A single nucleotide polymorphism in the IL1RL1 gene is associated with Behcet's disease in a Chinese Han population

AIM:To explore the association of single nucleotide polymorphisms(SNPs)in the IL33/IL1RL1 gene region with the susceptibility to Behcet’s disease(BD)in a Chinese Han ***:A total of eight SNPs in the candidate gene region(rs11792633,rs7025417,rs10975519 and rs1048274 in IL33;rs2310220,rs12712142,rs13424006 and rs3821204 in IL1RL1)were genotyped in783 BD patients and 701 healthy controls by the Sequenom Mass Array i PLEX ***:A statistically significant association was observed between IL1RL1 rs12712142 and BD *** frequency of IL1RL1 rs12712142 variant allele A was significantly lower in BD patients than that in controls(OR=0.8,95%CI:0.69-0.94,Pc=0.039);the genotype distribution(Pc=0.043)and additive and dominant genetic model analyses(OR=0.8,95%CI:0.69-0.94,Pc=0.040 and OR=0.72,95%CI:0.58-0.88,Pc=0.011)also indicated a strong association between rs12712142 and BD ***:This is the first study to reveal the association between IL1RL1 rs12712142 variant allele A and the decreased risk of BD in the Chinese Han population,indicating a protective role of IL1RL1 in the pathogenesis of BD.

Down-regulation of Risa improves podocyte injury by enhancing autophagy in diabetic nephropathy

Background: LncRNA AK044604(regulator of insulin sensitivity and autophagy, Risa) and autophagy-related factors Sirt1 and GSK3β play important roles in diabetic nephropathy(DN). In this study, we sought to explore the effect of Risa on Sirt1/GSK3β-induced podocyte ***: Diabetic db/db mice received Risa-inhibition adeno-associated virus(AAV) via tail vein injection, and intraperitoneal injection of lithium chloride(LiCl). Blood, urine, and kidney tissue samples were collected and analyzed at different time points. Immortalized mouse podocyte cells(MPCs) were cultured and treated with Risa-inhibition lentivirus(LV), EX-527, and LiCl. MPCs were collected under different stimulations as noted. The effects of Risa on podocyte autophagy were examined by qRT-PCR, Western blotting analysis, transmission electron microscopy,Periodic Acid-Schiff staining, and immunofluorescence ***: Risa and activated GSK3β were overexpressed, but Sirt1 was downregulated in DN mice and high glucosetreated MPCs(P<0.001, db/m vs. db/db, NG or HM vs. HG), which was correlated with poor prognosis. Risa overexpression attenuated Sirt1-mediated downstream autophagy levels and aggravated podocyte injury by inhibiting the expression of Sirt1(P<0.001, db/m vs. db/db, NG or HM vs. HG). In contrast, Risa suppression enhanced Sirt1-induced autophagy and attenuated podocyte injury, which could be abrogated by EX-527(P<0.001, db/db+Risa-AAV vs. db/db, HG+Risa-LV vs. HG). Furthermore, LiCl treatment could restore GSK3β-mediated autophagy of podocytes(P<0.001, db/db+LiCl vs. db/db, HG+LiCl vs. HG), suggesting that Risa overexpression aggravated podocyte injury by decreasing ***: Risa could inhibit autophagy by regulating the Sirt1/GSK3β axis, thereby aggravating podocyte injury in DN. Risa may serve as a therapeutic target for the treatment of DN.

Shape of slab solidification end under non-uniform cooling and its influence on the central segregation with mechanical soft reduction

In order to study the effect of continuous casting process parameters on the shape of slab solidification end under non-uniform cooling,a solidification model of a continuous-cast slab with non-uniform cooling condition was established with ProCAST *** model was verified by the results of nail shooting tests and the infrared temperature measurement *** characteristic parameters were defined to evaluate the uniformity of the shape of slab solidification *** results showed that the nonuniformity at the beginning and end of solidification,the solidification end length,and the solidification unevenness increased with the rise of casting *** each 10°C increase of superheat,the solidification unevenness increased by about ***,the effect of superheat on the solidification end length can be *** secondary cooling strength showed minimal effect on the nonuniformity at the beginning and end of *** the increase in secondary cooling intensity,the solidification end length decreased,but the solidification unevenness *** addition,the central segregation of the slab produced with and without the mechanical soft reduction(MSR)process was *** transverse flow of molten steel with low solid fraction influenced the central segregation morphology under MSR.

Elliptical inclusion in an anisotropic plane:non-uniform interface effects

We study the plane deformation of an elastic composite system made up of an anisotropic elliptical inclusion and an anisotropic foreign matrix surrounding the *** order to capture the influence of interface energy on the local elastic field as the size of the inclusion approaches the nanoscale,we refer to the Gurtin-Murdoch model of interface elasticity to describe the inclusion-matrix interface as an imaginary and extremely stiff but zero-thickness layer of a finite stretching *** opposed to isotropic cases in which the effects of interface elasticity are usually assumed to be uniform(described by a constant interface stretching modulus for the entire interface),the anisotropic case considered here necessitates non-uniform effects of interface elasticity(described by a non-constant interface stretching modulus),because the bulk surrounding the interface is *** this end,we treat the interface stretching modulus of the anisotropic composite system as a variable on the interface curve depending on the specific tangential direction of the *** then devise a unified analytic procedure to determine the full stress field in the inclusion and matrix,which is applicable to the arbitrary orientation and aspect ratio of the inclusion,an arbitrarily variable interface modulus,and an arbitrary uniform external loading applied *** non-uniform interface effects on the external loading-induced stress distribution near the interface are explored via a group of numerical *** is demonstrated that whether the nonuniformity of the interface effects has a significant effect on the stress field around the inclusion mainly depends on the direction of the external loading and the aspect ratio of the inclusion.

Effects of Monovacancy on Thermal Properties of Bilayer Graphene Nanoribbons by Molecular Dynamics Simulations

The monovacancy defect effect on thermal conductivity of bilayer graphene nanoribbons(BGNs)was investigated using non-equilibrium molecular dynamics(NEMD)simulations in this *** results demonstrate that the presence of monovacancy defect in BGNs reduces their thermal transport properties *** major finding of this work shows that the calculated thermal conductivity reduces approximately linearly with the raise of monovacancy *** contrast to the temperature-dependent thermal conductivity in perfect BGNs,the thermal conductivity of defected BGNs first increases and then decreases with the increasing *** addition,when the difference in the monovacancy density between two layers is larger,the thermal conductivity of BGNs is *** also calculated the phonon density of states,phonon relaxation time and participation ratio to provide a deeper understanding of the simulation *** investigation confirms that the BGNs-based nano-devices could be applied in thermal management by defect engineering.

Background:Non-alcoholic fatty liver disease(NAFLD)is a liver disorder characterized by the accumulation and degeneration of fat in the liver cells,a condition that may further deteriorate and lead to cirrhosis and liver *** studies showed that metabolic dysfunction can promote NAFLD *** Zhugan Decoction(LGZGD)has therapeutic effects on *** mechanism of LGZGD still remains *** study was to examine the impact of LGZGD on the metabolic processes involved in the development of ***:A mice model of NAFLD was treated with *** therapeutic potential of LGZGD was evaluated by assessing the activity of transaminases,lipids levels of blood,and pathological changes in the liver of the mice model of ***,this study also evaluated the influence of LGZGD on liver inflammation and oxidative ***:The results of untargeted metabolomics analysis showed that LGZGD reduced the disordered lipid metabolism in NAFLD *** improved the oxidative stress and also reduced the levels of pro-inflammatory cytokines in the *** metabolomics analysis of liver samples revealed that LGZGD treatment improved metabolic disorders,including alanine,aspartate,glutamate,glycerophospholipid metabolism,and citrate *** RT-qPCR and Western blot results showed that LGZGD could regulate the expression of key enzymes in the metabolic pathway of the citrate cycle,including ATP-citrate lyase(ACLY),alanine-glyoxylate aminotransferase-2(AGXT2),phosphatidylethanolamine N-methyltransferase(PEMT),and succinate dehydrogenase(SDH).Conclusion:We found that LGZGD can treat NAFLD by reducing inflammatory responses,inhibiting oxidative stress,regulating alanine,aspartate,glutamate,and glycerophospholipid metabolism,and citrate cycle pathways.

Comparison of D-flip-flops and D-latches:influence on SET susceptibility of the clock distribution network

As technology scales down, clock distribution networks(CDNs) in integrated circuits(ICs) are becoming increasingly sensitive to single-event transients(SETs).The SET occurring in the CDN can even lead to failure of the entire circuit system. Understanding the factors that influence the SET sensitivity of the CDN is crucial to achieving radiation hardening of the CDN and realizing the design of highly reliable ICs. In this paper, the influences of different sequential elements(D-flip-flops and D-latches, the two most commonly used sequential elements in modern synchronous digital systems) on the SET susceptibility of the CDN were quantitatively studied. Electrical simulation and heavy ion experiment results reveal that the CDN-SET-induced incorrect latching is much more likely to occur in DFF and DFF-based designs. This can supply guidelines for the design of IC with high reliability.

This paper is concerned with the bound of the cost of approximate controllability and null controllability of heat equations, i.e., the minimal Lp norm and L∞ norm of a control needed to control the system approximately or a control needed to steer the state of the system to zero. The methods we use combine observability inequalities, energy estimates for heat equations and the dual theory.
