Role of deubiquitinase JOSD2 in the pathogenesis of esophageal squamous cell carcinoma

BACKGROUND Esophageal squamous cell carcinoma(ESCC)is a deadly malignancy with limited treatment options.Deubiquitinases(DUBs)have been confirmed to play a crucial role in the development of malignant tumors.JOSD2 is a DUB involved in con-trolling protein deubiquitination and influencing critical cellular processes in cancer.AIM To investigate the impact of JOSD2 on the progression of ESCC.METHODS Bioinformatic analyses were employed to explore the expression,prognosis,and enriched pathways associated with JOSD2 in ESCC.Lentiviral transduction was utilized to manipulate JOSD2 expression in ESCC cell lines(KYSE30 and RESULTS )Preliminary research indicated that JOSD2 was highly expressed in ESCC tissues,which was associated with poor prognosis.Further analysis demonstrated that JOSD2 was upregulated in ESCC cell lines compared to normal esophageal cells.JOSD2 knockdown inhibited ESCC cell activity,including proliferation and colony-forming ability.Moreover,JOSD2 knockdown decreased the drug resistance and migration of ESCC cells,while JOSD2 overexpression enhanced these phenotypes.In vivo xenograft assays further confirmed that JOSD2 promoted tumor proliferation and drug resistance in ESCC.Mechanistically,JOSD2 appears to activate the MAPK/ERK and PI3K/AKT signaling pathways.Mass spectrometry was used to identify crucial substrate proteins that interact with JOSD2,which identified the four primary proteins that bind to JOSD2,namely USP47,IGKV2D-29,HSP90AB1,and PRMT5.CONCLUSION JOSD2 plays a crucial role in enhancing the proliferation,migration,and drug resistance of ESCC,suggesting that JOSD2 is a potential therapeutic target in ESCC.

Effectiveness of Acupoint Application in Patients with Pharyngeal Pain: Evidence from CHUNBO, A Prospective Real-World Study

Objective: To assess the outcomes after acupoint application in patients with pharyngeal pain in a real-world settings, and analyze the characteristics of effective population and prescription characteristics of acupoint application. Methods: Based on CHUNBO platform, patients with pharyngeal pain who were candidates for acupoint application on the basis of physician-evaluation, were enrolled in a nationwide, prospective, 69-week multicenter observational study from August 2020 to February 2022. Propensity score matching(PSM) was used to match the confounding factors and the association rules were used to analyze the characteristics of effective population and prescription characteristics of acupoint application. Outcome assessments included the disappearance rate of pharyngeal pain(within 3, 7, and 14 days), disappearance time of pharyngeal pain, as well as adverse events. Results: Of 7,699 enrolled participants, 6,693(86.9%) received acupoint application and 1,450(21.7%) with non-acupoint application. After PSM, there were 1,004 patients each in the application group(AG) and non-application group(NAG). The disappearance rate of pharyngeal pain in the AG at 3, 7, and 14 days were all higher than those in the NAG(Plication group with tonsil diseases was 2.19 times higher than that in the NAG(Plied to RN 8 most frequently(support 84.39%). A study of novel real-time power balance strategy with virtual asynchronous machine control for regional integrated electric-thermal energy systems

The development of regional integrated electric-thermal energy systems(RIETES) is considered a promising direction for modern energy supply systems. These systems provide a significant potential to enhance the comprehensive utilization and efficient management of energy resources. Therein, the real-time power balance between supply and demand has emerged as one pressing concern for system stability operation. However, current methods focus more on minute-level and hour-level power optimal scheduling methods applied in RIETES. To achieve real-time power balance, this paper proposes one virtual asynchronous machine(VAM) control using heat with large inertia and electricity with fast response speed. First, the coupling timescale model is developed that considers the dynamic response time scales of both electric and thermal energy systems. Second, a real-time power balance strategy based on VAM control can be adopted to the load power variation and enhance the dynamic frequency response. Then, an adaptive inertia control method based on temperature variation is proposed, and the unified expression is further established. In addition, the small-signal stability of the proposed control strategy is validated. Finally, the effectiveness of this control strategy is confirmed through MATLAB/Simulink and HIL(Hardware-in-the-Loop) experiments.

DC Traction Power Supply System Based on Modular Multilevel Converter Suitable for Energy Feeding and De-icing

A novel DC traction power supply system suitable for energy feeding and de-icing is proposed in this paper for an urban rail transit catenary on the basis of the full bridge submodule (FBSM) modular multilevel converter (MMC). The FBSM-MMC is a novel type of voltage source converter (VSC) and can directly control the output DC voltage and conduct bipolar currents, thus flexibly controlling the power flow of the urban rail transit catenary. The proposed topology can overcome the inherent disadvantages of the output voltage drop in the diode rectifier units, increase the power supply distance and reduce the number of traction substations. The flexible DC technology can coordinate multiple FBSM-MMCs in a wide area and jointly complete the bidirectional control of catenary power flow during the operation of the electric locomotive, so as to realize the local consumption and optimal utilization of the recovered braking energy of the train. In addition, the FBSM-MMCs can also adjust the output current when the locomotive is out of service to prevent the catenary from icing in winter. The working modes of the proposed topology are illustrated in detail and the control strategy is specially designed for normal locomotive operations and catenary de-icing. Simulation cases conducted by PSCAD/EMTDC validate the proposed topology and its control strategy.

Nowadays,the majority of the studies on the substitution are focused on cations(such as Y^(3+),Ti^(4+),P^(5+),etc.)in li_(1.3)Al_(0.3)Ti_(1.7)(PO_(4))_(3)(LATP),while there are few studies on the substitution of anion O^(2-).In this work,the modified LATP with a series of liCl(LATPClx,x=0.1,0.2,0.3,0.4)additives is prepared to enhance ionic conductivity.The successful introduction of Cl-makes the length of the c axis decrease from 20.822(2)to 20.792(1)Å,and the bulk conductivity of 2.13×10^(-3) S·cm^(-1) is achieved in LATPCl_(0.3).Moreover,the Al/Ti-O1/Cl1 and Al/Ti-O_(2)/Cl_(2) distance decrease,while the li1-O_(2)/Cl_(2) distance increases.lithium ions migrate more easily in the nanochannel of M3-M1-M3.In addition,the liCl additive increases the relative density and the grain boundary conductivity of LATPClx compounds.Naturally,a higher ionic conductivity of 2.12×10^(–4) S·cm^(-1) and a low activation energy of 0.30 eV are obtained in LATPCl_(0.3).Correspondingly,the symmetric cell exhibits a low overpotential of±50 mV for over 200 h in LATPCl_(0.3).The solid-state li|LATPCl_(0.3)|NCM811(NCM811=liNi0.8Co0.1Mn0.1O_(2))battery exhibits high initial capacity 185.1 mAh·g^(-1) with a capacity retention rate of 95.4%after 100 cycles at 0.5 C.This result suggests that liCl additive is an effective strategy to promote electrochemical properties of LATP solid electrolyte and can be considered for reference to other inorganic solid electrolytes systems.

Objective:This study aims to provide a theoretical foundation for the development and practical application of human immunodeficiency virus(HIV)/acquired immunodeficiency syndrome(AIDS)prevention and control policy systems in China by scrutinizing,analyzing,and synthesizing the evolution of Chinese policies in this domain.Methods:Utilizing grounded theory,we employed NVivo12 software to perform text content analysis,mining,and coding classification,along with visualization techniques,on policy texts sourced from Chinese government platforms,including the official website of China’s State Council.We considered four analytical dimensions—time,subject,type,and object.Results:The National Health Commission and the State Council emerged as the primary entities engaged in policymaking for HIV/AIDS in China.We identified four distinct stages in the evolution of these policies,culminating in a novel‘sun-shaped’HIV/AIDS prevention and control policy network model with an emphasis on‘knowledge,attitude,and practice’at its nucleus,which aligns with national conditions and societal progress.Furthermore,the focal groups of these policies have been dynamically refined and updated over time.Conclusions:Our findings introduce a‘sunshaped’HIV/AIDS prevention and control policy network model specific to China.We observed a conceptual policy shift towards prioritizing overall human health rather than confining the focus to disease treatment.Additionally,in light of China’s growing elderly population,the imperative to address HIV/AIDS prevention and control among older adults is an issue that warrants increased attention.

Based on the unified Hauser–Feshbach and exciton model,which can describe the particle emission processes between discrete energy levels with energy,angular momentum,and parity conservations,a statistical theory of light nucleus reaction(STLN)is developed to calculate the double-differential cross-sections of the outgoing neutron and light charged particles for the proton-induced^(6) li reaction.A significant difference is observed between the p+^(6) li and p+^(7) li reactions owing to the discrepancies in the energy-level structures of the targets.The reaction channels,including sequential and simultaneous emission processes,are analyzed in detail.Taking the double-differential cross-sections of the outgoing proton as an example,the influence of contaminations(such as^(1) H,^(7)li,^(12)C,and^(16)O)on the target is identified in terms of the kinetic energy of the first emitted particles.The optical potential parameters of the proton are obtained by fitting the elastic scattering differential cross-sections.The calculated total double-differential cross-sections of the outgoing proton and deuteron at E_(p)=14 MeV agree well with the experimental data for different outgoing angles.Simultaneously,the mixed double differential cross-sections of^(3) He andαare in good agreement with the measurements.The agreement between the measured data and calculated results indicates that the two-body and three-body breakup reactions need to be considered,and the pre-equilibrium reaction mechanism dominates the reaction processes.Based on the STLN model,a PLUNF code for the p+^(6) li reaction is developed to obtain an ENDF-6-formatted file of the double-differential cross-sections of the nucleon and light composite charged particles.

Pregnancy and the disease recurrence of patients previously treated for differentiated thyroid cancer:A systematic review and meta analysis

Background:Differentiated thyroid cancer(DTC)is commonly diagnosed in women of child-bearing age,but whether pregnancy influences the prognosis of DTC remains controversial.This study aimed to summarize existing evidence regarding the association of pregnancy with recurrence risk in patients previously treated for DTC.Methods:We searched PubMed,Embase,Web of Science,Cochrane,and Scopus based on the prespecified protocol registered at PROSPERO(CRD42022367896).After study selection,two researchers independently extracted data from the included studies.For quantitative data synthesis,we used random-effects meta-analysis models to pool the proportion of recurrence(for pregnant women only)and odds ratio(OR;comparing the risk of recurrence between the pregnancy group and the nonpregnancy group),respectively.Then we conducted subgroup analyses to explore whether risk of recurrence differed by response to therapy status or duration of follow-up time.We also assessed quality of the included studies.Results:A total of ten studies were included.The sample size ranged from 8 to 235,with participants’age at pregnancy or delivery ranging from 28 to 35 years.The follow-up time varied from 0.1 to 36.0 years.The pooled proportion of recurrence in all pregnant patients was 0.13(95%confidence intervals[CI]:0.06-0.25;I^(2):0.58).Among six included studies reporting response to therapy status before pregnancy,we observed a trend for increasingly higher risk of recurrence from excellent,indeterminate,and biochemically incomplete to structurally incomplete response to therapy(P trend0.05)or the OR of recurrence in studies with a comparison group(P>0.05).Two included studies that focused on patients with distant metastasis

Comparison of corrosion mechanisms of 510L low alloy steel treated by ACS and EPS techniques under various service environments

The corrosion mechanism of 510L low alloy steel treated by acid-cleaned surface(ACS)and eco-pickled surface(EPS)techniques in three simulated solutions(S0:atmospheric environment;S1:soil environment;S2:industrial environment)and the influence of interaction between different corrosive anions on corrosion were investigated.The results show that the total corrosion rates of samples in three simulated solutions were in order of S2>S0>S1,which is simultaneously correlated with initial corrosion dissolution processes as well as after the formation of corrosion products.HCO3−will inhibit the initial corrosion owing to the formation of films,whereas HSO3−will accelerate the dissolutions of the matrix based on the synergistic action of HSO3−and Cl−.On the other hand,there is no significant difference in corrosion rates between the samples treated by ACS and EPS techniques.The EPS technique that is safe,reusable and environmentally friendly can be further widely used in future work.

Background:Acute gouty arthritis(AGA)is an inflammatory joint disease with a high prevalence.Typical medical interventions,including nonsteroidal anti-inflammatory drugs,colchicine and glucocorticoids,can have serious adverse reactions.Huzhang Granule(HZG),a compound Chinese herbal medicine,has been used to treat AGA for more than 30 years with satisfactory effects and no significant adverse reactions.However,the efficacy and safety of HZG in AGA patients remains unknown.Objective:The present investigation was designed to examine the efficacy and safety profile of HZG in managing AGA patients.Design,setting,participants and interventions:The current study was conducted as a noninferiority,randomized controlled clinical trial on 180 eligible enrolled participants.Participants were randomly assigned into the HZG and etoricoxib groups.Treatments were administered for 5 d,during which the HZG group received HZG and placebo etoricoxib,while the etoricoxib group received etoricoxib and placebo HZG in the same ratio(1:1).Main outcome measures:The primary outcome was pain experienced by the patient in the gout-afflicted joint from days 2 to 5 of the treatment window.The pain level was measured via a visual analogue scale,ranging from 0 mm to 100 mm.The secondary outcomes comprised joint tenderness and swelling,reduction of inflammatory biomarkers,and the patient’s and investigator’s global evaluations of therapeutic response.Results:The mean reduction in pain was-51.22 mm(95%confidence interval[CI],[-53.42,-49.03]mm)for the HZG and-52.00 mm(95%CI,[-54.06,-49.94]mm)for the etoricoxib groups.The mean difference between the two groups was 0.78 mm(95%CI,[-2.25,3.81]mm).All additional efficacy endpoints,covering decreased inflammation and pain relief,yielded compelling proof of noninferiority.Patients in the HZG group exhibited a comparatively lower rate of adverse events compared to those in the etoricoxib group(4.44%vs 13.33%;P≤0.05).Conclusion:HZG and etoricoxib groups demonstrated similar levels of analgesic effectiveness
