Monitoring and identification of spoilage-related microorganisms in braised chicken with modified atmosphere packaging during refrigerated storage

This study mainly monitored the dominant bacterial populations and identified the spoilage-related microorganisms of braised chicken meat stored under different CO_(2)-modified atmosphere packaging(MAP)during refrigerated storage using a culture-dependent method and 16S rDNA identification.The quality changes and shelf life of the meat were also measured.The growth rate of total viable count(TVC)in braised chicken was slower with an increase of CO_(2) content in MAP,which also occurred in the remaining bacterial species monitored(lactic acid bacteria,Pseudomonas spp.,Brochothrix thermosphacta).The MAP exerted beneficial effects on the quality of braised chicken,as demonstrated by retarding the production of total volatile basic nitrogen(TVB-N)and delaying lipid oxidation(TBARS test).A total of 14 isolates were identified from braised chickens with different packaging at the end of storage,these included P.fragi(6 isolates),P.psychrophila(2 isolates),Enterococcus faecalis(3 isolates),B.thermosphacta(2 isolates),Staphylococcus equorum(1 isolate).

Effects of oxidation and precursors(lysine,glyoxal and Schiff base)on the formation of Nε-carboxymethyl-lysine in aged,stored and thermally treated chicken meat

Advanced glycation end products(AGEs)might pose health risks,and processing and storage could accelerate the generation of AGEs in meat.However,limited few reports indicated the changes of AGEs contents in meat during storage.In this study,the aim is to investigate the oxidation and precursors and their roles in the formation of N^(ε)-carboxymethyl-lysine(CML)in raw and cooked chicken meat after post-mortem ageing and storage.As post-mortem ageing and storage time increased,the CML content in cooked chicken breast significantly increased from 1.81 mg/kg to 2.00 mg/kg during 0-6 h,and then decreased from 2.00 mg/kg to 1.80 mg/kg during 6 h-1 day,finally increased again during 1-7 days,while the CML contents of raw and cooked leg significantly and continuously increased from 1.78 mg/kg to 2.08 mg/kg.Furthermore,CML was extremely positively correlated with fat oxidation(R^(2)=0.793,P<0.01),protein oxidation(R^(2)=0.917,P<0.01)and glyoxal(R^(2)=0.678,P<0.05),and was negatively correlated with lysine(R^(2)=0.536,P<0.05).No significant correlation was observed between the Schiff base and CML.

In-package cold plasma treatment of braised chicken: voltage effect

This study investigated the effect of different dielectric barrier discharge cold plasma(DBD-CP) treatment voltages on the microbiological growth and quality characteristics of braised chicken during storage at(4 ± 1) ℃. Argon-packed samples were subjected to DBD-CP treatment for 3 min at voltages of 50, 60 and 70 k V. As a result, there was no significant(P > 0.05) difference in L*, a*, b* and p H of braised chicken between control and treatment groups at the same storage time. However, the development of lipid oxidation in DBD-CP treatment samples was slower than that in air-packed samples. Compared to air-packed samples, the reduction of total viable count, yeasts and molds, lactic acid bacteria and Pseudomonads spp. in 70 k V treated samples on day 15 were 3.21, 2.41, 2.44 and 1.96 lg(CFU/g), respectively. The results indicate that DBD-CP treatment could extend the shelf life of braised chicken without negative impacts on quality characteristics.

An overview of rhenium effect in single-crystal superalloys

Nickel-based single-crystal superalloys are the key materials for the manufacturing and development of advanced aeroengines. Rhenium is a crucial alloying element in the advanced nickel-based single-crystal superalloys for its special strengthening effects. The addition of Re could effectively enhance the creep properties of the single-crystal superalloys; thus, the content of Re is considered as one of the characteristics in different-generation single-crystal superalloys. Owing to the fundamental importance of rhenium to nickel-based single-crystal superalloys, much progress has been made on understanding of the effect of rhenium in the single-crystal superalloys. While the effect of Re doping on the nickelbased superalloys is well documented, the origins of the socalled rhenium effect are still under debate. In this paper,the effect of Re doping on the single-crystal superalloys and progress in understanding the rhenium effect are reviewed. The characteristics of the d-states occupancy in the electronic structure of Re make it the slowest diffusion elements in the single-crystal superalloys, which is undoubtedly responsible for the rhenium effect, while the postulates of Re cluster and the enrichment of Re at the c/c0 interface are still under debate, and the synergistic action of Re with other alloying elements should be further studied.Additionally, the interaction of Re with interfacial dislocations seems to be a promising explanation for the rhenium effect. Finally, the addition of Ru could help suppress topologically close-packed（TCP） phase formation and strengthen the Re doping single-crystal superalloys.Understanding the mechanism of rhenium effect will be beneficial for the effective utilization of Re and the design of low-cost single-crystal superalloys.

Coriander(Coriandrum sativum L.)is recognized for its antioxidant property,as a kind of natural phenolic-rich ingredient.Polycyclic aromatic hydrocarbons(PAHs)present a class of heat-driven hazards in foods,especially the processed meat.In this study,the effect of coriander root and leaf extract on the formation and inhibition of PAH8 in roasted duck wings was firstly investigated.Coriander root extract(CRE)and coriander leaf extract(CLE)with five concentration groups(200,400,600,800,1000 mg/L)were prepared respectively to marinate the duck wings.CRE marinade exhibited greater inhibitory effect on PAH8 formation in roasted duck wings that ranged from 65.0%-87.4%.The electron spin resonance study indicated a significantly positive correlation between PAH8 and free radical level,suggesting the participation of radicals in PAHs formation.Also,it was speculated that the inhibitory effect on PAH8 was related to the phenolic compounds identified in coriander marinades.CRE made greater inhibitory effect on the formation of PAH8 and could be considered as a kind of natural source to mitigate PAHs in heat-processed meat products.

The effect of protein oxidation on the formation of advanced glycation end products after chicken myofibrillar protein glycation

Investigation that protein oxidation to the formation of advanced glycation end products(AGEs)after chicken myofibrillar protein glycation is limited.Models of protein oxidation induced by different concentrations of hydroxyl radicals(·OH)were developed after the chicken myofibrillar protein mild glycation(MPG).Results exhibited that levels of AGEs and surface hydrophobicity(H_(0))steadily increased with the a ddition of h ydrogen peroxide(H_(2)O_(2))concentration.However,levels of s ulfhydryl group,free amino group,and particle size gradually decreased with the H_(2)O_(2)concentration.The protein carbonyl value increased in H_(2)O_(2)concentration until 10 mmol/L.Pearson's correlation indicated that MPG structure modification(unfolding and degradation)induced by protein oxidation were significantly positively correlated with AGEs concentration(P<0.05).Finally,a mechanism was proposed to hypothesize t he effect of protein oxidation on the formation of AGEs under MPG conditions.

Etiological roles of core promoter variation in triple-negative breast cancer

Abnormal geneexpression playskeyrole in cancerdevelopment.Acorepromoter is located around the transcriptional start site.Through interaction between core promoter sequences and transcriptional factors,core promoter controls transcriptional initiation.We hypothesized that in cancer,core promoter sequences could be mutated to interfere the interaction with transcriptional factors,resulting in altered transcriptional initiation and abnormal gene expression and cancer development.We used triple-negative breast cancer(TNBC)as a model to test our hypothesis.We collected genome-wide core promoter variants from 279 TNBC genomes.After extensive filtering of normal genomic polymorphism,we identified 19,427 recurrent somatic variants in 1,238 core promoters of 1,274 genes and 1,694 recurrent germline variants in 272 core promoters of 294 genes.Many of the affected genes were oncogenes and tumor suppressors.Analysis of RNA-seq data from the same patient cohort identified increased or decreased gene expression in 439 somatic and 85 germline variantsaffected genes,and the results were validated by luciferase reporter assay.By comparing with the core promoter variation data from 610 unclassified breast cancer,we observed that core promoter variants in TNBC were highly TNBC-specific.We further identified the drugs targeting the genes with core promoter variation.Our study demonstrates that core promoter is highly mutable in cancer,and can play etiological roles in TNBC and other types of cancer through influencing transcriptional initiation.

Field implementation of enzyme-induced carbonate precipitation technology for reinforcing a bedding layer beneath an underground cable duct

A suitable bearing capacity of foundation is critical for the safety of civil structures.Sometimes foundation reinforcement is necessary and an effective and environmentally friendly method would be the preferred choice.In this study,the potential application of enzyme-induced carbonate precipitation(EICP)was investigated for reinforcing a 0.6 m bedding layer on top of clay to improve the bearing capacity of the foundation underneath an underground cable duct.Laboratory experiments were conducted to determine the optimal operational parameters for the extraction of crude urease liquid and optimal grain size range of sea sands to be used to construct the bedding layer.Field tests were planned based on orthogonal experimental design to study the factors that would significantly affect the biocementation effect on site.The dynamic deformation modulus,calcium carbonate content and longterm ground stress variations were used to evaluate the bio-cementation effect and the long-term performance of the EICP-treated bedding layer.The laboratory test results showed that the optimal duration for the extraction of crude urease liquid is 1 h and the optimal usage of soybean husk powder in urease extraction solution is 100 g/L.The calcium carbonate production rate decreases significantly when the concentration of cementation solution exceeds 0.5 mol/L.The results of site trial showed that the number of EICP treatments has the most significant impact on the effectiveness of EICP treatment and the highest dynamic deformation modulus(Evd)of EICP-treated bedding layer reached 50.55 MPa.The area with better bio-cementation effect was found to take higher ground stress which validates that the EICP treatment could improve the bearing capacity of foundation by reinforcing the bedding layer.The field trial described and the analysis introduced in this paper can provide a practical basis for applying EICP technology to the reinforcement of bedding layer in poor ground conditions.

Kinesin 26B modulates M2 polarization of macrophage by activating cancer-associated fibroblasts to aggravate gastric cancer occurrence and metastasis

BACKGROUND The regulatory effects of KIF26B on gastric cancer(GC)have been confirmed,but the specific mechanism still needs further exploration.Pan-cancer analysis shows that the KIF26B expression is highly related to immune infiltration of cancerassociated fibroblasts(CAFs),and CAFs promote macrophage M2 polarization and affect cancers’progression.AIM To investigate the regulatory functions of KIF26B on immune and metastasis of GC.METHODS We analyzed genes’mRNA levels by quantitative real-time polymerase chain reaction.Expression levels of target proteins were detected by immunohistochemistry,ELISA,and Western blotting.We injected AGS cells into nude mice for the establishment of a xenograft tumor model and observed the occurrence and metastasis of GC.The degree of inflammatory infiltration in pulmonary nodes was observed through hematoxylin-eosin staining.Transwell and wound healing assays were performed for the evaluation of cell invasion and migration ability.Tube formation assay was used for detecting angiogenesis.M2-polarized macrophages were estimated by immunofluorescence and flow cytometry.RESULTS KIF26B was significantly overexpressed in cells and tissues of GC,and the higher expression of KIF26B was related to GC metastasis and prognosis.According to in vivo experiments,KIF26B promoted tumor formation and metastasis of GC.KIF26B expression was positively associated with CAFs’degree of infiltration.Moreover,CAFs could regulate M2-type polarization of macrophages,affecting GC cells’migration,angiogenesis,invasion,and epithelial-mesenchymal transition process.CONCLUSION KIF26B regulated M2 polarization of macrophage through activating CAFs,regulating the occurrence and metastasis of GC.
