Bioenergetic alteration in gastrointestinal cancers:The good,the bad and the ugly

Cancer cells exhibit metabolic reprogramming and bioenergetic alteration,utilizing glucose fermentation for energy production,known as the Warburg ***,there are a lack of comprehensive reviews summarizing the metabolic reprogramming,bioenergetic alteration,and their oncogenetic links in gastrointestinal(GI)***,the efficacy and treatment potential of emerging anticancer drugs targeting these alterations in GI cancers require further *** review highlights the interplay between aerobic glycolysis,the tricarboxylic acid(TCA)cycle,and oxidative phosphorylation(OXPHOS)in cancer cells,as well as hypotheses on the molecular mechanisms that trigger this *** role of hypoxia-inducible transcription factors,tumor suppressors,and the oncogenetic link between hypoxia-related enzymes,bioenergetic changes,and GI cancer are also *** review emphasizes the potential of targeting bioenergetic regulators for anti-cancer therapy,particularly for GI *** the potential of targeting bioenergetic regulators for GI cancer therapy,the review categorizes these regulators into aerobic glycolysis/lactate biosynthesis/transportation and TCA cycle/coupled *** also detail various anti-cancer drugs and strategies that have produced pre-clinical and/or clinical evidence in treating GI cancers,as well as the challenges posed by these *** we highlight that understanding dysregulated cancer cell bioenergetics is critical for effective treatments,although the diverse metabolic patterns present challenges for targeted *** research is needed to comprehend the specific mechanisms of inhibiting bioenergetic enzymes,address side effects,and leverage high-throughput multi-omics and spatial omics to gain insights into cancer cell heterogeneity for targeted bioenergetic therapies.

Usefulness of fecal lactoferrin in predicting and monitoring the clinical severity of infectious diarrhea

AIM:To explore the value of fecal lactoferrin in predicting and monitoring the clinical severity of infectious ***:Patients with acute infectious diarrhea ranging from 3 mo to 10 years in age were enrolled,and one to three stool samples from each subject were *** parameters,including white blood cells /differential count,C-reactive protein,fecal mucus,fecal pus cells,duration of fever,vomiting,diarrhea and severity(indicated by Clark and Vesikari scores),were recorded and *** lactoferrin was determined by enzyme-linked immunosorbent assay and compared in different pathogen and disease *** estimating equations(GEE) were also used for ***:Data included 226 evaluations for 117 individuals across three different time *** lactoferrin was higher in patients with Salmonella(11.17 μg/g ± 2.73 μg/g) or Campylobacter(10.32 μg/g ± 2.94 μg/g) infections and lower in patients with rotavirus(2.82 μg/g ± 1.27 μg/g) or norovirus(3.16 μg/g ± 1.18 μg/g) *** of fecal lactoferrin were significantly elevated in patients with severe(11.32 μg/g ± 3.29 μg/g) or moderate(3.77 μg/g ± 2.08 μg/g) disease activity compared with subjects with mild(1.51 μg/g ± 1.36 μg/g) disease activity(P < 0.05).GEE analysis suggests that this marker could be used to monitor the severity and course of gastrointestinal infections and may provide information for disease ***:Fecal lactoferrin increased during bacterial infection and with greater disease severity and may be a good marker for predicting and monitoring intestinal inflammation in children with infectious diarrhea.

The effective construction of electromagnetic(EM)wave absorption materials with thin matching thickness,broad bandwidth,and remarkable absorption is a great solution to EM pollution,which is a hot topic in current environmental *** this study,N-doped reduced graphene oxide(N-rGO)was first prepared using a facile hydrothermal ***,high-purity 1T-MoS_(2)petals were homogeneously anchored to the wrinkled surface of N-rGO to fabricate 1T-MoS_(2)@N-rGO *** numerous electric di-poles and profuse heterointerfaces in 1T-MoS_(2)@N-rGO would induced the multiple reflection and scattering of EM waves in a distinct-ive multidimensional structure formed by two-dimensional N-rGO and 1T-MoS_(2)microspheres with plentiful thin nanosheets,remarkable conduction loss derived from the migration of massive electrons in a well-constructed conductive network formed by 1T-MoS_(2)@N-rGO,and abundant polarization loss(including dipolar polarization loss and interfacial polarization loss).All of these gave the 1T-MoS_(2)@N-rGO nanocomposites superior EM wave absorption *** effective absorption bandwidth of 1T-MoS_(2)@N-rGO reached 6.48 GHz with a relatively thin matching thickness of 1.84 mm,and a minimum reflection loss of-52.24 dB was achieved at 3.84 ***,the radar scattering cross-section reduction value of 1T-MoS_(2)@N-rGO was up to 35.42 dB·m^(2) at 0°,which further verified the huge potential of our fabricated 1T-MoS_(2)@N-rGO nanocomposites in practical applications.

Probiotic Bio-Three induces Th1 and anti-inflammatory effects in PBMC and dendritic cells

AIM:To investigate the immune response of peripheral blood mononuclear cells(PBMCs)and dendritic cells (DCs)that were stimulated by probiotic preparations. METHODS:PBMCs were isolated,cultured,and stimulated with Bio-Three(a mixture of Bacillus mesentericus, Clostridium butyricum and Enterococcus faecalis;105, 10 6 and 10 7 CFU/mL for 24 h).Cytokine production of (1)circulating PBMCs;(2)PBMCs stimulated by probiotic preparation;(3)monocyte-derived DCs;and(4)DC andT cell co-culture was determined by enzyme-linked immunosorbent *** analysis of circulating PBMCs was also investigated by flow *** was obtained from individuals who consumed Bio-Three (10 9 CFU/d ***,*** and ***) for 2 wk,or those who did not take probiotics orally. RESULTS:In culture supernatants,interferon-γ(IFN-γ) and interleukin(IL)-10 production increased,but IL-4 and tumor necrosis factor-α(TNF-α)production by PBMCs decreased after 1 and 2 wk of probiotic *** cytometry was also performed on day 14 and detected enhanced expression of CD11b,HLA-DR, CD4,CD45RA,CD25,CD44 and CD69 in response to ***,IL-10 and IL-12 were upregulated in supernatants of monocyte-derived DCs,and IFN-γand IL-10 were enhanced in supernatants of CD4 + T cells co-cultured with DCs. CONCLUSION:Bio-Three appeared to stimulate the Th1 immune response,downregulate pro-inflammatory cytokines(TNF-α)and upregulate anti-inflammatory cytokine(IL-10).Probiotics could be effective in activation of PBMCs and DCs.

Carbody sway behaviour of railway vehicles due to track irregularity from rail alternate side wear

Track irregularity from rail alternate side wear is manifested as uneven rail wear waveforms alternating in the left and right rails with equal intervals,which will cause carbody sway behaviour of railway vehicles and greatly influences the passenger *** this work,the carbody sway behaviour and mechanism due to track irregularity from rail alternate side wear and possible solutions to this issue were studied by the field testing and numerical calculation ***,the carbody sway of an urban rail transit train is introduced with full-scale field tests,through which the rail alternate side wear is characterized and the formatted track irregularity are ***,multibody vehicle dynamic models are developed to reproduce the carbody sway behaviour induced by the track irregularity from the rail alternate side *** creep forces acting on the wheel and rail are preliminarily discussed to study the influence of the carbody sway on the wear of the wheel flange and the rail ***,some potential solutions,*** the damping ratio of carbody rigid mode and rail grinding,are proposed to relieve this *** is concluded that an increased damping ratio of the carbody mode can alleviate the carbody sway and wheel–rail interactions,while properly maintaining track conditions can improve the vehicle performance.

Mixed matrix membranes(MMMs)are designed by incorporating microporous inorganic fillers into polymer matrix for the integration of inorganics’excellent gas separation performance with polymers’promising ***,due to the complex interphase interaction,the prevention of long-term aging is still challenging and the poor processability/recyclability of MMMs hinders their extensive ***,a coordination assembly protocol is developed for the fabrication of MMMs with both excellent processability/recyclability upon thermal annealing and promising dimension stability even under high gas *** bearing isophthalic acid(IPA)as side chains are applied to coordinate with Cu^(2+)and the assembled 2.5 nm microporous metal-organic polyhedra(MOP),{Cu_(24)IPA_(24)},serves as multi-functional cross-linkers for the polymer network *** MOPs’well-defined microporosity grants MMMs the capability for O_(2)/N_(2) separation while their activated dynamics at high temperatures(>120℃)endows polymer networks with excellent *** contrast to conventional supramolecular units,the MOPs possess unique logarithmic ligand-exchange kinetics and they show nearly frozen bonding dynamics at room temperature,ensuring promising dimensional stability to overcome the long-term aging and high gas pressure *** work not only provides fundamental understanding of the unique relaxation dynamics of complex systems with intertwined dynamics of supramolecular bonds and polymer chains,but also paves new avenues for the fabrication of robust and recyclable MMMs and elastomers.

Spatiotemporal evolution of ecological environment quality and its drivers in the Helan Mountain,China

Understanding the ecological evolution is of great significance in addressing the impacts of climate change and human ***,the ecological evolution and its drivers remain inadequately explored in arid and semi-arid *** study took the Helan Mountain,a typical arid and semi-arid area in China,as the study *** adopting an Enhanced Remote Sensing Ecological Index(ERSEI)that integrates the habitat quality(HQ)index with the Remote Sensing Ecological Index(RSEI),we quantified the ecological environment quality of the Helan Mountain during 2010-2022 and analyzed the driving factors behind the *** Component Analysis(PCA)was used to validate the composite ERSEI,enabling the extraction of key features and the reduction of redundant *** results showed that the contributions of first principal component(PC1)for ERSEI and RSEI were 80.23%and 78.72%,respectively,indicating that the ERSEI can provide higher precision and more details than the RSEI in assessing ecological environment ***,the ERSEI in the Helan Mountain exhibited an initial decline followed by an increase from 2010 to 2022,with the average value of ERSEI ranging between 0.298 and ***,the ERSEI showed a trend of being higher in the southwest and lower in the northeast,with high-quality ecological environments mainly concentrated in the western foothills at higher *** centroid of ERSEI shifted northeastward toward Helan County from 2010 to *** and digital elevation model(DEM)emerged as the primary drivers of ERSEI *** study highlights the necessity of using comprehensive monitoring tools to guide policy-making and conservation strategies,ensuring the resilience of fragile ecosystems in the face of ongoing climatic and anthropogenic *** findings offer valuable insights for the sustainable management and conservation in arid and semi-arid ecosystems.

Petrogenesis of late Cretaceous high Ba-Sr granodiorites,SE Lhasa block,China:implications for the reworking of juvenile crust and continental growth

The high Ba-Sr rocks can provide significant clues about the evolution of the continent lithosphere,but their petrogenesis remains *** the Late Cretaceous high Ba–Sr granodiorites in the SE Lhasa Block could potentially provide valuable insights into the continent evolution of the Qinghai-Tibet *** U–Pb ages suggest that the granodiorites were emplaced at 87.32±0.43 ***,the high Ba–Sr granodiorites are characterized by elevated K_(2)O+Na_(2)O contents(8.18-8.73 wt%)and K_(2)O/Na_(2)O ratios(0.99-1.25,mostly>1),and belong to high-K calc-alkaline to shoshonitic *** Yonglaga granodiorites show notably high Sr(653-783 ppm)and Ba(1346-1531 ppm)contents,plus high Sr/Y(30.92-38.18)and(La/Yb)_(N)(27.7-34.7)ratios,but low Y(20.0-22.8 ppm)and Yb(1.92-2.19 ppm)contents with absence of negative Eu anomalies(δEu=0.83-0.88),all similar to typical high Ba–Sr *** variable zirconεHf(t)values of-4.58 to+12.97,elevated initial^(87)Sr/^(86)Sr isotopic ratios of 0.707254 to 0.707322 and lowεNd(t)values of-2.8 to-3.6 with decoupling from the Hf system suggest that a metasomatized mantle source included significant recycled ancient *** occurrence of such high Ba–Sr intrusions indicates previous contributions of metasomatized mantle-derived juvenile material to the continents,which imply the growth of continental crust during the Late Cretaceous in the SE *** with regional data,we infer that the underplated mafic magma provides a significant amount of heat,which leads to partial melting of the juvenile *** melting of the metasomatized mantle could produce a juvenile mafic lower crust,from which the high Ba–Sr granitoids were derived from reworking of previous mafic crust during the Late Cretaceous(ca.100-80 Ma)in the SE Lhasa.
