Normal epigenetic inheritance in mice conceived by in vitro fertilization and embryo transfer

An association between assisted reproductive technology （ART） and neurobehavioral imprinting disorders has been reported in many studies, and it seems that ART may interfere with imprint reprogramming. However, it has never been explored whether epigenetic erros or imprinting disease susceptibility induced by ART can be inherited transgenerationally. Hence, the aim of this study was to determine the effect of in vitro fertilization and embryo transfer （IVF-ET） on transgenerational inheritance in am inbred mouse model. Mice derived from IVF-ET were outcrossed to wild-type C57BL/6J to obtain their female and male line F2 and F3 generations. Their behavior, morphology, histology, and DNA methylation status at several important differentially methylated regions （DMRs） were analyzed by Morris water maze, hematoxylin and eosin （H＆E） staining, and bisulfite genomic sequencing. No significant differences in spatial learning or phenotypic abnormality were found in adults derived from IVF （F1） and female and male line F2 and F3 generations. A borderline trend of hypomethylation was found in H19 DMR CpG island 3 in the female line-derived F3 generation （0.40±0.118, P=0.086）. Methylation status in H19/Igf2 DMR island 1, Igf2 DMR, KvDMR, and Snrpn DMR displayed normal patterns. Methylation percentage did not differ significantly from that of adults conceived naturally, and the expression of the genes they regulated was not disturbed. Transgenerational integrity, such as behavior, morphology, histology, and DNA methylation status, was maintained in these generations, which indicates that exposure of female germ cells to hormonel stimulation and gamete manipulation might not affect the individuals and their descendents.

Emodin regulating excision repair cross-complementation group 1 through fibroblast growth factor receptor 2 signaling

AIM: To investigate the molecular mechanisms underlying the reversal effect of emodin on platinum resistance in hepatocellular carcinoma. METHODS: After the addition of 10 μmol/L emodin to HepG2/oxaliplatin (OXA) cells, the inhibition rate (IR), 50% inhibitory concentration (IC 50 ) and reversal index (IC 50 in experimental group/IC 50 in control group) were calculated. For HepG2, HepG2/OXA, HepG2/OXA/T, each cell line was divided into a control group, OXA group, OXA + fibroblast growth factor 7 (FGF7) group and OXA + emodin group, and the final concentrations of FGF7, emodin and OXA in each group were 5 ng/mL, 10 μg/mL and 10 μmol/L, respectively. Single-cell gel electrophoresis was conducted to detect DNA damage, and the fibroblast growth factor receptor 2 (FGFR2), phosphorylated extracellular signal-regulated kinase 1/2 (p-ERK1/2) and excision repair cross-complementing gene 1 (ERCC1) protein expression levels in each group were examined by Western blotting. RESULTS: Compared with the IC50 of 120.78 μmol/L in HepG2/OXA cells, the IC 50 decreased to 39.65 μmol/L after treatment with 10 μmol/L emodin; thus, the reversal index was 3.05. Compared with the control group, the tail length and Olive tail length in the OXA group, OXA + FGF7 group and OXA + emodin group were significantly increased, and the differences were statistically significant (P < 0.01). The tail length and Olive tail length were lower in the OXA + FGF7 group than in the OXA group, and this difference was also statistically significant. Compared with the OXA + FGF7 group, the tail extent, the Olive tail moment and the percentage of tail DNA were significantly increased in the OXA + emodin group, and these differences were statistically significant (P < 0.01). In comparison with its parental cell line HepG2, the HepG2/OXA cells demonstrated significantly increased FGFR2, p-ERK1/2 and ERCC1 expression levels, whereas the expression of all three molecules was significantly inhibited in HepG2/ OXA/T cel

The hydrodynamic pressure is an important load on concrete face rockfill dams (CFRDs) subjected to earthquakes,the influence of which,however,is not clear as compared with that in concrete *** this paper,the coupling effect between the CFRDs and the reservoir water is studied based on two-dimensional finite element simulations by using a verified *** is found that neglecting the solid-fluid coupling effect not only results in an overestimation of the acceleration response within the rockfill materials but also makes an overestimation of the dynamical stresses in the concrete *** a reliable seismic response analysis of the CFRDs,therefore,the hydrodynamic pressure should be taken into account,particularly when the dam is subjected to a simultaneous excitation in both horizontal and vertical *** results show,however,that the compressibility of the water can be safely neglected in the seismic response analyses of the CFRDs even when the dam is as high as 300 m,except when the excitation is quite abundant in high frequency contents.

Oligo-lysine Induced Formation of Silica Particles in Neutral Silicate Solution

Oligo-（lysine） n （n = 1-4） containing different numbers of lysine residues was used to induce the condensation of silicic acid to form silica particles in neutral silicate solution. It was found that the condensation rate and the formation of silica particles are dependent on the number of lysine residues in an oligo-lysine. Oligo-lysine with more lysine residues can link more silicic acid together to form a matrix that promotes the effective aggregation of the condensed silica pieces to form large silica particles.

AN EXPERIMENTAL STUDY OF FLOW AROUND A BIO-INSPIRED AIRFOIL AT REYNOLDS NUMBER 2.0×10~3

The fluid flow around a bio-inspired airfoil with corrugated surfaces and its smooth counterpart at chord Reynolds number Re = 2.0×10^3. and different Angle-Of-Attack （AOA = 0°, 4°, 8° and 12°） were measured by using Particle Image Velocimetry （PIV）. The global characteristics of the fluid flow around two airfoils were analyzed by ensemble-averaged velocity field, distribution of reverse flow intermittency, and time-series flow visualizations. At AOA = 0°, no significant variation of the global flow patterns was recognized for both configurations. The statistical results of reverse flow intermittency results demonstrated that the protruding peaks of the corrugated airfoil delay flow separation occur at AOA = 4°. At large AOAs （8° and 12°）, however, the flow is massively separated in both configurations, the combination of large separation bubble above the corrugated airfoil and small recirculation zones in the upstream upper valley results in earlier separation of the flow. At AOA=8°, the wake region behind the corrugated airfoil is considerably shortened in comparison to the smooth one, indicating a remarkable reduction of the time-mean lift and drag forces, however, at AOA - 12°, the wake region behind the corrugated one is slightly larger than that behind the smooth one. For the case of AOA = 8° and 12°, the time-series flow visualizations demonstrate the intensified vortex shedding process of the corrugated airfoil, which would give rise to enhanced dynamic loading. Due to the fact that dragonfly wing is practically flexible, it is speculated that the wing structure of a gliding dragonfly might be sophisticatedly deformed in response to the periodic loading to reduce the drag.
