litter increasing with time.
