The hmr19 gene was cloned from Streptomyces hygroscopicus subsp. yingchengensis strain 10- 2., a bacterium strain producing agricultural antibiotics. Sequence similarity comparison indicates that hmr19 gene may encode a predicted protein with 14 putative transmembrane α-helical spanners, belonging to the drug:H+ antiporter-2.family of the major facilitator superfamily. The expression of hmr19 in the mycelium of strain 10-2. was detected by Western blotting analysis. Gene replacement technology was employed to construct an hmr19 disruption mutant. The growth inhibition test against different antibiotics indicated that the mutant strain was 5–2. fold more susceptible to tetracycline, vancomycin and mitomycin C than the parental wild type strain. The mutant took up tetracycline much faster and accumulated more antibiotics than the wild type strain 10-2.. While with the addition of an energy uncoupler, carbonyl cyanide m- chlorophenylhydrazone, the characteristics of the accumulation of [3H]tetracycline in these two strains were almost the same. It was thus concluded that hmr19 encoded a multidrug resistance efflux protein.

Dibromobiphenyl reacted with cynomethyl anion in ammonia under irradiation to form nucleophilic bis-substituted product in high yield without substantial monosubstituted product. Quantum yields for the formations of bis- and monosubstituted products were found to be 85.6 and 2.3×10-6 respectively, while the corresponding pseudo-first-order rates were 6.9×10-3 and 5.2.10-10 mol.L-1.S-1. Block up the possible electron transfer of 4-brome-4'-cyanomethylbiphenylyl radical anion to 4-cyanometbyl-biphenylyl radical and bromine ion.

The complexities of energy transfer mechanisms in the flagella of mammalian sperm flagella have been intensively investigated and demonstrate significant diversity across *** shuttles,particularly adenylate kinase(AK)and creatine kinase(CK),are pivotal in the efficient transfer of intracellular ATP,showing distinct tissue-and ***,the expression profiles of AK and CK were investigated in mice and found to fall into four subgroups,of which Subgroup III AKs were observed to be unique to the male reproductive system and conserved across *** AK8 and AK9 were found to be indispensable to male reproduction after analysis of an infertile male *** mouse models showed that AK8 and AK9 were central to promoting sperm *** combined with mass spectrometry revealed that AK8 and AK9 interact with the radial spoke(RS)of the *** of various human and mouse sperm samples with substructural damage,including the presence of multiple RS subunits,showed that the head of radial spoke 3 acts as an adapter for AK9 in the flagellar *** an ATP probe together with metabolomic analysis,it was found that AK8 and AK9 cooperatively regulated ATP transfer in the axoneme,and were concentrated at sites associated with energy consumption in the *** findings indicate a novel function for RS beyond its structural role,namely,the regulation of ATP *** conclusion,the results expand the functional spectrum of AK proteins and suggest a fresh model regarding ATP transfer within mammalian flagella.

Future optical clock networks will require high-precision optical time-frequency transfer between satellites and ground ***,due to atmospheric turbulence,satellite motion and time delay between the satellite–ground transmission links will cause spatial and temporal variations,respectively,resulting in the breakdown of the time-of-flight reciprocity on which optical two-way time-frequency transfer is ***,we experimentally simulate the atmospheric effects by two-way spatio-temporally separated links between two stationary terminals located 113 km apart and measure the effects for optical two-way time-frequency *** experiment shows that the effect on the link instability is less than 2.3×10^(−19) at 10,000 *** indicates that when the link instability of satellite-ground optical time-frequency transfer is on the order of 10^(−19),it is not necessary to consider the atmospheric non-reciprocity effects.

OBJECTIVE: To investigate the role of satellite glial cells in irritable bowel syndrome(IBS) and the effect of electroacupuncture(EA) at the Tianshu(ST2.) and Shangjuxu(ST37) combination. METHODS: A model for visceral hypersensitivity in IBS was induced through colorectal distension(CRD) stimulation. Clean-grade male Sprague-Dawley(SD) rats were randomly divided into four groups: a normal group(NG), a model group(MG), an electroacupuncture group(EA), and a glial cell inhibitor group(FCA). Bilateral EA(2.100 Hz, 1 mA, 30 min) was administered at the Tianshu(ST2.) and Shangjuxu(ST37) in week 6. Abdominal withdrawal reflex(AWR) scores were used to assess the behavioral response associated with visceral hyperalgesia, while hematoxylin-eosin staining was employed to evaluate pathological changes in the colon. The protein and mRNA levels of glial fibrillary acidic protein(GFAP) in the colon and colon-related dorsal root ganglion(DRG) were analyzed using immun-ofluorescence, immunohistochemistry, Western blotting, real-time polymerase chain reaction. The impact of EA on electrophysiological properties of colon-related DRG neurons was observed through whole-cell patch clamp analysis. RESULTS: EA significantly reduced the visceral pain behavior scores in rats with IBS in response to graded(2., 40, 60, 80 mm Hg) CRD stimulation. Additionally, EA downregulated the protein and mRNA expression levels of GFAP in the colon and colon-related DRG of rats with IBS. EA also regulated the resting membrane potential, rheobase and action potential of colon-related DRG neurons in rats with IBS. CONCLUSIONS: EA can regulate the excitatory properties of colon-related DRG neurons by downregulating the protein and mRNA expression of GFAP in the colon and colon-related DRG, indicating a potential neurobiological mechanism by which EA relieves visceral hypersensitivity in rats with IBS.

Ginsenoside Rb3 (GRb3) is an active ingredient extracted from Panax ginseng， which is known to enhance blood supply to heart and treat a variety of cardiac diseases. The aim of this study was to investigate the effect of GRb3 on myocardial injury induced by sleep deprivation and its mechanisms. GRb3 2. mg/kg group showed lower heart rate (438.2.± 10.06 bpm)， Creatine kinase (2..43 ± 2.85 U / mg prot) and Lactate dehydrogenase (412.9 ± 35.65 U / L) than SD group (592.0 ± 7.78 bpm， 44.18 ± 1.2. U / mg prot， 52..6 ± 38.18 U / L). GRb3 improved myocardial tissue damage and abnormal mitochondrial morphological changes. GRb3 also reduced the abnormal proteins expression of Atrial Natriuretic Peptide (ANP)， Sirtuin3 (SIRT3)， Tumor Protein P53 (P53)， Solute Carrier Family 7 (SLC7A11)， Glutathione peroxidase 4 (GPX4)， Transferrin Receptor Protein 1 (TFRC)， Acyl-CoA synthetase long chain family member 4 (ACSL4)， Heme Oxygenase-1 (HO-1)， Ferritin heavy polypeptide 1 (FTH1)， Ferroptosis suppressor protein 1 (FSP1). GRb3 ameliorated oxidative stress， such as Malondialdehyde (MDA，)， Glutathione (GSH)， and Superoxide dismutase (SOD). GRb3 also decreased Reactive Oxygen Species (ROS) content， restored cellular abnormal mitochondrial morphology and reversed the expression of ferroptosis-related proteins on H9c2.cells pretreated with Erastin. While the above effect could be reversed by 3-TYP. In summary， GRb3 exerts cardioprotective effects by regulating SIRT3/P53/SCL7A11/GPX4 axis to inhibit myocardial ferroptosis in sleep deprived mice. The findings imply that GRb3 holds promising potential for innovative approaches to safeguarding human cardiac health.
