Scan of the endogenous retrovirus sequences across the swine genome and survey of their copy number variation and sequence diversity among various Chinese and Western pig breeds

In pig-to-human xenotransplantation,the transmission risk of porcine endogenous retroviruses(PERVs)is of great ***,the distribution of PERVs in pig genomes,their genetic variation among Eurasian pigs,and their evolutionary history remain *** scanned PERVs in the current pig reference genome(assembly Build 11.1),and identified 36 long complete or near-complete PERVs(lc PERVs)and 23 short incomplete PERVs(si PERVs).Besides three known PERVs(PERV-A,-B,and-C),four novel types(PERV-JX1,-JX2,-JX3,and-JX4)were detected in this *** to evolutionary analyses,the newly discovered PERVs were more ancient,and PERV-Bs probably experienced a bottleneck~0.5 million years ago(Ma).By analyzing63 high-quality porcine whole-genome resequencing data,we found that the PERV copy numbers in Chinese pigs were lower(32.0±4.0)than in Western pigs(49.1±6.5).Additionally,the PERV sequence diversity was lower in Chinese pigs than in Western *** the lc PERV copy numbers,PERV-A and-JX2 in Western pigs were higher than in Chinese ***,Bama Xiang(BMX)pigs had the lowest PERV copy number(27.8±5.1),and a BMX individual had no PERV-C and the lowest PERV copy number(23),suggesting that BMX pigs were more suitable for screening and/or modification as xenograft ***,we identified 451 PERV transposon insertion polymorphisms(TIPs),of which 86 were shared by all 10 Chinese and Western pig *** findings provide systematic insights into the genomic distribution,variation,evolution,and possible biological function of PERVs.

Body mass index does not affect the survival of pancreatic cancer patients

AIM to evaluate the association of body mass index(b MI) with the overall survival of pancreatic ductal adenocarcinoma(PDAC) *** A retrospective analysis of PDAC patients diagnosed in the National Cancer Center of China between January 1999 and December 2014 was performed. these patients were categorized into four b MI groups(< 18.5, 18.5-22.9, 23-27.4 and ≥ 27.5 kg/m2). χ2 tests for comparison of the proportions of categorical variables, and Student's t-test or Mann-Whitney test for continuous variables were employed. Survival analysis was performed with the Kaplan-Meyer method. their HRs of mortality and 95%CIs were estimated using the Cox proportional hazards *** With a median age of 59.6 years(range: 22.5-84.6 years), in total 1783 PDAC patients were enrolled in this study. their mean usual b MI was 24.19 ± 3.53 for the whole cohort. More than half of the patients(59.3%) experienced weight loss during the disease onset and progression. Compared with healthy-weight individuals, newly diagnosed patients who were overweight or obese had more severe weight loss during their disease onset and progression(P < 0.001). Individuals who were overweight or obese were associated with positive smoking history(P < 0.001). A significant difference in comorbidity of diabetes(P = 0.044) and coronary artery disease(P < 0.001) was identified between high b MI and normal-weight patients. After a median follow-up of 8 mo, the survival analysis showed no association between b MI and the overall survival(P = 0.90, n = 1783). When we stratified the whole cohort by pancreatic cancer stage, no statistically significant association between b MI and overall survival was found for resectable(P = 0.99, n = 217), unresectable locally advanced(P = 0.90, n = 316) and metastatic patients(P = 0.88, n = 1250), respectively. the results did not change when we used the b MI at *** Our results showed no significance of b MI for the overall survival of PDAC pati

Microdissection of chromosome 7B of common wheat and cloning of low-copy specific DNA sequences

The 7B chromosome of common wheat was microdissected from pollen mother cells of the 7B monosomic line of common wheat cv. Chinese Spring (CS). After proteinase K and DNA topoisomerase Ⅰtreatments, the isolated chromosomes were subjected to 1—3 rounds of DOPPCR amplification, which produced continuous DNA fragments ranging from 150 to 700 bp. Genomic Southern hybridization confirmed that the PCR products were originated from the wheat genome. Cloning of portion ( > 200 bp) of the 3rd round DOP-PCR products (50 μL) could generate about 20 000 recombinant clones. Characterization of 50 randomly chosen clones indicated that 21 clones produced discrete PCR products with the size of 240—600 bp. Dot-blot hybridization showed that among the 21 clones, 11 (～ 55%) were of low-copy nature while 10 (～45%) were repetitive. Effect of Tanshinone IIA on High Glucose-induced Apoptosis and Oxidative Stress Damage in Vascular Endothelial Cells

Lysine, Methionine and Tryptophan Requirements of Beijing Ducklings of 0-2 Weeks of Age

A feed trial was conducted with a total of 1 134 Beijing ducklings to study the optimum level of dietary lysine （Lys）（0.95, 1.10, 1.25%）, methionine （Met）（0.26, 0.46, 0.66%） and tryptophan （Trp）（0.20, 0.30, 0.40%） for those ducklings during a phase of 0-2 weeks. Ducklings were randomly allotted to 27 groups according to a 3 × 3× 3 factorial arrangement and fed a basal corn-soybean-peanut meal diet containing 20.26% CP, 12.45 MJ kg^-1 ME. The results from this study indicate that Lys affected body weight （P〈0.01）, feed intake （0-14 d）（P〈0.01）, but had no effect on feed/gain （0-14 d）（P〉0.05）, uric acid concentration （P 〉 0.05）. Methionine influenced body weight （P 〈 0.01）, feed/gain （P 〈 0.05）, and feed intake （P 〈 0.01）. Tryphtophan had no effect on indices measured. The requirement of the Lys and Met for Beijing ducklings of 0-2 weeks of age were 1.10 and 0.46%. The requirement of Trp for Beijing ducklings of 0-2 weeks of age was not more than 0.20%.

How the Aggregates Determine Bound Rubber Models in Silicone Rubber? A Contrast Matching Neutron Scattering Study

The correlation between aggregates and bound rubber structures in silicone rubbers(S(phr)) with various silica fractions(ΦSi) has been investigated by contrast matching small-angle neutron scattering(SANS), swelling kinetics, and low-field nuclear magnetic resonance(NMR).Mixed solvents with deuterated cyclohexane fractions of 4.9% and 53.7% were chosen to match the scattering length densities of the matrix(SMP(phr)) and the filler(SMS(phr)), respectively. All the data consistently suggest that:(i) There is a critical threshold ΦSic between 10 and 30 phr;below ΦSic, the isolated aggregates are dominant, while beyond ΦSic, some rubber fraction is trapped among the agglomerate;(ii) ΦSiindependent thicknesses around 7.5 nm(NMR) and 8.6 nm(SANS) suggest that the bound rubber formation is determined by inherent properties of the components, and the power-law around 4.2 suggests an exponential changed gradient density of the bound rubber;(iii) SMS(80) presents a bicontinuous bound rubber with three characteristic lengths of 41, 100, and 234 nm. The expanded correlation length, a 20 nm smaller aggregate sizes suggest that such existent bicontinuous network in dry samples with less ΦSi is kind of impacted by swelling. With the obtained bound rubber models, the reinforcing mechanism of filled silicone rubber is elucidated.

Effects of Land Cover on Soil Organic Carbon Stock in a Karst Landscape with Discontinuous Soil Distribution

Land cover type is critical for soil organic carbon （SOC） stocks in territorial ecosystems. However, impacts of land cover on SOC stocks in a karst landscape are not fully understood due to discontinuous soil distribution. In this study, considering soil distribution, SOC content and density were investigated along positive successional stages （cropland, plantation, grassland, scrubland, secondary forest, and primary forest） to determine the effects of land cover type on SOC stocks in a subtropical karst area. The proportion of continuous soil on the ground surface under different land cover types ranged between 0.0% and 79.8%. As land cover types changed across the positive successional stages, SOC content in both the 0-20 cm and 20-50 cm soil layers increased significantly. SOC density （SOCD） within O-lOO cm soil depth ranged from 1.45 to 8.72 kg m^-2, and increased from secondary forest to primary forest, plantation, grassland, scrubland, and cropland, due to discontinuous soil distribution. Discontinuous soil distribution had a negative effect on 8OC stocks, highlighting the necessity for accurate determination of soil distribution in karst areas. Generally, ecological restoration had positive impacts on SOC accumulation in karst areas, but this is a slow process. In the short term, the conversion of croplandto grassland was found to be the most efficient way for SOC sequestration.

Concept of the solar ring mission:An overview

The concept of the Solar Ring mission was gradually formed from L5/L4 mission concept, and the proposal of its pre-phase study was funded by the National Natural Science Foundation of China in November 2018 and then by the Strategic Priority Program of Chinese Academy of Sciences in space sciences in May 2019. Solar Ring mission will be the first attempt to routinely monitor and study the Sun and inner heliosphere from a full 360-degree perspective in the ecliptic plane. The current preliminary design of the Solar Ring mission is to deploy six spacecraft, grouped in three pairs, on a sub-AU orbit around the Sun. The two spacecraft in each group are separated by about 30?and every two groups by about 120?. This configuration with necessary science payloads will allow us to establish three unprecedented capabilities:(1) determine the photospheric vector magnetic field with unambiguity,(2) provide 360-degree maps of the Sun and the inner heliosphere routinely, and(3) resolve the solar wind structures at multiple scales and multiple longitudes. With these capabilities, the Solar Ring mission aims to address the origin of solar cycle, the origin of solar eruptions, the origin of solar wind structures and the origin of severe space weather events. The successful accomplishment of the mission will advance our understanding of the star and the space environment that hold our life and enhance our capability of expanding the next new territory of human.
