Evaluation of the expression of sperm proteins in normozoospermic and asthenozoospermic men using monoclonal antibodies

Recent studies have shown that infertility affects estimated 15% of all couples. Male infertility is the primary or contributory cause in 60% of these cases. Consequently, the application of assisted reproduction is increasing. These methods could benefit from an extended evaluation of sperm quality. For this reason, we analyzed sperm proteins from 30 men with normal spermiograms and 30 men with asthenozoospermia. Ejaculates of both groups were tested by flow cytometry （FCM） and fluorescence with a set of well-characterized anti-human sperm Hs-monoclonal antibodies （MoAbs）, which were generated in our laboratory. No statistically significant differences were found between normospermics and asthenospermics in the expression of the sperm surface protein clusterin, evaluated with Hs-3 MoAb, and semenogelin, evaluated with Hs-9 MoAb. However, FCM revealed quantitative differences in the acrosomal proteins between normozoospermic and asthenozoospermic men, namely, in glyceraldehyde-3-phosphate dehydrogenase, evaluated with Hs-8 MoAb, valosin-containing protein, evaluated with Hs-14 MoAb, and ATP synthase （cAMP-dependent protein kinase II, PRKAR2A）, evaluated with MoAb Hs-36. Asthenozoospermic men displayed a highly reduced expression of intra-acrosomal proteins, with a likely decrease in sperm quality, and thus a negative impact on successful reproduction. Asthenozoospermia seems to be a complex disorder involving intra-acrosomal proteins.

Asian dust has been identified in subtropical soils of ***(Nd)and lead(Pb)elemental and isotopic geochemistry of soils in Southeast China were used to assess the significance of local versus extraneous *** εNd(0)values were close to the parent rocks(+2.9)in the young soils(NSJ);while their values were always negative(〜—3.7)in the old soils(OMJ),implying dust *** young soils contained less Asian dust contribution(<8%)while the old soils contained more than 50%Asian dust *** implied that dust contributions were gradually increasing with the soil development stage and Asian dust input had become the principal Nd source for the old soils in this *** the other hand,Pb excesses and low 207Pb/206Pb values(〜0.8503)in near-surface soils indicated a significant anthropogenic Pb addition onto surface *** close relationship between the Pb content and isotopic ratios in the soil profile indicated that the excessive lead in the surface soil was *** results suggested that Asian dust made up a significant fraction in the old soils,but that local sources(i.e.,basalt and anthropogenic)were not trivial in Southeast China.

Adverse variations of abiotic environmental cues that deviate from an optimal range impose stresses to *** stresses severely impede plant physiology and ***,such stresses dramatically reduce crop yield and negatively impact on ecosystem stability and *** components of abiotic stresses can be,for example,suboptimal temperature and osmotic perturbations,while representative chemical facets of abiotic stresses can be toxic ions or suboptimal nutrient *** sheer complexity of abiotic stresses causes a multitude of diverse components and mechanisms for their sensing and signal ***^(2+),as a versatile second messenger,plays multifaceted roles in almost all abiotic stress responses in that,for a certain abiotic stress,Ca^(2+)is not only reciprocally connected with its perception,but also multifunctionally ensures subsequent signal ***,we will focus on salt/osmotic stress and responses to altered nutrient availability as model cases to detail novel insights into the identity of components that link stress perception to Ca^(2+)signal formation as well as on new insights into mechanisms of Ca^(2+)signal ***,we will deduce emerging conceptual consequences of these novel insights and outline arising avenues of future research on the role of Ca^(2+)signaling in abiotic stress responses in plants.

Geographic variability in the alarm calls of the European ground squirrel

Geographic variability in vocalizations has been documented in many mammalian species. We examined to what extent it applies to the alarm calls of the European ground squirrel Spermophilus citellus. We recorded the calls of 82 adult individuals from 5 natural colonies in the Czech Republic and 24 adult individuals from an artificial seminatural colony located in a Czech zoo. The founders of this colony originated from 4 different natural colonies in the Czech Republic. Our results showed that there are hardly any differences in the acoustic structure of the alarm calls between male and female European ground squirrels. Discriminant function analysis showed the highest degree of discriminability for the most isolated sites (54-74% of individuals classified correctly), whereas the lowest degree of discriminability was found for 2 interconnected colonies (38-40% individuals classified correctly). Individuals from the artificial seminatural colony were often classified correctly to this colony (58% classified correctly);however, the precision of the classification was comparatively relatively low, that is, many individuals from other colonies were incorrectly classified into this seminatural colony. This likely corresponds to the different origins of its founders. These findings indicate that there is a rather substantial geographic variability in the alarm calls of the European ground squirrel, and our study highlights its possible impact on conservation measures such as establishing artificial colonies or reintroductions.

Statistically Extrapolated Nowcasting of Summertime Precipitation over the Eastern Alps

This paper presents a new multiple linear regression（MLR） approach to updating the hourly, extrapolated precipitation forecasts generated by the INCA（Integrated Nowcasting through Comprehensive Analysis） system for the Eastern *** generalized form of the model approximates the updated precipitation forecast as a linear response to combinations of predictors selected through a backward elimination algorithm from a pool of predictors. The predictors comprise the raw output of the extrapolated precipitation forecast, the latest radar observations, the convective analysis, and the precipitation analysis. For every MLR model, bias and distribution correction procedures are designed to further correct the systematic regression errors. Applications of the MLR models to a verification dataset containing two months of qualified samples,and to one-month gridded data, are performed and evaluated. Generally, MLR yields slight, but definite, improvements in the intensity accuracy of forecasts during the late evening to morning period, and significantly improves the forecasts for large thresholds. The structure-amplitude-location scores, used to evaluate the performance of the MLR approach,based on its simulation of morphological features, indicate that MLR typically reduces the overestimation of amplitudes and generates similar horizontal structures in precipitation patterns and slightly degraded location forecasts, when compared with the extrapolated nowcasting.

Equity crowdfunding and especially syndicate equity crowdfunding are gaining popularity in the startup financing *** equity crowdfunding possesses the overriding vision of liberating the market while at the same time creating many new opportunities for individual,semi-professional *** this context,the role of the syndicate lead investors is particularly crucial because they can act as a sign of trust and reduce problems of equity crowdfunding such as information asymmetry or principle-agent ***,the impacts of syndicate lead investors have not been sufficiently explored in the past.I thus focus on the influence of the level of network centrality and reputation of syndicate lead investors on the success of the equity crowdfunding campaign and the subsequent startup funding *** network centrality is represented by syndicate lead investors' centrality in an investor-based social ***,reputation is captured by three dimensions,namely the past financial performance,the number of followers on Angel List,and the ratio of repeated syndicate investments by individual investors with the same syndicate lead *** on 496 syndicated deals from the Angel List platform,I found that the level of network centrality of the syndicate lead investor and his/her reputation have significant influence on the amount of the equity crowdfunding *** addition,reputation also affects the odds of the appearance of a subsequent funding round of his/her syndicated startup *** interaction effect between both groups of independent variables could not be *** results conclude that a well-connected and highly respected syndicate lead investor is able to send more positive signals to individual investors in the equity crowdfunding campaign and in part also to other potential professional investors in a subsequent funding *** findings can be seen as proofs of the important role of the syndicate lead investors i

非晶态金属也称为金属玻璃或玻璃金属,是一种固体金属材料,通常是合金,具有无序的原子尺度结构。大多数金属在固态状态下都是结晶的,这意味着它们有高度有序的原子排列。非晶态合金目前最重要的应用是在工频和一些高频变压器、高效率、低磁化损耗的变压器中。非晶态合金材料因其技术应用前景广阔而受到人们的关注。本文采用二维单片测试仪和数据采集装置,测量了一维交变和二维旋转磁场中磁通密度B与磁场强度H的关系。提出了一种测量非晶合金材料中磁通量和磁场强度分量的方法。本文在50赫兹频率的不同外加磁场激励下,对非晶合金样品的B和H进行了测量。通过对测量结果的对比分析发现:在低通量密度(0.35 T)下的测量中,Bx-Hx和ByHy磁滞回线的形状几乎相同,在增加励磁电流后,两个方向的磁滞行为发生了变化。在测量磁通密度达到0.88 T时,这种现象更加明显,直到在y方向的磁通密度达到1.85T和X方向1.73 T,材料达到磁饱和值。本文采用Jiles-Atherton磁滞模型模拟了非晶态合金的磁滞特性。主要工作涉及用交变磁场模拟磁滞特性并与交变磁场测量B-H结果进行对比。仿真结果表明,JilesAtherton模型具有足够的精度,可以模拟该材料的一维磁滞特性。
